Sex-linked molecular markers and their application to endocrine disruption research in amphibians

Die weltweit mehr als 7500 Amphibienarten sind durch anthropogene Ursachen wie Habitatzerstörung, Krankheitsverbreitung, Klimawandel und Umweltverschmutzung in ihrem Bestand bedroht. Einige der Ursachen sind kaum erforscht, so die Verschmutzung aquatischer Ökosysteme durch endokrine Disruptoren (EDs), Substanzen, die mit dem Hormonsystem interagieren. Ausgehend von neuen molekularen Markern, welche die Ermittlung des genetischen Geschlechts erstmals bei einigen Hyliden und Bufoniden erlauben, wurde in der vorliegenden Arbeit auf die Wirkung des synthetischen Östrogens 17α- Ethinylestradiol (EE2)und des Weichmachers Bisphenol A (BPA) fokussiert. Für drei Bufonidenarten wurde zunächst die Geschlechtsgebundenheit von Mikrosatelliten getestet und ein XX/XY-System nachgewiesen. Diese und bereits etablierte Marker wurden anschließend in ein neu entwickeltes Versuchsdesign für ED-Studien integriert: Nach gleichzeitiger Aufzucht von Modell- (Xenopus laevis) und Nicht-Modell-Arten (Hyla arborea, Bufo viridis) unter EE2- bzw. BPA- Exposition wurde das genetische Geschlecht bestimmt und mit dem anatomisch und histologisch ermittelten phänotypischen Geschlecht erglichen. Die drei Anuren zeigten starke Empfindlichkeitsunterschiede gegenüber beiden EDs. Umweltrelevante Konzentrationen beeinflussten die somatische Entwicklung und führten zu artspezifischen Gonaden-Fehlbildungen. EE2 bewirkte zahlreiche partielle und komplette Geschlechtsumwandlungen, mit stärkeren Effekten bei X. laevis. Diese Arbeit zeigt somit, dass bereits niedrige EE2- und BPA-Konzentrationen zu starken Schädigungen führen können und die Substanzen aufgrund ihrer erheblichen aquatischen Präsenz als ernstzunehmende Faktoren der Amphibienkrise anzusehen sind. Die Ermittlung des genetischen Geschlechts wird als wichtig eingestuft, um verlässliche Aussagen über ED-Effekte zu treffen. Zudem sollten an der Modell-Art X. laevis gewonnene Erkenntnisse nicht vorbehaltlos auf andere Amphibienarten extrapoliert werden. / The more than 7500 known amphibian species are globally threatened, mainly due to anthropogenic causes like habitat destruction, dispersing diseases, climate change and environmental pollution. Some of the causes are barely investigated, e.g. the pollution of aquatic ecosystems with endocrine disrupting compounds (EDCs), substances that interfere with the hormone system. Based on new molecular markers, for the first time allowing genetic sexing in some hylids and bufonids, this thesis focused on the effects of the synthetic estrogen 7α-ethinylestradiol(EE2) and the plasticizer bisphenol A (BPA). Initially, several microsatellite markers were tested for sex-linkage in three bufonid species, and an XX/XY system could be revealed. Subsequently, these and other established markers were integrated into a newly developed experimental design for EDC-research: after simultaneous exposure of model (Xenopus laevis) and non-model species (Hyla arborea, Bufo viridis) to EE2 or BPA, metamorphs were genetically sexed. Anatomically and histologically determined phenotypic sexes were directly compared with the genetic sex of each individual. The three anurans showed striking differences in their susceptibilities in both EDCexperiments. Environmentally relevant concentrations affected the somatic development and led to species-specific gonadal anomalies. In addition, EE2 provoked high numbers of mixed sex and completely sex-reversed individuals, with more pronounced effects in X. laevis than in the two non-model species. This work shows that low concentrations of EE2 and BPA lead to severe damages. Due to their widespread presence in the aquatic environment, these substances might contribute to the worldwide amphibian crisis. To produce reliable results in EDC-studies, genetic sexing is considered important. Furthermore, findings gained with the model species X. laevis should not unreservedly be extrapolated to other amphibian species.

Stadt – Frau – Amerika

Ziel dieser Untersuchung ist es, feuilletonistische Entwürfe von „Amerika“ in der Zeit von 1918 bis 1938 zu analysieren. Der Amerika-Diskurs in der deutschen Presse diente mir dabei als Folie für eine weitere Untersuchung im Kontext der Ersten Tschechoslowakischen Republik. Der Hauptbeitrag der vorliegenden Arbeit liegt daher in einer gänzlich neuen Perspektive auf die Perzeption der USA sowie im Vergleich dieser tschechoslowakischen Sicht mit der deutschen. In den 1920er- und 1930er-Jahren vermochte insbesondere das Feuilleton als modernes urbanes Genre die aktuellen zeitgenössischen Diskurse einerseits aufzunehmen, andererseits mitzugestalten. Deshalb diente es mir als Quelle. In der vorliegenden Arbeit wurde die Modernisierung als komplexes Phänomen untersucht, das alle Bereiche des Alltags erfasste. Anhand ausgewählter Themenbereiche – Amerika, Stadt, Frau – wurde herausgearbeitet, wie sich die Zeitgenossen mit diesem Prozess auseinandersetzten. Da die Modernisierung mit Amerikanisierung gleichgesetzt wurde, ist der so genannte Amerikanisierungsdiskurs zentraler Bestandteil der Untersuchung. Amerika diente sowohl in der Weimarer Republik wie in der ČSR als Projektionsfläche für Wunschvorstellungen sowie für Probleme im Umgang mit der Modernisierung. Das Zentrum der Moderne war der urbane Raum. Daher konzentriert sich meine Arbeit auf die Großstädte Berlin und Prag. Als deren Bezugsgröße diente New York. Analysiert wurden die Selbst- und im Falle Berlins auch die Fremddarstellungen der Metropolen im Hinblick auf ihre voranschreitende Urbanisierung. Der gesellschaftliche Wandel manifestierte sich in den 1920er-Jahren am deutlichsten an der gesellschaftlichen Aufwertung der Frau; sie profitierte am meisten vom Prozess der Modernisierung. Die Weiblichkeitskonstruktionen widerspiegeln daher wichtige Aspekte der Auseinandersetzung mit der Modernisierung / Amerikanisierung. / The goal of this research is the analysis of perceptions about the American model of modernity in the period between 1918 and 1938. The debate about America in Germany serves as the template for its examination in the context of the first Czechoslovak Republic, which will present an entirely new perspective. The main contribution of this thesis is the removal of the one-sided approach to German perceptions of America by means of comparison and perspective from Czechoslovakia. In the ‘20s and ‘30s played in particular the feuilleton a crucial role in creating popular perceptions of society. As a modern urban genre it enabled both the depiction and the formation of contemporary discourse. The subject of this thesis is a study of modernization, a complex phenomenon that has touched upon all aspects of everyday life. The analysis of selected topics – the USA, the city, and the woman – shows how people responded to this process. Due to the fact that modernization became equated with Americanization, a discourse about the latter is central to this analysis. Both in the German Weimar Republic as well as in Czechoslovakia America was used as a backdrop for projecting; both the visions and challenges that occurred while dealing with modernization. The center of modernism was the urban environment. This fact has led the focus of this examination to the cities of Prague and Berlin, with the reference point for both being New York. This thesis analyzes the way these cities presented themselves from the point of view of urbanization – in the case of Berlin it also includes perception of the city by outsiders. In the ‘20s social change was predominantly manifest in the empowerment of women and it was also women who profited most from modernization of the social sphere. The evolution of the notion of femininity is thus reflected in important aspects of this confrontation with modernization / Americanization.

影響國中教師性別角色刻板化態度與兩性教育平等意識相關因素之研究 / The reserch on effective factors of junior high school teachers' attitude of gender stereotype and gender education equity

本研究從女性主義理論角度出發,主要研究目的在探討現今國中教師在教學互動過程裡,是否展現出性別角色刻板化態度?他們的態度否因其人口特質(如:性別、年齡、省籍、婚姻狀況)、師資培育訓練歷程、教學及行政經驗的不同而產生差異?上述的因素是如何影響教師的兩性平等教育意識(包括教學平權態度、教學性別區隔、女性主義理念)? 本研究係採用研究者自行編製的問卷作為研究工具,研究範圍則限台北市及台北縣國民中學,從研究者個人網絡選擇受訪者,再由受訪者代替研究者於授課學校進行隨機抽樣調查,總計有效樣本為401份。樣本資料經檢核登錄後,研究者以SAS統計分析軟體進行資料分析。 研究結果及主要發現如下: 一、男老師仍然掌控學校行政事權,女老師不僅較少參與行政工作,也因為傳統性別角色的影響(包括:母職及傳統女性角色的壓抑),她們的行政意願普遍低落。 二、教師的教學態度仍深為性別刻板角色所左右,包括教師仍強調複製兩性特質的教育功能及女性的母職角色。 三、教師仍容易以〝性別〞作為評估與詮釋學生行為的必然標準,.此外〝性別〞也是教師在控制學生及教室管理的重要方式;男教師的教學態度明額較不平權。 四、多數教師贊成教育機會平等,但對於〝女性主義教育理念〞(給予女性更多的教育資源以扭轉其弱勢地位)接受度低。 五、教師對於教職〝性別形象〞(女性化)的重視大過於〝專業能力〞;女教師的教職女性化刻板觀念較深。 六、性別社會化是影響教師〝性別角色刻板化〞及〝兩性教育平等意識〞的主因,女教師對於傳統性別角色的反省與兩性平權教育的思考均比男老師積極。 七、中壯年(40歲以上)教師不僅有明顯的性別偏見,他們對兩性教育的態度也較不平權;自然學科的教師也較贊同〝教育性別區隔〞,這表示他們在判斷學生的學習成就及傾向時,會有〝性別〞的預設立場;教學年數愈長的老師其性別角色的刻板化程度也愈高,他們也較贊同教育應有性別區隔。顯示教育的〝量〞(學習多久)對於個人性別平等意識的影響不如〝質〞(學習內容)來的重要。

本研究以電視新聞社會線為研究場域,旨在探討電視社會女記者與電視社會新聞之陰性化現象之間的關連。本研究採取質性研究的多元途徑,以參與觀察法、深度訪談法、案例分析法,探討兩大研究問題:一、近年來,電視社會新聞內容有沒有陰性化的傾向?1.社會新聞記者怎麼看待陰性的社會新聞主題?2. 社會新聞記者怎麼看待陰性的新聞報導角度?3.社會新聞記者採取什麼樣的工作倫理?二、電視新聞工作場域文化與社會女記者的新聞產製1. 社會線的文字記者需要具備什麼特質? 2. 社會線女記者的女性身份,是否影響:(1)社會線女記者線上關係之建立(2)社會線女記者在組織中的任務分配與評價(3)社會線女記者和受訪者的互動(4)社會線女記者社會新聞的角度? 在電視社會新聞的陰性化方面,本研究發現,在電視社會新聞內容方面,由於以市場為主要考量,軟性議題儘管增加,其份量和重要性仍低於感官化取材,而女性議題仍居少數。在電視社會新聞的處理方式與記者敘事方式上,趨向實用、貼近觀眾感受的陰性報導角度增加,但社會新聞仍以陽性化的感官處理為主要走向。 在電視社會新聞工作場域文化方面,研究發現組織當中父系意識型態的主導,與商業邏輯的感官化操作思維相結合。在社會女記者的新聞產製方面,女記者發展出中性化的性格特質與女性化的性別特質,並接受父系觀點對她們的觀視或期待,這是一種對場域文化的投降策略。因此,女記者的社會新聞報導與男性記者沒有差別。 / This study takes the impact of TV female journalist on the feminization of TV social news as the main inquiry. Adopting the channel of qualitative research, this study takes the participant-observation method, in-depth interview method, and case study method to discuss two significant issues. I. Is TV socail news feminized in the past few years? 1. How do journalists of TV social news view feminine topic of news reporting? 2. How do journalists of TV social news view feminine angle of news reporting? 3. What kind of working ethics do journalists of TV social news hold? II. The field culture of TV news work & the news production of female journalists 1. What are the conditions required for female journalists? 2. Does sex factor have influence on TV social news female journalists in aspects below: (1) their relationship with steady news source (2) the assignments and evaluation they get (3) their interation with news source (4) their angle of news? As to the phenomenon of the feminization of TV social news, we have found that market is the major consideration of news production. Though the soft news issues have increased, the amount and value of this kind of issues are still far less then those of sensational issues. Meanwhile, women issues are few. Concerning the style and the way of reporting of TV social news, it has tended to practical value and has become closer to the feelings of audience, which is a feminized new style. However, sensationlism remain the main mode that journalists deal with social news. In the aspects of the field culture of TV news work, we have found that the paternal hegemony is the leading power in newsroom, and it has been integrated with the sensational ways of newsmaking oriented towards commercial pursuits. As to female journalists, they practice neutralized characters and female sexual characteristics, and accepted the paternal gazing and expection imposed on them, which is considered as a strategy of surrender to the field culture. Therefore, there is no distinct difference between the news reporting of male and female journalists.

Escola complementar de Caxias : histórias da primeira instituição pública para formação de professores na cidade de Caxias do Sul (1930-1961)

O presente trabalho tem como propósito pesquisar e analisar a organização e instalação da primeira instituição pública, de caráter oficial, para formação de professores primários, em 1930, na cidade de Caxias do Sul. Escola Complementar de Caxias, posteriormente denominada Escola Normal Duque de Caxias. O estudo assentou-se na possibilidade de produzir um olhar sobre a instituição, através do inventário das práticas produzidas e produtoras dos sujeitos que vivenciaram a mesma e contribuíram para a construção das culturas escolares. Os sujeitos envolvidos no processo escolar são historicamente (re)lembrados, visto que eles se constituíram nas práticas e nos discursos. Como referencial teórico, recorre-se aos postulados da História Cultural e à tríade conceitual proposta por Roger Chartier, apropriação, representação e prática. A análise documental se deu pela interpretação dos documentos produzidos pela instituição, pelos relatórios da Intendência e pelo acervo do Arquivo Histórico Municipal João Spadari Adami. O texto está organizado em três capítulos, iniciando com as considerações acerca da cidade de Caxias, abordando aspectos históricos, desde a chegada dos primeiros imigrantes até a criação e instalação da Escola em 1930, contemplando ainda a história da educação escolarizada na cidade. Na sequência, analiso os sujeitos envolvidos no processo escolar, enfatizando o papel dos diretores (as), professores (as), e alunos (as). E, por fim, focalizam-se elementos das culturas escolares, sobretudo as práticas permeadas pelos saberes e discursos implicados no processo escolar. As práticas escolares, inicialmente, denotam características dos discursos escolanovista e médico, a partir de 1939, indicam forte influência do discurso de cunho nacionalista. Finally, it has been focused on the elements of school culture, mainly the practices related to knowledge and discourses implied in the schooling process. Initially, the school practices have demonstrated traits of the new school and medical discourses, and then, from 1939 on, they have indicated the strong influence of the nationalist discourse. Investigation of Wolbachia symbiosis in isopods and filarial nematodes by genomic and interactome studies / Étude des relations symbiotiques entre Wolbachia et les isopodes et nématodes par analyses génomiques et de l'intéractome

Les Wolbachia sont des alpha-proteobactéries présentes chez de nombreux arthropodes et nématodes filaires. Ces bactéries héritées maternellement induisent chez leurs hôtes des phénotypes allant du parasitisme au mutualisme, avec le long de ce continuum des phénotypes tels que la féminisation (F), l'incompatibilité cytoplasmique (IC) ou la mort des mâles. Wolbachia est ainsi un modèle particulièrement intéressant pour étudier les différents types de relations symbiotiques.Chez Brugia malayi, comme pour les autres nématodes filaires, Wolbachia vit en symbiose obligatoire avec son hôte. L'élimination de la bactérie par des traitements antibiotiques entraîne une perte de fertilité voire la mort du nématode. Chez l'isopode terrestre Armadillidium vulgare, Wolbachia induit la féminisation des mâles génétiques en femelles fonctionnelles entraînant des biais de sex-ratio vers les femelles dans la descendance.Pour comprendre les mécanismes impliqués dans ces deux symbioses, nous avons mis au point une nouvelle méthode de capture pour isoler l'ADN de Wolbachia et séquencer 8 souches de Wolbachia d'isopodes (F et IC). Une étude de génomique comparative a permis d'établir un premier pan-génome des bactéries du genre Wolbachia et d'identifier 2, 5 et 3 gènes présents seulement chez les souches mutualistes, féminisantes ou induisant la mort des mâles. L'expression des gènes potentiellement impliqués dans la féminisation ou le mutualisme a été étudiée au cours du développement de l'hôte. L'étude de l'interactome protéique bactérie-hôte a ensuite été initiée en utilisant comme appât des protéines bactériennes à domaines eucaryotes en vue d'identifier les cibles de Wolbachia chez l'hôte. / Bacteria of the genus Wolbachia are gram-negative alpha-proteobacteria present in many arthropods and filarial nematodes. These obligate intracellular bacteria are maternally inherited and induce a large number of phenotypes across the symbiosis continuum from mutualism to parasitism, including feminization (F), cytoplasmic incompatibility (CI) or male killing. Studying Wolbachia symbioses is therefore of particular interest in the investigation of symbiotic relationships.In Brugia malayi and other filarial nematodes, they are obligate leading to a loss of worm fertility, and eventual death upon their depletion with antibiotic. In arthropods, they rather are parasitic. In the isopod crustacean Armadillidium vulgare they cause feminization when present: genetic males develop as functional female leading to female biased sex-ratio progenies.In order to understand the molecular mechanisms of these two symbioses, we set up a new capture procedure to catch Wolbachia DNA and performed whole-genome sequencing on 8 Wolbachia strains, symbionts of isopods (F & CI). Comparative genomics led to the establishment of the Wolbachia pan-genome as well as the identification of phenotype related gene patterns. We identified 2, 5 and 3 genes that are only found in mutualist, feminizing and male killing strains, respectively. Ce travail vise à comprendre pourquoi et comment l'accordéon diatonique breton a connu une profonde transformation organologique et stylistique au cours des dernières décennies. L'évolution de l'instrument est ici le témoin des dynamiques historiques et sociales, que l'accordéon présente la particularité d'avoir intégrées dans sa forme-même du fait de sa rare plasticité, et qu'il permet d'éclairer. Statut des musiciens professionnels au sein d'une pratique restée massivement amateur, rôles et mécanismes de l'apprentissage d'un savoir traditionnel initialement basé sur l'oralité, fluctuation des attentes sociales à travers les générations successives, sont quelques-uns des aspects qui se dévoilent au fil de l'étude et qui se relient.À partir du constat d'une mutation organologique progressive, nous chercherons à comprendre les modifications profondes des fonctions sociales jouées par l'instrument. Un retour historique permettra de définir les intentions et le statut des « pionniers » du renouveau de l'accordéon diatonique breton dans les années 1970. En nous appuyant sur ce socle, nous montrerons comment l'accordéon en tant qu'outil est en interaction étroite et permanente avec le geste du musicien et avec son intention (produire des notes – et dans quelle fonction sociale), et combien le passage des générations a renouvelé le contexte d'exposition de l'accordéon et le statut des musiques bretonnes dites « à danser ». Ainsi, nous verrons que la modification progressive de l'intention a conduit à une modification de l'instrument, mais que celle-ci, en retour, fragilise l'efficacité des intentions antérieures. / This work aims to understand why and how the Breton diatonic accordion has undergone a profound organological and stylistic transformation during the last decades. The evolution of the instrument here reflects the historical and social dynamics, that the accordion has integrated into its very form because of its rare plasticity, bringing them into light. The status of professional musicians in a largely amateur context, the roles and mechanisms of learning traditional knowledge initially based on orality, the fluctuation of social expectations through successive generations, are some of the aspects that unfold over the course of the study and connect with one-another.Starting from the observation of a progressive organologic change, we will seek to understand the profound changes of the social functions played by the instrument. A historical overview will allow to define the intentions and the status of the "pioneers" of the revival of the Breton diatonic accordion in the 1970s. Based on this foundation, we will show how the accordion as a tool is in close and permanent interaction with the musician's gesture and with his intention (producing notes – and in what social function), and how much the passage of generations has renewed the context of use of the accordion and the status of Breton music said "to be danced". Thus, we will see that the progressive modification of the intention led to a modification of the instrument, but that this, in turn, weakens the effectiveness of the previous intentions.
