Étude du comportement thermique et sous irradiation du xénon dans l’oxycarbure de zirconium / Xenon behaviour in zirconium oxycarbide : effect of temperature and irradiation

Les réacteurs GEN IV (GFR) nécessitent l’emploi de matériaux d’enrobage ayant une bonne transparence aux neutrons, une conductivité thermique élevée et agissant comme barrière de diffusion pour les produits de fission. Le but de cette étude est de déterminer le rôle de la température et de l’irradiation sur le comportement du xénon implanté dans l’oxycarbure de zirconium (ZrCxOy). A cet effet, des poudres de deux stoechiométries ZrC0,95O0,05 et ZrC0,8O0,2 ont été synthétisées puis frittées par frittage flash, ou sous charge. Pour étudier le rôle de la fluence d’implantation sur la migration thermique du xénon dans le ZrC0.95O0.05, des ions 136Xe2+ ont été implantés à une énergie de 800 keV à trois fluences : 1015, 1016 et 1017 at/cm². Les échantillons ont ensuite été recuits sous vide secondaire dans une gamme de températures de 1500 à 1800°C. Les profils de distribution du xénon ont été mesurés par RBS ou par SIMS. Des analyses par MET, MEB, NBS et PAS-DBS ont été réalisées à chaque étape. Cette étude a montré qu’à 1015 et 1016 at/cm2 le xénon est piégé dans des bulles nanométriques dans les murs de dislocations. A 1017 at/cm2, la coalescence des bulles de plus grandes tailles conduit au relâchement du xénon aux joints de grains. Pour les échantillons de ZrC0,8O0,2, les recuits conduisent à une oxydation de surface corrélée à un important relâchement du xénon. Parallèlement, des expériences d’irradiation ont été effectuées sur la plateforme JANNUS et auprès du Tandem afin de déterminer le rôle respectif des dégâts balistiques et électroniques sur la migration du xénon. Nous avons observé que ces conditions d’irradiation n’entrainaient pas de migration du xénon / Refractory ceramics are considered for the GEN IV reactors (GFR). Transition metal carbides, like ZrC, are candidates as components for fuel elements owing to their good thermal stability and their neutronic performance. An extensive study was carried out to elucidate the role of temperature on the diffusion of xenon, an abundant and volatile radionuclide, in zirconium oxycarbide. For that purpose, dense zirconium carbide samples ZrC0.8O0.2 and ZrC0.95O0.05 were synthesized using Spark Plasma Sintering and Hot Pressing. 136Xe2+ ions were implanted at three fluencies: 1015, 1016 and 1017 at/cm2, at an energy of 800 keV. Thermal annealing were carried out under vacuum in a temperature range of 1500°C to 1800°C. The Xe distribution profiles were measured either by Rutherford Backscattering Spectrometry or by Secondary Ion Mass Spectrometry before and after the different treatments. Our results show that the ZrC0.8O0.2 stoichiometry is not stable at high temperature and for the ZrC0.95O0.05 stoichiometry, the Xe migration behaviour depends on the implantation fluence. The role of the implantation defects, their evolution during annealing and the trapping of Xe into bubbles was evidenced using Positron Annihilation Lifetime Spectroscopy and Transmission Electron Microscopy. In order to simulate the effects due to neutron irradiation, irradiation experiments were carried out at the JANNUS irradiation platform at CEA Saclay and the Alto Tandem accelerator at Orsay taking into account the respective roles of the ballistic and electronic processes. The present work relates to the transfer of the inspirational elements used in St. Vitus Cathedral in Prague focusing on the hanging bolts in the sacristy of the cathedral. Two architects, Matthias of Arras and Peter Parler, participated in the construction of St. Vitus Cathedral in the 14th. Century Both of these builders were inspired by the most important buildings of contemporary Europe and the experience they gained was incorporated in the Prague cathedral. Through thorough research of the life of Matthias of Arras I am able to describe this builder as a unique personality who used a bold modern style for the construction of Prague cathedral and laid the foundation for cathedral architecture in Bohemia. Subsequent analysis of selected buildings constructed in the south of France during the 13th. and the first half of the 14th. centuries reveals many features which exhibit common characteristics with elements used on St. Vitus Cathedral, thus indicating the sources of inspiration for Prague cathedral. Both builders work meets in a space called "Old Sacristy" decorated with two unique pendant bosses. Rôles des voies de signalisation à di-GMP cyclique chez Legionella pneumophila / Roles of cyclic di-GMP signaling pathways in Legionella pneumophila

Legionella pneumophila est une bactérie aquatique qui prolifère en se répliquant à l’intérieur de cellules amibiennes. Elle peut persister dans ces environnements en vivant en communauté sous forme de biofilms. L’inhalation par l’Homme d’eau contaminée, vaporisée par les réseaux d’eau chaude ou les tours aéro‐réfrigérantes, peut mener à l’infection des macrophages pulmonaires qui se traduit par une grave pneumonie appelée légionellose. Le di‐GMP cyclique (diGMPc) est impliqué, chez diverses espèces bactériennes, dans la transition entre les modes de vie mobiles et sessiles, et chez certains pathogènes, dans la régulation de la virulence. Mon travail de thèse vise à démontrer l’implication des voies de signalisation à diGMPc dans le contrôle de la virulence et de la formation de biofilms par L. pneumophila. Cette implication a été étudiée grâce à l’inactivation systématique de chacun des gènes codant les protéines du métabolisme du diGMPc chez la souche L. pneumophila Lens. Notre étude a révélé que trois de ces protéines, Lpl0780, Lpl0922 et Lpl1118, sont spécifiquement requises pour le contrôle de la virulence et, plus particulièrement, pour la survie précoce lors de l’infection de cellules‐hôtes via l’orchestration de la sécrétion de facteurs de virulence dans la cellule‐hôte. Lpl1118 participerait également à la biogénèse de la vacuole de réplication. Cinq autres de ces protéines sont impliquées dans la régulation de la formation et de l’architecture des biofilms. L’une d’elles est, plus particulièrement, requise pour la formation de biofilms en présence d’oxyde nitrique. Ces résultats contribuent à une meilleure compréhension de l’organisation complexe et spécifique des voies de signalisation à diGMPc chez L. pneumophila et pourraient permettre d’envisager une lutte plus efficace contre ce pathogène / Legionella pneumophila is a bacterium that proliferates in fresh water environments through the replication within amoebas. These bacteria can persist in these environments through biofilm formation. The inhalation of aerosolized contaminated water through hot water systems or cooling towers can induce the infection of human lungs, leading to a severe pneumonia called legionellosis. Cyclic di‐GMP (c‐di‐GMP) in involved, in various bacterial species, in the motility‐to‐sessility transition, and in some pathogens, in virulence control. My work aims to demonstrate the involvement of signaling pathways that use c‐di‐GMP in virulence control and biofilm formation of L. pneumophila. This involvement was investigated by systematically inactivating each gene encoding a c‐di‐GMP‐metabolizing enzyme in L. pneumophila Lens strain. Our work revealed that 3 of these proteins, Lpl0780, Lpl0922 and Lpl1118 are specifically involved in virulence control and, particularly, in the early survival during host cell infection through the orchestration of virulence factors secretion within host cell. Lpl1118 is particularly required for replicative vacuole biogenesis. Five other proteins, participate in the formation and architecture of biofilms. One of them is more specifically involved in biofilm formation in the presence of nitric oxide. These results help to better understand the complexity and the specificity of c‐di‐GMP signaling pathways in L. pneumophila and should allow the exploration of more effective ways to fight this pathogen

Novel roles for basement membrane collagens:isoform-specific functions of collagen XVIII in adipogenesis, fat deposition and eye development, and effects of the collagen IV-derived matricryptin arresten on oral carcinoma growth and invasion

Abstract Collagen XVIII is an evolutionarily conserved, ubiquitously-expressed basement membrane (BM) proteoglycan produced in three isoforms, the individual roles of which are largely unknown. The physiological in vivo roles of these collagen XVIII isoforms are studied here using novel genetically modified mouse strains deficient in either the short or the medium/long isoforms of the molecule. In addition, the effects of keratin-14-driven overexpression of the thrombospondin-1 (Tsp-1) –like domain, which is common to all three collagen XVIII isoforms, are studied. The findings underline the importance of the short collagen XVIII isoform in the eye, as its absence was sufficient to cause the aberrant vascularisation of the retina previously reported in mice lacking all isoforms of collagen XVIII. In addition, an excess of the collagen XVIII Tsp-1 domain led to serious eye abnormalities, possibly by interfering with the functions of the full-length collagen XVIII produced in mice. Collagen XVIII was also shown to contribute to adipogenesis in an isoform-specific manner, in that a lack of the medium/long isoforms of collagen XVIII led to impaired adipocyte maturation and the subsequent reduction in the adipocyte number induced liver steatosis and hypertriglyceridaemia. Hence this work establishes a new extracellular matrix ECM-directed mechanism contributing to control over the multistep adipogenesis programme and points to the functional consequences of its impairment for ectopic fat deposition. The enzymatic remodelling of ECM components results in molecules with novel biological activities. Arresten is a collagen IV(α1)-derived fragment with anti-angiogenic properties which was originally described as not having any direct anti-tumour effects on cancer cells themselves. The present data revealed novel inhibitory roles for arresten in oral squamous carcinoma cell proliferation, survival, motility and invasion. Since arresten is a potent inhibitor of angiogenesis, the data generated here further underline the possibility for using it as a therapeutic agent in cases of cancer. / Tiivistelmä Kollageeni XVIII on tyvikalvojen proteoglykaani ja yksi harvoista evoluutiossa konservoituneista kollageeneista. Se esiintyy elimistössä kolmena isomuotona, joiden biologiset tehtävät ovat vielä jokseenkin epäselviä. Tässä tutkimuksessa selvitettiin kollageeni XVIII:n isomuotojen fysiologista merkitystä hyödyntäen uusia hiirilinjoja, joilta kollageeni XVIII:n lyhyt tai kaksi pisintä varianttia oli geneettisesti inaktivoitu. Poistogeenisten hiirimallien rinnalle tehtiin kaikille varianteille yhteistä trombospondiini-1 (Tsp-1)-domeinia yli-ilmentävä hiirilinja. Tämän väitöskirjatutkimuksen avulla saatiin uutta tietoa kollageeni XVIII:n ja etenkin sen lyhimmän variantin tärkeästä roolista silmässä. Aikaisemmat tutkimukset ovat osoittaneet kollageeni XVIII:n puutteen häiritsevän silmän verkkokalvon verisuonituksen normaalia kehittymistä. Tässä työssä havaittiin, että pelkästään lyhyen isomuodon puute riitti altistamaan hiiret muutoksille verkkokalvon suonituksessa. Tsp-1-osan ylimäärän havaittiin lisäksi alistavan hiiret muutoksille silmän rakenteessa, mahdollisesti häiritsemällä silmässä jo olemassa olevan kollageeni XVIII:n toimintaa. Tässä työssä havaittiin myös uusi yhteys kollageeni XVIII:n ja rasvasolujen kypsymisen välillä. Verrokkihiiriin verrattuna muodostuvan rasvakudoksen havaittiin jäävän merkittävästi vähäisemmäksi poistogeenisillä hiirillä, joilta kollageeni XVIII:n pitkät isomuodot olivat geneettisesti inaktivoitu. Heikentynyt rasvakudoksen muodostuminen lisäsi triglyseridien kertymistä hiiren verenkiertoon ja maksaan. Tutkimustulos on merkittävä avaus soluväliaineen merkityksestä rasva-aineenvaihdunnalle ja kannustaa lisätutkimuksilla selvittämään, onko kollageeni XVIII:lla yhteys myös ihmisen metaboliseen oireyhtymään. Soluväliaineen komponenttien entsymaattinen muokkaus tuottaa usein molekyylejä, joilla on uusia isäntämolekyyleistä poikkeavia ominaisuuksia. Tässä työssä tutkittiin yhden tällaisen molekyylin, tyvikalvokollageenin IV hajoamistuotteen, arrestenin, suoria vaikutuksia syöpäsoluille. Arrestenin tiedettiin entuudestaan estävän syöpäkasvainten verisuonten uudismuodostusta koe-eläimillä. Työssä osoitettiin, että arresten vaikutti endoteelisolujen lisäksi myös itse syöpäsoluihin estäen niiden lisääntymistä ja vähentäen niiden elinkykyä ja liikkuvuutta, mikä tekee arrestenista entistä houkuttelevamman ehdokasmolekyylin lääkekehitystyöhön.

Pas de résumé en français / The goal of this research was the synthesis and spectroscopic, structural andphysical-chemical characterization of cyclic 6- and 7-membered hydroxamicacids, a tetrahydroxamic calix[4]arene-based tetrapodal receptor, and their metalcomplexes with trivalent and tetravalent metal cations. They were characterizedby several techniques such as 1H and 13C NMR, IR, and mass spectroscopies,single crystal X-ray analysis, and potentiometry. Cyclic hydroxamic acids arefound in a few mix siderophores but their coordination properties were stillunknown. The structural features of metal complexes formed with Fe(III),Ga(III), Ce(IV), Zr(IV), Hf(IV), U(IV) and U(VI) have been investigated both inthe solid state and in solution. The synthesis and complexation studies of anoriginal calix[4]arene-based tetrapodal receptor is described. Reactionparameters for all key steps in the synthetic route have been optimized. Thesingle X-ray crystal analysis of benzyl-protected receptor was obtained.Complexation studies with zirconium(IV) and hafnium(IV) evidenced theformation of two metal two ligand complexes rather than 1:1 species, whichwere shown to interact in solution with a third alkali cation

La littérature sapientielle sous Sanche IV (XIIIe - XIVe siècles, Castille) : écriture et enjeux / The wisdom literature under Sancho IV (Castile, 13th-14th century) : Writing and Stakes

En accord avec mes intérêts scientifiques, j’ai décidé, dans le cadre de ce travail doctoral, d’étudier un corpus correspondant règne de Sanche IV, roi de Castille et du León de 1284 à 1295, et constitué des quatre œuvres suivantes qui, à l'instar du Sendebar ou du Calila e Dimna antérieurs, exploitent toutes les ressources de l'exemplum afin d'enseigner certes mais aussi de captiver le lecteur : El Libro del Tesoro, El Lucidario, El libro del consejo e de los consejeros et Los Castigos de Sancho IV. Bien que chacun de ces livres ait été l’objet d’études ponctuelles intéressantes, ce groupement de textes n’avait, jusqu’à présent, bénéficié d’aucune grande étude monographique et appelait donc une analyse approfondie. Ces quatre ouvrages, d’origine spatiale et temporelle différente, se sont construits selon les mentalités et les pratiques de l’époque, c’est-à-dire en réutilisant, en retravaillant la matière des sources par des ajouts, des corrections, des suppressions en fonction des visées de l’auteur ou du promoteur de l’œuvre. Cette réappropriation des sources et des modèles correspond à une finalité plus ou moins évidente ou avouée, notamment si l’on en juge à l’aune du « molinisme », pensée politique du début du XIVe siècle. Les œuvres de cette période sont, en effet, soit écrites soit commanditées par le roi ou la régente, Marie de Molina. En outre, on connaît suffisamment le rôle joué par l’Archevêque de Tolède et l’importance de la pensée religieuse dans la constitution et la consolidation du « molinisme », pour parier sur l’existence d’un discours renvoyant à l’éthique chrétienne, à la morale ou à la philosophie. J’ai donc analysé la nature du projet moral, religieux et politique qui sous-tend cette littérature sapientielle. L’analyse des textes du corpus prend en compte, dans un premier temps, la structure des ouvrages et du cadre narratif, afin de définir la voix auctoriale, c’est-à-dire celle qui choisit sources et modèles et se les réapproprie : il s’agit de mettre en lumière un mode de pensée spécifique et des intentions particulières. Dans un deuxième temps, j’ai confronté ces textes aux contextes, notamment historique, si prégnant pendant le règne de Sanche IV et la régence, pour en extraire le sens politique et idéologique, moral, religieux ou philosophique. Enfin, une des finalités de ce travail de thèse est de comprendre comment, à partir de la réécriture des sources, traités, écrits religieux, auteurs classiques, exempla… s’est réalisé ce que l’on doit considérer comme un renouvellement de la production sapientielle. La thèse permet, grâce à une confrontation croisée entre histoire et littérature, de dégager l’évolution de la littérature sapientielle médiévale en castillan, et, parallèlement, de mettre en lumière la vision culturelle et morale, politique et sociétale promue par le roi Sanche IV et soutenue ensuite par Marie de Molina. / In line with my centres of scientific interest, I have decided to examine a corpus corresponding to the reign of Sancho IV, namely El Libro del Tesoro, El Lucidario, El libro del consejo e de los consejeros and Los Castigos of Sancho IV. Although interesting individual studies have been produced on each of these books, the collation of the four texts has not hitherto benefited from any in-depth monographic study. These four works, all of different origin in time and space, were put together according to the mind-sets and practices of the time, by revisiting the source material by means of additions, corrections and deletions according to the author’s intentions. Such reworking of the sources and models corresponds to a more or less self-evident or openly admitted purpose, given that these works were either written or commissioned by the king. It is therefore essential to question the nature of the political project underlying this sapential literature. The initial analysis of the texts takes account of their structure and narrative framework in order to define the voice of the author, that is to say who chooses and takes on the relevant sources and models. Secondly, these texts need to be set against their context, especially their historical context, in order to highlight their political, ideological, moral, religious or philosophical meaning. Finally, one of the aims of this thesis is to understand how, as from the-re-writing of the sources, treatises, scriptures, classic authors and other exempla, what can be considered as a renewal of sapiential or wisdom literature actually came about. Thanks to such a cross-matching of history and literature, the thesis should evidence the evolution of sapential literature and throw light upon the cultural, moral, political and social vision promulgated by king Sancho IV and upheld by Maria de Molina.

Aspects Of The Chemistry Of Oxovanadiulm(IV) Complexes Showing Photo-Induced Cytotoxicity And DNA Cleavage Activity

The present thesis deals with different aspects of the chemistry of oxovanadium(IV) complexes, their interaction with DNA and protein and photo-induced DNA and protein cleavage activity and photocytotoxicity. Chapter I presents a general introduction on various modes of interactions of organic compounds and transition metal complexes capable of targeting DNA leading to DNA strand scission, emphasizing particularly the photo-induced DNA cleavage activities for their potential application in PDT. The mechanistic pathways associated with the DNA cleavage are discussed. A comparison has been made on the advantages of photoactive metal complexes over organic conjugates. Objective of the present investigation is also dealt in this Chapter. Chapter II of the thesis deals with the synthesis, characterization, DNA binding and photo-induced DNA cleavage activity of ternary oxovanadium(IV) complexes of N-salicylidene-S-methyldithiocarbazate (salmdtc) and phenanthroline bases to explore the photo-induced DNA cleavage activity in UV-A light of 365 nm. Chapter III presents the synthesis, characterization, DNA binding and photo-induced DNA cleavage activity of ternary oxovanadium(IV) complexes containing N-salicylidene-L-methionate (salmet) and N-salicylidene-L-tryptophanate (saltrp) Schiff bases and phenanthroline bases. The objective of this work is to investigate the photo-induced DNA cleavage activity in near-IR light and to see the effect of pendant thiomethyl and indole moieties in the DNA cleavage reactions. Chapter IV deals with the synthesis, characterization, DNA binding, red-light induced DNA cleavage activity and photocytotoxicity of ternary oxovanadium(IV) complexes having N-salicylidene-L-arginine (sal-argH) and N-salicylidene-L-lysine (sal-lysH) Schiff bases and phenanthroline bases. The important results include the visible light-induced DNA cleavage activity and photocytotoxicity of the complexes in human cervical HeLa cancer cells. Chapter V describes the synthesis, characterization, DNA binding and photo-induced DNA and protein cleavage activity and photocytotoxicity of oxovanadium(IV) complexes containing bis(2-benzimidazolylmethyl)amine and phenanthroline bases. The significant results include DNA cleavage activity in near-IR light and photocytotoxicity of the dppz complex in non-small cell lung carcinoma/human lung adenocarcinoma A549 cells in visible light. Further, we have studied the protein cleavage activity of the complexes in UV-A light of 365 nm by using bovine serum albumin (BSA) and lysozyme. Finally, Chapter VI presents the binary oxovanadium(IV) complexes of phenanthroline bases. We have studied their synthesis, characterization, DNA binding and photo-induced DNA and protein cleavage activity and photocytotoxicity. Photocytotoxicity of dppz complex has been studied in human cervical HeLa cancer cells in visible light. Photo-induced protein cleavage activity of the complexes has been studied in UV-A light of 365 nm by using BSA and lysozyme. The references have been compiled at the end of each chapter and indicated as superscript numbers in the text. The complexes presented in this thesis are represented by bold-faced numbers. Crystallographic data of the complexes, characterized structurally by single crystal X-ray crystallography, are given in CIF format in the enclosed CD (Appendix-I). Due acknowledgements have been made wherever the work described is based on the findings of other investigators. Any omission that might have happened due to oversight or mistake is regretted.

L’objet de ce travail est d’étudier le procès fait à Bernat de Cabrera, conseiller de Pierre III de Catalogne (IV d’Aragon), pour trahison et lèse-majesté en 1364. Ce « procès » est conservé aux Archives de la Couronne d’Aragon mais a été édité en 1867. Des documents de la chancellerie de Pierre IV, quelques uns issus des fonds navarrais et, dans une moindre mesure, des sources narratives permettent des recoupements critiques. La volonté royale de justifier la condamnation à mort d’un privado issu de la haute noblesse par la compilation d’actes d’enquête, comme la réhabilitation de sa mémoire en 1381, interrogent sur la solidité des faits reprochés à Bernat de Cabrera, les méthodes judiciaires suivies, les intérêts en jeu dans le royaume d’Aragon et plus largement en Espagne durant la seconde moitié du XIVe siècle. La procédure comme les faits reprochés à celui qui meurt décapité le 26 juillet 1364 révèlent les enjeux diplomatiques à l’œuvre entre les royaumes d’Espagne durant la guerre castillano-aragonaise mais aussi plus largement en Europe, les nouveaux équilibres de pouvoirs entre noblesse, pouvoir urbain et pouvoir royal ainsi que les mutations d’un pouvoir royal mis en difficulté par des guerres incessantes et à la recherche de nouveaux soutiens. Les dépositions des témoins, bien qu’évidemment biaisées décrivent des comportements, évoquent des valeurs et aussi des attentes. Le travail essaie de cerner la trajectoire du comte d’Osona mais aussi des prisonniers et des intermédiaires dans une péninsule ibérique déchirée. L’étude porte donc aussi sur la confrontation des normes entre pouvoir et société. / The intention of this work is to study the proceedings against Bernat de Cabrera, advisor to Peter III of Catalonia (IV of Aragon) for treason and lese-majesty in 1364. These "proceedings" are kept in the Archives of the Crown of Aragon but were printed in 1867. Some documents from the Chancellery of Peter IV, some from the Navarrese funds and, in fewer numbers, some narrative sources enable critical cross checking. Fímbrias do tipo IV estão associadas a diversos fenótipos em bactérias gram-negativas, e o presente estudo consistiu na caracterização da fímbria Pil e investigação de seu papel na adesão bacteriana de isolados de EPEC atípica. Por PCR e RT-PCR foram investigadas a presença e a funcionalidade do operon Pil e os resultados demonstraram que este está sendo transcrito somente nos isolados BA558 e BA956. Os genes pilS e pilV foram clonados em vetor de expressão para obtenção das proteínas Pil recombinantes e produção de anticorpos policlonais. A análise qualitativa dos testes de inibição da adesão utilizando os soros anti-PilS e anti-PilV juntos demonstraram que o isolado BA558 apresentou mudança de fenótipo de adesão. Esses resultados nos permitem concluir que o operon Pil está funcional em BA558 e BA956, e a expressão da fímbria Pil nessas cepas não está relacionada à formação de biofilme e autoagregação, porém a proteína fimbrial PilS juntamente com a adesina PilV parecem exercer uma função acessória importante na interação de BA558 às células HEp-2. / Type IV fimbriae are associated with several phenotypes in gram-negative bacteria. The aim of this study was the characterization of the Pil fimbria and its role in the interaction of atypical EPEC isolates in bacterial adhesion. Using PCR and RT-PCR, we investigated the presence and functionality of the pil operon genes. The results showed that these genes are transcribed only in the BA558 and BA956 isolates. The pilS and pilV genes were cloned into an expression vector for recombinant proteins and polyclonal antibodies production. Qualitative analysis of the adherence inhibition assays using both rabbit sera changed to localized-like the phenotype of BA558 isolate adhesion. Together, these results allow us to conclude that the Pil operon is functional only in the BA558 and BA956 isolates and that the expression of Pil fimbriae in aEPEC is not related to biofilm formation and autoaggregation but, the fimbrial PilS protein together with PilV adhesin seem to play an important accessory function in the interaction between the BA558 and epithelial cells in vitro.

Novos agentes terapêuticos que preservem as células <font face=\"Symbol\">&#946 do pâncreas e o controle do peso são importantes para o diabetes melittus tipo 2 (DM-2), constituindo uma importante área de investimento farmacêutico. Com o objetivo de contribuir com a toxinologia comparada de venenos de répteis e com a eventual descoberta de novos agentes insulinotrópicos, o presente estudo realizou a prospecção de compostos hipoglicemiantes análogos à exendina-4 (isolada de lagartos Heloderma) ou inibidores da dipeptidil-peptidase IV (DPP-IV) no veneno da serpente Bothrops jararaca. A espectrometria de massas identificou uma K49 Fosfolipase A2 inédita neste veneno. Inibidores da DPP-IV não foram encontrados. Porém, a existência de frações polipeptídicas deste veneno sem similares estruturais descritos, sem efeito na pressão arterial média, imunologicamente similares à exendina-4 e com efeito hipoglicemiante e provável capacidade de ligação ao receptor do peptídeo glucagon-símile tipo 1, caracterizaram, pela primeira vez, a ação de veneno de serpente sobre o metaboloma. / New therapeutic agents that protect pancreatic <font face=\"Symbol\">&#946 cells and regulate body mass are important to diabetes mellitus type 2 (DM-2). The search for these agents is one of the main objectives of investments made by pharmaceutical industries. To contribute to the comparative toxinology studies of reptile venoms and to reveal new insulinotropic agents, the present investigation searched for hypoglycemiant compounds such as exendin-4 analogues (isolated from Heloderma lizards) or dipeptidyl-peptidase IV (DPP-IV) inhibitors in the venom of the snake Bothrops jararaca. The mass spectrometry analysis identified a novel K-49 Phospholipase A2 in this venom. DPP-IV inhibitors were not found. However, the existence of polypeptide fractions from this venom without structural similarity with reported compounds, without effect on mean arterial blood pressure and with immunological similarity with exendin-4 and probable ability to bind to glucagon-like peptide type 1 receptor, led to the first characterization of snake venom activity on metaboloma.
