Dans ce travail de thèse, des mélanges réactifs de polymères thermoplastiques immiscibles PMMA,PE et PS ont été réalisés dans un mélangeur (RMX®) qui génère principalement des écoulements élongationnels connus pour leurs pouvoirs distributifs et dispersifs même lorsque les composants ont une différence de viscosité importante. La polymérisation in-situ du styrène a été conduite par auto-polymérisation thermique et grâce à des amorceurs radicalaires. Différentes méthodes de préparations et paramètres de mélanges ont été testés puis évalués par l’analyse d’images de microscopie électronique. Les plus petits nodules de PS que nous avons obtenus ont un rayon moyen de l’ordre de 50 nm. Des mélanges binaires 90/10 à base de PS ou PE et d’un thermodur polyépoxyde (MDEA/DGEBA) ont également été réalisés. Les nodules sphériques les plus petits que nous avons obtenus ont un rayon moyen de l’ordre de 65 nm. / In this work, binary and ternary reactive blends based on immiscible thermoplastic polymers PMMA,PE and PS were realized. The in-situ polymerization of the styrene, precursor of PS, was led by thermal self-polymerization or still thanks to radical initiator. Blends were realized in a mixer named RMX who generates mainly extensional flows known for their distributive and dispersive skill even if components have an important viscosity difference. Various methods of preparation and parameters of mixtures were tested. Morphology was evaluated by the analysis of electronic microscopy images.The average radiuses of the dispersed PS phase were compared with those observed in the literature. The smallest that we obtained have an average radius of 50 nm. Binary mixtures 90/10 %m with PS or PE and with a thermodur polyepoxide (MDEA/DGEBA) were also realized. The smallest spherical nodules that we obtained have an average radius of the order of 65 nm.

Structure, morphology and performance relationships of organic photovoltaic devices : the block copolymer approachDeribew, Dargie Hailu 14 July 2013 (has links)
Ce travail se focalise sur l’étude de cellules solaires organiques modèles basées sur le mélange de poly(3-hexylthiophène) (P3HT) et de l'ester méthylique de l'acide [6,6]-phényl C61 butyrique (PCBM). La corrélation entre la morphologie de la couche active, les paramètres de mises en œuvre et le rendement photovoltaïque a été soigneusement étudiée afin d’obtenir l’optimisation de l’efficacité de tels dispositifs. Une méthode originale pour contrôler la séparation de phases dans ces mélanges a été proposée et consiste à l'intégration de copolymères blocs comme additifs. Trois copolymères séquencés ont été utilisés en tant qu’agents de nanostructuration et/ou d'agents de nucléation. Il a notamment été montré que l'incorporation de P3HT-b-PI permet l'augmentation du nombre de cristallites de P3HT tout en limitant l’agglomération du PCBM. D'autre part, l'incorporation de P3HT-b-P4VP dans les mélanges de P3HT:PCBM a permis de contrôler l'orientation des cristallites de P3HT, améliorant par ce fait le transport de charge dans les dispositifs. / This work investigates organic solar cells made of a blend of polymeric materials based on poly(3-hexylthiophene) (P3HT) and [6,6]-phenyl-C61-butyric acid methyl ester (PCBM) as model system. The correlation between the photovoltaic active layer morphology and the performance of the organic solar cell is thoroughly investigated. The chosen method for controlling phase separation in the polymeric blends is to incorporate block copolymers as additives. Three systematically selected block copolymers were used as nanostructuring and/or nucleating agents. Indeed, the incorporation of P3HT-b-PI induces the increase in the number of P3HT crystallites as well as suppresses the growth of PCBM aggregates. On the other hand, the incorporation of P3HT-b-P4VP into P3HT:PCBM decreases the crystallization of P3HT but increases its face-on orientation, a requirement for an enhanced charge transport in organic PV devices.

Structure et dynamiques de dispersions de gliadines de blé : effet de la concentration en protéines et de la température du solvant / Structure and dynamics of a wheat gliadins dispersions : effect of the protein concentration and solvent temperature.Boire, Adeline 14 February 2014 (has links)
De nombreuses études théoriques et expérimentales ont été menées au cours des 30 dernières années afin d'établir le lien entre les propriétés d'interaction des protéines, leurs transitions de phase et leur auto-assemblage. Des avancées significatives ont ainsi été permises grâce à l'application de concepts et méthodes de la physique des polymères et des colloïdes. Ces études ont, pour la majeure partie d'entre elles, été limitées à des protéines d'intérêt médical et à des protéines animales. Ce travail de thèse vise à appliquer ce type d'approche aux protéines végétales afin de mieux comprendre leurs propriétés d'interaction à l'origine de leurs propriétés fonctionnelles au sein des grains et dans les matrices alimentaires. Ce travail a été mené sur un isolat de protéines de réserve du blé composé principalement de la fraction monomérique: les gliadines. Nous avons étudié les transitions de phase des gliadines afin de mieux comprendre leurs propriétés d'interaction d'une part et les structures associées d'autre part. Dans un premier temps, une procédure d'extraction a été développée afin de travailler sur un isolat de composition contrôlée dont les masses moléculaires sont comprises entre 20 kDa et 300 kDa. Le comportement de phase de cet isolat a ensuite été étudié en diminuant la qualité du solvant. Nous avons ainsi déterminé le diagramme de phases (T-Φ), où T est la température et Φv la fraction volumique des gliadines. Cette étude a mis en évidence une séparation de phase de type liquide-liquide dans le système par diminution de la température. Une analyse détaillée de la répartition des protéines au sein des deux phases en fonction de leur masse moléculaire a permis d'identifier une masse moléculaire critique séparant des protéines de comportement de type colloïdal et des protéines de comportement de type polymérique. A partir du diagramme de phase, deux études structurales ont été effectuées. La première a étudié les cinétiques de séparation de phase lors de la diminution de la température pour caractériser la dynamique locale de séparation de phase et identifier les mécanismes qui génèrent les systèmes concentrés. Deux grands types de mécanismes de séparation de phase ont été identifiés : nucléation-croissance et décomposition spinodale. La seconde étude structurale a consisté à établir l'équation d'état pression osmotique vs concentration dans des conditions de bon solvant et à caractériser la structure des dispersions de protéines associée. La relation pression osmotique vs fraction volumique a permis de mettre en évidence l'existence de plusieurs régimes de structuration, associés à des changements de structure secondaire et de propriété rhéologique. La discussion générale permet de mettre en relation les propriétés thermodynamiques déduites de cette approche expérimentale et les changements structuraux observés à différentes échelles. / A substantial body of theoretical and experimental studies has been conducted over the last 30 years to establish the link between protein interaction properties, phase transitions and self-assembly. Both colloidal and polymer physics provide a new framework for understanding the driving force for proteins phase behaviour. Such studies have been limited to health-related proteins and to a few food proteins, mainly animal proteins such as casein, whey proteins. This thesis aims to apply this approach to plant proteins to better understand their interactions properties, at the basis of their functional properties within grains and food matrices. This work was carried out on a wheat storage protein isolate mainly composed of the monomeric fraction: gliadins.The objective of this PhD thesis is to investigate the phase transitions of wheat proteins to develop our knowledge on their interaction properties and the associated structures. We organized our experimental approach in five steps. First, we developed an extraction procedure to work on a protein isolate of controlled composition with molecular weight ranging from 20 to 300 kg mol-1. Then, we investigated the phase behaviour of the protein isolate by decreasing the solvent quality, here the temperature. We determined the T-Φ phase diagram, where T is the temperature and Φv the protein volume fraction, that maps the phase and structural transitions of the proteins. This study showed the existence of a liquid-liquid phase separation in the system upon a temperature decrease. We evidenced two different behaviours among proteins as a function of their MWs and highlighted a critical protein size above which the molecular weight is the key determinant of the protein properties. From the phase diagram, two structural studies were conducted. The first one studied the kinetics of phase separation upon temperature decrease to characterize the local dynamics of phase separation and to identify the mechanisms that generate concentrated systems. Two main mechanisms of phase separation have been identified: nucleation-growth and spinodal decomposition. The second one studied the effect of protein concentration on the multi-scale structure of wheat gliadins in good solvent. The integration of all these results allowed us to build the phase diagram of wheat gliadins, integrating thermodynamic and structural data.

Le contrôle de la microstructure des matériaux organiques est crucial pour des applications pratiques telles que la photonique, la biomédecine ou encore le domaine très dynamique de l'électronique organique. Les études récentes ont montré une possibilité de contrôler la structure des polymères à l'échelle nanométrique en utilisant l'auto-assemblage supramoléculaire sous confinement spatial. Bien que de nombreuses études ont déjà été effectuées dans ce domaine, plusieurs questions essentielles restent ouvertes. En particulier, il est important de comprendre comment les différents processus de formation structurale tels que la cristallisation, la formation d`une phase cristal liquide et la séparation de phases se déroulent sous confinement. Dans le présent travail, nous abordons l'effet du confinement à 1D et à 2D sur la formation de la structure pour une variété de systèmes, y compris les copolymères segmentés de poly(éther-ester-amide) (PEEA), les polymères cristaux liquides (CL) dont la chaîne principale appartient à la famille des poly(di-n-alkylsiloxane)s et des copolymères à bloc cristaux-liquides /semicristallins formés par complexation de poly(2-vinylpyridine-b-oxyde d'éthylène) (P2VP-PEO) avec un ligand cunéiforme, l'acide 4'-(3'',4'',5''-tris(octyloxy) benzamido) propanoïque. Recent studies have shown a possibility of tailoring the polymer structure on the nanoscale using supramolecular self-assembly under spatial confinement. Despite extensive studies already performed in this field, many questions remain open. In particular, it will be important to understand how different structure formation processes such as crystallization, LC-phase formation, microphase separation, and others occur under confinement. In the present work, we address the effect of 1D- and 2D-confinement on the structure formation for a variety of systems including segmented poly(ether-ester-amide) (PEEA) copolymers, main-chain liquid-crystalline (LC) polymers belonging to the family of poly(di-n-alkylsiloxane)s and liquid-crystalline/semicrystalline block copolymers formed through complexation of poly (2-vinylpyridine-b-ethylene oxide) (P2VP-PEO) with a wedge-shaped ligand, 4'-(3'',4'',5''-tris(octyloxy) benzamido) propanoic acid. In order to reveal the morphological diversity of the studied systems under confinement, the work was carried out on bulk materials and on thin films employing a battery of experimental methods. The concept of a self-incompatible solvent is introduced as a molecule composed of two parts (compound 1 and 2) with unfavourable interactions. A third compound will be readily dissolved in this solvent to diminish this unfavourable interaction by dilution. The more incompatible compounds 1 and 2 are, the stronger this behaviour is expected to be. In this work, ionic liquids comprising non-polar carbon chain and polar ionic group are chosen to serve as a model of self-incompatible solvent. The interactions parameters k of the ionic liquids with active ingredients are investigated to examine the effect of self-incompatibility of the ionic liquid molecule.
On the other hand, phase separation between compounds 1 and 2 will reduce the positive effect of self-incompatibility. The tendency of phase separation is increasing with increasing size of the two compounds. Thus, if compounds 1 and 2 are blocks tied together into a block copolymer, one expects a decreasing ability of the block copolymer to dissolve an active ingredient with increasing block length. In this work the ability of polybutadiene-block-poly(2-vinylpyridine) (PB-b-P2VP) block copolymers to dissolve the model compound anthracene is investigated. As expected, the solubility indeed decreases with increasing block length.

Combinatorial techniques, which overcome limitations of actual models of material research permitting to effectively address this large amount of variables, are utilized in this work to prepare combinatorial libraries of the blend of the biodegradable polymers Poly(e-caprolactone) and Poly(lactic acid). These libraries present continuous composition and temperature gradients in an orthogonal fashion that permit to obtain multiple surface morphologies with controllable microstructures due to the blends low critical solution phase behavior (LCST).
The goal of this study is to investigate the effect of surface morphology (surface chemical patterning and surface topography) on cell behavior. The varied surface topography of the libraries is used as a valuable tool that permits to assay the interaction between MC3T3-E1 cells and hundreds of different values of critical surface properties, namely, surface roughness and microstructure size. The outcome of this tool is a rapid screening of the effect of surface topography on cell behavior that is orders of magnitude faster than the standard 1-sample for 1 measurement techniques.
The results obtained show that cells are very sensitive to surface topography, and that the final effect of surface properties on cell function is intimately related with the stage of the cell developmental process. Meaning that, for example, areas with optimal characteristics to elicit enhancement of cell attachment is not necessarily the same that promotes cell proliferation.
This study imparts an improved understanding of an often neglected factor in biomaterials performance: surface morphology (particularly surface topography). The results provide a new insight into the importance of taking into consideration both chemistry and physical surface features for superior biomaterial design.

In Silizium-Nanokristall-Speichern werden im Gate-Oxid eines Feldeffekttransistors eingebettete Silizium Nanokristalle genutzt, um Elektronen lokal zu speichern. Die gespeicherte Ladung bestimmt dann den Zustand der Speicherzelle. Ein wichtiger Aspekt in der Technologie dieser Speicher ist die Erzeugung der Nanokristalle mit einerwohldefinierten Größenverteilung und einem bestimmten Konzentrationsprofil im Gate-Oxid. In der vorliegenden Arbeit wurde dazu ein sehr flexibler Ansatz untersucht: die thermische Ausheilung von SiO2/SiOx (x < 2) Stapelschichten. Es wurde ein Sputterverfahren entwickelt, das die Abscheidung von SiO2 und SiOx Schichten beliebiger Zusammensetzung erlaubt. Die Bildung der Nanokristalle wurde in Abhängigkeit vom Ausheilregime und der SiOx Zusammensetzung charakterisiert, wobei unter anderem Methoden wie Photolumineszenz, Infrarot-Absorption, spektroskopische Ellipsometrie und Elektronenmikroskopie eingesetzt wurden. Anhand von MOS-Kondensatoren wurden die elektrischen Eigenschaften derart hergestellter Speicherzellen untersucht. Die Funktionalität der durch Sputterverfahren hergestellten Nanokristall-Speicher wurde erfolgreich nachgewiesen. / In silicon nanocrystal memories, electronic charge is discretely stored in isolated silicon nanocrystals embedded in the gate oxide of a field effect transistor. The stored charge determines the state of the memory cell. One important aspect in the technology of silicon nanocrystal memories is the formation of nanocrystals near the SiO2-Si interface, since both, the size distribution and the depth profile of the area density of nanocrystals must be controlled. This work has focussed on the formation of gate oxide stacks with embedded nanocrystals using a very flexible approach: the thermal annealing of SiO2/SiOx (x < 2) stacks. A sputter deposition method allowing to deposit SiO2 and SiOx films of arbitrary composition has been developed and optimized. The formation of Si NC during thermal annealing of SiOX has been investigated experimentally as a function of SiOx composition and annealing regime using techniques such as photoluminescence, infrared absorption, spectral ellipsometry, and electron microscopy. To proof the concept, silicon nanocrystal memory capacitors have been prepared and characterized. The functionality of silicon nanocrystal memory devices based on sputtered gate oxide stacks has been successfully demonstrated.

Free surface films of binary liquid mixturesBribesh, Fathi January 2012 (has links)
Model-H is used to describe structures found in the phase separation in films of binary liquid mixture that have a surface that is free to deform and also may energetically prefer one of the components. The film rests on a solid smooth substrate that has no preference for any component. On the one hand the study focuses on static aspects by investigating steady states that are characterised by their concentration and film height profiles. A large variety of such states are systematically analysed by numerically constructing bifurcation diagrams in dependence of a number of control parameters. The numerical method used is based on minimising the free energy functional at given constraints within a finite element method for a variable domain shape. The structure of the bifurcation diagrams is related to the symmetry properties of the individual solutions on the various branches. On the other hand the full time dependent model-H is linearised about selected steady states, in particular, the laterally invariant, i.e.\ layered states. The resulting dispersion relations are discussed and related to the corresponding bifurcation points of the steady states. In general, the results do well agree and confirm each other. The described analysis is performed for a number of important cases whose comparison allows us to gain an advanced understanding of the system behaviour: We distinguish the critical and off-critical case that correspond to zero and non-zero mean concentration, respectively. In the critical case the investigation focuses on (i) flat films without surface bias, (ii) flat films with surface bias, (iii) height-modulated films without surface bias, and (iv) height-modulated films with surface bias. Each case is analysed for several mean film heights and (if applicable) energetic bias at the free surface using the lateral domain size as main control parameter. Linear stability analyses of layered films and symmetry considerations are used to understand the structures of the determined bifurcation diagrams. For off-critical mixtures our study is more restricted. There we consider height-modulated films without and with surface bias for several mean film heights and (if applicable) energetic bias employing the mean concentration as main control parameter.

Συσχετισμός δυναμικών ιδιοτήτων των οφθαλμικών ιστών και παθήσεων του οφθαλμού. Μη-επεμβατική διάγνωση με την χρήση τεχνικών σκέδασης φωτός laserΠέττα, Βασιλική 12 November 2007 (has links)
Λόγω της διαφάνειας των οφθαλμικών ιστών η σκέδαση φωτός αποτελεί ιδανικό εργαλείο για την ανίχνευση των αρχικών σταδίων ορισμένων παθολογικών τους καταστάσεων. Για παράδειγμα, η θόλωση του φακού των θηλαστικών λόγω ηλικίας ή/και άλλων εξωγενών αιτίων καλείται καταρράκτης. Ο καταρράκτης δεν μπορεί να διαγνωστεί κλινικά σε πρώιμο στάδιο με αποτέλεσμα την δημιουργία σοβαρών προβλημάτων στην όραση. Το γεγονός ότι το φως έχει την ικανότητα να ανιχνεύει τις μοριακές αλλαγές οι οποίες είναι πρόδρομα συμπτώματα του καταρράκτη αναδεικνύει την σημασία της έγκαιρης διάγνωσης στην αντιμετώπιση διάφορων οφθαλμικών παθήσεων. Ο φακός θεωρείται ως ένα πυκνό διάλυμα πρωτεϊνών (κρυσταλλίνες, ~40 % wt) σε νερό και η αδιαφάνεια η οποία αποτελεί την εκδήλωση του καταρράκτη προκαλείται ουσιαστικά από την συσσωμάτωση των πρωτεϊνών.
Στόχος αυτής της διατριβής είναι η διερεύνηση των μοριακών μεταβολών οι οποίες λαμβάνουν χώρα κατά την ανάπτυξη του καταρράκτη. Ιδιαίτερη σημασία δίνεται επίσης στην ανάπτυξη μιας μη-επεμβατικής μεθοδολογίας για έγκαιρη διάγνωση οφθαλμικών παθήσεων με τη βοήθεια της δυναμικής σκέδασης φωτός. Με την βοήθεια της τεχνικής αυτής, κατάλληλα τροποποιημένης για την μελέτη οφθαλμικών ιστών, μελετήθηκαν οι δυναμικές ιδιότητες των πρωτεϊνών χοίρειων φακών (π.χ. οι συντελεστές διάχυσης, η θερμοκρασιακή τους εξάρτηση σε διάφορα μέρη του φακού, κλπ.) χρησιμοποιώντας το πειραματικό μοντέλο του “ψυχρού” καταρράκτη. Στο μοντέλο αυτό η ελεγχόμενη ψύξη φακών επιφέρει βαθμιαία καταρρακτογένεση. Ιδιαίτερη έμφαση δόθηκε σε τέσσερα κυρίως είδη περαμάτων. (α) Μελέτη της εμφάνισης του ψυχρού καταρράκτη στον πυρήνα του φακού. (β) Μελέτη της επίδρασης του μήκους κύματος της ακτινοβολίας στην εμφάνιση και στην έκταση του φαινομένου του ψυχρού καταρράκτη. (γ) Μελέτη του φαινομένου του ψυχρού καταρράκτη κατά μήκος μιας διαμέτρου του φακού, δεδομένης της βαθμίδας συγκέντρωσης των πρωτεϊνών του φακού (μεγάλη συγκέντρωση στον πυρήνα και μικρή συγκέντρωση στην περιφέρεια του φακού). (δ) Μελέτη του επίδρασης της προθέρμανσης του φακού σε θερμοκρασίες υψηλότερες της φυσιολογικής στο φαινόμενο του ψυχρού καταρράκτη.
Τα βασικά συμπεράσματα της παρούσας διατριβής συνοψίζονται ως εξής. Υπάρχουν σαφείς συσχετισμοί μεταξύ των φασματικών χαρακτηριστικών (συναρτήσεις αυτοσυσχέτισης) και των ιεραρχικών σταδίων ανάπτυξης του καταρράκτη. Ποιοτικές και ποσοτικές αλλαγές στην θερμοκρασιακή εξάρτηση διαφόρων παραμέτρων, οι οποίες σχετίζονται με τις μοριακές διαμορφώσεις των αρχικών σταδίων του καταρράκτη, εμφανίζονται ήδη από τους 17 oC όπου ο πυρήνας του φακού είναι ακόμα διαυγής. Η χρήση ακτινοβολίας κοντά στο υπεριώδες μέρος τους φάσματος ενισχύει την ανάπτυξη του ψυχρού καταρράκτη στον πυρήνα του φακού. Ο ψυχρός καταρράκτης δεν αναπτύσσεται στην περιφέρεια του φακού. Η προθέρμανση του φακού σε συγκεκριμένη θερμοκρασία καθώς και ο χρόνος παραμονής σε αυτήν επηρεάζει σημαντικά την ανάπτυξη του ψυχρού καταρράκτη στον πυρήνα αλλά όχι στην περιφέρεια του φακού. Όλα τα παραπάνω δείχνουν πως η δυναμική σκέδαση φωτός μπορεί να παρέχει παραμέτρους οι οποίες μπορούν να χρησιμοποιηθούν με επιτυχία ως ευαίσθητοι και αξιόπιστοι δείκτες της έγκαιρης, μη-επεμβατικής, και in vivo διάγνωσης του καταρράκτη. / On account of the transparency of ophthalmic tissues, light scattering is an ideal tool for detecting the early stages of some of their pathological conditions. For example, the opacity of the mammalian lens due to age or other external causes is called cataract. Cataract cannot be detected clinically at early stages and as a result serious vision problems appear. The fact that, light has the ability to detect molecular changes that are related to the mechanism of cataract formation draws attention to the importance of early diagnosis in ophthalmic disorders. The lens can be considered as a dense colloidal protein dispersion (crystallins, ~ 40% wt) in water where the opacity that leads to cataract formation how its basis to the aggregation of proteins.
This dissertation is aimed at studying the molecular changes that take place upon cataract development. Particular emphasis is paid to the development of a non-invasive methodology for early diagnosis of ocular diseases with the aid of dynamic light scattering. By means of this technique, suitably modified for the study of ophthalmic tissues, the dynamic properties of the proteins of porcine lenses (e.g. diffusion coefficients and their temperature dependence at various parts inside the lens, etc.) were studied by using the experimental model of ‘cold’ cataract. In cold cataract the controlled cooling of the lens at temperatures below the physiological one induces gradual cataractogenesis. In particular, we focused on four kinds of experiments. (a) Detailed study on the cold cataract onset in the lens nucleus. (b) Study on the effect of the laser light wavelength in the onset and the extent development of cold cataract. (c) Study of the cold cataract effect along an equatorial diameter of the lens, considering the gradual concentration of the lens proteins (high protein concentration in the nucleus and low concentration in the cortex). (d) Study on the effect of thermal history, i.e. by warming up the lens at temperatures higher than the physiological one on the cold cataract effect.
The basic conclusions of the present dissertation are summarized as follows: There are clear correlations between the spectral characteristics (autocorrelation functions) and the hierarchical stages of the onset of cataract. Qualitative and quantitative changes in the temperature dependence of several parameters, which are related with the diffusive motions of proteins at the early stages of cataract, appear already at 17 oC while the nucleus is still clear and highly transparent. The use of laser radiation close to the ultraviolet part of the spectrum seems to enhance the formation of cold cataract in the lens nucleus. Cold cataract does not develop at the cortex of the lens, in view of the low protein concentration. The lens pre-heating at a certain temperature for various time periods affects significantly cold cataract formation in the lens nucleus but not in lens cortex. The above mentioned make clear that dynamic light scattering can indeed provide useful parameters that can be successfully used as sensitive and reliable indicators for the early, non-invasive diagnosis of cataract in mammalian lenses and in vivo.
