Säg Bob! : Sex sångare om nedsjungning

Detta är en kvalitativ studie med kvantitativa inslag med syftet att undersöka sångares upplevelser avnedsjungning efter högintensiv sång. Till en början har specifikt designade sångövningar förmedlats till sex medverkande sångare om fyra kvinnor och två män under individuella sånglektioner. Deltagarna har i en variant av crossover design skattat sin dagliga rösthälsa i EASE- självskattningsformulär för friska röster. Testperioden om tre veckor följdes sedan av kvalitativa intervjuer. Resultat visar att även om sångares vardag är komplex, i och med inre och yttre påverkande faktorer, har nedsjungning en upplevd effekt i avseendet förbättrad rösthälsa efter högintensiv sång. Detta gäller särskilt de kvinnliga sångarna som beskriver att nedsjungning bidragit till en mer samlad röst med minskad heshet och svullnad. Vidare rapporteras om större lätthet inför att sjunga i lägre delen av bröstregistret, en mer egaliserad röst och en förbättrad tal- och magstödsteknik tillsammans med större lätthet inför påföljande sångpass. Nedsjungning ses också ha skapat ett andrum i vardagen där minskad stress, prestationskrav och resultatinriktning omvänts till en samlad känsla av avslut. Likväl är begrepp som nedsjungning, att sjunga ned, kyla ned, eller att stretcha rösten nya begrepp som rekommenderas att vidare studeras och förmedlas till elever och studenter, framför allt då föreliggande studies resultat visar tydliga attitydskillnader avseende rösthälsa och röstvård emellan de olika könen. / This is a study regarding singers' experiences of a vocal-cool down routine preceded with a heavy vocal load. The study is mainly based on a qualitative approach with quantitative elements. At first individual singing lessons where held, whereas specifically designed vocal-cool down exercises were taught to six participating singers; four women and two men. With a modification of a crossover design the singers in this study have estimated their daily voice health in the Evaluation of the Ability to Sing Easily survey (EASE). After a three week period of tests the singers were interviewed. The result of this study shows that even if singers' everyday lives are complex due to internal and external factors, vocal- cool down where perceived as an improvement of voice health. It is mainly the female singers' that describes an increased vocal well-being, with less hoarseness and swollen vocal folds after a heavy vocal load, followed by a vocal -cool down routine. Furthermore a greater sense of ease was reported in terms of transitions between registers in the singing voice, where fatigue in the surrounding muscles also decreased alongside with a positive impact on the speaking voice, reaching lower notes in chest register and extended diaphragm breathing. The vocal cool-down routine itself also gave an unexpected finding in terms of being contemplative and relaxing for the singers; creating a breathing space in everyday life where stress, performance anxiety and other concerns where easier to let go of, with an overall feeling of closure. Vocal cool down, cooling down the voice, vocal stretch, or warming down your voice are all relatively new concepts that are recommended to be studied further, spread and taught to students and pupils, - especially as the result in this study indicates clear differences in attitudes regarding voice health in-between the male and female genders.

Implementing Educational Innovations: The case of the Secondary School Curriculum Diversification Programme in Lesotho

Philosophiae Doctor - PhD / Between 1974 and 1982 the MOE introduced in two phases the diversification programme [SSCDP] which sought to establish practical subjects in the secondary school curriculum. This study examines the sustainability of implementation efforts beyond project expiry. It was hypothesised that SSCDP is not working as originally intended. The broad research problem was framed thus: What implementation response arises from an open-ended innovation policy? Subsidiary questions are: 1. How far have the policy-makers communicated the meaning of SSCDP and what factors account for mismatches between policy intentions and innovation practice? 2. What is the response of Project schools and what factors explain variation in response? 3. What is their significance for the sustainability of SSCDP? The analysis draws key concepts from the innovation literature on models and strategies of planned change; relationships in the implementation hierarchy; determinants of and orientations to the implementation process. Centred around qualitative research methods, the investigation utilises data from project documents, semi-structured interviews and from observations during school visits. Findings show an overall low level of implementation that varies among project schools. This is attributed to: Poor interpretation of SSCDP goals; Deficiencies in the implementation management; Idiosyncratic school behaviours. The study concludes that the 'practitioner-policy-maker' discrepancy is significant, hence the gap between policy intents and innovation practice. The gap is not regarded so much as an ultimate failure of the programme but as a necessary condition that allows for mutual adaptation between the innovation and its setting. This is reflected in the varied patterns of implementation response, classified as the: faithful; negotiators; selective adaptors; expansionists; and reductionist. As a policy-oriented study aiming at providing an 'improvement value', the findings lead to a proposal of improvements in the strategies of managing change in three areas: shifting focus from an adoption to an implementation perspective. Recognising implementation as a process dependent on a mutual linkage relationship among participants. Recognising schools as important bearers of change. These three are crucial factors in the implementation-sustainability relationship.

Protein arginine methyltransferase 5 (PRMT5) is an essential regulator of the cellular response to ionizing radiation and a therapeutic target to enhance radiation therapy for prostate cancer treatment

Prostate cancer is one of the most frequently diagnosed cancers and failure to manage localized disease contributes to the majority of deaths. Radiation therapy (RT) is a common treatment for localized prostate cancer and uses ionizing radiation (IR) to damage DNA. Although RT is potentially curative, tumors often recur and progress to terminal disease. The cellular response to RT is multidimensional. For example, cells respond to a single dose of IR by activating the DNA damage response (DDR) to repair the DNA. Targeting proteins involved in the DDR is an effective clinical strategy to sensitize cancer cells to RT. However, multiple radiation treatments, as in fractionated ionizing radiation (FIR), can promote neuroendocrine differentiation (NED). FIR-induced NED is an emerging resistance mechanism to RT and tumors that undergo NED are highly aggressive and remain incurable.<br><br> Currently, the only clinical approach that improves RT for prostate cancer treatment is androgen deprivation therapy (ADT). ADT blocks androgen receptor (AR) signaling which inhibits the repair of DNA damage. In 2017, my lab reported that targeting Protein arginine methyltransferase 5 (PRMT5) blocks AR protein expression. Therefore, targeting PRMT5 may also sensitize prostate cancer cells to RT via a novel mechanism of action.<br><br> This dissertation focuses on the role of PRMT5 in the cellular response to IR and the goal of my work is to validate PRMT5 as a therapeutic target to enhance RT for prostate cancer treatment. I demonstrate that PRMT5 has several roles in the cellular response to IR. Upon a single dose of IR, PRMT5 cooperates with pICln to function as a master epigenetic activator of DDR genes and efficiently repair IR-induced DNA damage. There is an assumption in the field that the methyltransferase activity and epigenetic function of PRMT5 is dependent on the cofactor MEP50. I demonstrate that PRMT5 can function independently of MEP50 and identify pICln as a novel epigenetic cofactor of PRMT5. During FIR, PRMT5, along with both cofactors MEP50 and pICln, are essential for initiation of NED, maintenance of NED, and cell survival. Targeting PRMT5 also sensitizes prostate cancer xenograft tumors in mice to RT, significantly reduces and delays tumor recurrence, and prolongs overall survival. Incredibly, while 100% of control mice died due to tumor burden, targeting PRMT5 effectively cured ~85% of mice from their xenograft tumor. Overall, this work provides strong evidence for PRMT5 as a therapeutic target and suggests that targeting PRMT5 during RT should be assessed clinically.<br>

The National Curriculum Statement (NCS) heralds the beginning of a new curriculum for Grades 10 to 12 in South Africa. Underpinned by the South African Constitution, and based on the tenets of Outcomes-based Education, the NCS seeks to provide contextually-relevant education for all South African learners, so that they are able to embrace inevitable change. Although the NCS highlights the importance of assessment, an analysis of the English Home Language (EHL) NCS reveals tensions between policy and practice. This study attempts to contextualise the role of relevant assessment for the 21st century. It then proceeds to engage in a historical evaluation of assessment within the NSC in terms of how assessment was conducted in the former Natal Education Department, a liberal education department within former apartheid South Africa. The conclusions drawn from the evaluation are used to provide recommendations to relieve the tensions identified within the EHL NSC. / Educational Studies / M.Ed.
