Nature and origin of sedimentary deposits in the Ecuador subduction trench : paleoseismological implications / Nature et origine des dépôts sédimentaires de la fosse de subduction d’Equateur : implications paléosismologiques

La sédimentation marine récente dans les fosses de subduction est caractérisée par l'interstratification de sédiments hémipélagiques et de turbidites localement intercalées avec les coulées de débris, qui peuvent résulter de la destabilisation des pentes continentales par de tremblements de terre. La marge d’Equateur est constituée par une forte érosion tectonique qui contribue à la formation d'une fosse profonde remplie d'une suite complexe de faciès sédimentaires. La sédimentation par écoulements gravitaires est omniprésente le long de la marge et les faciès vont de dépôts de transport de masse d'épaisseur métriques latéralement continus à des turbidites d'épaisseur centimétriques isolées intercalées avec des couches d'hémipélagites, de volcanoclastiques et de téphras. Nous présentons l'interprétation de la bathymétrie, des profils sismiques à haute résolution et des données pétrophysiques des carottes sédimentaires. L'objectif de cette étude est de décrire la complexité morphologique à la frontière équatorienne de la plaque de Nazca où un ensemble d'aspérités marines profondes ont subducté à différentes échelles, et ses conséquences sur la distribution latérale des sédiments dans les différents sous-bassins. La marge équatorienne comprend trois segments géomorphologiques: Le segment nord, situé au nord de la crête Carnegie, est caractérisé par une large (5-10 km) et profonde fosse (3800-4000 m), une pente continentale ravinée et une plate-forme (10-40 km de large) avec subsidence active. Le segment central en face de la crête de Carnégie montre une fosse étroite (0-5 km de large) et peu profonde (3100-3700 m), la pente escarpée et ravinée, sans canyons, et plateau continental étroit de 15 à 40 km de large caractérisé par des zones d'affaissement et de soulèvement actifs. Enfin, le segment sud, situé au sud de la crête Carnegie, présente une large (5-10 km) et profonde fosse (4000-4700 m), une pente continentale pauvre en sédiments avec des systèmes de canyons bien définis et une large plate-forme de subsidence (20-50 km). La dynamique sédimentaire le long de la marge est évaluée par l'analyse de 15 carottes sédimentaires dont la description visuelle, les photographies à haute résolution, l'imagerie par rayons X, les données XRF et les propriétés pétrophysiques conduisent à l'identification de 11 faciès sédimentaires caractérisant 7 processus sédimentaires: dépôts de turbidite, hémipélagites, téphras, dépôts de coulées de débris, homogénites, des slumps et des dépôts de carbonate de ooze. Les âges des dépôts sont définis par la datation au radiocarbone des sédiments hémipélagites. Les âges vont de 500 à 48000 ans BP. Les profils sismiques à haute résolution permettent de définir 3 echo-faciès: transparent, stratifiés et chaotiques. Le facies transparent est principalement associé aux dépôts d'homogénites, le facies stratifié est associé aux dépôts interstratifiés turbiditique-hémipélagique et le facies chaotique est associé à des dépôts gravitaires grossiers. Le remplissage de la fosse représente un enregistrement lacunaire mais important de l'histoire de la marge de subduction. De grandes coulées de débris se déplaçant vers l'est dans les deux séquences inférieures du remplissage de la fosse sont initiées le long de la paroi extérieure de la fosse, le long de grandes failles normales dues à la flexion de la plaque océanique subductante. Les sédiments de la séquence supérieure du remplissage qui nappent la fosse sont plus largement fournis par la paroi interne de la fosse mais avec un fort contrôle de la ride de Carnegie. En conséquence, la profondeur, la fréquence, l'épaisseur, la composition et la disposition latérale des dépôts sédimentaires varient grandement entre le nord et le sud. Les grands méga-lits simples, les slumps, les coulées de débris et les homogénites sont situés dans les segments nord et sud. The active margin of Ecuador comprises tectonic erosion that contributes to the formation of a deep trench filled by a complex suite of sedimentary facies. Gravity flow sedimentation is ubiquitous along the margin and facies range from laterally continuous m-thick mass transport deposits to isolated cm-thick turbidites intercalated with hemipelagite, volcanoclastics and tephra. In this study we show interpretation of swath bathymetry, high-resolution seismic profiles and petrophysical data from cores. The objective is to describe the morphologic complexity on the Ecuadorian border of the Nazca plate where a set of deep marine asperities is subducting at different scales, and their consequences on the distribution of sediments in the different sub-basins. Ecuadorian margin comprises three geomorphological segments: The northern segment, northward of the Carnegie Ridge, is characterized by a wide (5-10 km) and deep trench (3800 – 4000 m), a gentler gullied continental slope and a shelf (10-40 km wide) with active subsidence. The central segment facing the Carnegie Ridge, is strongly influenced by the subduction of the Carnegie ridge which induces a narrow (0–5 km wide) and shallow trench (3100 – 3700 m depth), a steep and gullied slope with no canyons and a 15–40 km wide shelf characterized by areas with active subsidence and uplift. Finally, the southern segment, southward of the Carnegie Ridge, presents a wide (5–10 km) and deep (4000–4700 m) trench, a starved continental slope with well-defined canyon systems and a wide subsiding shelf (20–50 km). The sedimentary dynamics along the margin is evaluated by the analysis of 15 cores. Visual description, high-resolution photographs, X-Ray imagery, XRF data and petrophysical properties led to the identification of 11 sedimentary facies that characterize seven sedimentary processes: turbidites, hemipelagites, tephras, debris flows, homogenites, slumps, and ooze carbonate deposits. Age of the deposits is defined by radiocarbon age dating of hemipelagic sediments. Ages range from 500 to 48,000 years BP. High-resolution seismic profiles allow definition of three echo-facies: transparent, layered and chaotic. Transparent echo-facies is mainly associated to homogenite deposits, layered echo-facies is associated to the turbiditic-hemipelagic interbedded deposits and chaotic echo-facies is associated to reworked gravity flow deposits. The trench fill represents a lacunar but important record of the subduction margin history. Large eastward debris flows in the lower two sequences of the trench fill are provided by the trench outer wall as a results of slope failures along normal faults due to the downward bending of the oceanic plate. The sediment of the upper sequence of the trench fill draping the trench floor, are largely provided by the inner trench wall strongly controlled by the Carnegie Ridge. As a result, depth, frequency, thickness, composition and lateral disposition of the deposits vary greatly from those at north and south. The large, simple mega-beds like slump, debris flows and homogenites are located at the northern and southern segments. They were triggered by large regional faults in the North and enhanced by the activity of sets of splay faults in the South overhanging the seafloor at the slope toe. Small-size, fluid rich events were triggered by subduction of isolated seamounts at the edges of the Carnegie Ridge due to frequent but small destabilizations of an inner trench wall preconditioned by the impacts of successive seamounts. Sets of partly volcanoclastic turbidites in central segment might have been triggered by the complex interaction of slope and continental shelf deformation by seamount subduction

For example, new knowledge is available on the variant versions of the Septuagint and application of the literary concept of intertextuality has yielded significant results in recent biblical scholarship. However, considering that 1 Peter arguably uses the Old Testament in the most condensed manner of all the New Testament writings, it is surprising that relatively few recent studies have been carried out on the use of the Old Testament in 1 Peter. As a partial corrective to this situation, this in-depth study is focused on the use of the Psalms in 1 Peter, utilizing an integrated hermeneutical procedure that combines traditional grammatical-historical analysis with the state of the art on New Testament use of the Old Testament. The aim is to clarify the hermeneutical implications of the use of the Psalms in 1 Peter for Old Testament and New Testament exegesis within the Reformed tradition today. Thus, a comprehensive exegetical approach is systematically applied to each pericope of 1 Peter and to every relevant Psalm passage in order to establish a valid foundation for understanding how the author interprets the Psalms and how the Psalm references function within the argument of 1 Peter. In addition, intertextual resonance is considered as an effective means to enhance understanding of the function and effect of Psalm references as intended by the author, and to determine the possible unintended effect of Psalm references upon the addressees and later readers. Furthermore, consideration of reader response sometimes makes it possible to identify echoes of Psalm passages not likely intended by the author, but nevertheless, very likely to have come to the minds of the recipients with significant effect. One conclusion of this study is that there is no compelling evidence that the author’s interpretation of the Psalms ignored the intended meaning of the Psalm in its own context, or merely reflected contemporary Second Temple interpretations. Furthermore, it has been possible to refine the criteria for identifying and classifying Psalm allusions rather than merely following broad, predetermined criteria. Thus, a distinction is made between specific allusion, in which case the author’s argument depends upon recognition of specific Psalm passages, and general allusion, in which case the author’s argument depends upon a concept recognizably derived from the Psalms but not limited by the wording of specific references. This study also demonstrates that the use of the Psalms in 1 Peter is based upon a strong sense of solidarity with believers of the past, especially as they expressed their responses to God in the context of suffering. In particular, compared with other New Testament writings, 1 Peter stands out for the fact that the majority of its Psalm allusions are used for the purpose of developing and reapplying significant themes. A synchronic approach to the Serek ha-Yahad (1QS) : from text to social and cultural context

The purpose of this study is to investigate the social and cultural contexts of 1QS (Serek ha-Yahad) by means of a textual study. The analysis of the text is performed in a synchronic perspective. This means that lexical choices, grammatical forms, references, topics, themes, and intertextuality are analyzed text-internally. By doing so, this study sheds new light on old questions of textual cohesion and coherence, questions that until now have been dealt with mostly from a diachronic perspective. The text analysis entails investigation in view of three interrelated dimensions of language function: ideational, interpersonal, and textual. These imply language as transmitting information, creating and sustaining relations, and functioning to organize itself into cohesive units. Although applying some of the terminology from the field of text linguistics (SFL), the focus in this study is on what a text means rather than why. This means that the semantic-pragmatic aspects of language are of foremost interest here. The analysis is performed from bottom and up, then from top down again. Words, phrases, and sentences are investigated up to the broadest linguistic level, namely, to the semantic discourse itself. With an understanding of the larger discourse at hand thanks to this analysis of textual cohesion and coherence, textual details are once again revisited and interpreted anew. In this work, 1QS is analyzed from beginning to end—chronologically, so to say. Then, at the end of each major section, the discourse is analyzed overall. Following the text analysis, conclusions of the investigations are presented. The conclusions argue that the hierarchal structure of the community and its stringent regulations are to be understood as a corrective in response to corrupt society. It is also argued that language in 1QS has a performative function. Rather than describing the way things are, it aspires to evoke the ideal society. Instead of understanding 1QS and the community mirrored in it as a deviant group with little or no contact with the surrounding world, it is then understood as a potent contribution to late Second Temple Jewish discourse concerning how to create a just society and a sanctifying cultic practice. In the final chapter, the insights gained from textual analysis of 1QS are brought into encounter with the theoretical framework posited by French historian and philosopher René Girard (1923–2015). In light of Girard’s philosophy, the hierarchal organization of the community (the Yahad) as well as its regulations can be interpreted as an effort to prevent a mimetic crisis. The function of the scapegoat in 1QS is discussed in light of Girard’s grand theory of the mechanisms of scapegoating in all societies. The study closes with the tentative hypothesis that the community in 1QS deconstructs the scapegoating mechanism by taking the role of the scapegoat upon itself.

Hydrology and Bed Topography of the Greenland Ice Sheet : Last known surroundings

The increased temperatures in the Arctic accelerate the loss of land based ice stored in glaciers. The Greenland Ice Sheet is the largest ice mass in the Northern Hemisphere and holds ~10% of all the freshwater on Earth, equivalent to ~7 metres of global sea level rise. A few decades ago, the mass balance of the Greenland Ice Sheet was poorly known and assumed to have little impact on global sea level rise. The development of regional climate models and remote sensing of the ice sheet during the past decade have revealed a significant mass loss. To monitor how the Greenland Ice Sheet will affect sea levels in the future requires understanding the physical processes that govern its mass balance and movement. In the southeastern and central western regions, mass loss is dominated by the dynamic behaviour of ice streams calving into the ocean. Changes in surface mass balance dominate mass loss from the Greenland Ice Sheet in the central northern, southwestern and northeastern regions. Little is known about what the hydrological system looks like beneath the ice sheet; how well the hydrological system is developed decides the water’s impact on ice movement. In this thesis, I have focused on radar sounding measurements to map the subglacial topography in detail for a land-terminating section of the western Greenland Ice Sheet. This knowledge is a critical prerequisite for any subglacial hydrological modelling. Using the high-resolution ice thickness and bed topography data, I have made the following specific studies: First, I have analysed the geological setting and glaciological history of the region by comparing proglacial and subglacial spectral roughness. Second, I have analysed the subglacial water drainage routing and revealed a potential for subglacial water piracy between adjacent subglacial water catchments with changes in the subglacial water pressure regime. Finally, I have looked in more detail into englacial features that are commonly observed in radar sounding data from western Greenland. In all, the thesis highlights the need not only for accurate high-resolution subglacial digital elevation models, but also for regionally optimised interpolation when conducting detailed hydrological studies of the Greenland Ice Sheet. / De ökade temperaturerna i Arktis påskyndar förlusten av landbaserad is lagrad i glaciärer och permafrost. Grönlands inlandsis är den största ismassan på norra halvklotet och lagrar ca 10% av allt sötvatten på jorden, vilket motsvarar ca 7 meter global havsnivåhöjning. För ett par decennier sedan var inlandsisens massbalans dåligt känd och antogs ha liten inverkan på dagens havsnivåhöjning. Utvecklingen av regionala klimatmodeller och satellitbaserad fjärranalys av inlandsisen har under de senaste decenniet påvisat en betydande massförlust. För att förutse vilken inverkan inlandsisen har på framtida havsnivåhöjningar krävs en förståelse för de fysikaliska processerna som styr dess massbalans och isrörelse. I de sydöstra och centrala västra delarna av inlandsisen domineras massförlusten av dynamiska processer i isströmmar som kalvar ut i havet. Massförlusten i de centrala norra, sydvästra och nordöstra delarna domineras av isytans massbalans. Ytterst lite är känt om hur det hydrologiska systemet ser ut under inlandsisen; hur väl det hydrologiska systemet är utvecklat avgör vattnets påverkan på isrörelsen. I denna doktorsavhandling har jag använt markbaserade radarmätningar för att kartlägga den subglaciala topografin för en del av den västra landbaserade inlandsisen. Denna kunskap är en viktig förutsättning för att kunna modellera den subglaciala hydrologin. Med hjälp av rumsligt högupplöst data över istjockleken och bottentopografin har jag gjort följande specifika studier: Först har jag analyserat de geologiska och glaciologiska förhållandena i regionen genom att jämföra proglacial och subglacial spektralanalys av terrängens ytojämnheter. Sedan har jag analyserat den subglaciala vattenavrinningen och påvisat en potential för att avrinningsområdena kan ändras beroende på vattentryckförhållandena på botten. Slutligen har jag tittat mer i detalj på englaciala radarstrukturer som ofta observerats i radardata från västra Grönland. Sammanfattningsvis belyser avhandlingen behovet av inte bara noggranna rumsligt högupplösta subglaciala digitala höjdmodeller, utan även regionalt optimerad interpolering när detaljerade hydrologiska studier ska utföras på Grönlands inlandsis.

Génération et reconnaissance de rythmes au moyen de réseaux de neurones à réservoir

Le programme que nous avons développés pour ce travail est en language Python. / Les réseaux de neurones à réservoir, dont le principe est de combiner un vaste réseau de neurones fixes avec un apprenant ne possédant aucune forme de mémoire, ont récemment connu un gain en popularité dans les communautés d’apprentissage machine, de traitement du signal et des neurosciences computationelles. Ces réseaux qui peuvent être classés en deux catégories : 1. les réseaux à états échoïques (ESN)[29] dont les activations des neurones sont des réels 2. les machines à états liquides (LSM)[43] dont les neurones possèdent des potentiels d’actions, ont été appliqués à différentes tâches [11][64][49][45][38] dont la génération de séquences mélodiques [30]. Dans le cadre de la présente recherche, nous proposons deux nouveaux modèles à base de réseaux de neurones à réservoir. Le premier est un modèle pour la reconnaissance de rythmes utilisant deux niveaux d’apprentissage, et avec lequel nous avons été en mesure d’obtenir des résultats satisfaisants tant au niveau de la reconnaissance que de la résistance au bruit. Le second modèle sert à l’apprentissage et à la génération de séquences périodiques. Ce modèle diffère du modèle génératif classique utilisé avec les ESN à la fois au niveau de ses entrées, puisqu’il possède une Horloge, ainsi qu’au niveau de l’algorithme d’apprentissage, puisqu’il utilise un algorithme que nous avons spécialement développé pour cette tache et qui se nomme "Orbite". La combinaison de ces deux éléments, nous a permis d’obtenir de bons résultats, pour la génération, le sur-apprentissage et l’extraction de données. Nous pensons également que ce modèle ouvre une fenêtre intéressante vers la réalisation d’un orchestre entièrement virtuel et nous proposons deux architectures possibles que pourrait avoir cet orchestre. Dans la dernière partie de ce travail nous présentons les outils que nous avons développés pour faciliter notre travail de recherche. / Reservoir computing, the combination of a recurrent neural network and one or more memoryless readout units, has seen recent growth in popularity in and machine learning, signal processing and computational neurosciences. Reservoir-based methods have been successfully applied to a wide range of time series problems [11][64][49][45][38] including music [30], and usually can be found in two flavours: Echo States Networks(ESN)[29], where the reservoir is composed of mean rates neurons, and Liquid Sates Machines (LSM),[43] where the reservoir is composed of spiking neurons. In this work, we propose two new models based upon the ESN architecture. The first one is a model for rhythm recognition that uses two levels of learning and with which we have been able to get satisfying results on both recognition and noise resistance. The second one is a model for learning and generating periodic sequences, with this model we introduced a new architecture for generative models based upon ESNs where the reservoir receives inputs from a clock, as well as a new learning algorithm that we called "Orbite". By combining these two elements within our model, we were able to get good results on generation, over-fitting and data extraction. We also believe that a combination of several instances of our model can serve as a basis for the elaboration of an entirely virtual orchestra, and we propose two architectures that this orchestra may have. In the last part of this work, we briefly present the tools that we have developed during our research.

Neuro-inspired computing enhanced by scalable algorithms and physics of emerging nanoscale resistive devices

<p>Deep ‘Analog Artificial Neural Networks’ (AANNs) perform complex classification problems with high accuracy. However, they rely on humongous amount of power to perform the calculations, veiling the accuracy benefits. The biological brain on the other hand is significantly more powerful than such networks and consumes orders of magnitude less power, indicating some conceptual mismatch. Given that the biological neurons are locally connected, communicate using energy efficient trains of spikes, and the behavior is non-deterministic, incorporating these effects in Artificial Neural Networks (ANNs) may drive us few steps towards a more realistic neural networks. </p> <p> </p> <p>Emerging devices can offer a plethora of benefits including power efficiency, faster operation, low area in a vast array of applications. For example, memristors and Magnetic Tunnel Junctions (MTJs) are suitable for high density, non-volatile Random Access Memories when compared with CMOS implementations. In this work, we analyze the possibility of harnessing the characteristics of such emerging devices, to achieve neuro-inspired solutions to intricate problems.</p> <p> </p> <p>We propose how the inherent stochasticity of nano-scale resistive devices can be utilized to realize the functionality of spiking neurons and synapses that can be incorporated in deep stochastic Spiking Neural Networks (SNN) for image classification problems. While ANNs mainly dwell in the aforementioned classification problem solving domain, they can be adapted for a variety of other applications. One such neuro-inspired solution is the Cellular Neural Network (CNN) based Boolean satisfiability solver. Boolean satisfiability (k-SAT) is an NP-complete (k≥3) problem that constitute one of the hardest classes of constraint satisfaction problems. We provide a proof of concept hardware based analog k-SAT solver that is built using MTJs. The inherent physics of MTJs, enhanced by device level modifications, is harnessed here to emulate the intricate dynamics of an analog, CNN based, satisfiability (SAT) solver. </p> <p> </p> <p>Furthermore, in the effort of reaching human level performance in terms of accuracy, increasing the complexity and size of ANNs is crucial. Efficient algorithms for evaluating neural network performance is of significant importance to improve the scalability of networks, in addition to designing hardware accelerators. We propose a scalable approach for evaluating Liquid State Machines: a bio-inspired computing model where the inputs are sparsely connected to a randomly interlinked reservoir (or liquid). It has been shown that biological neurons are more likely to be connected to other neurons in the close proximity, and tend to be disconnected as the neurons are spatially far apart. Inspired by this, we propose a group of locally connected neuron reservoirs, or an ensemble of liquids approach, for LSMs. We analyze how the segmentation of a single large liquid to create an ensemble of multiple smaller liquids affects the latency and accuracy of an LSM. In our analysis, we quantify the ability of the proposed ensemble approach to provide an improved representation of the input using the Separation Property (SP) and Approximation Property (AP). Our results illustrate that the ensemble approach enhances class discrimination (quantified as the ratio between the SP and AP), leading to improved accuracy in speech and image recognition tasks, when compared to a single large liquid. Furthermore, we obtain performance benefits in terms of improved inference time and reduced memory requirements, due to lower number of connections and the freedom to parallelize the liquid evaluation process.</p>

Ce travail se place dans le contexte des recherches sur les mémoires quantiques pour la lumière. L’information quantique est stockée dans un état de superposition atomique, dont la durée de vie détermine le temps maximum de stockage.On s’intéresse particulièrement aux matériaux capables de capturer la lumière par excitation résonnante d’une raie d’absorption, puis de conserver l’information quantique dans un état de superposition du fondamental électronique.Dans Tm3+:YAG, l’information est enregistrée dans un état de spin nucléaire. Cependant le champ magnétique qui lève la dégénérescence nucléaire entraîne les différents spins à des vitesses de précession différentes, ce qui tend à détruire l’aimantation initiale, porteuse de l’information.Une étude quantique du cristal est réalisée lors du premier chapitre de ce manuscrit. Les trois chapitres suivants traitent des différents mécanismes conduisant au déphasage des spins nucléaires. On y trouvera différente analyses théoriques qui seront confirmées par un ensemble de résultats expérimentaux, ainsi qu’une description détaillée du dispositif expérimental. Enfin le dernier chapitre, prospectif, exploite les outils développés au cours de la thèse pour préserver les cohérences optiques. Il présente quelques résultats expérimentaux prometteurs sur l’allongement du temps de vie de ces cohérences optiques. / This work takes place in the context of research about quantum memories for light. The quantum information is stored in an atomic superposition state whose lifetime sets the maximum storage time. We are particularly interested in materials which are able to hold the light by resonant excitation of an absorption line, preserving the quantum information in a superposition state of the electronicfundamental.n Tm3+:YAG the information is stored in a nuclear spin state. However, the magnetic field which lifts the nuclear degeneracy generates different precession speeds of the spins. This destroys theinitial magnetization carrier of the information.In the first chapter of this thesis, a quantum analysis of the crystal is done. The following three chapters are devoted to different mechanisms to control the nuclear spins dephasing. There it ispossible to find different theoretical analysis which will be confirmed by a series of experimental measurements, including an extended description of the set-up. Finally, the last chapter presentsthe different techniques used to preserve the optical coherence. Promising experimental measurements are presented to extend the life time of the optical coherences.

Actuellement, des méthodes quantitatives de résonance magnétique nucléaire (RMN) offrent des biomarqueurs qui permettent la réalisation d’études longitudinales pour le suivi de l’évolution des maladies neuromusculaires et des essais thérapeutiques de manière non-invasive. A la différence de la dégénérescence graisseuse, les processus d’inflammation/œdème/nécrose et fibrose sont des signes d’activité des maladies et leurs quantifications constitueraient ainsi de biomarqueurs parfaitement adaptés pour le suivi thérapeutique. Ce travail de thèse a consisté à mettre en place des méthodologies quantitatives plus précises et adaptées à l’étude clinique du muscle pour : (i) détecter et quantifier des sites d’activité de maladies par la cartographie T2 de l’eau ; (ii) identifier les différents processus pathophysiologiques qui sont à l’origine des altérations du T2 ; et (iii) détecter et quantifier la fibrose musculaire. Nous avons implémenté deux méthodes pour la quantification du T2 de l’eau dans le muscle. La première est basée sur une séquence d’écho de spin du type CPMG, où les signaux provenant des protons des lipides et de l’eau sont acquis simultanément et séparés à postériori par un traitement tri-exponentiel qui exploite la différence entre les T2 qui caractérisent les signaux de l’eau et de la graisse. La deuxième technique est basée sur une séquence de « partially spoiled steady state free precession (pSSFP) ». Différemment de la première technique qui nécessite un traitement assez élaboré sur des images acquises à 17 temps d’écho différents, dans la pSSFP la cartographie T2 est extraite à partir de deux séries de données 3D. L’acquisition 3D est compatible avec des techniques de sélection spectrale de l’eau, ce qui évite la contamination par les signaux des lipides. Les deux méthodes ont été validées expérimentalement chez des malades et des sujets sains et ont démontré leur capacité à détecter et quantifier des sites d’activité de maladies. Ces deux travaux font l’objet de deux publications dans des journaux scientifiques internationaux : Azzabou, de Sousa, Araujo, & Carlier, 2014. Journal of Magnetic Resonance Imaging. Malgré le fait de permettre la détection des sites d’activité de maladies, la mesure mono-exponentielle du T2 de l’eau par imagerie reste non-spécifique vis-à-vis des processus physiologiques à l’origine de l’augmentation du T2. Il est connu que la relaxation T2 du muscle squelettique n’est pas mono-exponentielle. Cela est interprété comme une conséquence de la compartimentation anatomique de l’eau tissulaire. Nous avons mis au point une méthode pour l’acquisition localisée de données CPMG. Cette technique permet l’acquisition des données dans des conditions nécessaires pour la réalisation de traitements multi-exponentiels précis. Ce travail nous a permis d’établir un modèle de compartimentation qui explique parfaitement la relaxation T2 dans le muscle. Il a fait l’objet d’un article publié dans le « Biophysical Journal » (Araujo, Fromes & Carlier 2014. New Insights on skeletal muscle tissue compartments revealed by T2 NMR relaxometry. (In press)). Les essais réalisés chez des sujets malades suggèrent un grand potentiel pour l’application de la méthode dans des études cliniques. La formation de la fibrose commence avec une accumulation excessive de tissu conjonctif intramusculaire (TCIM). Nous avons exploité la technique « Ultrashort Time-to-Echo » (UTE) pour essayer de détecter et caractériser le signal du TCIM. Dans une première étude, nous avons caractérisé in vivo une composante à T2 court (~500 µs) dans le muscle, et nous avons trouvé des indices qui suggèrent qu’elle représente le TCIM. Dans une deuxième étude, nous avons mis au point une méthodologie qui a permis d’imager cette composante à T2 court dans le muscle pour la première fois. / Current quantitative nuclear magnetic resonance (NMR) technics offer biomarkers that allow performing non-invasive longitudinal studies for the follow up of therapeutic trials in neuromuscular disorders (NMD). In contrast to fat degeneration, the mechanisms of inflammation/oedema/necrosis and fibrosis are characteristic signs of disease activity, which makes their quantification a promising source of crucial biomarkers for longitudinal studies. This thesis work consisted on the implementation of more precise quantitative NMR methods adapted to the clinical study of skeletal muscle (SKM) for : (i) detection and quantification of sites of disease activity by T2-mapping of muscle water ; (ii) investigation of the different pathophysiological mechanisms underlying T2 alterations ; and (iii) Detection and quantification of muscle fibrosis. We implemented two methods for T2 mapping of muscle water. The first one is based on a multi-spin-echo sequence du type CPMG. In this method the 1H-NMR signals from water and lipids are acquired simultaneously. The acquired data are fitted to a tri-exponential model, in which water and fat signals are separated by exploring the T2 difference between water and fat. This method allows extraction of muscle water T2-value in the presence of fat infiltration. The second method is based on a « partially spoiled steady state free precession » (pSSFP) sequence. In contrast to the first method, which demands a sophisticated post-treatment of images acquired at 17 different echo-times, with the pSSFP a T2-mapping is extracted from two 3D data sets. 3D acquisition is compatible with spectrally selective water excitation, which eliminates signal contribution from lipids. Both methods were validated experimentally on patients and healthy subjects. The results demonstrated their capacity to detect and quantify disease activity sites. Although it was shown to reveal disease activity, mono-exponential T2 of muscle water is non-specific to what concerns the mechanisms underlying its alterations. It has been long known that T2 relaxation in SKM tissue is multi-exponential. This is currently accepted to reveal anatomical compartmentation of myowater. We implemented a method for localized spectroscopic CPMG acquisition. CPMG data respect echo-time sampling and signal to noise ration limits for allowing robust multiexponential analysis. This work allowed us to establish a compartmentation model that perfectly explains the multi-exponential T2 relaxation observed in SKM tissue. This work was published in the « Biophysical Journal » (Araujo, Fromes & Carlier 2014. New Insights on skeletal muscle tissue compartments revealed by T2 NMR relaxometry. (In press)). Pilot studies performed in patients show promising results and suggest potential application of the method in clinical studies. Fibrosis starts with an excessive accumulation of intramuscular connective tissue (IMCT). We have explored the « Ultrashort time to echo » (UTE) method with the aim to detect and characterize the signal from IMCT. In a first study we characterized in vivo a short T2 component (~500 µs) in SKM, and we collected evidences suggesting that this component might reflect IMCT. Then we implemented a methodology that allowed imaging this short component in SKM tissue for the first time.
