Subprodutos de acerola como fontes de compostos fenólicos em leites fermentados probióticos / Acerola by-products as sources of phenolic compounds in probiotics fermented milks.

Subprodutos de frutas são rotineiramente descartados pelas indústrias. Porém, são ricos em compostos bioativos, podendo ser utilizados como ingredientes em produtos funcionais, promovendo a saúde e minimizando o impacto ambiental. O objetivo deste trabalho foi estudar o potencial funcional de subprodutos desidratados de acerola e de laranja, como fontes de compostos fenólicos, e desenvolver leites fermentados probióticos adicionados deste resíduo, avaliando suas características físico-químicas durante o armazenamento sob refrigeração (28 dias; 4 ± 1 °C), bem como o impacto das condições gastrointestinais sobre os flavonoides e as cepas probióticas. Os subprodutos foram obtidos em indústrias de processamento de frutas do estado de São Paulo, e foram realizadas as seguintes análises para caracterizá-los: composição centesimal, teores de vitamina C, minerais, fibras alimentares, compostos fenólicos totais e proantocianidinas, capacidade antioxidante in vitro e perfil cromatográfico de flavonoides (CLAE). Foram elaboradas quatro formulações de leites fermentados: F0 (controle), sem adição de resíduo de acerola (RA); F2, com 2% de RA; F5, com 5% de RA; F10, com 10% de RA. Adicionou-se a cultura probiótica ABT-4 nos produtos, constituída de duas cepas probióticas: Bifidobacterium animalis subsp lactis Bb-12 e Lactobacillus acidophilus La-5, além da cultura starter Streptococcus thermophilus. As seguintes análises foram realizadas com as formulações de leites fermentados, durante o armazenamento sob refrigeração (28 dias, 4 ± 1 °C): composição centesimal, pH, acidez, viabilidade dos microrganismos, teor de compostos fenólicos totais (CF), cor e textura instrumentais. Além disso, os leites fermentados foram submetidos a condições gastrointestinais simuladas in vitro, para avaliação do impacto na viabilidade das cepas probióticas e nos compostos fenólicos. O RA mostrou-se excelente fonte de vitamina C (605 mg/100 g b.u.), além de apresentar melhor capacidade antioxidante in vitro do que o RL. Proantocianidinas foram encontradas apenas no RA, na concentração de 617 &#181;g EC/g b.s. O teor de compostos fenólicos totais do RA (3240 &#181;g EAG/100 g b.s.) foi 3,6 vezes maior que o do RL. Os principais compostos fenólicos encontrados no RA foram: derivados de quercetina, procianidina B1, rutina, e derivados de caempferol. No RL, foram identificados: naringenina, sinensetina, homorientina, isovitexina e derivados de ácido clorogênico. Os subprodutos estudados apresentaram elevados teores de fibras totais (acima de 60%) e proteínas totais (RA: 10,4%; RL: 9,9%), além de reduzido teor de lipídeos totais (RA: 1,6%; RL: 2,6%). Os principais minerais identificados em ambos os resíduos foram: potássio, magnésio, cálcio e fósforo. Quanto às quatro formulações de leites fermentados, estas apresentaram baixo teor de lipídeos totais (menor que 1%), e o teor de proteínas totais variou entre 3,9 e 5,1 g/100 g, estando de acordo com a legislação vigente para este tipo de produto. O pH das formulações F0 (controle) e F2 manteve-se estável significativamente (p > 0,05) ao longo do período de armazenamento sob refrigeração (28 dias; 4 ± 1 °C), e das outras formulações sofreu pequena queda, mesmo assim mantendo-se acima de 4,5. A acidez das formulações, que variou entre 0,92 a 1,28 mg de ácido lático/g, aumentou entre os dias 1 e 14 de armazenamento, depois se manteve até o final da vida de prateleira. O RA não interferiu de maneira negativa nas populações de microrganismos analisadas durante o armazenamento, já que as formulações F2, F5 e F10 mantiveram suas populações em torno de 8 log UFC/g. Quanto ao teor de CF, as amostras diferiram significativamente entre si (p < 0,05), sendo que F0 apresentou teor em torno de 5 vezes inferior a F10 (21,13 e 101,13 &#181;g EAG/100 g, respectivamente, no dia 1). A cor dos produtos se manteve até o final da vida de prateleira, e diferiram significativamente (p < 0,05) entre si. O RA influenciou pouco nos parâmetros de textura dos leites fermentados, mas a formulação controle foi a única que perdeu adesividade. Após a fase gástrica da digestão simulada in vitro, no 7° dia de armazenamento, as populações de bactérias probióticas diminuíram drasticamente (quedas em torno de 3 a 5 log UFC/g), e após a fase entérica não foram detectadas contagens. Por outro lado, os flavonoides encontrados nos leites fermentados adicionados de RA aumentaram em torno de 2 a 5 vezes, após a fase gástrica, mantendo-se ou sofrendo pequena queda após fase entérica. Estes resultados mostram que o pó de subprodutos de acerola é um valioso ingrediente a ser utilizado em alimentos funcionais, pois é rico em vitamina C, fibras e compostos fenólicos, agregando valor nutricional, além de servir como antioxidante natural. Seus flavonoides parecem ser altamente resistentes aos ácidos e sais da digestão, podendo, assim, exercer efeitos positivos sobre a saúde. / Fruits by-products are routinely discarded by industries. However, they are rich in bioactive compounds, and can be used as ingredients in functional foods, promoting health and minimizing environmental impact. The objective of this study was to investigate the functional potential of acerola and orange dehydrated by-products, as sources of phenolic compounds, and to develop probiotic fermented milks suplemented with this residues, evaluating its physico-chemical characteristics during refrigerated storage (28 days, 4 ± 1 °C), as well as the impact of gastrointestinal conditions on flavonoids and probiotic strains. The by-products were obtained from fruit processing industries of São Paulo, and the following analyzes were performed to characterize them: contents of moisture, ash, lipids, proteins, vitamin C, minerals, dietary fibers, total phenolic compounds and proanthocyanidins, antioxidant capacity in vitro and flavonoids chromatographic profile (HPLC). Were elaborated four formulations of fermented milks: F0 (control), without addition of acerola residue (AR); F2, with 2% AR; F5, with 5% AR; F10, with 10% AR. Was used the probiotic culture ABT-4, composed of two probiotic strains, Bifidobacterium animalis subsp lactis Bb-12 and Lactobacillus acidophilus La-5, in addition to the starter culture Streptococcus thermophilus. During the refrigerated storage (28 days, 4 ± 1 °C), the following analyzes were performed with the fermented milks: contents of moisture, ash, lipids and proteins, pH, acidity, viability of microorganisms, total phenolic compounds (PC), instrumental color and texture. In addition, the fermented milks were submitted to in vitro simulated gastrointestinal conditions to evaluate the impact on the viability of probiotic strains and phenolic compounds. AR presented excellent content of vitamin C (605 mg/100 g), in addition to presenting better antioxidant capacity in vitro than orange residue (OR). Proanthocyanidins were found only in AR (617 &#181;g CE/g). The PC content of AR (3240 &#181;g GAE/100 g) was 3.6 higher than in OR. The phenolic compounds identified in AR were quercetin-3-rhamnoside, rutin and others quercetin derivatives, procyanidin B1 and kaempferol derivatives. In OR, were identified naringenin, sinensetin, homorientin, isovitexin and chlorogenic acid derivatives. The by-products studied showed high total fibers content (above 60%) and total proteins (AR: 10.4%, OR: 9.9%), as well as reduced total lipids content (AR: 1.6%; OR: 2.6%). Both residues showed high levels of potassium, calcium, magnesium and phosphorus. The four formulations of fermented milks presented low total lipids content (below 1%), and the total proteins content ranged from 3.9 to 5.1 g/100 g, being in agreement with the legislation. The pH of F0 (control) and F2 formulations remained stable (p > 0.05) throughout the refrigerated storage period (28 days, 4 ± 1 °C), and the other formulations showed a small decreased, even thus remaining above 4.5. The acidity of the formulations, ranging from 0.92 to 1.28 mg of lactic acid/g, increased between days 1 and 14 of storage, then remained until the end of shelf life. The AR did not negatively interfere in the populations of microorganisms analyzed during storage, since the formulations F2, F5 and F10 maintained their populations around 8 log CFU/g. Regarding PC content, the samples differed significantly (p < 0.05), with F0 being about 5 lower than F10 (21.13 and 101.13 &#181;g GAE/100 g, respectively, in the day 1). The instrumental color of the products remained until the end of shelf life, and differed significantly (p < 0.05) from each other. The AR influenced a little in the texture parameters of the fermented milks, but the control formulation was the only one that lost adhesiveness. After the gastric phase of the simulated digestion in vitro, on the 7th day of storage, the populations of probiotic bacteria decreased dramatically (of 3 to 5 log CFU/g), and after the enteric phase no colonies were detected. On the other hand, the flavonoids found in the fermented milks that were suplemented with AR increased from 2 to 5 times, after the gastric phase, maintaining or suffering small decreased after enteric phase. These results show that acerola by-products powder is a valuable ingredient to be used in functional foods because it is rich in vitamin C, dietary fibers and phenolic compounds, adding nutritional value, and serving as a natural antioxidant. Its flavonoids appear to be highly resistant to the acids and salts of digestion and can thus have positive effects on health.

(Acai) has demonstrated different biological activities, such as antioxidant, anti-inflammatory, antihypertensive, antinociceptive and antineoplastic. In this new study, the aim was to analyze the antineoplastic activity of fractions derived from hydroalcoholic extract of Euterpe oleracea Mart. seed in cell line MCF-7, and to identify the compounds responsible for the antineoplastic action by mass spectrometry using electrospray ionization source, and a ciclotrônico analyzer coupled to a Fourier transform (ESI-FT-ICR MS). The MCF-7 cell line was treated with 10, 20, 40 and 60 μg L-1 with fractions hexane (FH), chloroform (FC) and ethyl acetate (FAE) of the hydroalcoholic extract acai seed for 24, 48 and 72 hours. After treatment the cell viability was measured using the assay with 3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT) and the cell type of death was assessed using annexin - Iodide propidium (PI) assay. Data were analyzed statistically by analysis of variance (ANOVA) or Student's t test, us appropriate. It was observed that all the fractions caused significant reduction of cell viability of MCF-7, but the FAE was the most cytotoxic (p <0.001). In the test Annexin-PI, there was no significant labeling annexin but increased PI staining was significant (p <0.001). This study showed that the FAE was more effective in reducing cell viability, following necroptose mechanism in MCF-7 cell. The FAE is composed of epicatechin, proanthocyanidin A2 and trimeric and tetrameric procyanidins. / A semente de Euterpe oleracea Mart. (açaí) tem demonstrado diferentes atividades biológicas, tais como: antioxidante, anti-inflamatória, anti-hipertensiva, antinociceptiva e antineoplásica. Neste estudo, o objetivo foi analisar a atividade antineoplásica de frações do extrato hidroalcoólico da semente de Euterpe oleracea Mart. na linhagem celular MCF-7, assim como identificar os compostos responsáveis pela ação antineoplásica por espectrometria de massas utilizando fonte de ionização por eletrospray e um analisador ciclotrônico acoplado a uma transformada de Fourier (ESI- FT-ICR MS). A linhagem MCF-7 foi tratada com 10, 20, 40 e 60 μg L-1 com as frações hexânica (FH), clorofórmica (FC) e de acetato de etila (FAE) do extrato hidroalcoólico da semente do açaí por 24, 48 E 72 horas. Após o tratamento a viabilidade celular foi mensurada usando o ensaio com 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) e o tipo de morte celular foi avaliado com o ensaio Anexina - Iodeto de propídeo (PI). Os dados foram analizados estatisticamente por análise de variância (ANOVA) ou por teste T student, quando apropriado. Foi observado que todas as frações causaram redução significativa da viabilidade celular da MCF-7, porém a FAE foi a mais citotóxica (p < 0,001). No ensaio de anexina-pi, não foi observado marcação significativa para anexina porém o aumento da marcação de PI foi significativo (p<0,001). O presente estudo demonstrou que a FAE foi a mais eficaz na redução da viabilidade celular, seguindo mecanismo de necroptose na célula MCF-7. A FAE é composta por epicatequina, proantocianidina A2 e procianidinas trimérica e tetramérica.

Chá mate (Ilex paraguariensis): compostos bioativos e relação com atividade biológica / Mate tea (Ilex paraguariensis): bioactive compounds and relationship with biological activity

Introdução: A erva mate (Ilex paraguariensis St. Hilaire) é uma espécie nativa do Brasil, Paraguai e Argentina e tem importante apelo social, econômico e cultural. É utilizada na preparação de vários tipos de bebidas, como chimarrão, tererê e chá-mate, consumidas em toda a América Latina. O chá mate, assim como o chimarrão, apresenta potencial protetor à saúde devido à atividade antioxidante, ente outras, conforme demonstrado em diversos estudos. A alta concentração de compostos bioativos, como as saponinas, os compostos fenólicos e as metilxantinas respondem, em conjunto, pela sua atividade biológica. Objetivos: a) validar metodologia para isolar e quantificar as principais classes de compostos bioativos do chá mate (compostos fenólicos, xantinas, sapogeninas e melaninas); b) avaliar se há diferenças nos teores desses compostos nas diferentes marcas deste produto comercializado em São Paulo; c) avaliar a atividade biológica de cada classe por diferentes métodos in vitro. Metodologia: Foram analisados três lotes das três marcas de chá mate 100 por cento presentes em hipermercados no município de São Paulo. A validação da metodologia para quantificação, empregando CLAE, consistiu no cálculo da exatidão (recuperação), repetibilidade e sensibilidade para os compostos: ácido caféico, ácido-5-cafeoilquínico, cafeína, teobromina, acido ursólico e ácido oleanólico. Foram determinados os teores de fenólicos totais por método espectrofotométrico com o reagente Folin-Ciocalteu. As frações de fenólicos e xantinas, sapogeninas e melaninas foram isoladas e testadas quanto à atividade antioxidante pelo: sistema de oxidação -caroteno e ácido linoléico, teste da capacidade de absorbância de oxigênio radical ORAC (Oxygen Radical Absorbance Capacity), capacidade de seqüestrar o radical DPPH, e atividade ligante de ácidos biliares (ácidos glicocólico, cólico, deoxicólico e taurocólico). Resultados: Os coeficientes de determinação das curvas de calibração foram maiores que 0,99. A recuperação dos compostos variou de 84,2 a 115,88por cento e o coeficiente de variação médio foi 5 por cento. Uma xícara de chá mate (182 mL) apresentou, em média, 161,96 mg de fenólicos totais; 14,71 mg de ácido clorogênico (5CQA); 3,13 mg de ácido caféico ; 2,74 mg de teobromina e 15,16 mg de cafeína. Os teores de ácido ursólico e ácido oleanólico no chá mate (sachês), foram, em média 82,60 g/g e 54,08 g/g, respectivamente. As concentrações dos compostos variaram significativamente entre as marcas analisadas. A atividade antioxidante do chá mate (69,2 por cento de inibição da oxidação pelo sistema -caroteno e ácido linoléico; DPPH (IC50) = 0,014 mgSS/mL; ORAC = 9997molTrolox/xícara de chá), diferiu significativamente (p<0,05) entre as marcas apenas para o IC50. Para as frações houve diferença estatística apenas para a porcentagem de inibição da oxidação do ácido linoléico para o extrato melânico. Verificou-se correlação linear (p<0.05) positiva ou negativa entre a atividade antioxidante e a concentração de alguns compostos bioativos. A capacidade ligante de ácidos biliares foi semelhante ao controle positivo colestiramina para o chá mate, FS e FFX. O EM demonstrou baixa ou nenhuma capacidade de si ligar aos ácidos biliares. Conclusões: O chá mate é uma excelente fonte de compostos bioativos com atividade biológica. Ocorreu variação da concentração de compostos bioativos em função da marca analisada. Essa variação, no entanto, não influenciou na atividade antioxidante da bebida, conforme avaliada pelos métodos ora utilizados. / Introduction: Yerba maté (Ilex paraguariensis St. Hilaire) is a spontaneous species that grows in Brazil, Argentina and Paraguay and several beverages (chimarrão, terere, maté tea) are produced from its leaves. Maté tea, one of I. paraguariensis beverages present antioxidant activity and other biological effects. Compounds such as aponins, polyphenols and methylxanthines, which are present in considerable amounts, are responsible for the biological effects. Objectives: The aims of this work were: a) to validate analytical methodologies to evaluate the main bioactive compounds from maté tea (phenolic compounds, xanthines, sapogenins and melanins); b) to evaluate if there are differences on these compounds contents among maté tea brands commercialized in São Paulo; c) to evaluate biological activity of each class of compounds by different in vitro methodologies. Methods: Three lots from three commercial brands of maté tea present in 100 per cent of the main supermarkets in São Paulo city were analysed. HPLC methodology validation was assessed by determining accuracy (recovery), repeatability and sensibility (linearity, limits of detection and quantitation) for caffeic acid, 5- caffeoylquinic acid, caffeine, theobromine, ursolic and oleanolic acids. Total phenolic content was assessed with the Folin-Ciolcalteu´s reagent. Maté tea phenolic and methylxanthines (FFX), sapogenins (FS) and melanins (EM) fractions were isolated and tested for theirs in vitro antioxidantactivity by the following methods: -carotene and linoleic acid system, ORAC (Oxygen Radical Absorbance Capacity), radical scavenging capacity (DPPH). The ability of binding bile acids (glycocholic, cholic, taurocholic and deoxycholic acids) was also assayed. Results: Calibration curves determination coefficient (r 2) were higher than 0,99 for all compounds. Recovery ranged from 84,2 to 115,88per cent and repeatability average was 5per cent. One maté tea cup (182 mL) contains in average 161,96 mg5CQA of total phenolic compounds, 14,71 mg of chlorogenic acid (5CQA), 3,13 mg of caffeic acid, 2,74 mg of theobromine and 15,16 mg of caffeine. The roasted leaves contains 82,60 and 54,08 g/g of ursolic and oleanolic acids in average, respectively. Compounds contents varied significantly (p<0,05) among brands. The antioxidant activity assays showed no significant difference (p>0,05) among brands to maté tea, except for DPPH (IC50), and the average results for the maté tea are the following: 69,2 per cent in -carotene and linoleic acid system, DPPH (IC50) = 0,014mgSS/mL; ORAC = 9997,7molTrolox/tea cup. Antioxidant activity of the isolated fractions showed significant difference only in -carotene and linoleic acid system to melanic extract. A positive or negative correlations between the antioxidant activity and the bioactive compounds contents was observed (p<0.05). Maté tea, FS and FFX bile acid binding capacity were similar to the positive control cholestyramine. EM showed very little or no bile acid binding capacity. Conclusions: Maté tea is a source of bioactive compounds. Although concentration can vary significantly from brand to brand, this fact did not affect maté tea antioxidant activity evaluated by the methods used in this study.

Effets athéroprotecteurs de la curcumine et d’extraits riches en polyphénols d’Antirhea borbonica et de Doratoxylon apetalum / Atheroprotective effects of curcumin and polyphenol rich extracts of Antirhea borbonica and Doratoxylon apetalum

Les maladies cardiovasculaires représentent la première cause de mortalité en France avec une prévalence encore plus importante à La Réunion. La plupart des décès sont attribuables aux cardiopathies ischémiques due à la rupture d'une plaque d'athérosclérose et à la formation d'un caillot entrainant une ischémie cardiaque. L'oxydation des lipoprotéines de basse densité (LDL) et la dysfonction de l'endothélium représentent des étapes importantes dans la déstabilisation de la plaque, d'où l’intérêt de rechercher un traitement capable de diminuer l'oxydation des LDL ou de protéger les cellules endothéliales de leurs effets cytotoxiques. Les polyphénols sont des molécules antioxydantes très présentes dans le règne végétal. Des études préliminaires du laboratoire sur des adipocytes humain on montré que des extraits riches en polyphénols d'Antirhea borbonica et de Doratoxylon apetalum, deux espèces respectivement endémique et indigène de l'Île de La Réunion, possédaient des propriétés antioxydantes et anti-inflammatoires (Marimoutou, M et al. 2015). Le But de cette thèse était, dans un premier temps, de tester les capacités antioxydantes et cytoprotectrices d'extraits riches en polyphénols de ces deux espèces sur des cellules endothéliales. Puis dans un second temps nous avons testé l'extrait de Doratoxylon apetalum sur un modèle murin d'athérosclérose (souris ApoE KO) pour évaluer son effet anti-inflammatoire sur les plaques d'athérome. / Cardiovascular diseases are the leading cause of death in France with an even higher prevalence in Reunion. Most deaths are due to ischemic heart disease because of atherosclerotic plaque rupture and thrombus formation leading to cardiac ischemia. The oxidation of low density lipoproteins (LDL) and endothelial dysfunction represent important steps in the destabilization of plaque, hence the interest of a treatment capable of reducing the oxidation of LDL or of protecting endothelial cells from their cytotoxic effects. Polyphenols are antioxidant molecules very present in the plant kingdom. Preliminary studies on human adipocytes have shown that polyphenol-rich extracts of Antirhea borbonica and Doratoxylon apetalum, respectively endemic and indigenous species of Reunion Island, had antioxidant and anti-inflammatory properties (Marimoutou , M et al., 2015). The aim of this thesis was, first, to test the antioxidant and cytoprotective capacities of polyphenol rich extracts of these two species on endothelial cells. Then in a second time we tested the extract of Doratoxylon apetalum on a murine model of atherosclerosis (ApoE KO mice) to evaluate its anti-inflammatory effect on atheroma plaques.

Mécanismes de la perte osseuse et musculaire avec l'âge et étude du potentiel de l'huile d'olive, de la vitamine D et du DHA pour une stratégie nutritionnelle de prévention

Les sociétés économiquement développées sont confrontées à la problématique du vieillissement de leur population associée à la recrudescence des pathologies dégénératives. Parmi les manifestations du vieillissement, l'atteinte de l'appareil locomoteur (ostéoporose et sarcopénie) est considérée comme un problème majeur de santé publique. De ce fait, le développement de mesures préventives permettant de préserver la santé des séniors constitue un véritable enjeu de santé publique. La recherche scientifique fournit de nombreuses preuves concernant l'importance de l'alimentation dans la diminution ou l'exacerbation de certaines maladies. Ces observations confirment le vieux concept soutenant que l'adoption d'une alimentation équilibrée permet le maintien d'un état de santé satisfaisant, en particulier avec l'avancée en âge. A ce jour, le potentiel de l'huile d'olive, en tant que telle ou comme vecteur de nutriments lipophiles d'intérêt pour la prévention nutritionnelle de l'ostéoporose et de la sarcopénie, n'a jamais été testé. La prévalence de l'ostéoporose étant très supérieure chez la femme ménopausée qui souffre également d'une perte musculaire accélérée, cette population a été ciblée dans la présente étude. Dans une première étape, des résultats originaux ont été obtenus concernant l'effet de l'ovariectomie sur le métabolisme musculaire. Ainsi, notre travail montre qu'après ablation des ovaires, la synthèse protéique musculaire est réduite, en association avec une diminution d'un marqueur de la protéolyse de ce tissu. Pris dans son ensemble, ces résultats démontrent un ralentissement du renouvellement protéique musculaire suite à l'ovariectomie. En termes de mécanismes mis en jeu, une augmentation de l'infiltration lipidique tissulaire a été observée. En parallèle du ralentissement du renouvellement musculaire après ovariectomie, une augmentation du renouvellement osseux, classiquement décrit dans la littérature, a été mis en évidence. Dans un second temps, l'effet de l'huile d'olive et plus particulièrement des polyphénols présents dans cet aliment en association avec la vitamine D et l'acide ocosahexaénoïque et leur éventuelle synergie a été évalué à l'aide d'un modèle d'ostéoporose post-ménopausique. Des souris ovariectomisées ont été soumises à une des 4 huiles suivantes : huile de maïs, huile d'olive raffinée, huile d'olive extra vierge ou huile de poisson. Pour chaque huile, un groupe supplémentaire de souris ovariectomisées a reçu de l'huile enrichie en vitamine D. Cette étude a permis de mettre en évidence un effet protecteur de l'association de l'huile d'olive extra vierge et de la vitamine D sur la perte osseuse induite par l'ovariectomie chez la souris. En effet, l'accélération du renouvellement osseux consécutive à la carence estrogénique est freinée par cette association de nutriments, qui diminuent également l'expression des marqueurs du stress oxydant et de l'inflammation. Les résultats obtenus sont en cours de validation chez l'Homme grâce à une investigation clinique réalisée chez des femmes ménopausées. Celle-ci a été mise en place au cours de ces travaux de thèse. / Economically developed societies face the problem of an aging population combined with the increase in degenerative diseases. Among the signs of aging, musculoskeletal pathologies (osteoporosis and sarcopenia) are considered major public health problems. Therefore, the development of preventive measures to preserve seniors’ health is a real public health issue. Scientific research provides ample evidence of the importance of diet in diseases reduction or exacerbation. These observations confirm the old concept arguing that the adoption of a balanced diet helps to maintain a healthy life, especially with advancing age. To date, the potential of the olive oil, as such or as a vector of lipophilic nutrients, for the prevention of osteoporosis and sarcopenia, has never been tested. As the prevalence of osteoporosis is much higher in postmenopausal women who also suffer from an accelerated muscle mass loss, this population was targeted in this study. In a first step, original results were obtained regarding the effect of ovariectomy on muscle metabolism. Thus, our work showed that after removal of the ovaries, muscle protein synthesis was reduced in association with a decrease in the expression of a proteolysis marker of this tissue. Taken together, these results suggest a slowing down of muscle protein turnover after ovariectomy. In terms of mechanisms, an increase in lipid tissue infiltration was observed. The slowdown in muscle protein renewal after ovariectomy was associated with an increased bone turnover, classically described in the literature. In a second step, the effect of olive oil and more particularly its polyphenols in combination with vitamin D and docosahexaenoic acid and their possible synergy was assessed using a postmenopausal osteoporosis model. Ovariectomized mice were subjected to one of the following oils: corn oil, refined olive oil, extra virgin olive oil or fish oil. Four additional groups of ovariectomized mice received the oil fortified with vitamin D. This study demonstrated a protective effect of the combination of virgin olive oil and vitamin D on bone loss induced by ovariectomy in mice. Indeed, the acceleration of the bone turnover induced by the estrogen deficiency was inhibited by this combination of nutrients, which also decreased the expression of oxidative stress and inflammation markers. These results are currently being validated in a clinical investigation conducted in postmenopausal women, which has been established during this thesis work.

Impact des flavanones des agrumes sur la prévention de l'athérosclérose et mécanismes d'action mis en jeu / Impact of citrus flavanones on atherosclerosis prevention and underlying mecanisms involved

Les maladies cardiovasculaires représentent l’une des principales causes de mortalitédans le monde et leur prévention constitue un enjeu majeur de santé publique. Les études épidémiologiques montrent que la consommation de fruits et légumes est associée à un moindre risque cardiovasculaire. Ces effets pourraient être expliqués par leur richesse en micronutriments et plus particulièrement en polyphénols. La consommation de flavanones, une classe de polyphénols présente spécifiquement et en grandes quantités dans les agrumes, a été inversement associée au risque d’accidents coronaires et d’accidents vasculaires cérébraux. Des données expérimentales obtenues in vivo et in vitro suggèrent que les flavanones (hespéridine ou naringine) possèdent des propriétés hypolipémiantes, hypotensives et anti-inflammatoires. Ces effets pourraient notamment être médiés par une modulation de l’expression de gènes par les flavanones. Cependant jusqu’à présent, l’action anti-athérogène des flavanones in vivo n’a été explorée qu’à des doses supra-nutritionnelles et les mécanismes responsables de ces effets sont encore largement méconnus. Par ailleurs, les données mécanistiques in vitro sont peu informatives, car obtenues avec des formes de flavanones non présentes dans l’organisme. Les objectifs de cette thèse ont donc été : (1) d’évaluer l’impact d’une consommation de flavanones (naringine et hespéridine) à dose nutritionnelle sur le développement de la lésion athéromateuse chez différents modèles murins d’hypercholestérolémie et d’appréhender les mécanismes moléculaires mis en jeu par une approche transcriptomique ; (2) de déterminer l’impact des métabolites circulants sur la fonction des cellules endothéliales et d’identifier les mécanismes moléculaires sous-jacents. In vivo, nous avons montré que seule la supplémentation en naringine (0,02%), la flavanone majoritaire du pamplemousse, réduisait la progression des lésions athéromateuses dans un modèle murin d’athérosclérose induite par l’alimentation (souris C57Bl/6J sous régime riche en graisse et en cholestérol). Cet effet était associé à une réduction des concentrations plasmatiques de non HDL-Chol et de biomarqueurs de la dysfonctionendothéliale, mais semblait indépendant d’une modulation du statut anti-oxydant. Une analyse transcriptomique pangénomique de l’aorte de ces animaux, a montré, que la supplémentation en naringine induisait une variation de l’expression de gènes impliqués dans des processus, tels que l’adhésion cellulaire ou l’organisation du cytosquelette, qui sont impliqués dans la migration trans-endothéliale des leucocytes. Ces résultats mettent en évidence de nouvelles cibles moléculaires d’action des flavanones qui ont été plus largement abordées in vitro. Les données obtenues in vitro montrent que l’exposition de cellules endothéliales(HUVEC) à des concentrations physiologiques de métabolites circulants de la naringénine(dérivés glucuronidés) ou de l’hespérétine (dérivés glucuronidés et sulfatés) diminue l’adhésion des monocytes (U937) aux cellules endothéliales, une étape clé de la migrationtrans-endothéliale. En accord avec ces résultats, une analyse TaqMan Low Density Array a montré que l’exposition des cellules endothéliales aux métabolites circulants de flavanones affectait l’expression de gènes impliqués dans l’inflammation, le chimiotactisme, l’adhésion ou encore la migration trans-endothéliale des leucocytes. Pris dans leur ensemble, les résultats de ce travail de thèse montrent que les effets antiathérogènesdes flavanones ne découlent pas exclusivement d’un effet hypolipémiant, mais aussi d’une action directe sur des étapes clés du développement de l’athérosclérose au niveau de l’endothélium, en particulier les processus d’adhésion et migration trans-endothéliale. / Cardiovascular disease is a major cause of mortality worldwide and its prevention is a major public health issue. Epidemiological studies have shown that fruit and vegetables consumption is associated with a lower cardiovascular risk. These effects could be explainedby their richness in micronutrients, especially polyphenols. Consumption of flavanones, a class of polyphenols present specifically and in large amounts in citrus fruits, was inversely associated with risk of coronary events and stroke. Experimental data obtained in vivo and invitro suggest that the flavanones (hesperidin or naringin) have lipid-lowering, hypotensive and anti-inflammatory properties. These effects could be mediated via modulation of gene expression by these bioactives. However, the anti-atherogenic effect of flavanones in vivo has been only explored at supra-nutritional doses and the mechanisms responsible for these effects are largely unknown. Furthermore, in vitro mechanistic data are questionable as they have been obtained using native forms of flavanones which are not present in the body.The objectives of this thesis were: (1) to assess the impact of consumption of flavanones (naringin and hesperidin) at nutritional doses on the development of atherosclerotic lesions indifferent mouse models of hypercholesterolemia and decipher molecular mechanisms brought into play, using a transcriptomic approach, (2) to determine the impact of physiological concentrations of plasma flavanone metabolites on endothelial cell function and theunderlying molecular mechanisms. In vivo, we showed that only naringin supplementation (0.02%), the major flavanonein grapefruit, reduced the progression of atherosclerotic lesions in a mouse model of dietinduced atherosclerosis (C57BL/6J mice fed an enriched diet in fat and cholesterol). Thiseffect was associated with a reduction in plasma non-HDL-Chol and biomarkers ofendothelial dysfunction, but appeared independent of a modulation of antioxidant status. Agenome-wide transcriptome analysis of the aorta of these animals showed that naringin supplementation modulated expression of genes involved in processes such as cell adhesion and cytoskeleton organization; these latter being involved in leukocytes trans-endothelialmigration. These results reveal new molecular targets of action of flavanones that have beenfurther studied in vitro.The in vitro data showed that exposure of endothelial cells (HUVEC) to physiological concentrations of circulating metabolites of naringenin (glucuronides) or hesperetin(glucuronides and sulfate) decreased the adhesion of monocytes (U937) to endothelial cells, a key step in the trans-endothelial migration. Consistent with these results, a TaqMan Low Density Array analysis showed that exposure of endothelial cells to these flavanone metabolites affected the expression of genes involved in inflammation, chemotaxis, adhesionas well as leukocytes trans-endothelial migration. Overall, the results of this work show that the anti-atherogenic effect of flavanones is not exclusively derived from lipid-lowering effect, but also due to a direct action on the endothelium by modulating key processes of atherosclerosis development, particularly adhesion and trans-endothelial migration.

Study of the Electron-Transfer properties of phenolics and their relationship with the biological activity on cancer cells

During aerobic cell metabolism, molecular oxygen is reduced to water through electron-transfer reactions, and the oxygen not completely reduced is transformed into a set of neutral and free radical molecules with high oxidant ability, generally denominated Reactive Oxygen Species (ROS). To regulate the cellular ROS content, organisms are endowed with an efficient endogen antioxidant system. The physiological ROS levels may be excessively increased by different factors including unbalanced diets, ionizing radiations, and tobacco smoke among others, leading to the oxidative stress, term referred to the imbalance between ROS production and its neutralization by the endogenous antioxidant system. More importantly, in vitro and in vivo studies reveal the relationship between suffering oxidative stress and the development of cardiovascular and respiratory diseases, diabetes, and cancer. To prevent and treat the oxidative stress and its adverse effects, exogenous, readily oxidizable molecules may help the organism to neutralize ROS into less dangerous species. The phenolic compounds or (poly)phenols, molecules very abundant in our diet (i.e. fruits, vegetables, and beverages such as tea and wine) may be responsible for this antioxidant activity of fruits and vegetables. Currently, (poly)phenols are regarded as natural antioxidant molecules with outstanding beneficial effects, including anti-aging activity, and the prevention of cancer and diabetes. The antioxidant activity of (poly)phenols is mainly associated with their radical scavenging activity, action conferred by the transfer of a hydrogen atom or an electron to a free radical, rendering a less reactive molecule. On the other hand, chemical probes and in vitro studies have demonstrated that some highly reacting (poly)phenols are able to generate small quantities of ROS. Phenolic compounds have shown antiproliferative activity which may be caused inter alia by their ability to scavenge or generate toxic radicals. To further understand the connection between the redox reactivity of (poly)phenols and their biological actions, studies with more sensitive and selective chemical probes may help to clarify the role of redox reactions in the physiological actions of phenolics and their metabolites. In this thesis, we have focused on the utilization of two stable radicals synthesized in our laboratory, the tris(2,4,6-trichloro-3,5-dinitropehnyl)methyl (HNTTM) and the tris(2,3,5,6-tetrachloro-4-nitrophenyl)methyl)radical (TNPTM), as chemical probes to determine the electron-transfer activity of dietary (poly)phenols and some metabolites. The different reducing potential of the two stable radicals facilitates the quantitative evaluation of the radical scavenging capacity of each (poly)phenol, as well as the establishment of the most reactive moieties. TNPTM is a useful tool to determine the most reactive (poly)phenols as electron transfer donors, (poly)phenols that cannot be differentiate with any other chemosensor. The results obtained are compared with two well-established methods for the quantification of electron-transfer capacity. The action of these (poly)phenols on cell cultures of a colon cancer cell line is also presented, showing a correlation between those (poly)phenols detected with TNTPM and with those that produce the highest antiproliferative activity. This cell line is particularly relevant because dietary (poly)phenols are in contact with epithelial cells of this kind during their transit along the digestive tract and may exert some preventive action on colon cancer. In addition, a chemoenzymatic strategy to prepare glucuronated metabolites of (-)-epigallocatechin-3-O-gallate (EGCG), the most abundant and active (poly)phenol of green tea, was attempted, obtaining the acetylated and methylated precursor of the EGCG-4’’-glucuronide, the most abundant EGCG glucuronide obtained in the human metabolism. The final conjugated, the EGCG-4’’-glucruonide, was obtained albeit with not enough quantity to be purified. / "Estudi de la Transferència Electrònica de compostos fenòlics i la seva relació amb l’activitat biològica en cèl•lules canceroses" En el metabolisme i respiració dels organismes aeròbics, l’oxigen és utilitzat com a receptor electrònic reduint-se principalment a aigua. Una petita part d’aquest oxigen no és totalment reduint obtenint-se un conjunt d’espècies radicalàries i no radicalàries (ROS). Aquestes espècies en ser altament reactives poden malmetre macromolècules, activitat que està relacionada amb l’aparició de malalties com el síndrome metabòlic i el càncer. Per aquest motiu els organismes han desenvolupat un sistema de regulació de la seva concentració. En determinades situacions, aquest sistema no és suficient i per tant, antioxidants exògens poden ajudar a aquest sistema endògen. Els polifenols molècules abundants en la nostra dieta (fruita i verdures) són els principals candidats, i s’ha demostrat un efecte preventiu i terapèutic en la salut per la seva part. L’efecte beneficiós exercit pels polifenols pot tenir lloc per dos mecanismes de reacció; per transferència d’hidrogen i per transferència electrònica. Actualment, no hi ha cap mètode prou eficaç que ens permeti determinar la acció del polifenols exercida per transferència electrònica, mecanisme que també habilita als polifenols per a formar ROS. En el nostre laboratori hem sintetitzat dos radicals lliures estables, els radicals tris(2,4,6-tricloro-3,5-dinitrofenil)metil (HNTTM) i el tris(2,3,5,6-tetracloro-4-nitrofenil)metil (TNPTM) com a quimiosensors de transferència electrònica. Aquests radicals ens permeten mesurar la activitat antiradicalària dels polifenols, i el TNPTM ens permet detectar a aquells polifenols amb una reactivitat per transferència electrònica més elevada i que no poden ser diferenciats amb altres mètodes. S’han fet estudis d’antiproliferació per part dels polifenols en cèl•lules de càncer de còlon HT-29 i s’ha demostrat que els polifenols amb més activitat antiproliferativa són els que tenen activitat front al TNPTM. Per tant, el radical TNPTM permet determinar als polifenols amb una elevada reactivitat per a cedir electrons i es suggereix que els mecanismes de transferència electrònica juguen un paper important en els mecanismes d’ antiproliferació en cèl•lules de càncer de còlon HT-29. S’ha proposat metodologia sintètica per a obtenir els principals glucuronats de l’epigal•locatequin-3-O-gal•lat (EGCG) el més abundant i més actiu dels polifenols del te verd. S’ha obtingut el precursor acetil•lat i metil•lat del glucuronat majoritari de l’EGCG obtingut en el metabolisme humà, l’EGCG-4’’-glucurònid i s’ha obtingut el conjugat final però no amb una quantitat suficient com per a ser aïllat.

The allergic disease of asthma is characterized by an infiltration of inflammatory cells to the lung, a process co-ordinated by T-helper (TH) cells. The TH2 cytokine Interleukin (IL)-4 promotes infiltration of eosinophils to sites of inflammation. Eosinophil-selective chemoattractant cytokines (eg. eotaxins) are synthesized by lung epithelial cells. Eotaxin-3 is expressed at high levels in the asthmatic lung, predominantly after IL-4 stimulation. Eotaxin-3 is therefore a marker of inappropriate airway inflammation. Polyphenolic (PP) compounds found in high concentrations in berries may have beneficial effects in inflammatory conditions. Plant and Food Research produced high-PP extracts of blackcurrant (BC) cultivars that were tested for inflammation modulating effects. Since high doses of PPs have been shown to cause cell death, we tested two BC cultivars at a range of concentrations in a cell viability (WST-1) assay. While no toxic effects were attributable to the BC extracts (1-50µg/ml), a dose-related trend in cell death was observed and therefore 10µg/ml was chosen for further experiments Ten BC cultivars were compared for efficacy by measuring eotaxin-3 production in IL-4 stimulated human lung epithelial (A549) cells in vitro. Cells were incubated with BC extracts (10µg/ml) and IL-4 (10ng/ml) for 24 hours. The supernatants were then quantified for eotaxin-3 levels by an enzyme-linked immunosorbent assay (ELISA). All ten BC extracts reduced eotaxin-3 levels after stimulation with IL-4, and six BC extracts were effective by statistically significant levels (P<0.05), (BC cultivars -01, -02, -03, -05, -09 & -10). Of those, BC extracts of four cultivars demonstrated a reduction of more than 65% from the IL-4 stimulated control. In addition, a positive trend in inflammation modulation vs. one anthocyanin (ACN) in the BC extracts was shown. This study has demonstrated the beneficial inflammation modulatory effects of polyphenolic BC extracts, which could be related to cyanidin 3-O-rutinoside content. These results may have therapeutic potential for asthma.
