Substratinduzierte Differenzierung von Endothelzellen

Der Erfolg neuer Strategien in der Regenerativen Medizin und im Tissue Engineering hängt maßgeblich von einem gut entwickeltem vaskulären Netzwerk ab, welches die auf den Implantaten wachsenden Zellen und Gewebe versorgen. Oberflächeneigenschaften der Implantate sowie die Präsentation verschiedener Liganden für extrazelluläre Matrixproteine spielen bei der Besiedlung der Implantate, als auch bei der Bildung versorgender Blutgefäße durch die Endothelzellen eine wesentliche Rolle. In dieser Arbeit konnte durch Variation der Anbindungsstärke (kovalent oder physisorptiv) des extrazellulären Matrixproteins Fibronektins an die MSA-Copolymere der Einfluss des Aufbaus der extrazellulären Matrix auf das Differenzierungsverhalten der Endothelzellen gezeigt werden. Auch die initiale Konzentration von Adhäsionsproteinen an der Substratoberfläche zeigte sich bedeutend für das Verhalten der Zellen. Optimal für eine gute Adhäsion, native Entwicklung und Kapillarbildung der Endothelzellen war die stabile (kovalente) Anbindung weniger Adhäsionsproteine (hier Fibronektin) an die Substratoberfläche, so dass die Zellen problemlos adhärieren konnten. Erfolgte die weiter Proteinadsorption an die Oberflächen in einem nativen Zustand (hier auf den hydrophilen Oberflächen) so waren die Endothelzellen in der Lage, die extrazelluläre Matrix zu reorganisieren und ein dem in vivo Zustand ähnlicher Aufbau der extrazellulären Matrix konnte realisiert werden. Dies ermöglichte den Zellen wiederum ein natürliches Verhalten. Die Ausbildung einer moderaten Anzahl von Adhäsionsstellen der Zellen, sowie der in vivo ähnliche Aufbau der Adhäsionspunkte ermöglichte den Zellen einen eher lockeren Kontakt zum Substrat. Daher waren sie sehr flexibel in ihrer Morphologieanpassung. Unter diesen Bedingungen war es möglich, dass die Endothelzellen bei Stimulierung der Angiogenese kapillarähnliche Strukturen ausbildeten. Die Verwendung dreidimensionaler Zellkulturträger zeigte eine Unterstützung der Kapillarbildung der Endothelzellen in Abhängigkeit unter den beschrieben Bedingungen. / The success of tissue engineering strategies using artificial scaffolds crucially depends on a controlled formation of well-developed vascular networks in growing tissues. The presentation of extracellular matrix ligands on scaffolds is often envisioned as an appropriate strategy to support capillary formation. We show that the control of primary coupling mode — covalent versus physisorbed — as well as of secondary interactions of cell-secreted extracellular matrix proteins have a strong impact on endothelial cell development. A set of maleic anhydride copolymer thin films was used as planar model substrates. They exhibit a switchable mode of primary matrix coupling combined with a gradation of secondary matrix–substrate interactions due to a variation of surface hydrophobicity and polarity. We found that the cells adhere in a more native state at a low amount of covalent primary coupled fibronectin ligands in conjunction with weak interactions of secondarily adsorbed adhesion ligands on hydrophilic surfaces. These substrates allow for a formation of capillary-like networks of endothelial cells. High ligand densities and strong secondary hydrophobic interactions inhibit a pronounced capillary formation. The composition and structure of the formed extracellular matrix correlates well with the specific integrin expression pattern. From these results it is concluded that the formation of blood capillaries in artificial scaffolds can be triggered by controlling primary and secondary coupling of cell adhesion ligands to implant materials. 2

A Comparative Study on the Hydrolysis of Acetic Anhydride and N,N-Dimethylformamide: Kinetic Isotope Effect, Transition-State Structure, Polarity, and Solvent Effect

Recent studies have shown that general-base assisted catalysis is a viable mechanistic pathway for hydrolysis of smaller anhydrides. Therefore, it is the central purpose of the present work to compare and contrast the number of hydrogen atoms in-flight and stationary in the transition state structure of the base-catalyzed mechanisms of 2 hydrolytic reactions as well as determine if any solvent effects occur on the mechanisms. The present research focuses on the hydrolytic mechanisms of N,N-dimethylformamide (DMF) and acetic anhydride in alkali media of varying deuterium oxide mole fractions. Acetic anhydride has been included in this study to enable comparisons with DMF hydrolysis. Comparative studies may give synergistic insight into the detailed structural features of the activated complexes for both systems. Hydrolysis reactions in varying deuterium oxide mole fractions were conducted in concentrations of 2.0M, 2.5M, and 3.0M for DMF and 0.10M for acetic anhydride at 25°C. Studies in varying deuterium mole fractions allow for proton inventory analysis, which sheds light on the number and types of hydrogen atoms involved in the activated complex. For these systems, this type of study can distinguish between direct nucleophilic attack of the hydroxide ion on the carbonyl center and general-base catalysis by the hydroxide ion to facilitate a water molecule attacking the carbonyl center. The numerical data are used to discuss 3 possible mechanisms in the hydrolysis of DMF.

Comportements thermomécaniques de polymères chargés selon différents chemins de déformation et traitements thermiques / Thermomechanical behaviours of filled polymers along various deformation paths and thermal treatments

Le centre de recherche ArcelorMittal de Montataire développent de nouvelles solutions acier innovantes associant métal et polymère. Pour les ailes de voiture, le composite retenu est un matériau multicouche composé d’une lame d’acier sur laquelle est déposé un film mince de polypropylène choc chargé avec des particules minérales par l’intermédiaire d’une fine couche de polypropylène fonctionnalisé par le greffage d’anhydride maléique. Afin de prévoir et de connaitre le comportement de la partie organique du matériau lors de sa mise en forme par emboutissage et à posteriori de prédire l’état de ses propriétés mécaniques lors de son utilisation, la détermination des lois de comportement mécanique vrai et intrinsèque sur le modèle de la loi G’sell et Jonas est nécessaire. Ces lois sont définies selon trois chemins de déformation : la traction uniaxiale, le cisaillement simple et la traction plane. Les micromécanismes de déformation de la microstructure semi-cristalline des différentes formulations des matériaux selon leur mode de sollicitation mécanique ont été étudiés. Les résultats obtenus Post Mortem et In Situ ont permis la description qualitative et quantitative des évolutions des principales modifications microstructurales. Ces dernières diffèrent avec l’ajout de charges minérales. Deux nouvelles méthodes, la Tomographie X et la spectroscopie Raman permettent la détermination de la déformation volumique dans le cas de matériau de géométrie fine (300 µm). Le retrait lors d’un cycle thermique est étudié. Les influences du chauffage, de la formulation et de la microstructure (orientation des chaînes macromoléculaires et endommagement volumique) sont décrites / The ArcelorMittal research centre of Montataire elaborates innovative steel / polymer products. In the case of automotive fenders, the composite is a multilayered material. A thin impact polypropylene film is laminated on steel using a thin layer of a functionalized polypropylene. Mineral particles are added to improve stiffness. In order to predict and understand the behaviour of the organic layer all along its production process and finally to be able to characterize the state of its mechanical properties in use, the determination of the true and intrinsic mechanical behaviour laws according to the G’sell and Jonas model is necessary. These laws are obtained for three different mechanical paths: uniaxial tensile, simple shear and plane tensile. The deformation micromechanisms of the impact polypropylene semi-crystalline microstructure which depend on the materials formulations and on the mechanical path used are studied. Post Mortem and In Situ results give qualitative and quantitative description of the main microstructural modifications. Two new methods, X Tomography and Raman spectroscopy allow the quantification of the volume deformation which is developed during tensile tests. They are mainly available for very thin samples. X radiography and VideoTraction™ are not suitable anymore for this kind of geometry. Finally, the thermo-mechanical phenomenon of shrinkage which occurs during thermal treatment above the material melting point is analysed. Influences of the heating conditions, of the material formulations and of the material microstructure are described. Etude des propriétés électriques et thermiques de matériaux composites à matrice époxy-anhydride pour l'isolation haute tension / Study of electrical and thermal properties of epoxy-anhydride composite materials for high voltage insulation

L’avènement des énergies renouvelables, notamment offshore, et la nécessité de transporter l’électricité sur des distances toujours plus grandes tout en réduisant les pertes en ligne requièrent la mise en place d’un nouveau réseau électrique plus performant, le supergrid. L’amélioration des sous-stations ultra haute tension en courant alternatif (UHVAC) de type poste sous enveloppe métallique (PSEM), i.e. la réduction de leur empreinte au sol ou leur montée en tension, s’inscrit parmi les défis engendrés par le développement du supergrid. L’amélioration de la tenue aux contraintes électrothermiques des isolants solides employés dans les PSEM a été identifiée comme le principal verrou technologique pour le perfectionnement de ces appareillages, déjà pleinement maîtrisés en HVAC. Les travaux présentés dans ce manuscrit ont été motivés par la nécessité de développer un matériau isolant électrique plus performant que les matériaux existant actuellement sur le marché. L’un des matériaux couramment employés pour la fabrication d’isolants solides pour PSEM, une matrice époxy-anhydride chargée d’alumine micrométrique, a servi de référence commerciale à notre étude. Suite à une étude bibliographique, nous avons choisi de conserver la matrice époxy du système commercial de référence et de jouer sur les charges inorganiques employées pour optimiser les propriétés du matériau isolant. Les travaux présentés mettent en évidence l’influence de la nature des charges inorganiques (alumine ou nitrure de bore hexagonal), de leur facteur de forme (quasi sphérique ou lamellaire) et de leur fraction volumique sur la conductivité thermique, le coefficient d’expansion thermique, les propriétés mécaniques dynamiques, les propriétés diélectriques, la conductivité électrique à haute tension (DC) et la rigidité diélectrique (AC) des matériaux composites. L’étude expérimentale des relations structure-propriétés est complétée par un travail de modélisation des propriétés diélectriques et de la conductivité thermique des matériaux composites. / The integration of renewable energies to the power grid requires its modification in order to ensure its stability, security and efficiency. Improving ultra-high voltage alternative current (UHVAC) gas insulated substations (GIS), e.g. reducing their size or increasing their voltage, is one of the challenges induced by the development of the future power grid, the supergrid. Increasing the ability of solid insulators used in such equipment to withstand electro-thermal stress has been identified as the main obstacle to overcome. The work presented in this manuscript has been motivated by the necessity to develop more efficient electrical insulating materials compared to commercially available ones. An epoxy-anhydride matrix filled with micron sized alumina, often used to produce GIS solid insulators, has been used as a reference for this study. We decided to keep the matrix of the reference material throughout our work and to concentrate on the filler influence in order to optimize the properties of the composites. The impact of the nature of the filler (alumina or hexagonal boron nitride), its shape factor (platelets or almost spherical particles) and its volume fraction upon thermal conductivity, coefficient of thermal expansion, dynamic mechanical properties, dielectric properties, high voltage direct current (DC) conductivity and AC breakdown strength have been highlighted. The experimental study of structure-property relationships is completed by dielectric properties and thermal conductivity modelling using the effective medium theory.

Towards new catalytic systems for the formation of methyl methacrylate from methyl propanoate

The two stage Lucite Alpha Process for the industrial manufacturing of methyl methacrylate (MMA) represents one of the most efficient technologies currently available for the large scale production of this important chemical commodity. The second stage of this process, which involves the condensation of methyl propanoate (MeP) with formaldehyde over a heterogeneous fixed bed catalyst, however, still shows great scope for improvement. Herein the development of a novel homogeneous catalytic system that would promote the condensation of either propanoic acid or MeP with formaldehyde is explored. Since C–C bond forming reactions which proceed via C–H activation pathways typically display high atom efficiency, our efforts were particularly focussed on employing a functionalisation strategy that is mediated by C–H activation. In the case of propanoic acid, the possibility of achieving regioselective α-methylenation by linking the substrate to phosphorus was evaluated. Thus, a series of acyloxyphosphines and acylphosphites derived from either propionic acid or phenylacetic acid was prepared and, where stability allowed, fully characterised. Some of the resultant simple mixed anhydrides posed problems relating to their stability, and the stabilisation of such ligand systems by using electronic and / or steric effects was therefore explored. In addition, the coordination chemistry and in solution behaviour of Rh(I) and Ru(II) complexes containing these ligands was examined. Similar to the free ligands, complexes derived from these mixed anhydrides rearranged in solution via a number of decomposition pathways, with the specific pathway dependent on the nature of the auxiliary ligands. For most of these complexes, however, ligand decarbonylation was the route of preference for decomposition. Despite the instability of these complexes, a selection of Rh(I) mixed anhydride complexes were assessed for their potential as C-H activation catalysts in reactions aimed at the α-methylenation of saturated carboxylic acids. Furthermore, the stabilisation of Rh(I) mixed anhydride complexes with chelating auxilary ligands, such as bisphosphines or N-substituted diphosphinoamines, was explored. In particular, a series of new Rh(I) mixed anhydride complexes containing dppe, dppb and dppbz as secondary ligands were prepared and the effects of these secondary ligands on the in solution stability of these complexes assessed. As MeP represents the final product in the first stage of the Alpha process and not propanoic acid, the utilisation of PNP iridium pincer complexes in the regioselective sp³ C–H activation of MeP and related esters was also examined. The factors that govern the regioselectivity of such reactions were of great interest to us and, in particular, the effects of water on the reactivity and regioselectivity of these reactions were explored. For MeP, preferential C–H activation of the methoxy group was found to proceed under anhydrous conditions and the catalytic functionalisation of this site with ethene using this activation approach was considered. Formaldehyde, employed in the second stage of the Alpha process, is a difficult substance to manufacture and handle, especially on a large scale. A preliminary study on the in situ production of anhydrous formaldehyde via the catalytic dehydrogenation of methanol was therefore performed. During this study, catalytic systems based on carbonate salts and / or transition metal complexes were considered. In the hope of reducing the number of steps required in the production of MMA, a new one-pot cascade reaction for the indirect α-methylenation of MeP with methanol was developed. Although the production of MMA using this system only proceeded with low efficiency, the obtained results serve as an important proof of concept for future developments in this area. Finally, the capacity of a series of simple bases to catalyse the condensation of MeP with formaldehyde was assessed as part of a fundamental study directed towards determining the factors that govern the efficiency of this reaction. In addition, the extent to which each base effects the deprotonation in the α-position of MeP was determined with the aid of deuterium labelling experiments. Similarly, using sodium propanoate as model base a rough estimate of the kinetics of deprotonation could be made based on the degree of deuterium incorporation over time. These studies suggested that the low efficiency of this condensation reaction is not caused by ineffective deprotonation but rather by the weak nucleophilicity of the generated carbanion. For this reason, attempts to increase the electrophilicity of formaldehyde through Mannich-type condensations reactions involving secondary amine and carboxylic acid additives were made.

These organic compounds are generated during thermal degradation of polymers such as polyurethane (PUR) or epoxy.</p><p>Isocyanates, amines and anhydrides are airway irritants known to cause occupational asthma. Some of the compounds are listed as human carcinogens. Many workers are exposed.</p><p>Isocyanates and anhydrides are reactive and needs to be immediately derivatized during sampling. Methods have been developed for determination of airborne isocyanates, aminoisocyanates and anhydrides using di-n-butylamine (DBA) as reagent to form stabile urea derivatives or amide derivatives. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) enabled detection limits as low as 10 attomoles. A nitrogen-selective LC-detector enabled quantification of DBA-derivatives in reference solutions. </p><p>A novel sampler is presented. The sampler consists of a denuder in series with a three-stage cascade impactor and an end filter. The sampler made it possible to reveal the distribution of isocyanates between gas and different particle size fractions. During thermal degradation of PUR, isocyanates were associated to particle size fractions (<1 µm) that may penetrate to the lower airways. The distribution during 8 minutes changes noticeably. Aromatic isocyanates become associated to small particles (<1 µm). As a reference method, air-sampling was performed using an impinger filled with di-n-butylamine (DBA) in toluene, connected in series with a glass fiber filter. There was a good agreement between the denuder-impactor sampler and the reference method.</p>

The thesis entitled “Chemistry of Tetrathiomolybdate: Applications in Organic Synthesis” is divided in to six chapters Chapter 1: Synthesis of -amino disulfides, cystines and their direct incorporation into peptides mediated by tetrathiomolybdate In this chapter, we report a simple method for direct access to β-amino disulfides by regioselective ring opening of sulfamidates with benzyltriethylammonium tetrathiomolybdate [BnEt3N]2MoS4. The versatility of this reaction has been shown by preparing a number of β-amino disulfides having different N-protecting groups and the stability of these protecting groups under the reaction conditions has been evaluated. This methodology is also extended to the synthesis and direct incorporation cystine and 3, 3′-dimethyl cystine derivatives into peptides. Chapter 2: Unusual reactivity of tetrathiomolybdate: A new entry to the synthesis of b-aminothiols In this chapter, we disclose a simple and highly efficient method for the synthesis of β and γ-amino thiols via regioselective ring opening of sulfamidates with tetrathiomolybdate 1. The scope and generality of this methodology has been exemplified by synthesizing a carbohydrate derived β-aminothiol. This methodology has also been extended to the synthesis of isocysteine derivatives in optically pure form. Chapter 3: Part 1: Synthesis of β-aminodiselenides via sequential one-pot, multistep reactions mediated by tetrathiomolybdate In this chapter, we have demonstrated that a variety of N-alkyl-β-aminodiselenides can be synthesized in high yield from appropriate sulfamidates under mild reaction conditions using potassium selenocyanate and tetrathiomolybdate [BnEt3N]2MoS4 via a sequential one-pot multistep process. The compatibility of different protecting groups under the reaction conditions has been discussed. Chapter: 3 Part 2: Synthesis of unnatural seleno amino acids and their direct incorporation into peptides In this chapter, we have demonstrated the first and general method for the synthesis of selenocystine, 3, 3'-dialkylselenocystine, isoselenocystine and their direct incorporation into peptides using a one-pot multistep reaction strategy mediated by tetrathiomolybdate. Chapter 4: Synthesis and functionalization of cysteine, selenocysteine and their derivatives via the formation of unsymmetrical disulfide and sulfur-selenium bond. In this chapter, we present a novel one-pot multi component strategy for the synthesis and functionalization of cysteine, selenocysteine and their derivatives via unsymmetrical disulfides and sulfur-selenium bond formation. Chapter 5: Part 1: A novel method for the synthesis of thioacetates employing benzyltriethylammonium tetrathiomolybdate and acetic anhydride In this chapter, we report a simple and efficient methodology for the synthesis of thioacetates using benzyltriethylammonium tetrathiomolybdate [BnEt3N]2MoS4 and acetic anhydride as the key reagents, starting from alkyl halides in a multi step, tandem reaction process. The application of this methodology for the synthesis of orthogonally protected cysteine derivatives and anomeric β-thioglycosides has also been demonstrated. Chapter 5: Part 2: One-pot synthesis of β-aminothioacetates using benzyltriethyl-ammonium tetrathiomolybdate and acetic anhydride. In this chapter, we have demonstrated a simple and efficient method for the synthesis of β-amino thioacetates and pseudo thioinositol derivatives, via ring opening of aziridines and aziridino epoxides using tetrathiomolybdate 1 and acetic anhydride as key reagents. Chapter 6: Simple and efficient synthesis of allo and threo-3, 3'-dimethylcystine derivatives in optically pure form In this chapter, we have presented a simple and efficient methodology for the synthesis of allo-3,3'-dimethylcystine and threo-3,3'-dimethylcystine derivatives in optically pure form using L-threonine as the chiral pool and benzyltriethylammonium tetrathiomolybdate 1 as the key reagent. Aprovechamiento integral del Cannabis sativa como material de refuerzo/carga del polipropileno

En este trabajo se ha estudiado el potencial tanto los filamentos de cáñamo como de la cañamiza como refuerzo/carga del polipropileno. La modificación de estos materiales se realiza para lograr una mayor compatibilidad con la matriz polimérica. Se evaluaron las propiedades mecánicas de las resistencias a tracción e impacto, de los materiales compuestos reforzados tanto de filamento como de cañamiza. Los filamentos de cáñamo poseen suficiente capacidad de refuerzo en los materiales compuestos basado en polipropileno debido a sus propiedades intrínsecas, siendo una buena alternativa como material de refuerzo. Así, la adición de MAPP (polipropileno modificado con anhídrido maleico) conduce a materiales compuestos con unas resistencias a tracción de hasta el 70% de las que se obtienen con compuestos de PP reforzados con fibra de vidrio. Mientras que la cañamiza ha actuado como una carga en la matriz, incrementado significativamente la rigidez de los materiales compuestos. / In this work, the potential of hemp strands as well as hemp straw, to be used as reinforcement/filler of polyprppylene has been studied. The modification of these materials ha been carried out to obtain a greater compatibility with the polymer matrix. The mechanical properties of the obtained materials were evaluated under tensile, flexural and impact stresses. The hemp strands showed a sufficient capacity of reinforcement in the polypropylene-based composite due to their intrinsic properties, being a good alternative like reinforcement agent. Thus, the addition of MAPP (polypropylene modified with maleic anhydride) leaded to composites with a value of ultimate tensile strength that achieved 70% of the value of materials formulated with PP and fibreglass. In the particular case of the addition of hemp straw, this component acted as a filler of the polymer matrix, increasing the rigidity of the composite but with a limited influence in the ultimate tensile strength.
