Deformačně napěťová analýza TEP kyčelního kloubu – typ Santori / Stress - strain analysis of total hip replacement - type Santori

Submitted Diploma thesis deals with stress-strain analysis of deformation proximal end of femur with applied total hip joint endoprosthesis (replacement) – shortcut type. To identify deformation and tensity (stress) was used computational simulation by method of final elements. Have been created two computational models TEP- type Santori and type DePuy Proxima. Geometry model Santori was created on low level model geometry through the use of X-ray photograph. Principle of geometry model type DePuy Proxima was real Femoral stem endoprosthesis which was scanned on scanner ATOS. Geometry of both these replacements were set up in program Rhinoceros 4.0 and then execute in program CatiaV5R17. Data for geometry model of femur were gained from CT chains. Material model of femur have been crated in two variants. The first one looks at structure bone tissues and the second one were created by Gruen´s zones. Femoral Stem was weighted by static equivalent resultant force acting in hip joint. Computational model of system and self solution, including depiction results, was done by ANSYS Workbench 11.0 for four model variants.

This diploma thesis deals with hip endoprothesis, primarily with endoprothesis conservative Mayo. A part of this paper considers a study aimed at an anatomy, types and modern trends in endoprothetic. On the basis of this study, gained CT data and the help of systems Ansys Workbench, Rhino Ceros and Catia there was made a model of proximal part of femur and TEP Conservative Mayo. A computational model was build up afterwards, which was used for a stress-train analysis.

Abstract The aim of the present thesis was to choose and adjust a suitable methodology for counting particles in 3D space, which would be suitable for unbiased estimation of chloroplast number in needle mesophyll cells. The disector method was used for the first time to determine the number of chloroplasts. This method enables unbiased estimation of chloroplast number in needle volume from optical sections captured from fresh free-hand sections by confocal microscope. The sections did not need any pre-processing. Another aim was to compare selected photosynthetic and anatomical characteristics of sun and shade Norway spruce needles, which were grown under different CO2 concentration. The trees were grown for eight years in ambient (during the experiment increasing from 357 up to 370 µmol CO2 ∙ mol-1 ) CO2 concentration or elevated (700 µmol ∙ mol-1 ) CO2 concentration in special glass domes on an experimental research site of the Institute of Systems Biology and Ecology, Academy of Sciences of the Czech Republic at Bílý Kříž in Moravskoslezské Beskydy mountains. Étude comparative des lésions cérébrales dans deux maladies démyélinisantes pédiatriques récurrentes : la sclérose en plaques et la maladie associée aux anticorps anti- glycoprotéine oligodendrocytique de myéline

Les syndromes démyélinisants acquis (SDA) pédiatriques sont un groupe de maladies qui affectent la substance blanche (SB) et la substance grise (SG) du système nerveux central (SNC) chez les enfants, et qui partagent certaines caractéristiques et mécanismes pathologiques. Les SDA peuvent être monophasiques ou récurrents. Les SDA comprennent des maladies telles que l'encéphalomyélite aiguë disséminée (EMDA), les troubles du spectre de la neuromyélite optique (TS-NMO), la sclérose en plaques (SEP) et le syndrome démyélinisant récurrent avec anticorps contre la glycoprotéine oligodendrocytique de myéline (anticorps anti-MOG). Ce dernier syndrome, appelé aussi maladie MOG+, a été reconnu récemment comme une entité distincte faisant partie des maladies démyélinisantes récurrentes chez les enfants. La maladie MOG+ présente des caractéristiques semblables à celles de la SEP; en effet, certains cas ont été déjà considérés comme une forme « atypique » de SEP. La maladie MOG+ et la SEP partagent des lésions dans la SB du SNC, mais la SEP est caractérisée aussi par des lésions corticales (LCs) cérébrales, insuffisamment étudiées dans la maladie MOG+. Par conséquent, le but de cette recherche a été de comparer les caractéristiques démographiques et des lésions cérébrales visibles sur des études d’imagerie par résonance magnétique (IRM) chez les enfants atteints de SEP et ceux atteints de la maladie MOG+. Pour atteindre cet objectif, nous avons utilisé des scans IRM 3T, incluant les contrastes pondérés T1, FLAIR et des images de transfert de magnétisation (ITM) de 14 enfants atteints de SEP et 13 enfants atteints de la maladie MOG+. Nous avons mesuré le nombre des LCs, le volume des lésions dans la SB et les valeurs normalisées d’ITM dans les LCs et les lésions de la SB. Nos résultats ont montré que les enfants atteints de la maladie MOG+ étaient plus jeunes au début de la maladie et que celle-ci présentait une durée plus longue que la maladie du groupe SEP. Quant aux lésions cérébrales, les LCs étaient présentes dans la maladie MOG+, mais leur nombre était significativement plus élevé dans le groupe SEP. Cependant, les valeurs normalisées d’ITM dans ces lésions (valeurs qui sont sensibles à la quantité de myéline) n'étaient pas significativement différentes entre les deux groupes. En plus, le volume des lésions de la SB était significativement plus élevé dans le groupe SEP et les valeurs normalisées d’ITM dans ces lésions, significativement inférieures comparativement à la maladie MOG+, témoignant ainsi d’une démyélinisation plus sévère et des différences potentielles dans les mécanismes de démyélinisation. / Pediatric Acquired Demyelinating Syndromes (ADS) are a group of diseases that affect the white matter (WM) and gray matter (GM) of the central nervous system (CNS) in children and that share similar pathological characteristics and mechanisms. ADS can be monophasic or recurrent. The ADS include diseases like acute disseminated encephalomyelitis (ADEM), neuromyelitis optic spectrum disorders (NMO-SD), multiple sclerosis (MS) and Relapsing Myelin Oligodendrocyte Glycoprotein (MOG) syndrome or MOG+ disease, which has been recently recognised as a distinct pathology and is part of the relapsing ADS in children. MOG+ disease shares features with MS; indeed, some MOG+ cases have been considered as an “atypical” form of MS until recently. Both MOG+ disease and MS present lesions in the WM of the CNS. MS is also characterized by focal brain cortical lesions (CL), which have not been extensively studied in MOG+ disease yet. For this reason, the aim of this research project was to compare the demographic and brain magnetic resonance imaging (MRI) characteristics of children with MS and children with MOG+ disease. To achieve our goal, we used 3T MRI including T1-weighted, FLAIR and magnetization transfer ratio (MTR) contrasts of 14 MS participants, and 13 relapsing MOG+ participants. CL counts, WM lesion volumes, normalized MTR values in CLs, and WM lesions were compared across groups. Our results show that children with MOG+ disease were younger at disease onset and had a longer disease duration compared to the MS group. CL were present in MOG+ participants, but counts were significantly higher in the MS group. However, their normalized-MTR values, which are sensitive to myelin, were not significantly different between both groups. WM lesion volumes were significantly higher in the MS group, but their normalized MTR values were significantly lower than in MOG+ WM lesions, likely reflecting more severe demyelination and potential differences in the demyelinating mechanism.

La présence des artistes chinois en France durant la première moitié du XXe siècle s’est fixée de manière exceptionnelle et durable à l’École des beaux-arts de Paris, au point qu’on puisse, à partir du dépouillement des archives nationales (cote AJ52), parler d’un « phénomène chinois à l’École des beaux-arts de Paris ». Ce phénomène a engagé plus de 130 élèves chinois inscrits dans les galeries et dans les ateliers de peinture et de sculpture entre 1914 et 1955. Aussi, cette présence à l’École des beaux-arts constitua une caractéristique essentielle du mouvement des artistes chinois en France et plus largement en Occident. À ce titre, ce phénomène a joué un rôle important dans l’évolution du champ artistique chinois moderne, sur le plan social,technique et artistique à travers un processus de « transfert culturel ». Ce phénomène fut possible grâce à une relation privilégiée qui exista entre la France et la Chine au début du XXe siècle (le « dialogue entre deux républiques »). Mais l’École des beaux-arts fut aussi un espace de concurrence entre les différentes tendances artistiques modernes chinoises dont beaucoup des chefs de file passèrent par les ateliers de l’École. Parmi eux,Xu Beihong (1895-1953) occupa une place particulière car il développa une stratégie sociale et artistique cohérente qui posait comme fondamentales la formation artistique académique et l’expérience à l’École des beaux-arts de Paris. Cette expérience enrichie par la maîtrise du dessin académique, de l’anatomie artistique et de la peinture d’histoire, fut orientée vers une production inédite à bien des égards en Chine, à l’huile et à l’encre. Aussi, après une période consensuelle des années 1910 aux années 1920, il semble qu’à partir des années 1930, le phénomène chinois à l’École des beaux-arts de Paris alimenta essentiellement le pôle éducatif et artistique de Xu Beihong en France et en Chine. Ce phénomène chinois à l’École des beaux-arts de Paris,attaché à la formation académique et à l’art académique français, fut un élément dynamique dans l’élaboration de la modernité artistique en Chine au XXe siècle. / The presence of Chinese artists in France during the first half of the Twentieth Century was an exceptional and enduring phenomenon at the National School of Fine Arts of Paris (École nationale des Beaux-Arts de Paris). Based on the analysis of the documents from the French National Archives, the number of Chinese students was so substantial that it deserves to be called as the 'Chinese phenomenon at the École des Beaux-Arts'. Between 1914 and 1955, more than 130 Chinese students enrolled at the 'Galeries' (preparatory training in drawing) and at the painting and sculpture studios called 'Ateliers'. This situation at the École des Beaux-Arts essentially reflected the movement of Chinese artists in France and more widely in the West. It played an important role in the changing field of the modern Chinese art, socially, technically and artistically ,through a process of "Cultural Transfer" and was made possible by the privileged relationship between France and China at the beginning of the Twentieth Century (the "Dialogue between two Republics"). Nevertheless, the École des Beaux-Arts also became an area of competition between the various modern Chinese artistic tendencies, as many leaders of different groups studied at the workshops of the École des Beaux-Arts. Amongthem, Xu Beihong (1895-1953), who developed a coherent social and artistic strategies, was especially significant. Xu received fundamental academic artistic training at the École des Beaux-Arts in Paris. Xu’s experience, enriched by his mastery of academic drawing, artistic anatomy and history painting, made his artistic production unprecedented in many respects of Chinese art, in oil and in ink. In addition, after a consensual period from the 1910s to the 1920s, it seems that from the 1930s, the Chinese phenomenon at the École des Beaux-Arts in Paris mainly fostered Xu’s central position in educational and artistic camps inFrance and China. This Chinese phenomenon at the École des Beaux-Arts in Paris, which is attached to academic training and to French academic art, was a dynamic element in the elaboration of artistic modernityin Twentieth Century China.

Modelling cortical laminae with 7T magnetic resonance imaging

To fully understand how the brain works, it is necessary to relate the brain’s function to its anatomy. Cortical anatomy is subject-specific. It is character- ized by the thickness and number of intracortical layers, which differ from one cortical area to the next. Each cortical area fulfills a certain function. With magnetic res- onance imaging (MRI) it is possible to study structure and function in-vivo within the same subject. The resolution of ultra-high field MRI at 7T allows to resolve intracortical anatomy. This opens the possibility to relate cortical function of a sub- ject to its corresponding individual structural area, which is one of the main goals of neuroimaging. To parcellate the cortex based on its intracortical structure in-vivo, firstly, im- ages have to be quantitative and homogeneous so that they can be processed fully- automatically. Moreover, the resolution has to be high enough to resolve intracortical layers. Therefore, the in-vivo MR images acquired for this work are quantitative T1 maps at 0.5 mm isotropic resolution. Secondly, computational tools are needed to analyze the cortex observer-independ- ently. The most recent tools designed for this task are presented in this thesis. They comprise the segmentation of the cortex, and the construction of a novel equi-volume coordinate system of cortical depth. The equi-volume model is not restricted to in- vivo data, but is used on ultra-high resolution post-mortem data from MRI as well. It could also be used on 3D volumes reconstructed from 2D histological stains. An equi-volume coordinate system yields firstly intracortical surfaces that follow anatomical layers all along the cortex, even within areas that are severely folded where previous models fail. MR intensities can be mapped onto these equi-volume surfaces to identify the location and size of some structural areas. Surfaces com- puted with previous coordinate systems are shown to cross into different anatomical layers, and therefore also show artefactual patterns. Secondly, with the coordinate system one can compute cortical traverses perpendicularly to the intracortical sur- faces. Sampling intensities along equi-volume traverses results in cortical profiles that reflect an anatomical layer pattern, which is specific to every structural area. It is shown that profiles constructed with previous coordinate systems of cortical depth disguise the anatomical layer pattern or even show a wrong pattern. In contrast to equi-volume profiles these profiles from previous models are not suited to analyze the cortex observer-independently, and hence can not be used for automatic delineations of cortical areas. Equi-volume profiles from four different structural areas are presented. These pro- files show area-specific shapes that are to a certain degree preserved across subjects. The Hair Bundle: Fluid-Structure Interaction in the Inner Ear

A multitude of processes cooperate to produce the sensation of sound. The key initial step, the transformation from mechanical motion into an electrical signal, takes place in highly specialized mechanosensitive organelles that are called hair bundles due to their characteristic appearance. Each hair bundle comprises many apposed cylindrical stereocilia that are located in a liquid-filled compartment of the inner ear. The viscous liquid surrounding the hair bundle dissipates energy and dampens oscillations, which poses a fundamental physical challenge to the high sensitivity and sharp frequency selectivity of hearing. To understand the structure-function relationship in this complex system, a realistic physical model of the hair bundle with an appropriate representation of the fluid-structure interactions is needed to identify the relevant physical effects. In this work a novel approach is introduced to analyze the mechanics of the fluid-structure interaction problem in the inner ear. Because the motions during normal mechanotransduction are much smaller than the geometrical scales, a unified linear system of equations describes with sufficient accuracy the behavior of the liquid and solid in terms of a displacement variable. The finite-element method is employed to solve this system of partial differential equations. Based on data from the hair bundle of the bullfrog's sacculus, a detailed model is constructed that resolves simultaneously the interaction with the surrounding liquid as well as the coupling liquid in the narrow gaps between the individual stereocilia. The experimental data are from high-resolution interferometric measurements at physiologically relevant amplitudes in the range from a fraction of a nanometer to several tens of nanometers and over a broad range of frequencies from one millihertz to hundred kilohertz. Different modes of motion are analyzed and their induced viscous drag is calculated. The investigation reveals that grouping stereocilia in a bundle dramatically reduces the total drag as compared to the sum of the drags on individual stereocilia moving in isolation. The stereocilia in a hair bundle are interconnected by oblique tip links that transmit the energy in a sound to the mechanotransduction channels and by horizontal top connectors that provide elastic coupling between adjacent stereocilia. During hair-bundle deflections, the tip links induce additional drag by causing small but very dissipative relative motions between stereocilia; this effect is offset by the horizontal top connectors that restrain such relative movements, assuring that the hair bundle moves as a unit and keeping the total drag low. In the model the stiffness of the links, the stiffness of the stereocilia, and the geometry are carefully adjusted to match experimental observations. The references are stiffness and drag measurements, as well as the coherence measurements for the bundle's opposite edges, both with and without the tip links. The results are further validated by a comparison with the relative motions measured in a sinusoidally stimulated bundle for the distortion frequencies at which movements are induced by the nonlinearity imposed by channel gating. The model of the fluid-structure interactions described here provides insight into the key step in the perception of sound and the method presented provides an efficient and reliable approach to fluid-structure interaction problems at small amplitudes. / Bei der Hörwahrnehmung eines Klangs spielen viele komplexe Prozesse zusammen. Der Schlüsselprozess, die Umwandlung mechanischer Schwingungsbewegung in elektrische Signale, findet in den Haarbündeln im Innenohr statt. Diese Haarbündel sind hoch entwickelte mechanosensitive Organellen, bestehend aus vielen nahe beieinander stehenden Stereozilien umgeben von Flüssigkeit. Die beträchtliche Viskosität dieser Flüssigkeit führt zur Energiedissipation und zur Schwingungsdämpfung, was im Gegensatz zur bekannten hohen Empfindlichkeit und der ausgezeichneten Frequenzselektivität der Hörwahrnehmung steht. Um die Komponenten des Haarbündelsystems in ihrem funktionalen Zusammenspiel besser zu verstehen, bedarf es eines wirklichkeitsgetreuen Modells unter Einbeziehung der Wechselwirkung zwischen Flüssigkeit und Struktur. Mit dieser Arbeit wird ein neuer Ansatz vorgestellt, um die Mechanik der Fluid-Struktur-Wechselwirkung im Innenohr zu analysieren. Da die Bewegungen bei der normalen Mechanotransduktion wesentlich kleiner als die geometrischen Abmessungen sind, ist es möglich, das Verhalten von Fluid und Struktur in Form der Verschiebungsvariable in einem linearen einheitlichen System von Gleichungen ausreichend genau zu beschreiben. Dieses System von partiellen Differentialgleichungen wird mit der Finite-Elemente-Methode gelöst. Basierend auf experimentell ermittelten Daten vom Haarbündel des Ochsenfrosches wird ein detailliertes Modell erstellt, welches sowohl die Interaktion mit der umgebenden Flüssigkeit als auch die koppelnde Flüssigkeit in den engen Spalten zwischen den einzelnen Stereozilien erfasst. Die experimentellen Daten sind Ergebnisse von hochauflösenden interferometrischen Messungen bei physiologisch relevanten Bewegungsamplituden im Bereich von unter einem Nanometer bis zu mehreren Dutzend Nanometern, sowie über einen breiten Frequenzbereich von einem Millihertz bis hundert Kilohertz. Das Modell erlaubt die Berechnung der auftretenden viskosen Widerstände aus der numerischen Analyse der verschiedenen beobachteten Bewegungsmoden. Es kann gezeigt werden, dass durch die Gruppierung zu einem Bündel der Gesamtwiderstand drastisch reduziert ist, im Vergleich zur Summe der Widerstände einzelner Stereozilien, die sich individuell und unabhängig voneinander bewegen. Die einzelnen Stereozilien in einem Haarbündel sind durch elastische Strukturen mechanisch miteinander verbunden: Die Energie des Schalls wird durch schräg angeordnete sogenannte Tiplinks auf die mechanotransduktiven Kanäle übertragen, wohingegen horizontale Querverbindungen die Stereozilien direkt koppeln. Während der Haarbündelauslenkung verursachen die Tiplinks zusätzlichen Widerstand durch stark dissipative Relativbewegungen zwischen den Stereozilien. Die horizontalen Querverbindungen unterdrücken diese Bewegungen und sind dafür verantwortlich, dass sich das Haarbündel als Einheit bewegt und der Gesamtwiderstand gering bleibt. Die Steifigkeit der Stereozilien und der Verbindungselemente sowie deren Geometrie sind in dem Modell sorgfältig angepasst, um eine Übereinstimmung mit den Beobachtungen aus verschiedenen Experimenten zu erzielen. Als Referenz dienen Steifigkeits- und Widerstandsmessungen, sowie Kohärenzmessungen für die gegenüberliegenden Außenkanten des Bündels, die jeweils mit und ohne Tiplinks durchgeführt wurden. Darüberhinaus sind die Ergebnisse durch den Vergleich mit experimentell beobachteten Relativbewegungen validiert, die das Haarbündel infolge von sinusförmiger Anregung bei Distorsionsfrequenzen zeigt. Diese haben ihren Ursprung in dem nichtlinearen Prozess des öffnens von Ionenkanälen. Das entwickelte Modell eines Haarbündels liefert neue Einblicke in den Schlüsselprozess der auditiven Wahrnehmung. Zur Behandlung von Problemen der Fluid-Struktur-Wechselwirkungen bei kleinen Amplituden hat sich der hier ausgearbeitete Ansatz als effizient und zuverlässig erwiesen.

Les sutures crâniennes ont été régulièrement étudiées au fil des siècles puisque déjà Vésale établissait une relation entre l’âge et la synostose suturale.<p><p>Leur imprécision a quelque peu confiné l’observation de ces sutures dans un certain oubli, justifié en partie.<p><p>Il est clair que leur fiabilité quant à l’estimation de l’âge au décès reste discutable et ce, pour diverses raisons.<p><p>Leur observation est difficile et dès lors sujette à subjectivité dans l’appréciation de leurs stades de fusion.<p><p>De plus, leur apparence sur le crâne sec peut être altérée par divers artéfacts de conservation (cire, vernis…).<p><p>Outre l’observation des classiques sutures ectocrâniennes de voûte et l’utilisation des méthodes habituelles en la matière (méthode de Acsádi et Nemeskéri, méthode de Masset), nous avons orienté notre étude vers des sutures peu voire pas exploitées, à savoir les sutures palatines, les sutures fronto-naso-maxillaires et les sutures de l’os zygomatique.<p><p>Ces trois groupes suturaux ont la particularité d’évoluer de manière très lente vers la fusion, à tel point que peu d’individus en présentent une oblitération complète.<p><p>Face au vieillissement de la population et de par notre expérience médico-légale et anthropologique d’étude de pièces osseuses de personnes âgées, nous avons estimé qu’il était utile de se pencher sur des collections particulières de sujets d’âge avancé, afin d’apprécier l’évolution morphologique de ces sutures faciales.<p><p>Si certaines personnes fort âgées gardent malgré tout des caractéristiques suturales peu évoluées, il existe dans l’ensemble, une progression suturale quasiment constante en fonction de l’âge.<p><p>Nous avons tenté de la cerner, en attribuant à ces sutures, des degrés bien définis de cette progressive fusion et par là, l’aboutissement à un coefficient moyen d’oblitération suturale, se traduisant aisément en pourcentage d’oblitération ou pouvant être introduit dans des équations de régression.<p><p>Tout en connaissant les limites de cette approche, nous pouvons estimer qu’elle peut rendre des services lors de l’étude de restes humains squelettisés, notamment s’ils appartiennent à des personnes fort âgées, dès lors qu’à ces stades de vieillissement, peu de méthodes restent encore applicables. D’autre part, l’approche en pourcentage d’oblitération suturale rend également des services lorsque les crânes étudiés sont fragmentés. C’est la situation que nous rencontrons actuellement lors de l’étude d’une très importante collection anthropologique à l’Institut royal des Sciences naturelles de Belgique.<p><p>Cette observation suturale pourra également – à l’avenir – trouver un terrain d’approche fort utile par l’étude des sutures en CT-Scan ou en micro-CT.<p><p>Enfin, grâce à ces techniques modernes d’imagerie médicale, les sutures trouvent un regain d’intérêt dans une application toute particulière, qui est celle de l’identification comparative, puisqu’il apparaît que le « dessin sutural » s’avère tout à fait propre à chaque individu.<p><p>/<p><p>Cranial sutures were regularly studied during centuries since Vésale already established a relationship between age at death and sutural fusion. <p><p>Their inaccuracy somewhat confined the observation of these joints in a certain lapse of memory, partly justified. <p><p>Various reasons clearly indicate that their reliability for age at death estimation remains debatable. <p><p>Their observation is difficult and consequently prone to subjectivity in the appreciation of their stages of fusion. <p><p>Moreover, their appearance on dry cranium can be modified by various artefacts from preserving methods (wax, varnished…). <p><p>In addition to the traditional observation of ectocranial sutures (with the usual methods such as Acsádi and Nemeskéri method and Masset method), we have directed our study towards not much exploited sutures, namely the palatine sutures, the fronto-naso-zygomatic sutures and the sutures of the zygomatic bone. <p><p>These three sutural groups are characteristic by evolving very slowly to fusion, and than, only few individuals present a complete obliteration of the latter. <p><p>Because of general population ageing and our medicolegal and anthropological experience of skeletal remains in elderly, we estimated the usefulness in studying particular collections of old people, in order to appreciate the morphological evolution of these facial joints. <p><p>Despite some very old people who present little evolved sutural characteristics, we consider as a whole, an almost constant sutural progression according to age. <p><p>We tried to define degrees of this progressive fusion and by the way, the result with a sutural obliteration average coefficient, can be easily translate as a percentage obliteration or able to be introduced into regression equations. <p><p>The limits of this approach are well known but we estimate that it is possible to help the examination of human skeletal remains in elderly, although at these stages of ageing, some methods remain still applicable. In addition, the approach expressed as a percentage of sutural obliteration is also very helpful when craniums are fragmented. This situation is currently observed on the anthropological collections at the Royal Belgian Institute of Natural Sciences. <p><p>Furthemore in the future, the sutural observation will find an interesting and useful approach by the study of the joints in CT-Scan or micro-CT. <p><p>At least, thanks to these modern techniques of medical imagery, the sutures find a renewed interest in a very particular application, such as comparative identification.
