Cellules souches cancéreuses : ontologie et thérapies / Cancer stem cells : ontology and therapies

Une nouvelle théorie du cancer s’est récemment imposée dans la communauté scientifique. Selon cette dernière, les cancers se développeraient à partir d’une sous-population bien particulière de cellules cancéreuses, appelées « cellules souches cancéreuses » (CSC). Les partisans de la théorie des CSC soutiennent que les rechutes seraient causées par ces cellules, plus aptes à échapper aux thérapies classiques. En conséquence, ils soutiennent que l’élimination de toutes les CSC, dans un cancer donné, est nécessaire et suffisante pour guérir le patient. Dans cette thèse, je propose d’examiner cette stratégie thérapeutique de ciblage des CSC et je montre que sa capacité à guérir les cancers dépend de la façon dont on envisage la nature de la propriété souche. En effet, les cellules souches cancéreuses sont définies par la possession de la propriété souche, c’est-à-dire par leur capacité à s’auto-renouveler et à se différencier. Cependant, cette propriété elle-même reste obscure quant à sa nature. S’agit-il d’une propriété catégorique ou d’une disposition ? Une cellule non-souche (cancéreuse ou non) peut-elle acquérir la propriété souche et sous quelle condition ? En me basant sur une analyse de la littérature scientifique, je montre que quatre conceptions distinctes de la nature de la propriété souche sont aujourd’hui possibles et que, si la théorie des CSC est vraie, déterminer la nature exacte de la propriété souche est capital pour le traitement des cancers. / A new theory of cancer has recently gained importance in the scientific community. According to this theory, cancers develop from a particular sub-population of cancer cells, named “cancer stem cells” (CSCs). The proponents of the CSC theory argue that relapses are caused by CSCs because they escape classical therapies. Consequently, they claim that eliminating all the CSCs of a given cancer is a necessary and sufficient condition to cure the patient. In this dissertation, I scrutinize this therapeutic strategy and I argue that its ability to cure cancers will depend on our understanding of the nature of stemness. Indeed, cancer stem cells are characterized by this property, that is, the capacity to self-renew and to differentiate. However, the nature of stemness is rather obscure. Is it a categorical property or a disposition? Can a non-stem cell (cancerous or not) acquire stemness, and under which conditions? On the basis of analysis survey of the scientific literature, I distinguish four possible concepts of the nature of stemness. I contend that if the CSC theory is true, determining the exact nature of stemness is essential for cancers treatments.

A fundamentação moral do direito na filosofia de Kant

Esta tese tem o objetivo de investigar o estatuto dos princípios fundamentais do direito, apresentados por Kant na Doutrina do direito, parte constitutiva da obra Metafísica dos costumes, a fim de mostrar como Kant fundamenta a sua teoria do direito racional. A questão fundamental no que concerne à teoria do direito de Kant, e que há muitos anos suscita o debate entre os intérpretes, diz respeito à possibilidade ou não do direito encontrar os fundamentos de seus conceitos e princípios fundamentais na teoria moral elaborada por ele na Fundamentação da metafísica dos costumes e na Crítica da razão prática. Recentemente pode-se observar um crescente número de estudiosos que afirmam a independência da Doutrina do direito da teoria moral kantiana e, consequentemente, a independência dos princípios a priori do direito do imperativo moral. Contrariamente a essa posição, esta tese procura mostrar que Kant fundamenta o direito em sua teoria moral - o direito pressupõe a teoria moral e seu princípio fundamental, o imperativo moral, - e que essa é a única maneira de interpretarmos a Doutrina do direito se não quisermos fazer afirmações que entrem em contradição com o pensamento do próprio autor. A tese que afirma a fundamentação moral do direito se baseia em duas afirmações, que encontram respaldo nos textos de Kant. São elas: a) as leis jurídicas são uma espécie de leis morais, cujo princípio último é o imperativo categórico – os princípios a priori do direito derivam do princípio supremo da moral; b) o uso da coerção externa para obrigar outrem a cumprir um dever jurídico é moralmente justificável, o que significa que esse tipo de constrangimento imposto ao arbítrio é autorizado por uma lei moral. A tese tem, enfim, o objetivo de reconstruir os argumentos de Kant para mostrar que buscar na sua filosofia moral os fundamentos da teoria do direito está totalmente de acordo com seus textos e com o seu pensamento sistemático. / This thesis aims to investigate the nature of fundamental principles of Right, presented by Kant in the Doctrine of Right, a constituent part of the work Metaphysic of Morals, in order to show how Kant grounded his theory of rational Right. The fundamental question regarding the Kant‟s theory of Right, and that for many years raises debate among interpreters, concerns the possibility or not of the Right to find the foundations of its concepts and principles in moral theory elaborated by him in the Groundwork the Metaphysics of Morals and Critique of Practical Reason. Recently one can observe a growing number of scholars who assert the independence of the Doctrine of Right of Kantian moral theory and accordingly the independence of a priori principles of Right of moral imperative. Contrary to this position, this thesis aims to show that Kant bases the Right on his moral theory - the Right presuppose the moral theory and its fundamental principle, the moral imperative - and that is the only way to interpret the doctrine of Right if not want to make statements that come into conflict with the author's own thought. The thesis argues that the moral foundation of Right rests on two assertions that are supported in the writings of Kant. They are: a) juridical laws are a kind of moral law whose ultimate principle is the Categorical imperative - the a priori principles of Right derived from the supreme principle of morality; b) the use of external coercion to compel another to fulfill a duty legal is morally justifiable, meaning that this type of constraint imposed on the choice is authorized by a moral law. In short, the thesis has the objective of reconstructing Kant's arguments to show that find in his moral philosophy the foundations of the theory of Right is fully consistent with his writings and his systematic thinking.

In my diploma thesis entitled Implementing the Gold Rule of Morality in Crisis Planning for Population Protection, I will try to draw on the ideas and conclusions of my bachelor thesis, in which I investigated whether members of the Integrated System Can use the golden rule of morality in an emergency. I have come to the conclusion that the members of the individual components of the integrated rescue system are guided by the valid legislation and orders of the commander of the intervention. Nevertheless, they would welcome in certain situations the possibility of deciding according to the golden rule of morality, which briefly tells us: "I do not want to do the other to you." Translated into situations in the event of an extraordinary event may mean deciding in tedious situations so, as if you were the Rescued and not the savior. The aim of my thesis is to find out whether it is possible to implement this idea in the crisis plans themselves and, above all, whether the intervening members of the integrated rescue system themselves would use this rule in certain situations. The association of two seemingly incompatible worlds seems at first glance absurd, but research and interviews with individual members of the integrated system have convinced me that the philosophical questions and the name of Imanuel Kant can very closely mingle with the world of extraordinary events and the salvation of Human lives, health and property of the population. I would very much like to work with this bachelor thesis together with my previous bachelor thesis, tried to create a unified whole as an idea that can motivate us to think whether it is necessary to change the current state of legislation or whether the legislation itself is set as a form of golden rule of morality .

In this works Kant introduced the respect as a practical feeling that is produced by the consciousness of moral law, the only one able to show practical freedom as autonomy of the will. Beside the respect Kant presents another practical feeling, namely, the humiliation. Both concepts operate as two forces of the spirits that are exercised by the will face the law. Multiple Imputation for Two-Level Hierarchical Models with Categorical Variables and Missing at Random Data

abstract: Accurate data analysis and interpretation of results may be influenced by many potential factors. The factors of interest in the current work are the chosen analysis model(s), the presence of missing data, and the type(s) of data collected. If analysis models are used which a) do not accurately capture the structure of relationships in the data such as clustered/hierarchical data, b) do not allow or control for missing values present in the data, or c) do not accurately compensate for different data types such as categorical data, then the assumptions associated with the model have not been met and the results of the analysis may be inaccurate. In the presence of clustered/nested data, hierarchical linear modeling or multilevel modeling (MLM; Raudenbush & Bryk, 2002) has the ability to predict outcomes for each level of analysis and across multiple levels (accounting for relationships between levels) providing a significant advantage over single-level analyses. When multilevel data contain missingness, multilevel multiple imputation (MLMI) techniques may be used to model both the missingness and the clustered nature of the data. With categorical multilevel data with missingness, categorical MLMI must be used. Two such routines for MLMI with continuous and categorical data were explored with missing at random (MAR) data: a formal Bayesian imputation and analysis routine in JAGS (R/JAGS) and a common MLM procedure of imputation via Bayesian estimation in BLImP with frequentist analysis of the multilevel model in Mplus (BLImP/Mplus). Manipulated variables included interclass correlations, number of clusters, and the rate of missingness. Results showed that with continuous data, R/JAGS returned more accurate parameter estimates than BLImP/Mplus for almost all parameters of interest across levels of the manipulated variables. Both R/JAGS and BLImP/Mplus encountered convergence issues and returned inaccurate parameter estimates when imputing and analyzing dichotomous data. Follow-up studies showed that JAGS and BLImP returned similar imputed datasets but the choice of analysis software for MLM impacted the recovery of accurate parameter estimates. The purpose of the study is to understand, from interviews with four teachers, how they relate to students with or in reading and writing difficulties. Furthermore, the purpose is to investigate whether the teachers in the study regard students as either with reading or writing difficulties or in reading and writing difficulties. The transcribed interview answers are interpreted and analyzed based on the categories of relational and categorical approaches. There are teachers who carry on both approaches. Teachers 'approaches in these regards have consequences for teachers' practical organization of teaching. A relational approach tends to be reflected in if teachers prefer to include students in reading and writing difficulties in regular teaching, while teachers with a more categorical approach to students with reading and writing difficulties tend to advocate that these students benefit best from alternative teaching methods that involve that the students are excluded from the rest of the class and the regular teaching. From the present study it appears that two teachers assume a relational perspective, and the other two carry both a relational and a categorical perspective at the same time, one teacher leans more on one perspective with elements of the other and vice versa. The teachers in the study mention the importance of social interaction at school and how it benefits learning. Therefore, this study's analysis is rooted in the sociocultural learning theory. Overall, collected data suggest that teachers and special educators and specialist teachers need to collaborate more within the classroom framework to create an inclusive environment.

Autour du lambda-calcul avec constructeurs / On the lambda calculus with constructors

Le lambda calcul avec constructeurs (de Arbiser, Miquel et Rios) est une extension du lambda calcul avec un mécanisme de filtrage. Le filtrage à la ML y est décomposé en deux étapes: une analyse de cas sur des constantes (telle l'instruction «case» de Pascal), et une commutation de l'application avec la construction de filtrage. Cette règle de commutation entre deux constructions de natures différentes induit une géométrie de calcul surprenante, a priori incompatible avec les intuitions habituelles de typage. Cependant il a été montré que ce calcul est confluent, et vérifie la propriété de séparation (à la Böhm). Cette thèse propose un système de types du polymorphique pour ce calcul, et décrit ensuite un modèle de réalisabilité, qui adapte les candidats de réductibilité de Girard au lambda calcul avec constructeurs. La normalisation forte du calcul typé et l'absence d'erreur de filtrage lors de l'évaluation en découlent immédiatement. Nous nous intéressons ensuite à la sémantique du lambda calcul avec constructeurs non typé. Une notion générique de modèle catégorique pour ce calcul est définie, puis un modèle particulier (le modèle syntaxique dans la catégorie des PERs) est construit. Nous en déduisons un résultat de complétude. Enfin, nous proposons une traduction CPS du lambda calcul avec constructeurs dans le lambda calcul simplement typé avec paires. Le lambda calcul avec constructeurs peut ainsi être simulé dans un calcul bien connu, et cette traduction nous permet aussi de transformer tout modèle par continuation en modèle du lambda calcul avec constructeurs. Une équation catégorique caractéristique de ces modèles apparait alors, qui permet de construire des modèles non syntaxiques (dans les domaines) de Scott du lambda calcul avec constructeurs. / The lambda calculus with constructors was introduced by Arbiser, Miquel and Rios in the early 2000's as an extension of lambda calculus with pattern matching features. It decomposes the pattern matching à la ML into a case-analysis on constant constructors (in the spirit of the case instruction in Pascal), and a commutation rule between case construction and application. This commutation rule between two different kinds of constructions designs a surprising computational behaviour, a priori} not compatible with usual typing intuitions. However the whole calculus was proved confluent, and it enjoys the separation property (a version of Böhm's lemma).In this thesis we propose a polymorphic type system for this calculus, and we develop a realisability model, based on Girard's reducibility candidates. This leads to a strong normalisation result for the typed calculus, and guaranties that the type system prevents match failure. Next we focus on semantics for the untyped calculus. We first define a generic notion of models for the lambda calculus with constructors in Cartesian closed categories. We then establish the syntactic model in the category of PERs, and deduce a completeness result from it.Finally, we consider a translation of the lambda calculus with constructors into the pure lambda lambda calculus relying on continuation passing style techniques. This enables the simulation of the lambda calculus with constructors by a well known calculus, and provides a transformation of every continuation model into a model of the lambda calculus with constructors. Thereby a categorical equation characteristic of these models appears, which enables the construction of non syntactic models in Scott's domains.
