Sistemas de produção com rotação de culturas, coberturas e herbicidas para redução e prevenção do desenvolvimento de plantas daninhas resistentes ao Glifosato / Agricultural production systems with crop rotation, cover crops and herbicides for reduction and prevention of development of weeds resistant to Glyphosate

O aparecimento de plantas daninhas resistentes ao glifosato tem colocado em risco a utilização de culturas que têm a tecnologia RR. A fim de verificar o desenvolvimento de plantas daninhas resistentes ao glifosato, um experimento de campo foi realizado na estação experimental AgroMarochi localizada em Ponta Grossa, estado do Paraná, de 2009 a 2012, com o objetivo de avaliar diferentes sistemas de produção, incluindo a rotação de culturas, culturas de cobertura e aplicação de herbicidas com diferentes mecanismos de ação. O delineamento experimental foi em blocos casualizados, com dezesseis tratamentos e três repetições. Em função dos resultados, conclui-se que: (i) a rotação de culturas e de ingredientes ativos de herbicidas proporcionam melhor desempenho agronômico e econômico (menor risco de aparecimento de plantas daninhas resistentes e maiores produtividades), (ii) os sistemas 3 (soja/ trigo/ soja/ aveia /milho /trigo) e 4 (soja/ azevém/ soja/ azevém/ milho/ aveia), com 33% da área com rotação de cultura com milho na safra de verão, são os mais rentáveis independentemente dos cenários 1 e 2 do VPL (Valor Presente Líquido), (iii) os tratamentos com manejo padrão de herbicidas contribuíram no aumento da população das plantas daninhas (leiteiro, guanxuma, erva-quente, azevém - resistente e não resistente ao Glifosato e buva - resistente e não resistente ao Glifosato), e apresentaram menor rentabilidade, (iv) os manejos com rotação de herbicidas propiciaram a redução da população de plantas daninhas, (v) a cultura de aveia contribui para a redução de plantas daninhas, como azevém e buva resistentes e não resistentes ao glifosato, principalmente, (vi) área em pousio contribui para o aumento da população de plantas daninhas, como azevém e buva resistentes e não resistentes ao glifosato, principalmente, e (vii) a aplicação de Glifosato associado a Atrazina em pós emergência (tratamentos 6, 7, 11 e 14: sistemas de rotação de culturas 2, 3, 4 e 5, respectivamente) não diferiu da aplicação de Tembotrione associado a Atrazina (tratamentos 8, 9, 12 e 15: sistemas de rotação de culturas 2, 3, 4 e 5, respectivamente) na cultura de Milho RR sobre a população de azevém e buva resistente e não resistente ao glifosato. / The emergence of weeds resistant to Glyphosate has been putting at risk the use of crops that have the RR technology. In order to check the development of weeds resistant to Glyphosate, a field experiment was carried out at AgroMarochi experimental station located in \'Ponta Grossa\', State of Paraná, from 2009 to 2012, with the purpose of evaluating different agricultural production systems, including crop rotation, cover crops and application of herbicides with different mechanisms of action. The experimental design was a randomized blocks, with sixteen treatments and three replications. In light of the results, it is concluded that: (i) rotation of crops and of active ingredients of herbicide provides better agronomic and economic performance (lower risk of emergence of resistant weeds and higher yields), (ii) the systems 3 (soybean/ wheat/ soybean/ oat/ maize/ wheat) and 4 (Soybean/ ryegrass/ soybean/ ryegrass/ maize/ oat), with 33% of the area with crop rotation with maize in the summer season, are the most profitable regardless of the scenarios 1 and 2 of the NPV (Net Present Value), (iii) treatment with standard management of herbicides contributed in increasing the population of weeds (Euphorbia heterophylla, Sida sp., Spermacoce latifolia, Lolium multiflorum - resistant and not resistant to Glyphosate, Conyza spp. - resistant and not resistant to Glyphosate), and had lower profitability, (iv) the management with rotation of herbicides favored reducing weed populations, (v) the oat crop helps in reducing weeds, as ryegrass (Lolium multiflorum) and Conyza spp. resistant and not resistant to Glyphosate, mainly, (vi) fallow area contributes to the increasing population of weeds, as ryegrass and Conyza spp. resistant and not resistant to Glyphosate, mainly, and (vii) the application of Glyphosate associate to atrazine in post emergency (treatments 6, 7, 11 and 14: crop rotation systems 2, 3, 4 and 5, respectively) not differ from application of Tembotrione associate to Atrazine (treatments 8, 9, 12 and 15: crop rotation systems 2, 3, 4 and 5, respectively) in RR maize crop over weed population of ryegrass and Conyza spp. resistant and not resistant to Glyphosate.

Molecular Marker Applications in Oat (Avena Sativa L.) Breeding and Germplasm Diagnostics

The ability to identify germplasm and select traits accurately is fundamental to successful plant breeding. Pedigrees and molecular markers facilitate these processes; however misleading experimental results can occur when incorrect relationships and/or cultivar names are recorded. Molecular markers can identify these inconsistencies, and with advances in genotyping technology these diagnostics can be done faster and more objectively. This study aimed to develop molecular marker assays and graphical genotyping methodologies for cultivar identification, seed purity assessment and trait selection in oat (Avena sativa L.). KBioscience’s Allele-Specific PCR (KASP™) and genotyping-by-sequencing (GBS) technologies were applied to a set of current Canadian oat cultivars to evaluate their utility for identifying cultivars and detecting intra-cultivar variation. Both KASP™ and GBS detected different extents of heterogeneity among a set of 160 seeds that originated from four seed sources of four cultivars. In both cases, the detected variation did not appear to be limited to a specific cultivar or seed source, reinforcing that all cultivars are heterogeneous. Graphical genotyping localized heterogeneity to specific chromosome regions, thereby distinguishing physical contamination from true genetic heterogeneity and heterozygosity. Pre-existing genotype data for 700 oat cultivars and breeding lines were also used to construct graphical genotypes for pedigree validation and discovery of potential sources for favourable quantitative trait loci (QTL) alleles. This methodology used historical QTLs and anchoring markers to identify 25 putative “high oil” allele carriers. The results from this study will provide diagnostic tools for cultivar identification and pedigree validation, in addition to meaningful information about existing heterogeneity and possible QTL locations in current cultivars.

HUMAN AND ANIMAL HEALTH RISK ASSESSMENT OF MYCOTOXIN MIXTURES IN MAIZE: FROM FUNGAL PRODUCTION AND OCCURRENCE TO HARMONISED RISK CHARACTERISATION

Maize is the principal staple food/feed crop exposed to mycotoxins, and the co-occurrence of multiple mycotoxins and their metabolites has been well documented. Dietary (co)-exposure to mycotoxins is associated with human and animal health concerns as well as economic losses. The present thesis aims to apply a holistic approach for the risk assessment of mycotoxin mixtures in food and feed, i.e. from fungal production and occurrence to harmonised risk characterisation. This was done in three folds. Firstly, available environmental, ecological, and agronomic factors that may affect the relative abundance of co-occurring mycotoxins in the contaminated crops were collected from peer-reviewed literature, with focus on maize (Chapter I). Secondly, (co-)occurrence data on mycotoxins in core cereals was extracted from available articles in the scientific literature and analysed to estimate potential pattern of co-exposure in humans and animals (Chapter II). Finally, Chapter III investigates the applicability of the EFSA guidance to multiple mycotoxins through a scenario of possible co-exposure in humans and animals, using maize as a case study. in English The apoplastic barriers of the root (endodermis and exodermis) represent an important regulatory mechanism for the uptake of water and nutrients from the environment, ensuring its selectivity. In addition, both layers respond to stress factors by altering its rate and degree of cell wall modification, which affects the transport properties of the root and represents adaptive plants to high heterogeneity of the soil environment. Apoplastic barriers also respond to the availability of mineral nutrients. This issue has recently been intensively studied and a number of ambiguities persist. Interestingly, the deficiency of some mineral nutrients stimulates the differentiation of barriers, while the deficiency of other mineral nutrients delays the differentiation. In addition, different plant species react differently to the deficiency of the same element. Another interesting aspect is the fact that the reaction of the endodermis and exodermis is localized and takes place mainly in that part of the root system which is directly exposed to the stress factor. This phenomenon has been observed with cadmium toxicity, but more recently with local nutrient deficiencies (nitrogen and potassium) in Zea mays. The world is facing a major climate threat where carbon dioxide emissions are in focus. At the same time, society requires new buildings and developed infrastructure. Concrete is one of the most common building materials and 30 billion tonnes are produced annually, which corresponds to about 6 percent of the world's carbon dioxide emissions. The majority of carbon dioxide emissions occur during cement production. Sweden's long-term climate goal is that by 2045 there will be no net emissions to the atmosphere. To achieve this, a major change is required in the concrete and cement industry. Carbon dioxide emissions can be reduced by reducing the amount of cement in concrete and replacing the reduced amount with a supplement. Materials with pozzolanic properties have been shown to work well as binders in concrete. Pozzolan materials contain a high content of silica which reacts well with calcium hydroxide in cement. Oat and rice husk ash that is burned under the right conditions has shown good pozzolanic properties. The purpose of this thesis is to optimize a concrete recipe that will result in reduced carbon dioxide emissions by reducing the amount of cement. The reduced amount of cement is replaced with rice husk ash. The report is based on laboratory work where concrete recipes are mixed, measured for density, tested for set and spread measurements and cast into cubes for later tensile strength testing. The results from the laboratories showed that rice husk ash as a binder in concrete provides high early compressive strength, high density through particle compaction and becomes self-compacting without stone separation. One conclusion that can be drawn is that rice husk ash as a supplement to cement can reduce the amount of carbon dioxide emissions by 54 percent. / Världen står inför ett stort klimathot där koldioxidutsläppen står i fokus. Samtidigt är samhället i behov av nya bostäder och utvecklad infrastruktur. Betong är ett av de vanligaste byggnadsmaterialen och det produceras årligen 30 miljarder ton, vilket motsvarar cirka 6 procent av världens koldioxidutsläpp. Majoriteten av koldioxidutsläppen sker vid cementtillverkningen. Sveriges långsiktiga klimatmål är att år 2045 inte ha några nettoutsläpp till atmosfären. För att uppnå detta krävs en stor förändring i betong- och cementindustrin. Koldioxidutsläppen kan minskas genom att reducera mängden cement i betongen och ersätta den reducerade mängden med ett supplement. Material med puzzolana egenskaper har visat sig fungera bra som bindemedel i betong. Puzzolana material innehåller en hög halt av kiseldioxid som reagerar bra med kalciumhydroxid i cement. Havre- och risskalaska som bränns under rätt process har uppvisat bra puzzolana egenskaper. Syftet med det här examensarbetet är att optimera ett betongrecept som ska resultera i minskade koldioxidutsläpp genom att reducera mängden cement. Den reducerade mängden cement ersätts med risskalaska. Rapporten bygger på laborationer där betongrecept blandas, mäts för densitet, testas för sätt- och utbredningsmått samt gjuts i kuber för senare tyckhållfasthetsprover. Resultatet från laborationerna visade att risskalaska som bindemedel i betong ger hög tidig tryckhållfasthet, hög densitet genom partikelpackning samt blir självkompakterande utan att stenseparation sker. En slutsats som kan dras är att risskalaska som supplement till cement kan reducera mängden koldioxidutsläpp med 54 procent.

Use of Agricultural Wastes as Supplementary Cementitious Materials / Användning av jordbruksavfall som kompletterande cementmaterial

Global cement production is continuously increasing from 1990 till 2050 and growing particularly rapidly in developing countries, where it represents a crucial element for infrastructure development and industrialisation. Every tonne of ordinary Portland cement (OPC) produced releases, on average, about 800 kg of CO2 into the atmosphere, or, in total, the overall production of cement represents roughly 7% of all man-made carbon emissions. The present paper aims to deepen the re-use of agricultural solid waste materials as partial replacement of OPC, which can positively contribute to the sustainability of the concrete industry because of their availability and environmental friendliness. In particular, rice-husk ash (RHA) and oat-husk ash (OHA), burned under the right conditions, can have a high reactive silica content, representing very potential pozzolans. The mechanical and physical characteristics of both materials are investigated to evaluate the influence on concrete properties. Subsequently, using the environmental product declarations (EPDs) of the material used, a comparative environmental impact analysis between RHA concrete and ordinary concrete having the same resistance class, is presented. It is concluded that the use of RHA as supplementary cementitious material can serve a viable and sustainable partial replacement to OPC for the reduction of CO2 emissions and global warming potential. / Den globala cementproduktionen ökar från 1990 till 2050 och växer särskilt snabbt i utvecklingsländer, där den utgör en viktig del för infrastrukturutveckling och industrialisering. Varje ton vanligt portlandcement (OPC) släpper i genomsnitt ut cirka 800 kg koldioxid i atmosfären, och, totalt, representerar den totala cementproduktionen ungefär 7% av alla koldioxidutsläpp från mänsklig verksamhet. Det här examensarbetet syftar till att fördjupa kunskapen om och därmed i förlängningen återanvändningen av fasta avfallsmaterial från jordbruket som delvis ersättning av OPC, vilket kan bidra till hållbarheten i betongindustrin på grund av deras tillgänglighet och miljövänlighet. I synnerhet kan risskalaska (RHA) och havreskalaska (OHA), som bränns under rätt process, ha en hög reaktiv kiseldioxidhalt, vilket representerar mycket potentiella puzzolaner. De mekaniska och fysiska egenskaperna hos båda materialen har undersökts för att utvärdera deras inverkan på betongegenskaper. Därefter presenteras en jämförande miljökonsekvensanalys mellan RHA-betong och OPC-betong med samma motståndsklass med användning av miljövarudeklaration (EPD) för det använda materialet. Man drar slutsatsen att användningen av RHA som alternativt bindemedel (SCM) till OPC kan hjälpa till att minska koldioxidutsläppen och den globala uppvärmningspotentialen.

GIS-based MCDA for Integrated Risk Assessment and Prioritization of High-Risk Areas in the Göta River Valley

Climate change will bring more precipitation in Northern Europe and Sweden.Increasing precipitation will cause more floods, which will increase landslide risks. Inthe Göta river valley there are high landslide risks due to the thick layer of clay andquick clay present there. The river valley has been the home of numerous industriesfor a long time, which unfortunately have left large areas with contaminated soils alongthe river. The Göta river is the only outlet for Sweden's largest lake, called LakeVänern. With increasing precipitation, the water level of the lake will rise. This willlead to a larger tapping of the lake, to not cause floods around the lake. A largertapping will lead to higher flood and landslide risks in the river valley and thusincreasing the risk for contaminated soils entering the river.This study seeks to provide a comparison of two analyses where flood risk classes andheight above water level, functioning as flood uncertainties, as well as contaminatedsoil criteria are used in one of the analyses but not the other, to see if there is a difference.The study also seeks to show where along the river there are high risk areas inneed for climate adaptation measures to reduce risks.The method used was a multi-criteria decision analysis (MCDA) with the Analytichierarchy process (AHP) technique, to include several criteria in the analysis, pairwisecompare the criteria, and calculate their relative weights. The method was utilizedwith Geographic Information System software and AHP software. The robustness ofthe analyses were controlled by performing a One-Factor-at-a-Time sensitivityanalysis.The result of this study was the findings of four areas with high risk values along theGöta river. Three of those areas were found in both performed MCDA's. The fourtharea was only found in the second MCDA where the three additional criteria wereincluded in the analysis.The conclusion of this study is that the areas with the highest risks in relation to flood,landslide, and contamination risks are located in the localities of Lilla Edet, Älvängen,Sure, Göta, and in the city of Gothenburg. Also, there is a difference in the resultswhen including the three additional factors, another area is highlighted as having highrisks. The method used in this study makes it clear that the usage of five or eight criteriacan produce results that are similar to each other. However, by using eight criteriainstead of five and thus including additional risks that are connected with each other,there is an impact on the result, which becomes more precise.

Les répercussions de l’insuffisance rénale chronique sur le transport des médicaments

L’insuffisance rénale chronique (IRC) affecte 13 % de la population américaine et son incidence ne cesse d’augmenter. Malgré un ajustement des doses de médicaments administrés en fonction du taux de filtration glomérulaire du patient urémique, près de 40 % des patients reçoivent une dose trop élevée en raison de modifications de l’élimination extrarénale des médicaments chez ces patients. Il est connu que l’IRC affecte l’élimination métabolique des médicaments par les cytochromes P450 et les enzymes de biotransformation de phase II. Nous avons aussi démontré, chez le rat, que l’IRC affecte l’expression et l’activité de transporteurs de médicaments intestinaux entraînant une augmentation de la biodisponibilité de certains médicaments. On retrouve des transporteurs de médicaments dans de nombreux organes comme le foie, les reins et la barrière hématoencéphalique (BHE) où ils jouent des rôles importants dans les éliminations biliaire et rénale et la pénétration des médicaments au cerveau. Le but de ce travail était de mesurer, chez des rats néphrectomisés, les impacts de l’IRC sur l’expression protéique et génique et l’activité des transporteurs de médicaments hépatiques, rénaux et cérébraux. Les transporteurs étudiés sont de la famille des transporteurs ABC (P-glycoprotéine, multidrug-resistance related protein, breast cancer resistance protein) ou des solute carriers (organic anion transporter, organic anion transporting protein). Aussi, une étude réalisée chez l’humain visait à évaluer la pharmacocinétique de deux médicaments : la fexofénadine, un médicament majoritairement transporté, et le midazolam, un substrat du cytochrome P450 3A4, chez des sujets dialysés. Nos résultats montrent que, chez le rat, l’IRC entraîne des modulations de l’expression des transporteurs d’influx et d’efflux hépatiques pouvant entraîner des diminutions du métabolisme hépatique et de l’excrétion biliaire des médicaments. Dans le rein, nous avons démontré des modulations de l’expression des transporteurs de médicaments. Nous avons aussi démontré que l’IRC diminue l’élimination urinaire de la rhodamine 123 et favorise l’accumulation intrarénale de médicaments transportés comme la benzylpénicilline et la digoxine. À la BHE, nous avons démontré des diminutions de l’expression des transporteurs de médicaments. Toutefois, nous n’avons pas observé d’accumulation intracérébrale de trois substrats utilisés (digoxine, doxorubicine et vérapamil) et même une diminution de l’accumulation intracérébrale de la benzylpénicilline. Il semble donc que, malgré les modulations de l’expression des différents transporteurs de médicaments, l’intégrité et la fonction de la BHE soient conservées en IRC. Chez l’humain, nous avons démontré une augmentation de la surface sous la courbe de la fexofénadine chez les sujets dialysés, comparativement aux témoins, suggérant une altération des mécanismes de transport des médicaments chez ces patients. Nous n’avons, toutefois, pas observé de modification de la pharmacocinétique du midazolam chez les patients dialysés, suggérant une activité métabolique normale chez ces patients. Un ou des facteurs s’accumulant dans le sérum des sujets urémiques semblent responsables des modulations de l’expression et de l’activité des transporteurs de médicaments observées chez le rat et l’humain. Ces travaux mettent en évidence une nouvelle problématique chez les sujets urémiques. Nous devons maintenant identifier les mécanismes impliqués afin d’éventuellement développer des stratégies pour prévenir la toxicité et la morbidité chez ces patients. / Chronic renal failure (CRF) affects 13% of the American population and its incidence is rising. Despite dose adjustment of drugs administered to CRF patients according to their glomerular filtration rate, nearly 40% of patients receive up to 6,45-times the recommended dose due to modifications in the extra-renal elimination of drugs. It is known that CRF affects the metabolic elimination of drugs via cytochrome P450s and Phase II biotransformation enzymes. Also, we showed modulations in the expression and activity of intestinal drug transporters in CRF rats that could lead to increases in the bioavailability of drugs. Drug transporters are expressed in various organs including the liver, the kidneys and the blood-brain barrier (BBB) where they play important roles in the biliary and renal elimination, and the brain penetration of drugs. The objective of this work was to measure, using a rat model of CRF, the impacts of CRF on the protein and mRNA expression and the activity of liver, kidney and brain drug transporters. We studied ABC transporters (P-glycoprotein, multidrug-resistance related protein, breast cancer resistance protein) and solute carriers (organic anion transporters, organic anion transporting proteins). Also, a study conducted in human aimed to evaluate the pharmacokinetics of two drugs: fexofenadine, a transported drug, and midazolam, a substrate of cytochrome P450 3A4, in dialyzed patients. In rats, our results show modulations in the expression and activity of hepatic influx and efflux drug transporters that could lead to decreases in the hepatic metabolism and biliary excretion of drugs. In the kidney, we demonstrated modulations in the expression of drug transporters in CRF rats. We also demonstrated that CRF causes a reduction of the urinary elimination of rhodamine 123, a P-glycoprotein substrate, and the intra-renal accumulation of at least two transported drugs: benzylpenicillin and digoxin. Finally, we demonstrated decreases in the expression of influx and efflux drug transporters at the BBB of CRF rats. However, these decreases did not correlate with in vivo changes since BBB permeability of benzylpenicillin was decreased in CRF rats while digoxin, doxorubicin and verapamil permeabilities were unchanged. It thus appears that, even with decreased drug transporters, BBB integrity and function is conserved in CRF. In human, we showed an increase in the area under the curve of fexofenadine in dialyzed subjects compared to healthy controls, suggesting alterations of drug transport mechanisms in these patients. However, we observed no modifications in the pharmacokinetics of midazolam in dialyzed patients, suggesting a normal metabolic activity in these patients. Results from in vitro studies suggest that one or many uremic factors accumulating in the serum of uremic rats and patients are responsible for the observed modulations in drug transporter expression and activity observed in rat and human. This work demonstrates the impacts of CRF on the expression and activity of drug transporters and how they could affect drug pharmacokinetics in patients. Now, the mechanisms leading to these modulations need to be identified in order to eventually develop strategies to prevent drug toxicity and morbidity in uremic patients.
