Characterization of a novel lysine acetylation site in the N-terminal domain of the centromeric histone variant Cse4 in Saccharomyces cerevisiae

Die zentromerischen Regionen eukaryotischer Chromosomen sind notwendig für die Segregation der Schwesternchromatiden während der Zellteilung. In den Nukleosomen in diesen Regionen ist das kanonische Histon H3 durch die zentromerische Histon H3 Variante CENP-A ersetzt. Diese wird in Saccharomyces cerevisiae Cse4 genannt. Indem CENP-A die geordnete Assemblierung einzelner Untereinheiten des Kinetochors vermittelt, spezifiziert es die zentromerische Chromosomenregion und ist damit essentiell für die korrekte Segregation der Chromosomen während der Zellteilung. Kürzlich wurde gezeigt, dass Cse4 posttranslational modifiziert wird. Von Bedeutung für diese Arbeit sind die in der essentiellen Domäne des N-terminus liegenden Methylierung von Arginin 37 und die Acetylierung von Lysin 49, die durch phänotypische Suppression eine genetische Interaktion und eine antagonistische Funktion bei der epigenetischen Regulation in der korrekten Assemblierung der Kinetochoruntereinheiten zeigen. In dieser Arbeit wurde gezeigt, dass die Acetylierung an Cse4K49 von der Histonacetyltransferase Gcn5 abhängt, und dass dieses Enzym Komponenten des SAGA-Komplexes, aber nicht des ADA-Komplexes benötigt. Außerdem konnte in dieser Arbeit gezeigt werden, dass die Acetylierung von K49 in der frühen S-Phase ansteigt und zum Ende der S-Phase abnimmt. Es konnte weiterhin eine biochemische Interaktion der N-terminalen Domäne von Cse4 mit der COMA-Untereinheit Ctf19, der zentralen Region des Kinetochors, nachgewiesen werden, möglicherweise mit einer Präferenz zu einer nicht acetylierten Form von Cse4K49. In dieser Arbeit konnte gezeigt werden, dass der Transkriptions-Co-Aktivator Komplex SAGA eine Funktion an der zentromerischen Region in S. cerevisiae aufweist. Des Weiteren ist die Acetylierung an Cse4K49 in die Rekrutierung der Kinetochoruntereinheit Ctf19 involviert, und gibt damit einen Hinweis auf einen epigenetischen Regulationsmechanismus während der Chromosomensegregation. / The centromeric regions of eukaryotic chromosomes are essential for proper chromosome segregation. The nucleosomal composition in these regions differs from that of canonical nucleosomes in that histone H3 is replaced by the H3 variant CENP-A, which is termed Cse4 in Saccharomyces cerevisiae. CENP-A is the most important epigenetic mark on centromeres and essential for accurate kinetochore establishment at centromeric sites, which in turn are necessary to connect the centromeric sites to the microtubules. Whereas the histone fold domain of Cse4 shares a sequence homology with canonical histone H3, the N-terminal domain is rather long as compared to canonical histone tails and the centromeric histone variants of higher eukaryotes. One part of this flexible, positively charged histone tail has been shown to represent an essential N-terminal domain (END). Lysine 49 was defined as an acetylation site in this region, but the modification remains to be characterized in detail. In this study, we found that the Cse4K49 acetylation is dependent on the histone acetyltransferase Gcn5, and that the enzyme in this context acts within the SAGA/SLIK complex, whereas an involvement of the smaller ADA complex was not observed. Furthermore, we addressed the cell-cycle dependence of the K49 acetylation and found an increase in early S-phase and a decrease in late S-phase, whereas the R37 methylation persisted throughout S-phase. Ctf19 was found to bind acetylated and unacetylated Cse4K49 with a preference for unacetylated Cse4. The findings presented here show that the transcriptional co-activator complex SAGA functions at centromeric regions in S. cerevisiae, and that the Cse4K49 acetylation has an influence on the recruitment of the kinetochore subunit Ctf19. This suggests an epigenetic regulatory mechanism involving a specific acetylation on the N-terminus of Cse4 in chromosome segregation.

Nachweis Proteinkinase C abhängig exprimierter Gene in Astrozytomen

Die Proteinkinase C (PKC) ist eine wichtige Signaltransduktionskomponente, deren Aktivierung die Expression zahlreicher Gene induziert und zur Zelldifferenzierung und Zellproliferation führt. Ein besonders hohes Expressionsniveau der PKC findet man in vielen Tumoren. So korreliert in malignen Gliazellen das Expressionsniveau der PKC mit deren Wachstumsgeschwindigkeit. Es wird angenommen, daß die aktivierte PKC eine wichtige Rolle in der Tumorpromotion hat. In der vorliegenden Arbeit wurde untersucht, ob in Astrozytomzellinien Gene zu finden sind, die nach PKC-Aktivierung durch den Phorbol-Ester TPA differentiell exprimiert werden. Zunächst wurden kultivierte Zellen der Astrozytomzellinie LN-405 mit TPA respektive dem PKC-Inhibitor Chelerythrin behandelt. Nach Gewinnung der mRNA aus der zuvor isolierten RNA wurden in einem mehrstufigem PCR-Verfahren (SSH) cDNA-Abschnitte gewonnen, die zu vermeintlich differentiell exprimierten Genen gehören. Diese cDNA-Abschnitte wurden in Plasmid-Vektoren eingefügt, kloniert und zur Bestimmung sequenziert. Um falsch positive Sequenzen zu erkennen, wurden die zuvor radioaktiv markierten cDNA-Abschnitte mit Northernblots hybridisiert. Gleichzeitig ließ sich so ein zeitabhängiger Anstieg der Expression nach PKC-Stimulation untersuchen. Durch den PCR-Select-Assay (SSH) konnten insgesamt 11 Gene gefunden werden, die sich in der radioaktiven Northernblot-Hybridisierung, als nach PKC-Aktivierung differentiell exprimiert, darstellen ließen. Dabei bestätigt der gefundene Zusammenhang zwischen PKC-Aktivierung und differentieller Exprimierung bei fünf der 11 Gene (IL-8, Calpain, Interferon-gamma Rezeptor 2, Methionin Adenosyltransferase, beta-2 adrenerger Rezeptor) Ergebnisse anderer Autoren, wobei dieser Zusammenhang nur bei zwei Genen (IL-8 und Calpain) auch in Astrozytom- bzw. Gliom-Zellen schon früher gezeigt werden konnte. Sechs Gene (M-Phase Phosphoprotein-1, ect2-Onkogen, ERM-Gen, Ornithin-Decarboxylase-Antizym 2, MHC-bindendes Protein 2, Sequenz aus Cosmid F0811) wurden in der vorliegenden Arbeit erstmalig als PKC-abhängig exprimiert beschrieben. Die gefundenen Gene haben auf verschiedene Funktionen der Zellen Einfluß. So beeinflussen sie die Regulation des Zellzyklus (MPP1, ect2-Oncogen), die Immunregulation (MBP-2, IL-8, Interferon-gamma Rezeptor 2), die Signaltransduktion (beta-2 AR), die Transkription (ERM-Gen), die Proteinsynthese (ODC-Antizym, MAT), die Wachstumskontrolle (ODC-Antizym) und die Regulation der PKC selbst (Calpain). Für fünf Gene läßt sich ein eindeutiger Zusammenhang mit der Tumorpromotion herstellen: IL-8 (Angioneogenese), MBP-2 (Immunsuppression), ERM-Gen (Transkriptionspromotion), MAT (allgemein fördernder Einfluß auf den Metabolismus) und ect2-Oncogen (Oncogen). / The protein kinase C (PKC) is one of the major signal transduction systems and its activation leads to the induction of the expression of several genes, to cell differentiation and cell proliferation. Very high expressed PKC are found in many tumors. In malignant glia cells the expression of PKC correlates with their proliferation rate. The PKC activity has an important role for the tumor promotion. The object of this paper, was to investigated, if there are genes differentialy expressed after activation of PKC through the phorbol-ester TPA in astrocytoma cell lines. The astrocytoma cell line LN-405 was incubated with TPA and the PKC-inhibitor chelerythrine respectively. After isolation of RNA and mRNA the suppression subtractive hybridization (SSH) was used to isolate differentially expressed cDNA fragments. These cDNA fragments were inserted into the T/A cloning vector, cloned and sequenced. To detect false positives the cDNA fragments were analysed with northern blot technique. Examined was also a time-dependent acceleration of expression after TPA treatment. 11 genes were detected by suppression subtractive hybridization, showing differentially expressed in the northern blot hybridization. Five of the genes were found differentially expressed after PKC activation before (IL-8, calpain, interferon gamma receptor 2, beta-2-adrenergic receptor, methionine adenosyltransferase alpha), two of these genes (IL-8, calpain) also in astrocytoma- and glioma-cells respectively. Six genes (M-phase phosphoprotein 1, ect2-onkogene, erm gene, ornithine decarboxylase antizyme 2, MHC binding protein 2, sequence from Cosmid F0811) were described as PKC dependent expressed for the first time. The genes detected influence several cell functions. They are involved in cell-cycle regulation (MPP1, ect2-oncogene), immuneregulation (MBP-2, IL-8, interferon gamma receptor 2), signal transduction (beta 2 adrenergic receptor), transcription (erm-gene), synthesis of proteins (ODC-antizyme 2, MAT), growth control (ODC-antizyme) and regulation of PKC (Calpain). Five genes show a clear connection to tumor promotion: IL-8 (angioneogenesis), MPB-2 (immunesuppression), erm gene (promotion of transcription), MAT (promotion of metabolism) and ect2-oncogene (oncogene).

Unterschiedliche Autoregulation am PU.1 Lokus in B -Zellen und myeloischen Zellen

Als Schlüsselfaktor des hämatopoietischen Systems spielt PU.1 eine ent-scheidende Rolle in der Entwicklung der meisten hämatopoietischen Li-nien. Das PU.1 Expressionslevel bestimmt das Differenzierungspotential hämatopoietischer Stammzellen und Vorläufer. In den unterschiedlichen Zelltypen werden verschiedene Expressionsstärken etabliert. Wie diese zelltypischen Expressionslevel vonPU.1 generiert werden, ist bisher weit-gehend unbekannt. In der vorliegenden Doktorarbeit wurde mit Hilfe eines transgenen Maus-modells die cis-regulatorische Einheit von PU.1 definiert, um mit nachfol-genden molekularbiologischen und genomweiten Ansätzen Mechanismen der zellspezifischen Regulation von PU.1 zu aufzuzeigen. Die Definition der cis-regulatorischen Einheit von PU.1 erfolgte mit Hilfe eines transgenen Mausmodells, welches ein humanes PU.1 BAC Kons-trukt trägt. Es konnte gezeigt werden, dass humanes und murines PU.1 substituierbar sind und den gleichen Regulationsmechanismen unterlie-gen. Mit Hilfe genomweite DNaseI hypersensitivity Analysen, Methylie-rungs- und Bindungsstudien konnte ein neuer Regulationsmechanismus beschrieben werden, der eine spezifische kombinatorische Interaktion verschiedener cis-regulatorischer Elemente erfordert. Durch Reportergenassays in verschiedenen Zelltypen war es möglich, einen myeloischen Enhancer zu identifizieren. Es konnte gezeigt werden, dass PU.1 mit zelltyp-spezifischen Transkriptionsfaktoren interagiert, um unterschiedliche Bindungsmuster an seinen regulatorischen Elementen zu etablieren. Dadurch kommt es zu den spezifischen Expressionstärken von PU.1 / The transcription factor PU.1 occupies a central role in controlling myeloid and early B cell development and its correct lineage-specific expression is critical for the differentiation choice of hematopoietic progenitors. However, little is known of how this tissue-specific pattern is established. We previously identified an upstream regulatory cis-element (URE) whose targeted deletion in mice decreases PU.1 expression and causes leukemia. We show here that the URE alone is insufficient to confer physiological PU.1 expression, but requires the cooperation with other, previously unidentified elements. Using a combination of transgenic studies, global chromatin assays and detailed molecular analyses we present evidence that PU.1 is regulated by a novel mechanism involving cross-talk between different cis-elements together with lineage-restricted autoregulation. In this model, PU.1 regulates its expression in B cells and macrophages by differentially associating with cell-type specific transcription factors at one of its cis-regulatory elements to establish differential activity patterns at other elements.

It presents the reading - made by teachers during a reading event - of an Arnaldo Jabor s chronicle, which pressuposts were the indetermination of feelings and reading as a social co-construction process at interactionism. Therefore, the use of verbal protocol in group was detached at data collection. The study focused itself of which kinds of relations are established between the metaphors found in the chronicle and the readers voices trying to make sense. The readers that take part of this research are teachers that work at high school of a public school in the state of São Paulo. The empirical material was collected and analyzed in an ethnographic projection. The results shown that the metaphors reading were constructed into the interaction process with basis in relations established in knowledge acquired previously by readers, through the experience with other sources of knowledge structured in language. Thus, this study presented, by means of co-construction transacted in text meanings, the reading as an active process in which the communicative competence is used for the participants of this research, at the text comprehension / Este trabalho é uma pesquisa empírica sobre a leitura de metáforas contidas em uma crônica jornalística, objetivando verificar quais tipos de relações se estabelecem entre a metáfora e as manifestações dos leitores no processo de interpretação. Esta investigação insere-se na área de Língua Portuguesa. Apresenta a leitura de uma crônica de Arnaldo Jabor, realizada por professores, durante um evento de leitura, tendo por pressupostos a indeterminação dos sentidos e a leitura como um processo de co-construção social no enfoque interacionista. Destaca-se assim, na coleta de dados, o uso do protocolo verbal em grupo. O estudo centrou-se nos tipos de relações que se estabelecem entre as metáforas contidas na crônica e as vozes dos leitores tentando fazer sentido. Os leitores que participaram da pesquisa são professores do ensino médio de uma escola pública do Estado de São Paulo. O material empírico foi coletado e analisado em uma perspectiva etnográfica. Os resultados mostraram que a leitura de metáforas construiu-se no processo de interação com base em relações estabelecidas nos conhecimentos adquiridos anteriormente pelos leitores, por meio da experiência com outras fontes de conhecimento previamente estruturado pela linguagem. Portanto, esta pesquisa apresentou, por meio da co-construção negociada dos sentidos do texto, a leitura como um processo ativo em que a competência comunicativa é utilizada, pelos participantes da pesquisa, na interpretação do texto

A repetição como estratégia argumentativa na narrativa de Gil Gomes

Corpus consists of an oral narrative, produced by the well-known reporter Gil Gomes that, in order to ensure the interactivity with the audience, articulates different linguistic elements that establish sense relations and ensure the textual progression. We verify that the progression and retroaction movement in the texts is only possible by means of the cohesive procedures needed to establish sense relations. Among the procedures, we highlight the Repetition, as an important articulator that corroborates in the textual formulation and promotes the progression of the narrative flow. In order to study it, we have based on Marcuschi s (1992, 2006) and Koch s (2003) studies. In the text we verified that repetition is not restrained to the tautological occurrence of a single word, sentence or expression. Rather than a simple orality formulator process, repetition has a significant argumentative role. Koch (2003, p. 127): says one repeats as a mean to beat inside the interlocutor s mind until this latter allows to be persuaded . Ruled in a fait-diver narrative, O maníaco da Dutra presents the story of a maniac obsessed by the sexual degeneration. In that study, we noticed that Repetition is a resource very much used as argumentative element of fixation and warning, whilst during the broadcasting of the radiophonic narrative, the speaker, by means of the repetition, not only drives the audience towards the memorization of the crime particularities (rape) but also warns him/her about the rapist s modus operandi / Esta dissertação insere-se na linha de pesquisa da Análise da Conversação, Linguística Textual e Análise do Discurso, e tem como objetivo analisar os recursos lingüísticos essenciais no estabelecimento da coesão textual, como a repetição, a referência, a paráfrase etc, articulados na produção de uma crônica policial radiofônica. O corpus constitui-se de uma narrativa oral, produzida pelo prestigiado repórter Gil Gomes que, para garantir a interatividade com o ouvinte, articula diversos elementos lingüísticos que estabelecem relações de sentido e asseguram a progressão textual. Verificamos que o movimento de progressão e retroação no texto só é possível mediante os procedimentos coesivos necessários para estabelecer relações de sentido. Dentre os procedimentos, pontuamos a Repetição como importante articulador que corrobora na formulação textual e promove a progressão do fluxo narrativo. Para estudá-la, baseamo-nos nos estudos de Marcuschi (1992, 2006) e Koch (2003). No texto, verificamos que a repetição não está restrita à ocorrência tautológica de uma palavra, de uma frase ou de uma expressão. Mais do que um simples processo formulativo da oralidade, a repetição cumpre um importante papel argumentativo. Para Koch (2003, p. 127): repete-se como meio de martelar na mente do interlocutor até que este se deixe persuadir . Pautada em um fait-divers, a narrativa O maníaco da Dutra apresenta como enredo a historia de um maníaco obcecado pela tara sexual. Nesse estudo, constatamos que a Repetição é um recurso muito utilizado como elemento argumentativo de fixação e de alerta, uma vez que, durante a transmissão da narrativa radiofônica, o locutor, por meio da repetição, não só conduz o ouvinte à memorização das particularidades do crime (estupro) como também o alerta sobre o modo de agir do estuprador

Besides standing out as a painter and sculpturer, this memory collector is also a notable curator, carver, designer, printmarker, set designer, museologist, collector, cultural enticer. As a sculpturer, he applies constructivist code and makes use of contemporary material and procedure along with archaic primitive features. Emanoel Araújo is a black man who has made a name in art and culture. His first experience in a museum was as the director of Museu de Arte da Bahia, moreover he has headed Pinacoteca do Estado de São Paulo for ten years, where he could promote the leading expositions in the international circuit. Administrator of various initiatives, he worked as Secretary of Culture in São Paulo city. After researches, publications and exhibitions about the historical, cultural and artistic heritage of the black people in Brazil, Emanuel Araújo granted 1100 pieces from his personal collection under a free-lease agreement and founded Museu Afro Brasil. About the methodology used, the study began with analyses of documents in public archives in Bahia and São Paulo, as well as private documents belonging to the artist, family members and friends. After compiling the documents, oral history techniques were used; hence the interview was the basis for data collection in order to rebuild Emanuel Araújo’s memory. Besides numerous meetings with the subject of this survey himself, 70 more interviews were made with representatives of all segments related to Emanuel Araújo as relatives, friends, gallerists, cultural enticers and administrators, editors, teachers, artists, religious people, politicians, people directly related to plastic and visual art, museum employees from Bahia and São Paulo, among others. The results were qualitatively analyzed and showed that, by elaborating Emanuel Araújo’s memory, his successful journey couldn’t protect him from racism for being black. Besides the direction of Museu Afro Brasil, aimed to research, conservation and exhibition of objects concerning the afro-Brazilian cultural universe, the sculpturer writes articles, produces books and goes on with the production of sculptures, undertaking the curatorship of expositions in Brazil and abroad. As a conclusion, the discussion has enabled the comprehension of the sculpturer’s role as a depositary of the Afro-Brazilian cultural legacy in order to decrease racism and try to break resistance towards the black culture / Este estudo tem por objetivo geral conhecer, analisar e difundir os conhecimentos do negro a partir da memória, vida e obra de Emanoel Araújo, tendo o africanismo como plausibilidade para a diminuição do racismo e da discriminação. Afora se destacar na pintura e na escultura, esse “garimpeiro de memórias”, notabiliza-se como curador, entalhador, desenhista, gravurista, cenógrafo, museólogo, colecionador, fomentador e agitador cultural. Como escultor, emprega em sua arte uma linguagem construtivista e usa materiais e procedimentos contemporâneos com características arcaicas e primitivas. A história de Emanoel Araújo é a história de um negro que, por seu talento e esforço pessoal, consagrou-se no mundo das artes e da cultura. Sua experiência museal se inicia como diretor do Museu de Arte da Bahia. Nos dez anos à frente da Pinacoteca do Estado de São Paulo, promoveu as exposições mais concorrentes do circuito internacional. Gestor de uma série de iniciativas foi secretário da cultura do município de São Paulo. Após realizar pesquisas, publicações e mostras sobre a herança histórica, cultural e artística do negro do Brasil, Emanoel Araújo cedeu, em regime de comodato, 1100 peças de seu acervo particular e criou, em São Paulo, o Museu Afro Brasil. Na metodologia, o estudo iniciou-se com uma análise documental em arquivos públicos da Bahia e de São Paulo, como também de arquivos particulares do artista, de parentes e de amigos. Após a condensação deste arcabouço documental, empregou-se técnicas da História Oral tendo a entrevista como matéria prima na coleta de dados para reconstruir a memória de Emanoel Araújo. Além dos múltiplos encontros com o sujeito da pesquisa, foram realizadas 70 entrevistas com informantes representativos de todos os segmentos como parentes, amigos, galeristas, fomentadores e gestores culturais, editores, professores, artistas, religiosos, pessoas ligadas diretamente à arte plástica e visual, políticos, funcionários de museus, da Bahia e de São Paulo dentre outros. Os resultados foram analisados de forma qualitativa e demonstraram que, ao elaborar as memórias de Emanoel Araújo, sua trajetória de sucessos não impediu que ele vivenciasse o racismo por ser negro. Percebe-se que além da direção do Museu Afro Brasil, voltado à pesquisa, conservação e exposição de objetos relacionados ao universo cultural afro-brasileiro, o escultor escreve artigos, produz livros e prossegue com sua produção escultórica empreendendo a curadoria de exposições no Brasil e no exterior. Por conclusão, a discussão possibilitou compreender a figura do escultor como depositário do legado cultural afro-brasileiro a fim de reduzir o racismo, para buscar romper a resistência diante da cultura negra

A ruína da tradição em Walter Benjamin e a prosa poética de Charles Baudelaire

Walter Benjamin identified this scenario in The storyteller essay, which, by taking into account the overcoming of traditional experience by individual life experience, makes headway for his analysis of Charles Baudelaire s choc experience . Through this analysis, Benjamin reveals a number of facets inherent to the poet s thinking, both in Baudelaire s art critique and in his poetry production , as being proposals of what would be the ideal attitude capable of promoting a fruitful relationship between the individual and the modern city. This attitude would cause the individual to abandon his passiveness of character and would supply him with instruments to fully and wholly become part of the crowd without having to rely singly and solely on his own individual life experience. As an example of this new attitude in the work of Baudelaire, one finds Les Petits Poèmes en Prose. These poems are considered a supplementary part of Les Fleurs du Mal, and instead of singing to the end of an era however, the fifty fragments go one step further toward the insertion of poetry in the city s day-to-day, thus freeing it from former and ancient contingencies, or more precisely, from rhyme and verse / Esta dissertação aborda o declínio da tradição na modernidade como ponto de partida para a busca de outras expressões adequadas a um novo tempo. A constatação desse cenário por Walter Benjamin no ensaio O narrador, tendo em vista a superação da experiência tradicional pela vivência, abre caminho para sua análise sobre a experiência do choc em Charles Baudelaire. Por meio desta, o filósofo expõe diversas facetas inerentes ao pensamento do poeta, tanto no âmbito da crítica de arte quanto em sua produção poética, como recomendações a um ideal de postura para o indivíduo a fim de que este possa se relacionar de maneira frutífera com a cidade moderna. Tal postura o faria abandonar a passividade de seu caráter, dando-lhe instrumentos para se inserir na multidão de maneira plena, sem ter como único respaldo a própria vivência. Como exemplo dessa nova postura na própria obra do poeta francês, encontra-se os Petits Poèmes en Prose. Considerados complementares a Les Fleurs du Mal, ao contrário de cantarem o fim de uma era, porém, seus cinqüenta fragmentos dão um passo adiante rumo à inserção da poesia no cotidiano da cidade, livrando-a de antigas contingências; mais precisamente, da rima e do verso

Concepção de experiência em Walter Benjamin

A presente dissertação tem por objetivo apresentar a degradação da experiência humana na modernidade a partir do diagnóstico feito por Walter Benjamin. Para tanto, num primeiro momento, parte-se de um rápido percurso da história da filosofia a fim de delinear o conceito de experiência e analisar um dos primeiros textos em que Benjamin trata diretamente do assunto, “Sobre um programa da filosofia vindoura”. Num segundo momento, busca-se demonstrar como vai acontecendo a decadência da experiência a partir do ensaio “Experiência e pobreza”, em que Benjamin considera que aos pobres de experiência resta apenas assumir uma nova barbárie. O tema também aparece em “O narrador”, em que o autor demonstra o aniquilamento de uma experiência, outrora sinônimo de sabedoria e autoridade, consolidada por meio de sua transmissão de geração em geração, própria de uma organização coletiva, comunitária, ritualística e artesanal. As formas narrativas correspondentes são sintomáticas deste processo de esfacelamento da experiência: a narração - que vigorava no solo de um tempo onde ainda tinha-se tempo para contar e ouvir histórias - fora substituída pelo romance, caracterizador dos indivíduos isolados e solitários, e este, por sua vez, substituído pela informação jornalística, forma narrativa fragmentada e desconexa. Analisar a questão da experiência (erfahrung) em Walter Benjamin é contrapô-la à noção de vivência (erlebnis). Na esfera da vivência, saturada de eventos e sensações, resta ao ser humano a capacidade de reagir a esses estímulos (reportando à noção de choque em Freud). A memória (e seu correlato – o esquecimento) é imprescindível à experiência, mas perante os choques, o ser humano só armazena suas vivências na camada mais superficial da consciência, impossibilitando recursos para a experiência estética ou poética. Num terceiro momento, procura-se, nesse estudo, fazer um diálogo entre as constatações e os conceitos da filosofia da experiência benjaminianas com algumas imagens literárias, buscando, na literatura, elementos que vicejam a reflexão filosófica. O conto de Jorge Luis Borges - “Funes, o memorioso” - e o trecho de “Cem anos de solidão”, de Gabriel Garcia Marquez, sobre a doença do esquecimento, são preciosos mananciais para esse fim. Por último, problematiza-se a constatação da degradação da experiência na modernidade. Discute-se a possibilidade da realização da experiência, bem como o resgate de seu aniquilamento provocado pelas idiossincrasias da contemporaneidade e, ainda, quais espaços propiciariam este possível resgate. / This thesis presents the degradation of human experience based on Walter Benjamin’s diagnosis of it. In order to do that, firstly a brief overview of the history of philosophy is presented so as to delineate the concept of experience and analyze one of the first texts in which Benjamin directly deals with the subject (“Sobre um programa da filosofia vindoura” – “On the Program of the Coming Philosophy”). Secondly, the decadence of experience is demonstrated in the essay “Experiência e pobreza” (“Experience and Poverty”), in which Benjamin considers that the poor of experience are left with only taking on a new barbarism. That theme is also dealt with in “O narrador” (“The Narrator”), in which the author demonstrates the annihilation of an experience once synonymous with wisdom and authority, consolidated through its transmission from generation to generation and proper to a collective, communitarian, ritualistic, and artisanal organization. The corresponding narrative forms are symptomatic of that process of experience ruin: narration, which thrived at a time when one had time to tell and listen to stories, was replaced by the novel, a characterizer of isolated and lonely individuals. The latter, in its turn, was replaced by journalistic information, a fragmented and disconnected narrative form. Analyzing the issue of enduring experience (erfahrung) in Walter Benjamin means opposing it to the notion of fleeting experience/ephemera (erlebnis). In the sphere of fleeting experience/ephemera, saturated with events and sensations, the human being is left with the capacity to react to those stimuli (reminding us of Freud’s notion of shock). Memory (and its correlate – oblivion) is essential to enduring experience, but in the presence of shocks, the human being only stores his fleeting experiences/ephemera on the most superficial layer of consciousness, precluding resources for the aesthetic or poetic experience. At a third moment, this study promotes a dialogue between the realizations and concepts mentioned above and some literary images, searching in literature for elements which cause philosophic reflection to bloom. Jorge Luis Borges’ short story “Funes, o memorioso” (“Funes the Memorious”) and the extract from Gabriel García Marquez’s Cem anos de solidão (One Hundred Years of Solitude) about the disease of forgetfulness are precious sources for that end. Lastly, the realization of the experience ruin in modernity is discussed. Is it still possible to have an enduring experience and rescue it from its annihilation caused by the idiosyncrasies of contemporary times? What possible spaces would allow that rescue?

Dança-teatro com jovens : uma proposta pedagógica inspirada em processos de criação de Pina Bausch

O mote central da investigação é a dança-teatro e suas possibilidades pedagógicas. O estudo tem por objetivo extrair princípios pedagógicos em dança-teatro a partir de premissas de criação da coreógrafa alemã Pina Bausch e do documentário Sonhos em Movimento (2010), dirigido por Anne Linsel e Rainer Hoffmann, que mostra o desenvolvimento do trabalho de Bausch com jovens, na remontagem do espetáculo Kontakthof (2008). De modo geral, pretende-se evidenciar a dança-teatro como elemento de conhecimento humano, através da educação. Como campo teórico relacionado à dança-teatro são estudados diversos autores brasileiros e estrangeiros que abordam o processo cênico de Bausch. Referente à noção de “juventudes”, são trazidos autores que discutem a relação do jovem com seu meio social, sua criação de identidade e sua corporeidade. Para tratar de conceitos pedagógicos são utilizados escritos de Jacques Rancière, Fernando Becker, Vera Lúcia Bertoni dos Santos, João Pedro Gil e Odailso Berté, dentre outros. Através dos depoimentos dos jovens participantes do documentário pode-se identificar que, mesmo sem este fim, a proposta de Bausch possibilita neles uma nova visão de si perante seu cotidiano a partir da entrada no universo da arte, gerando construção de conhecimento por meio da poética artística. Para finalizar o estudo foi desenvolvida uma oficina com jovens, que gerou o compartilhamento das descobertas da investigação, possibilitando o relato dos participantes acerca de seus aprendizados junto à proposta pedagógica em dança-teatro. / The central theme of the investigation is Dance Theatre and its pedagogical possibilities. This study aims to extract the pedagogical principles of Dance Theatre from creation premises of the German choreographer Pina Bausch and from the documentary Dancing Dreams (2010), directed by Anne Linsel and Rainer Hoffmann, which shows the development of Bausch's work with young people in the recreation of the spectacle Kontakthof (2008). In general, this investigation intends to consolidate Dance Theatre as an element of human knowledge, through education. As theoretical field related to Dance Theatre, several Brazilian and foreign authors dedicated to Bausch's scenic process are studied. Concerning the notion of "youths", we brought authors that discuss young people's relations with their social environment, with the creation of their identity and with their corporeity. To deal with Pedagogy concepts it is used writings of Jacques Rancière, Fernando Becker, Vera Lúcia Bertoni dos Santos, João Pedro Gil, Odailso Berté, among others. Through the testimonies of the young participants of the documentary it can be identified that, even without that intention, Bausch's proposition promotes a new view of themselves towards their daily lives, through their entrance in the Art world, generating the construction of knowledge through the artistic poetics. To end the study, there was a workshop with young people, which made it possible to share the research findings, allowing the participants to report their learnings along with the pedagogical proposition of Dance Theatre.
