In the research developed from the principles of the complexity of Morin (2002, 2007, 2008), we recognize a world in transformation in which the instrumental rationality - characteristic of modernity - loses strength and comes to live with new approaches of knowledge. These approaches analyze society, organizations, likings, and relationships between subjects, in environments marked by complexity and uncertainty. We investigate, through qualitative and exploratory research, the contributions that different areas of knowledge, such as Linguistics, Sociology, and Philosophy, contribute to the understanding of dialogue. In the space of practices, through empirical research, we analyze, interpret and (re) connect the dialogue assumed in the 2014 Sustainability Report and practiced in the virtual environment of the social media of the mining company Vale. As a result, we come to understand the dialogue provided by the interactions between the subjects as the space of transformation itself. We present dialogical dialogue as the proposition of a process that can happen from spontaneous and/or strategically managed interactions in the communication that occurs in the context of the organizations, transcending the still present and predominant instrumental-informational dimension. / Religar conhecimento e tecer junto o que emergiu de diferentes espa?os de pesquisa para poder compreender as manifesta??es poss?veis do di?logo na comunica??o estrat?gica foi o desafio estabelecido neste trabalho, com fins de alcan?ar contribui??es cient?ficas para o campo da comunica??o no contexto das organiza??es. Na pesquisa desenvolvida a partir dos princ?pios da complexidade de Morin (2002, 2007, 2008), (re) conhecemos um mundo em transforma??o no qual a racionalidade instrumental ? caracter?stica da modernidade ? perde for?a e passa a conviver com novas abordagens do conhecimento. Abordagens essas que analisam a sociedade, as organiza??es, a gest?o, os v?nculos e as rela??es entre sujeitos, em ambientes marcados pela complexidade e pela incerteza. Investigamos, por meio de pesquisa qualitativa e explorat?ria, as contribui??es que diferentes ?reas do conhecimento como a Lingu?stica, a Sociologia e a Filosofia aportam sobre a compreens?o do di?logo. IQG-607 is an analog of isoniazid (INH). According to several experiments carried out by our research group, IQG-607 showed both in vitro and in vivo anti-TB activity. Initial studies showed that the compound INCT-TB551 (a quinoline derivative) also presents in vitro anti-TB activity. The aim of this study was to develop analytical methods by high performance liquid chromatography (HPLC-UV) for quantification of IQG-607 and INCT-TB551 in mice plasma, and therefore to perform pharmacokinetic studies. The analytical method for the determination of IQG-607 in mice plasma showed linearity (r = 0.9992) in 0.5? 50 ?g/mL concentration range. Intra- and inter-day precision was < 15%, and the recovery ranged from 92.07 to 107.68%, showing that the method provides a precise and accurate analysis of the compound. In addition, IQG-607 was stable in plasma for at least 30 days at ?80 ?C, and after plasma processing, for 4 h in the auto-sampler (6 ?C) and maintained on ice (recovery > 85%). The applicability of the method for pharmacokinetic studies was determined after intravenous (i.v.) and oral (fasted and fed conditions) administrations to mice. IQG-607 levels in plasma were quantified at time points for up to 2.5 h. A short half-life (t1/2) (1.14 h), a high clearance (CL) (3.89 L/h/kg), a moderate volume of distribution at steady state (Vdss, 1.22 L/kg), were observed after i.v. (50 mg/kg) administration. Similar results were obtained for oral administration (250 mg/kg) under fasted and fed conditions. The oral bioavailability (F), approximately 4%, was not altered by feeding. Plasma protein binding was 88.87 ? 0.9%. Recently, experiments in mice infected with M. tuberculosis have shown that the compound INCT-TB551 has no in vivo activity, unlike previous in vitro activity studies. An analytical method was developed for the determination of INCT-TB551 in mice plasma to assess compound absorption, if any, after oral administration. The analytical method presented linearity from 0.1-10 ?g/mL (r = 0.9999). After development of the analytical method, the compound was orally administered in mice and the plasma levels were quantified at time points for up to 1 h. The compound INCT-TB551 was detected in the plasma of animals, which indicated that INCT-TB551 was absorbed. Although absorbed when orally administered, the compound is not exhibiting activity against M. tuberculosis in this animal model. This finding may be associated with the formation of inactive metabolites and/or the plasma concentration of INCT-TB551 achieved may be inadequate to exert its therapeutic effect. However, these points require further investigations. The protocols described here may serve as support to initiate pharmacokinetic studies of promising compounds, collaborating to advance in earlier stages of drug development. / A tuberculose (TB) ? uma doen?a infecto-contagiosa, causada principalmente pelo Mycobacterium tuberculosis, respons?vel por um n?mero consider?vel de mortes em todo o mundo. O composto IQG-607 ? um an?logo da isoniazida (INH) que, de acordo com diversos experimentos realizados pelo grupo de pesquisa envolvido no seu estudo, apresenta atividade anti-TB in vitro e in vivo. Estudos iniciais do mesmo grupo demonstraram que o composto INCT-TB551 (um derivado quinol?nico) tamb?m apresenta promissora atividade anti-TB. O objetivo deste estudo foi desenvolver metodologias anal?ticas por cromatografia l?quida de alta efici?ncia (CLAE-UV) para a quantifica??o do composto IQG-607 e do INCT-TB551, em plasma de camundongos, para a realiza??o de estudos de farmacocin?tica. O m?todo anal?tico para a determina??o do composto IQG-607 em plasma de camundongos apresentou linearidade (r = 0.9992) na faixa de 0.5?50 ?g/mL. Os valores de precis?o intra e interensaio (CV < 15%) e de recupera??o (92.07-107.68%) demonstram que o m?todo ? preciso e exato para a an?lise do composto. Al?m disso, o IQG-607 se mostrou est?vel no plasma por at? 30 dias, quando armazenado no freezer -80 ?C, e est?vel ap?s o tratamento da amostra do plasma, por at? 4 horas (h) na bancada (gelo) e no autosampler do equipamento (6 ?C). A aplicabilidade do m?todo anal?tico para o estudo de farmacocin?tica foi determinada ap?s a administra??o i.v. e oral em camundongos (animais em jejum e animais alimentados). As concentra??es plasm?ticas de IQG-607 foram quantificadas por at? 2,5 h ap?s a administra??o nos animais. Quando administrado pela via i.v. foi observado um tempo de meia vida (t1/2) curto (1,14 h), elevado clearance (CL) (3,89 L/h/kg), e um moderado volume de distribui??o (Vdss) (1,22 L/kg). Resultados similares foram obtidos ap?s a administra??o oral (250 mg/kg) em camundongos que estavam em jejum ou alimentados. A biodisponibilidade oral (F) foi de aproximadamente 4 %, valor que n?o foi alterado pela alimenta??o. A taxa de liga??o a prote?nas plasm?ticas do IQG-607 ? de aproximadamente 88 ? 0.9%. Experimentos recentes em camundongos infectados com M. tuberculosis demonstraram que o composto INCT-TB551 n?o apresentou atividade neste modelo in vivo, ao contr?rio dos estudos de atividade in vitro realizados anteriormente. Ap?s o m?todo ter sido padronizado, o composto foi administrado por via oral em camundongos, e as concentra??es plasm?ticas foram quantificadas por at? 1 h ap?s a administra??o. The piece of work adopts as its methodology the qualitative research that contain two sources of collecting data: psychologist Field Daily and the semi-structured Interview with the school educators. The competence to instantiate a set of communicative procedures is taken as a component of human rationality that meets a key role of regulating our cognitive environment (a set of mental states, centrally assumptions and emotions), in order to maximize practical goals and sociability. The linguistic-inferential approach offered here for such scope of rationality covers two levels: cognitive and practical, treated hierarchically, according to the assumed regulations. We consider that the cognitive apparatus (the inferential, representational and metarepresentational basis), along with the linguistic apparatus (computation plus interpretable expressions), allows us to operate from the most basic levels of linguistic processing to higher levels (where agents consider assumptions about other minds). In the practical domain, we consider that the linguistic and communicative behavior is used by agents to affect mental states and others? courses of action, thus being in the basis of our social cognition. In this scenario, we not only interact with agents, but we also create a social agency via language. We, therefore, consider a communicative agency framework in which acts are performed within a dialogical structure. The general thesis is that communication requires the use of skills that incorporate practical rationality parameters. This regulation would be dependent on a cognitive and practical structure of agency in which human cognition represents three types of agents: individuals, group members and groups (collectives or representatives). Each of these levels presents characteristic features of communicative agency. In all of them, however, there is the possibility of disagreement among agents, cognitive or practical, in dialogue situations. We illustrate this aspect with a scenario of conflict between agents that are supposed to reach a peace agreement. The illustrative analysis focuses on real negotiation dialogues between group members and representatives of the State of Israel and of Palestine. We observe how practical goals of agents of these types regulate their cognitive and dialogical goals. As a result, we present an alternative proposal to the standard scenario of negotiation, or conflict mediation. As a theoretical benefit, ad hoc pragmatic issues (relevance to the individual qua agent, conflicts between agents) are given prominence and effective treatment. As a practical benefit, the model can be applied to the area of conflict mediation, given the downsizing of a biosocial disposition: our cognitive states are particularly affected by stimuli from a class of agents (artists), with potential effect on individual and collective agencies. / Este trabalho tem como tema ag?ncia comunicativa humana. A compet?ncia para instanciar um conjunto de procedimentos comunicativos ? tomada como um componente da racionalidade humana que cumpre a fun??o central de regula??o de nosso ambiente cognitivo (um conjunto de estados mentais, destacadamente suposi??es e emo??es) tendo em vista maximizar metas pr?ticas e sociabilidade. A abordagem lingu?stico-inferencial desse escopo de racionalidade que oferecemos abarca dois n?veis: cognitivo e pr?tico, tratados hierarquicamente em fun??o das regula??es supostas. Primeiramente, consideramos que o aparato cognitivo (base inferencial, representacional e matarrepresentacional), junto com o aparato lingu?stico (computa??o mais express?es interpret?veis), nos permite operar do n?vel mais b?sico de processamento lingu?stico a n?veis mais altos (em que se consideram suposi??es sobre outras mentes). No dom?nio pr?tico, por sua vez, consideramos que o comportamento lingu?stico-comunicativo ? usado por agentes para agir sobre estados mentais e/ou cursos de a??o de outros, estando na base de nossa cogni??o social. Nesse dom?nio, n?o apenas interagimos com agentes como tamb?m criamos uma ag?ncia social via linguagem. Consideramos, assim, um quadro de ag?ncia comunicativa em que atos s?o performados dentro de uma estrutura dial?gica. A tese geral ? a de que a comunica??o exige o uso de habilidades que incorporam par?metros de racionalidade pr?tica. Essa regula??o estaria dependente de uma estrutura cognitiva e pr?tica de ag?ncia, segundo a qual a cogni??o humana representaria agentes de tr?s tipos: indiv?duos, membros de grupos (integrantes) e grupos (agentes coletivos ou representantes). Cada um desses n?veis apresenta tra?os de ag?ncia comunicativa caracter?sticos. Em todos eles, por?m, h? a possibilidade do desacordo, cognitivo ou pr?tico, em situa??es de di?logo. A an?lise ilustrativa se foca em di?logos reais de negocia??o entre membros de grupos e representantes do Estado de Israel e da Palestina que est?o encarregados do processo de paz. Since both inferential processes, natural dialogue, politeness, swearing and social networking sites have heteromorphic elements, they are complex problems, hard to be treated in a disciplinary way and, therefore, to be argued that interdisciplinary approaches are more interesting. It was assumed, therefore, being the Metatheory of Interfaces (CAMPOS, 2004a, 2007a, 2007d), the external interfaces are between Linguistics, Cognition, Communication and Computing (with respect to the Web), while the interior is among morphology, syntax, lexicology, semantics and pragmatics. This study aimed to: i. demonstrate the importance of internal and external interfaces consistent for the study of social networks on the Internet, ii. describe politeness present in social networks via the inferential perspective (GRICE, 1991; LEVINSON, 2000; SPERBER & WILSON, 1995). iii. to illustrate the expressiveness of bad words through a pragmatic perspective of rhetoric, and iv. emphasize its value in interdisciplinary research &#8213; for example, the link between language and emotion. Considering the importance pointed out by Pinker (2008) and others to study the "language as a window into human nature," it is argued that due to the location of common language and conscious thought to be concentrated in the neocortex, also linguistic politeness, being the result of rationalization (BROWN and LEVINSON, 1987; ESCANDELL-VIDAL, 1995), is in the same region. However, the taboo words appear in the limbic system (PINKER, 2008), thus presenting a more direct link with emotions and hence greater effectiveness in the expression of these. Given this difference and considering the principle of non-trivial connectivity (COSTA, in press), politeness and bad words were treated as a counterpoint and addressed predominantly in a perspective pragmatic-inferential, because if the semantics value is relatively the same, the pragmatic weight varies according to, for example, the intention and form. We chose to use dialogue taken from social networking sites because they have the possibility of huge corpus analysis, in which interactions are spontaneous. It has been corroborated that the rhetorical value of politeness and bad words is linked to the emotional aspect (PINKER, 2008; JAY, 2009). / Neste trabalho, foi proposta uma perspectiva interdisciplinar de ret?rica lingu?stica, predominantemente pragm?tico-inferencial, em que a forma tem efeito sobre o conte?do,1 atrav?s de discuss?o acerca de polidez e palavr?es se usando di?logos naturais e digitais, retirados de redes sociais na internet (Facebook, Youtube, Twitter), para ilustra??o. Objetivou-se: i. demonstrar a import?ncia de interfaces internas e externas bem constru?das para o estudo das redes sociais na internet; ii. descrever implicaturas de polidez (BRONW e LEVINSON, 1987; ESCANDELL-VIDAL, 1995; JARY, 1998) presente nas redes sociais atrav?s de processos inferenciais (GRICE, 1991; LEVINSON, 2000; SPERBER e WILSON, 1995). iii. ilustrar a expressividade dos palavr?es atrav?s de uma perspectiva pragm?tica de ret?rica; e iv. enfatizar seu valor em pesquisas interdisciplinares como, por exemplo, a liga??o entre linguagem e emo??o. Considerando-se a import?ncia apontada por Pinker (2007) e outros de estudar a linguagem como uma janela para a natureza humana, argumenta-se que devido ? localiza??o da linguagem comum e do pensamento consciente ser concentrada no neoc?rtex, tamb?m a polidez lingu?stica, por ser fruto de racionaliza??o (BROWN e LEVINSON, 1987; ESCANDELL-VIDAL, 1995), encontra-se nessa mesma regi?o. Contudo, os palavr?es surgem no sistema l?mbico (PINKER, 2008), assim apresentando liga??o mais direta com emo??es e, consequentemente, maior efetividade na express?o dessas. Dada essa diferen?a e considerando-se o Princ?pio da Conectividade N?o Trivial (COSTA, no prelo), polidez e palavr?es foram tratados como contraponto e abordados, predominantemente, numa perspectiva pragm?tico-inferencial, pois se a carga sem?ntica ? relativamente a mesma, o peso pragm?tico ? vari?vel conforme, por exemplo, as inten??es e a forma. Optou-se por utilizar di?logos retirados de redes sociais na internet, porque eles apresentam a possibilidade de an?lise de um corpus gigantesco, em que as intera??es s?o espont?neas. A situation analysis of the scientific production concerning psychologist s social commitment can aid the historical constitution of this theme and also subsidize reflections about the course of this profession in Brazil. In order to contribute to this debate, the theme social commitment of psychologist was analyzed in the scientific literature concerning psychologist profession in the country. Specifically, papers about the profession that mentioned the theme social commitment were characterized and their approach of this matter was analyzed. In order to accomplish these goals, two research stages were fulfilled: first, in a systematic search for scientific literature in internet databases, studies about the psychologist profession in the country were gathered; in the second stage, scientific papers relating to social commitment in their titles, abstracts or keywords were selected. 61 papers were retrieved, organized in electronic database and full-text analyzed, based in two axis: scientometric and thematic. The papers were identified, in general, as recent, of theoretic character and aligned with several Psychology subareas; mainly produced in public universities, in country s southwest; were of conjunctural interest to the authors research projects, individually published, by professors in touch with post-graduation; published in open access journals, with high Qualis evaluation. It was verified that there is no consensus on the meaning of the theme, prevailing definitions concerning the understanding of the social reality, to the majority of people or to a progressive-aligned professional performance. Are adduced as committed agents: the psychologist (sometimes together with other kinds of professionals or with users), the academia, and the representative entities of the profession. Depending on the subareas and the nature of study, all papers mentioned at least one of the following criteria: expansion of target-audience, renewal of practices, political direction, defined theoretical approach or adequate technical competence, sometimes relating each other. In short, psychologist s social commitment is a contradictory issue which have earned projection in scientific literature, reiterating the historical custom of evaluate the psychologist profession. As conclusion, it is regarded as indispensable to intensify the reflection about the role whose psychologist plays in a class-cleaved society and the limits and potentials of his performing in that framework. / Uma an?lise de conjunto da produ??o cient?fica sobre compromisso social do psic?logo pode auxiliar a constitui??o hist?rica sobre o tema e subsidiar reflex?es acerca dos rumos dessa profiss?o no Brasil. A fim de contribuir para esse debate, o tema compromisso social do psic?logo foi analisado na literatura cient?fica sobre a profiss?o de psic?logo no pa?s. Especificamente, caracterizou-se artigos cient?ficos sobre a profiss?o, que mencionam o tema compromisso social; e analisou-se como esse assunto ? abordado nos documentos investigados. Para isso, realizou-se duas etapas de pesquisa: primeiro, numa busca sistem?tica de literatura cient?fica em bases de dados na internet, recolheu-se estudos sobre a profiss?o de psic?logo no pa?s; na segunda etapa, selecionou-se artigos cient?ficos relacionados a compromisso social nos t?tulos, nos resumos ou nas palavras-chave. Recuperou-se 61 artigos, que foram organizados em bancos eletr?nicos e analisados na ?ntegra, com base em dois eixos: cientom?trico e tem?tico. Identificou-se que os artigos, em geral, s?o recentes, de car?ter te?rico, alinhados a diversas sub?reas da Psicologia; produzidos em universidades p?blicas, no Sudeste do pa?s; de interesse conjuntural nos projetos de pesquisa dos autores, publicados individualmente, por professores vinculados ? p?s-gradua??o; veiculados em peri?dicos em acesso aberto, com elevada avalia??o Qualis. Verificou-se que n?o h? consenso acerca do significado do tema; prevalecendo defini??es concernentes ? compreens?o da realidade social, ? considera??o das necessidades da maioria das pessoas, ou a uma atua??o progressista. S?o citados como atores comprometidos: o psic?logo (por vezes, em conjunto com outros profissionais ou o p?blico-alvo); a academia; e as entidades representativas da profiss?o. A depender das sub?reas e da natureza dos estudos, todos os artigos mencionam pelo menos um dos crit?rios, por vezes relacionados entre si: expans?o do p?blico-alvo, renova??o das pr?ticas, dire??o pol?tica, aporte te?rico definido, ou adequada compet?ncia t?cnica. To that end, this work presents the results obtained through a quantitative and qualitative research, by the analysis of results obtained through free association from a word inducer, and by discourse analysis. In this sense, the present study also reflects on the role of the psychologist working on socio-education from the adolescents? theoretical foundations and knowledge about the work of this professional. The research involved the participation of 102 adolescents in the socio-educational system and the results reveal that the Social Representation of the Psychologist is of a professional who helps, answers, talks, who relieves the adolescents? distress during the period of stay in DEGASE and who the adolescents recognize by name / Alicer?ado na Teoria das Representa??es Sociais, o presente estudo teve como objetivo primordial a identifica??o das representa??es que os adolescentes em interna??o provis?ria e em cumprimento de medidas socioeducativas de interna??o e semiliberdade t?m do Psic?logo que atua no Departamento Geral de A??es Socioeducativas do Rio de Janeiro (DEGASE). Com este intento, o estudo apresenta o resultado obtido atrav?s de uma pesquisa de cunho quantitativo e qualitativo, pela an?lise dos resultados obtidos atrav?s de associa??o livre a partir de uma palavra indutora e pela an?lise do conte?do do discurso dos adolescentes. Neste sentido, o estudo tamb?m ? reflete sobre o papel do psic?logo no trabalho socioeducativo a partir dos embasamentos te?ricos e dos saberes dos adolescentes acerca das a??es deste profissional. CrawLogo: An Experiment in End-User Programming for Web-Enabled Applications

<p>With the rise of the Web, there is more interest among end-users to create different kinds of software that use elements from the Web or allow for networked interaction between users. Currently however, many available tools for this purpose are hard to use or lack a sufficient level of expressiveness. To provide new insights on the construction of tools that allow end-users to create their own Web-enabled software, this thesis explores design issues and consequences of applying the Turtle metaphor from the Logo-programming language to an end-user programming environment for Web-enabled applications. </p><p>In order to explore this, CrawLogo was created - a programmable end-user tool that supports the creation of Web-enabled applications using a Turtle-like control metaphor and language adapted from Logo. As a proof-of-concept, several Web-enabled applications were created using this new tool, including CrawLogo Pong, a somewhat alternative version of Atari’s classic Pong game, and a collaborative browsing environment, in which users can browse the Web together. The resulting CrawLogo environment allows for creating Web-enabled applications that - using more traditional programming languages - would be quite complex and require deep technical programming skills. Further, while utilizing a Turtle-like control metaphor in CrawLogo allows for the creation of some new types of applications and some new ways of interacting with the Web, it also raises new problems such as how to successfully design within the CrawLogo metaphor and how to create a meaningful representation of Turtle- geometry-based navigation on the Web.</p>

A Study in RNA Bioinformatics : Identification, Prediction and Analysis

<p>Research in the last few decades has revealed the great capacity of the RNA molecule. RNA, which previously was assumed to play a main role only as an intermediate in the translation of genes to proteins, is today known to play many important roles in the cell in addition to that as a messenger RNA and transfer RNA, including the ability to catalyze reactions and gene regulations at various levels.</p><p>This thesis investigates several computational aspects of RNA. We will discuss identification of novel RNAs and RNAs that are known to exist in related species, RNA secondary structure prediction, as well as more general tools for analyzing, visualizing and classifying RNA sequences.</p><p>We present two benchmark studies concerning RNA identification, both de novo identification/characterization of single RNA sequences and homology search methods.</p><p>We develope a novel algorithm for analysis of the RNA folding landscape that is based on the nearest neighbor energy model adopted in many secondary structure prediction programs. We implement this algorithm, which computes structural neighbors of a given RNA secondary structure, in the program RNAbor, which is accessible on a web server.</p><p>Furthermore, we combine a mutual information based structure prediction algorithm with a sequence logo visualization to create a novel visualization tool for analyzing an RNA alignment and identifying covarying sites.</p><p>Finally, we present extensions to sequence logos for the purpose of tRNA identity analysis. We introduce function logos, which display features that distinguish functional subclasses within a large set of structurally related sequences, as well as the inverse logos, which display underrepresented features. For the purpose of comparing tRNA identity elements between different taxa we introduce two contrasting logos, the information difference and the Kullback-Leibler divergence difference logos.</p>
