This bachelor's thesis is about exploring what rhetorical power a daily newspaper’s front page might possess in affecting a nation’s emotion with regards to the portrayal of news. More specifically, we study how the news about the war in Ukraine presented on the front pages of Swedish newspapers may contribute to the sense of alarm regarding the current situation in Sweden. The material consists of four front covers from two newspapers, two from Svenska Dagbladet and two from Aftonbladet, from the 25th and 26th of February, the first two days after the war broke out. Our theoretical framework aims to propose a nuanced multimodal view on pathosanalysis that is based on the combined insights from linguistics and argumentation studies on the perspective of emotions with our own proposal for a visual addition. The analysis shows how emotions are conveyed through the linguistic structure of the headlines, the choice of images and the overall composition of the front page. We were able to provide potential links from the emotions conveyed in the material to the unease experienced in Sweden during that time even though further research is required for verification. The results also indicate an interrelation between objectivity and the need for sales in newspaper front cover with regards to prior research on magazine covers.

Macromolecular Reactions and Sensory Perception at the Air-Water-Human Interface

During 20th century main concern was to have sanitary water flowing through the tap. In 21st century constant supply of safe drinking water is common at any home in USA. Hence consumers pay attention to aesthetic quality of tap water. Odorous algal metabolites in source water and metals introduced to drinking water due to corrosion of pipes in the distribution system cause taste, odor and color problems, and result in complaints and perception of tap water as unhealthy. Millions of dollars are spent each year by water industry to address and prevent these issues. This research focused on some of the taste-and-odor issues associated with drinking water. First aim was to understand when geosmin, 2-MIB, and nonadienal become detectable, employing two-resistance mass transfer theory to determine the concentration of odorants in bathroom air. Results showed that water temperature and odorant concentration in water play an important role. Next focus was to develop an international odor standard to be used for training of sensory analysis panelists. There are many sensory methods to monitor drinking water to detect the off-flavors however an odor standard has been missing. Hexanal was studied with trained flavor profile analysis panels and was proposed as an ideal odor reference standard to be used for training and sensory assessment of water samples. Main focus was to understand metallic flavor of drinking water caused by iron and copper. It was shown that metallic sensation has taste and retronasal components creating the flavor and humans are very sensitive to it. Occurrence of lipid oxidation in the oral cavity was shown when metals were ingested, that produces carbonyls which are responsible for the metallic flavor. Antioxidants and chelators were investigated to study prevention of lipid oxidation and, chelators were determined to be more effective. Oral epithelial cell cultures were developed as a model for oral cavity to further investigate lipid oxidation and effectiveness of the antioxidants and chelators. This dissertation is a result of inter-disciplinary work and possibly a good example for how problems may be solved by incorporating different methods and point of views from several disciplines. / Ph. La deuxième Intifada dans les caricatures éditoriales de quotidiens allemands, canadiens, américains, israéliens et palestiniens : une analyse comparative de la prise de position à travers les représentations visuelles

Ce mémoire propose une analyse comparative de la caricature éditoriale dans des quotidiens de l'Allemagne, du Canada, des États-Unis, d'Israël et de la Palestine. L'étude examine la prise de position des dessins de presse pour ou contre une des parties impliquées dans la Deuxième Intifada du conflit israélo-palestinien. De plus, la recherche analyse la variation des représentations visuelles de nature politique et culturelle selon les quotidiens d'orientations politiques diverses et de différents types d'entreprise. L'étude s'est effectuée à partir d'une analyse de contenu des caricatures selon des méthodes quantitatives et qualitatives. Les résultats mettent en évidence que l'origine du pays de publication est plus décisive dans la prise de position de la caricature éditoriale que l'orientation politique ou le type d'entreprise du quotidien ; de plus, les représentations politiques sont plus fréquentes que celle de la culture, indiquant une vision de la Deuxième Intifada comme un conflit politique plutôt qu'ethnique.

社群媒體已經深刻地改變了我們的生活,並且對於企業行銷上產生了重大的影響。 這項研究的動機來自於社群媒體 Facebook 在台灣的普及性。此外,價值共創的概念,導致消費者忠誠度的重生,他們可以參與價值鏈的任何階段。我們的目標是將價值共創概念運用於Facebook粉絲專頁經營,探索如何達到最大化的行銷效益。 我們的研究數據來自於台灣資策會,結果顯示:一個粉絲專頁如果擁有越高的價值共創程度,確實能夠提高品牌行銷的效益。具體而言,我們發現粉絲專頁若提高內容多樣性和提供的回饋種類,更容易增加品牌的知名度。用戶原創內容程度越高,更可能增加使用者與品牌的互動,若要求使用者刻意推薦,則是更降低他們與品牌的互動。 此外,當粉絲年齡在大於或等於25歲時,以及粉絲專頁的粉絲總數越多時,價值共創程度對於行銷效益的顯著性會更明顯。 / Social media has profoundly changed our lives and is having a major impact on marketing today. This research is motivated by the tremendous growth of a social computing platform, Facebook. Besides, the co-creation concept leads to a rebirth of customer loyalty. Customers, in other words, can get involved at just any stage of the value chain. We aim to apply the co-creation concept in the Facebook fan page to explore what and how co-creation in social media marketing is effective. Our study has collected data from Taiwan’s Government research institute – Institute for Information Industry. The results of our findings show that the co-creation level in the Facebook marketing campaigns can truly increase brand’s performance. In detail, we found that kind of information and form of exchange are more likely to increase the brand’s awareness. Type of user created content is more likely to increase the brand’s engagement while way of advocacy is more likely to decrease the brand’s engagement. In addition, co-creation level is more significantly related to campaign’s performance when the fans are over (or equal to) 25 years old, and is more significantly related to campaign’s performance when the fan page has more fans than others.

檔案館運用Facebook粉絲專頁互動經營之研究 / A Study of the Public Communication and Operating Status of Archives with Facebook

網路技術的快速發展,使人與社會的接觸從實體走入虛擬,人與人之間的交流互動,可以透過社群媒體與網站進行意見的交流與資訊,政府單位、典藏機構與公眾人物等具有群眾號召力之訊息發布者,莫不透過網路社群的功能,達成資訊的成功傳遞。典藏機構是一般大眾最為信賴的知識傳播者,隨著社群網站成為主流的資訊中介站,檔案館也開始在2008年設立Facebook粉絲專頁。有鑑於Facebook具有資訊傳播效益,能提升檔案館與使用者意見的直接溝通。本研究旨在瞭解國內外檔案館Facebook粉絲專頁的互動經營模式,透過內容分析、深度訪談與問卷調查法,以不同的角色探討檔案館Facebook的經營策略與網路使用者的互動需求。 本研究以媒介豐富度的指標為基礎架構,透過內容分析法,觀察國內外6所檔案館Facebook粉絲專頁2015年10月至12月間579則貼文的發布頻率、主題內容與互動率,另輔以深度訪談法,探討檔案館Facebook經營者的貼文回饋速度、多元化內容提示與語言多樣性,瞭解國內檔案館Facebook粉絲專頁的經營維護政策。最後採問卷調查法,分析123份有加入檔案館Facebook之使用者對於該社群網站經營狀況的看法,及其認為檔案館最需要改進的地方。 研究結果發現,雖然檔案館的Facebook發文頻率偏高,但卻無法引發使用者共鳴,同時,使用者也認為檔案館之留言回饋率過低,不符需求。由內容分析與深度訪談結果得知,檔案館Facebook粉絲專頁經營者希望借助該社群網站的易用性與即時性,進行該館活動的推廣,而從Facebook頁面中,則可以發現互動率最高的媒體類型分別為相片、影片與連結。問卷調查部分,檔案館的Facebook使用者較能接受口語化的撰寫方式,且其亦會依據貼文主題的不同,給予不同程度的互動。本研究建議檔案館Facebook可提供豐富的多媒體貼文內容,透過符合使用者需求的內容包裝方式,讓兩者的互動更為積極。 / With the advent of the Internet, social networks afford an opportunity to make information much more broadly available and make direct contact between administrators such as governments, libraries, archives, museums and internet users. Library, archives and museums are the most trusted information suppliers. With the concept of becoming a communication intermediate, archives around the world started to create Facebook Pages since 2008 for extending communication channel with users of archives. The purpose of this research is to investigate the public communication and operating status of archives on Facebook. This research collected data from analyzing the media richness on the Facebook posts from US National Archives (U.S.), The National Archives (U.K.), National Archives of Australia and 3 public archives in Taiwan during October 1 to December 31, 2015. Besides, also used in-depth interview with archivists who managed the Facebook Pages to understand the operating policies of archives. Furthermore, questionnaire survey was conducted to capture users’ access behavior and analyzed the users’ barriers. According to the results, most of the users stated that, although archives have tried very hard to keep up with the trend, failure on reaching consensus between users’ need and administrative costs and effectiveness were a big problem. Facebook followers considered insufficient communicating cannels and lacked of immediate feedback reduced the satisfaction on the presentation of Archives’ Facebook. With content analysis, this study found that the media which could cause highly interaction were photos, videos and links, even the spoken text markup language were more acceptable by Internet users. This study suggests that archives should deliberate to provide more posts with multiple types of clues. Through the process of modifying original transactional messages, the communication between archives and users will be more effective.

The purpose of this study was to illustrate how the Swedish tabloid press construct their sports sections front pages and what they present on these pages. A second purpose, and by the first following, was to investigate the items these newspapers offer the audience to add to their daily agendas. The aim to show a bigger picture of the front pages content was reached by implementing a content analysis of 196 front pages from the two largest tabloid newspapers in Sweden, Aftonbladet and Expressen, during a period of four years. The study examined 98 items per newspaper and since the aim was to examine sports journalism the units were chosen from the two newspapers sports sections, Sportbladet and SportExpressen. The great conclusions that were made during this process were the differences in between genders, the dominating sports and what big part sponsors have in sports. The most common front page shows a Swedish man playing football in the age between 20-35. A less common page is woman, playing any sport. She is only found on nine of a hundred front pages. As in the case of men, football is the dominating sport in Swedish tabloid press. On second place we found ice hockey. What we also could find was the big part sponsors have in sports. We saw the logos of sponsors on every second front page.

Enhanced gel electrophoresis (GE) and inductively coupled plasma-mass spectrometry (ICP-MS) based methods for the identification and separation of proteins and peptides

The main focus of the PhD study was to develop new gel electrophoresis and ICP-MS based methods to analyze a wide variety of the bio-molecules such as proteins, phosphoproteins and metalloproteins etc. The tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis (tricine-SDS-PAGE) method is commonly used to resolve low molecular mass proteins, however, it requires a high percentage gel and a very complicated procedure to achieve this separation. This study describes a modification to tricine-SDS-PAGE to make it more effective for the separation of smaller proteins and for coupling to ICP-MS. The modified method employs low percentage PAGE gels and low reagent concentrations that provide efficient separations, good quantitation and low matrix levels that are compatible with ICP-MS. This modified method was applied to analyze phosphopeptides. Phosphopeptides are very small in size and difficult to separate using the other techniques such as Laemmli SDS-PAGE, original tricine-SDS-PAGE, immobilized metal affinity chromatography (IMAC), size exclusion chromatography (SEC) etc. In this study a simplified procedure is described based on modifying the original tricine-SDS-PAGE method. A comparative study showed that this modified method successfully resolved a digest mixture of very low to high molecular mass phosphopeptides/peptides. In off-line coupling of this method with ICP-MS, much better recoveries of the peptides from the gel were obtained as compared to traditional methods which indicate the compatibility of this modified method for quantitative studies. An on-line coupling of the modified system with ICP-MS was also demonstrated and it was applied for the separation, detection and quantification of phosphopeptides. Another application of this modified system was the separation of serum proteins. Blood serum contains five major protein groups i.e., albumin, alpha-1 globulin, alpha-2 globulin, beta globulin and gamma globulin. The separation of these five major proteins in a single gel is difficult to achieve using traditional methods. The modified system was shown to be superior for the separation of these serum proteins in a 7% (m/v) native-PAGE gel and a cellulose acetate membrane. A further study was carried out into controlling the factors that cause metal loss and protein fragmentation in SDS-PAGE. Using a reducing sample buffer, and heating to high temperatures (90-100ºC) in alkaline or acidic conditions may cause protein fragmentation and decrease the metal binding affinity. 70ºC was found suitable to prepare the sample at neutral, alkaline or acidic pH as no fragmentation observed. To prevent metal loss, the binding constant (log K) values of metal-amino acids, play the major role. Those metals which have high binding affinities with the amino acids in proteins can also be affected by the variation of the pH so prior information about pH to maintain the binding constant values is essential to minimize metal loss. This was observed in the loss of zinc, and to a lesser extent copper from human serum albumin (HSA) as measured by inductively coupled plasma mass spectrometry (ICP-MS). The method described above was applied for the separation and quantification of the serum proteins obtained from age-related macular degeneration (AMD) patients (where the AMD patients were from Moorfields Eye Hospital, London). Zn and Cu were quantified employing external calibration. Zn concentration showed variation whilst Cu did not show any significant variations in samples from AMD patients. A brief study of the interaction of cisplatin and oxaliplatin with HSA and transferrin was also performed. Cisplatin bound much faster than oxaliplatin with HSA. After 24 hours incubation, cisplatin showed a decrease in signal intensity which indicates that cisplatin binding decreases with time. Cisplatin binding with transferrin as compared to HSA was not significant, which could be the result of unstable Pt-transferrin complex formation. Oxaliplatin did not show high binding to either protein, perhaps due to the presence of the bulky, non polar DACH ligand.

Purification of mitochondrial RNase P in A. nidulans

Résumé La ribonucléase P (RNase P) est une ribonucléoprotéine omniprésente dans tous les règnes du vivant, elle est responsable de la maturation en 5’ des précurseurs des ARNs de transfert (ARNts) et quelques autres petits ARNs. L’enzyme est composée d'une sous unité catalytique d'ARN (ARN-P) et d'une ou de plusieurs protéines selon les espèces. Chez les eucaryotes, l’activité de la RNase P cytoplasmique est distincte de celles des organelles (mitochondrie et chloroplaste). Chez la plupart des espèces, les ARN-P sont constituées de plusieurs éléments structuraux secondaires critiques conservés au cours de l’évolution. En revanche, au niveau de la structure, une réduction forte été observé dans la plupart des mtARN-Ps. Le nombre de protéines composant la RNase P est extrêmement variable : une chez les bactéries, environ quatre chez les archéobactéries, et dix chez la forme cytoplasmique des eucaryotes. Cet aspect est peu connu pour les formes mitochondriales. Dans la plupart des cas, l’identification de la mtRNase P est le résultat de longues procédures de purification comprenant plusieurs étapes dans le but de réduire au minimum le nombre de protéines requises pour l’activité (exemple de la levure et A. nidulans). Cela mène régulièrement à la perte de l’activité et de l’intégrité des complexes ribonucléo-protéiques natifs. Dans ce travail, par l’utilisation de la technique de BN-PAGE, nous avons développé une procédure d’enrichissement de l’activité RNase P mitochondriale native, donnant un rendement raisonnable. Les fractions enrichies capables de cette activité enzymatique ont été analysées par LC/MS/MS et les résultats montrent que l’holoenzyme de la RNase P de chacune des fractions contient un nombre de protéines beaucoup plus grand que ce qui était connue. Nous suggérons une liste de protéines (principalement hypothétiques) qui accompagnent l’activité de la RNase P. IV De plus, la question de la localisation de la mtRNase P de A. nidulans a été étudiée, selon nos résultats, la majorité de la mtRNase P est attachée á la membrane interne de la mitochondrie. Sa solubilisation se fait par l’utilisation de différents types de détergent. Ces derniers permettent l’obtention d’un spectre de complexes de la RNase P de différentes tailles. / Abstract RNase P is a ribonucleo-protein complex (an RNA enzyme or ribozyme) that cleaves 5’ leader sequences of precursor tRNAs and a few other small RNAs. It occurs in all three domains of life, Bacteria, Archaea and Eukarya, with the latter containing distinct nuclear and organellar (mitochondrial or plastid) activities. In most instances, the complex contains a single, well-conserved RNA subunit that carries the active center of the enzyme. Yet, compare to bacterial and nuclear P RNA, most mtP RNAs are structurally highly reduced. The number of P proteins is highly variable: one in Bacteria, about four in Archaea, and ten in the cytoplasmic form of Eukarya. Much less is known in the case of mitochondria. MtRNase P is usually purified by using numerous separation steps that include unphysiological conditions, leading to complexes having a minimum number of subunits (e.g., in yeast and Aspergillus nidulans), that often loose their activity. Here, using BN PAGE, we have developed an enrichment procedure for A. nidulans mtRNase P that avoids some of the most disruptive conditions. The protein composition of active fractions was identified with LC/MS/MS, indicating that the RNase P holoenzyme is much larger than previously thought. Finally, the question of mtRNase P localization within mitochondria was investigated, by tracing its RNA subunit by RT PCR. We found that mtRNase P of A. nidulans is a predominantly membrane-attached enzyme; it is in part solubilized by detergents such as digitonin and Triton.

American literature and verse advanced in dialectal writing during the late-nineteenth century. Charles Chesnutt’s “The Goophered Grapevine” (1887), “Po’ Sandy” (1888), and “Hot-Foot Hannibal” (1899); Joel Chandler Harris’ Uncle Remus: His Songs and His Sayings (1881); Thomas Nelson Page’s “Marse Chan” (1884); and Mark Twain’s “Sociable Jimmy” (1874) and “A True Story, Repeated Word for Word as I Heard It” (1874) provided diverse dialect representations. Dialect expanded into poetry with James Whitcomb Riley’s “She ‘Displains’ It” (1888), “When the Frost is on the Punkin” (1882), and “My Philosofy” (1882) and Paul Laurence Dunbar’s “The Spellin’ Bee” (1895), “An Ante-Bellum Sermon” (1895), and “To the Eastern Shore” (1903). Dialect styles and how they conveyed political or social perspectives are assessed. Correspondence between late-nineteenth century literary figures as well as periodical reviews reveal attitudes toward the use of dialect. Reader responses to dialect based on their political or social interpretations are explored.

The thesis "Gender stereotypes in the communication of retail chains on Facebook" deals with the topic of gender stereotyping in the Facebook network. In the theoretical part of the thesis, the theoretical foundations are analyzed. The introductory part of the thesis introduces new media, their theoretical basis and history, and then defines the social network Facebook, on which this work focuses. The thesis focuses on gender and gender stereotypes, specifically the gender stereotyping of women. The aim of this thesis is to transfer the outlined theoretical basis, which focuses primarily on new media and gender stereotypes, into practice and to analyze the presence of female gender stereotypes on Facebook pages on the Czech market that primarily target the female audience. Specifically, it is examined whether and how retail chains use female gender stereotypes to communicate, frequency and what types of female stereotypes are most common, and whether the use of gender stereotyping of women changes over time. At the same time, the work also focuses on women's autostereotypes, specifically how women respond to the presence of female gender stereotypes in Facebook communication and whether there is a visible difference from communication without using gender stereotyping of women.
