Weight Estimation through Frequency Analysis

The weight of a heavy duty vehicle plays an important role when dealing with different control systems. Examples of control units in a truck that need this parameter are the ones used to control the brakes, the engine and the gearbox. An accurate estimation of the weight leads not only to a more fuel efficient and safer transport, but also assures the driver that current law limits are not exceeded. The weight can be estimated with pretty good accuracy if the truck is equipped with air suspension. In trucks that lack this type of suspension other methods are used to estimate the weight. At present these methods are inaccurate. In this thesis a new method where the weight is to be estimated through frequency analysis of the truck's driveline is developed and evaluated.

Molecular-cytogenetic analysis of repetitive sequences in genomes of Beta species and hybrids / Molekular-cytogenetische Analyse der repetitiven Sequenzen in Genomen von Beta Arten und Hybriden

The elucidation of the composition and organization of genomes of higher plants is a fundamental problem of modern molecular biology. The genus Beta containing 14 species assigned to the sections Beta, Corollinae, Nanae and Procumbentes provides a suitable system for the comparative study of the nuclear genomes. Sugar beet Beta vulgaris has a genome size of 758 Mbp DNA with estimated 63 % repetitive sequences and the number of chromosomes n=9. The wild beet Beta procumbens is an important natural pool of resistance against pests and tolerance to unfavorable growth conditions. The subject of this research was the isolation and description of new repetitive DNA families from genomes of this Beta species. This work presents the molecular investigation and cytogenetic characterization by high-resolution multicolor fluorescent in situ hybridization (FISH) of the satellite and dispersed repetitive sequences in wild and cultivated beet species and in their hybrids. New repetitive sequences were isolated from the B. procumbens genome. The AluI restriction satellite repeats pAp11 are 229-246 bp long and form subfamilies. The satellite is amplified in the section Procumbentes, but also found in distantly related section Beta. Thus, pAp11 is probably an ancient component of Beta genomes. It could be the ancestor of the satellite subfamily pEV4 in B. vulgaris based on sequence analysis, Southern hybridization and comparative FISH. pAp11 was found at centromeric and a few intercalary sites in B. procumbens and formed intercalary blocks on B. vulgaris chromosomes where it co-localized with pEV4. These remarkable differences in the chromosomal position of pAp11 between Procumbentes and Beta species indicate that both satellites were likely involved in the expansion or rearrangement of the intercalary heterochromatin of B. vulgaris. Other two sequence families characterized on molecular, genomic and chromosomal levels are the non-homologous repeats pAp4 and pAp22, 1354 and 582 bp long. They have a dispersed organization in the genome and are widely scattered along B. procumbens chromosomes. pAp4 and pAp22 are specific for the section Procumbentes and can be used as DNA probes to discriminate parental genomes in interspecific hybrids. High-resolution FISH on meiotic chromosomes showed that the both sequences mostly co-localize. The PCR analysis of their flanking regions revealed that pAp22 is a part of a Long Terminal Repeat (LTR) of an Athila-like env-class retrotransposon. This is the first indication that the retrovirus-like DNA elements exist in Beta. An ancient family of subtelomeric satellite DNA pAv34 was isolated from all four sections of the genus Beta and from spinach, a related Chenopodiaceae. Five clones were analyzed from each of the five species. The genomic organization and species distribution of the satellites were studied by sequencing and Southern hybridization. The repeating units in all families are 344-362 bp long and share 46.2-98.8 % similarity. Each monomer consists of two subunits SU1 and SU2 of 165-184 bp. The maximum likelihood and neighbor joining analyses of the 25 subtelomeric satellite monomers and their subunits indicated, that the duplication leading to the emergence of the 360 bp satellite should have occurred early in the phylogeny. The two directions of diversification are the clustering of satellites in two groups of subunits SU1 and SU2 and the arrangement of satellite repeats in section-specific groups. The comparative chromosomal localization of the telomeric repeat, pAv34 and rDNA was investigated by multicolor FISH. B. vulgaris chromosome termini showed unique physical organization of telomeric repeat and the subtelomeric satellite, as studied by high-resolution FISH on extended DNA fibers. The estimated length of the telomeric array was 0.55 - 62.65 kb, the length of pAv34 was 5.0-125.25 kb, the spacer between these sequences spanned 1.0-16.60 kb. Eight various classes of repeats were used to characterize the minichromosomes of the sugar beet fragment addition lines PRO1 and PAT2 by comparative multi-color FISH. The study allowed to propose a schematic pattern of repetitive DNA organization on the PRO1 and PAT2 minichromosomes. PRO1 has an acrocentric minichromosome, while PAT2 possesses a metacentric or submetacentric chromosome fragment. The functional integrity of the fragment addition line centromeres was confirmed by an immunostaining localization of the proteins specific to the active kinetochore. The serine 10-phosphorylated histone H3 was detected in pericentromeric regions of the PRO1 chromosomes. The microtubuli attachment sites were visualized as parts of kinetochore complexes.

Identitet, makt och matematiska begrepp i åtgärdsprogram : Diskursanalys av elevens behov och åtgärd i matematik

I vår studie intar vi en hermeneutisk ansats och genomför en textanalys. I vår studie tolkar, analyserar och kategoriserar vi innehållet i åtgärdsprogrammets behov och åtgärd. Utifrån diskurser konstruerar vi en ram som hjälper oss att tolka formuleringar i åtgärdsprogram. Syftet med denna studie är att undersöka hur identitet, makt och matematiska begrepp synliggörs i åtgärdsprogram. I studien undersöker vi i vilken omfattning begreppen ovan förekommer i behov och åtgärder för årskurs 6 och 7. Vi utgår från ett diskursivt perspektiv mot makt och identitet. Vår studie visar att koppling mellan behov och åtgärd saknas i vissa åtgärdsprogram. Tillskriva identitet har den största andelen för åtgärder. Matematiska begrepp har störst andel för upptagna behov. Makt (makt genom kunskap och titel) har lägst förekomst i upprättade åtgärder. Det är varje skolas diskurs och dess skolpersonal som avgör på vilket sätt och i vilket forum som arbetet kring elevens behov fungerar. På samma sätt är den skillnad som existerar mellan olika upprättade åtgärdsprogram ett uttryck för den kultur eller diskurs som råder just nu, lokalt på skolan. Detta är avgörande om åtgärdsprogrammet används som ett fungerande verktyg. / In our study we take a hermeneutical approach and interpret text. In our study we interpret, analyse, and categorize the content of the IEP´s needs and arrangements. Based on discourses we construct a framework that helps us interpret phrasings in IEPs. The purpose of this study is to investigate how identity, power and mathematical concepts are made visible in the text of IEPs. In this study, we investigate to what extent the concepts above occur in the needs and arrangements for grades 6 and 7. We start from a discursive perspective on power and identity. Our study shows that the connection between needs and arrangements is missing in some IEPs. Attributing identity has the largest share of arrangements. Mathematical concepts have the greatest proportion of occupied needs. Power (power through knowledge and title) has the lowest occurrence of written arrangements. It is the discourse of every school and its school personal that decides in what direction and in what context the work toward the needs of the student. In the same way is the difference that exist between different IEPs an expressian in the culture that is dominant at schools. This is crusial if the IEPs is working as a functioning tool.

MonoGenjin : En praktisk undersökning av 2D spelmotors- och spelutveckling

Spelindustrin är gigantisk och kontinuerligt växande, där detta inkluderar spelutveckling samt spelande av spel. Detta medför att en betydande mängd akademiker i sina yrkesliv kommer att beröra ämnesområdet spelutveckling. Trots detta faktum finns det en avsaknad av akademisk litteratur som behandlar ämnesområdet spelutveckling, specifikt med ett IS-perspektiv. För att fylla avsaknaden ämnar denna undersökning utöka det specifika konceptet inom spelutveckling som är “skapande av en 2D-spelmotor med tillhörande spel”. Att spela innebär hedonistiska upplevelser. Gäller denna hedonism också vid spelutveckling? Vilka utmaningar och vilken problematik uppstår vid utveckling av en spelmotor? För att utreda detta används en kvalitativ undersökning med en induktiv metodik via forskning genom design (eng. Research through Design, RtD) tillsammans med kollaborativ autoetnografi (Collaborative Autoethnography, CAE). Detta skapar ett unikt perspektiv som används för att producera välmotiverad vägledning för introduktion till ämnesområdet. Datainsamling utgörs av loggbok och dagbok där dessa används för att reflektera kring problem, utmaningar, framgångsfaktorer samt hedonistiska upplevelser inom processen. Undersökningens bakomliggande teori är baserat på Hedonic-motivation system adoption model (HMSAM) samt kriterier för utvärdering av ett lyckat RtD-projekt. Resultaten visar att spelutveckling är en komplicerad process, där egenskaper av spelutveckling identifierats och som bildat riktlinjer som förbereder akademiker inför framtida forskning och arbete. / The gaming industry is gigantic and continuously growing, which includes game development and gaming. This means that a significant number of academics will touch on the subject area of game development during their professional lives. Despite this fact there is a lack of academic literature which deals with the subject area of game development, specifically with an IS perspective. To fill the gap, this study aims to expand the specific concept in game development that is “creation of a 2D game engine with an associated game”. Gaming involves hedonistic experiences. Is this also associated with game development? What challenges and problems arise when developing a game engine? Answering this involves a qualitative study with an inductive methodology via Research through Design (RtD) together with Collaborative Autoethnography (CAE). This creates a unique perspective that is used to produce well-motivated guidance for introduction to the subject area. Data collection consists of a journal and diary where these are used to reflect on problems, challenges, success factors and hedonistic experiences within the process. The study’s underlying theory is based on the Hedonic-motivation system adoption model (HMSAM) and criterias for evaluating successful RtD projects. The results show that game development is a complicated process, where characteristics of game development have been identified which developed guidelines that prepare academics for future research and work.

Denna artikel har som ändamål att undersöka hur ett företag kan förmedla sina värderingar om hållbarhet inte bara genom ord och texter utan också genom grafisk design och visuell kommunikation. Artikeln djupdyker i grafiska profiler och identiteter och undersöker hur man med hjälp av dem kan visuellt uttrycka ett företags fokus på och värderingar kring hållbara tillverkning, förpackning samt hållbarhet för företaget och dess anställda. Artikeln bidrar med forskning som kan hjälpa designers att kommunicera specifika värderingar, i denna kontext hållbarhet, genom grafik samt visuella element, och dessutom hur man potentiellt kan förhålla sig till ämnen som “greenwashing”. Resultaten baseras på relevant litteratur i området, information samlad ifrån metoder som fokusgruppdiskussion och internetenkät, samt en visuell gestaltning i form av en grafisk profil för ett mock-up företag. / Awareness of the environment and achieving a more sustainable way of life has increased among the general public and companies for several years. The purpose of this article is to investigate how a company might convey its values ​​of sustainability not only through words and texts but also through graphic design and visual communication. The article delves into graphic profiles and brand identities and examines how they can be used to visually express a company's focus on, and values ​​around sustainable manufacturing, packaging, and sustainability for the company and its employees. The article contributes research that could help designers, in the context of sustainability, deliver specific messages and values through graphics and visual elements, as well as how one can potentially approach topics such as "greenwashing". The results are based on relevant literature in the area, information gathered from methods such as focus groups and surveys, as well as a practical design in the form of a graphic profile for a mock-up company.

Computational mapping of regulatory domains of human genes

Ljudski genom sadrži milijune regulatornih elemenata - enhancera - koji kvantitativno reguliraju ekspresiju gena. Unatoč ogromnom napretku u razumijevanju načina na koji enhanceri reguliraju ekspresiju gena, području još uvijek nedostaje pristup koji je sustavan, integrativan i dostupan za otkrivanje i dokumentiranje cis-regulatornih odnosa u cijelom genomu. Razvili smo novu računalnu metodu - reg2gene - koja modelira i integrira aktivnost enhancera~ekspresije gena. reg2gene sastoji se od tri glavna koraka: 1) kvantifikacija podataka, 2) modeliranje podataka i procjena značaja, i 3) integracija podataka prikupljenih u reg2gene R paketu. Kao rezultat toga, identificirali smo dva skupa enhancer-gen interakcija (EGA): fleksibilni skup od ~ 230K EGA (flexibleC) i strogi skup od ~ 60K EGA (stringentC). Utvrdili smo velike razlike u prethodno objavljenim računalnim modelima enhancer-gen interakcija; uglavnom u lokaciji, broju i svojstvima definiranih enhancera i EGA. Izveli smo detaljno mjerenje performansi sedam skupova računalno modeliranih EGA-a, ali smo pokazali da se niti jedan od trenutno dostupnih skupova referentnih podataka ne može koristiti kao referentni skup podataka "zlatnI standard". Definirali smo dodatni referentni skup pozitivnih i negativnih EGA -a pomoću kojih smo pokazali da stringentC ima najveću pozitivnu prediktivnu vrijednost (PPV). Pokazali smo potencijal EGA-a za identifikaciju genskih meta nekodirajucih SNP-ova. Proveli smo funkcionalnu analizu kako bismo otkrili nove genske mete, pleiotropiju enhancera i mehanizme aktivnosti enhancera. Ovaj rad poboljšava naše razumijevanje regulacije ekspresije gena posredovane enhancerima. / Das menschliche Genom enthält Millionen von regulatorischen Elementen - Enhancern -, die die Genexpression quantitativ regulieren. Trotz des enormen Fortschritts beim Verständnis, wie Enhancer die Genexpression steuern, fehlt es in diesem Bereich immer noch an einem systematischen, integrativen und zugänglichen Ansatz zur Entdeckung und Dokumentation von cis-regulatorischen Beziehungen im gesamten Genom. Wir haben eine neuartige Methode - reg2gene - entwickelt, die Genexpression~Enhancer-Aktivität modelliert und integriert. reg2gene besteht aus drei Hauptschritten: 1) Datenquantifizierung, 2) Datenmodellierung und Signifikanzbewertung und 3) Datenintegration, die in dem R-Paket reg2gene zusammengefasst sind. Als Ergebnis haben wir zwei Sätze von Enhancer-Gen-Assoziationen (EGAs) identifiziert: den flexiblen Satz von ~230K EGAs (flexibleC) und den stringenten Satz von ~60K EGAs (stringentC). Wir haben große Unterschiede zwischen den bisher veröffentlichten Berechnungsmodellen für Enhancer-Gene-Assoziationen festgestellt, vor allem in Bezug auf die Lage, die Anzahl und die Eigenschaften der definierten Enhancer-Regionen und EGAs. Wir führten ein detailliertes Benchmarking von sieben Sets von rechnerisch modellierten EGAs durch, zeigten jedoch, dass keiner der derzeit verfügbaren Benchmark-Datensätze als "goldener Standard" verwendet werden kann. Wir definierten einen zusätzlichen Benchmark-Datensatz mit positiven und negativen EGAs, mit dem wir zeigten, dass das stringentC-Modell den höchsten positiven Vorhersagewert (PPV) hatte. Wir haben das Potenzial von EGAs zur Identifizierung von Genzielen von nicht-kodierenden SNP-Gene-Assoziationen nachgewiesen. Schließlich führten wir eine funktionelle Analyse durch, um neue Genziele, Enhancer-Pleiotropie und Mechanismen der Enhancer-Aktivität zu ermitteln. Insgesamt bringt diese Arbeit unser Verständnis der durch Enhancer vermittelten Regulierung der Genexpression in Gesundheit und Krankheit voran. / Human genome contains millions of regulatory elements - enhancers - that quantitatively regulate gene expression. Multiple experimental and computational approaches were developed to associate enhancers with their gene targets. Despite the tremendous progress in understanding how enhancers tune gene expression, the field still lacks an approach that is systematic, integrative and accessible for discovering and documenting cis-regulatory relationships across the genome. We developed a novel computational approach - reg2gene- that models and integrates gene expression ~ enhancer activity. reg2gene consists of three main steps: 1) data quantification, 2) data modelling and significance assessment, and 3) data integration gathered in the reg2gene R package. As a result we identified two sets of enhancer-gene associations (EGAs): the flexible set of ~230K EGAs (flexibleC), and the stringent set of ~60K EGAs (stringentC). We identified major differences across previously published computational models of enhancer-gene associations; mostly in the location, number and properties of defined enhancer regions and EGAs. We performed detailed benchmarking of seven sets of computationally modelled EGAs, but showed that none of the currently available benchmark datasets could be used as a “golden-standard” benchmark dataset. To account for that observation, we defined an additional benchmark set of positive and negative EGAs with which we showed that the stringentC model had the highest positive predictive value (PPV) across all analyzed computational models. We reviewed the influence of EGA sets on the functional analysis of risk SNPs and demonstrated the potential of EGAs to identify gene targets of non-coding SNP-gene associations. Lastly, we performed a functional analysis to detect novel gene targets, enhancer pleiotropy, and mechanisms of enhancer activity. Altogether, this work advances our understanding of enhancer-mediated gene expression regulation in health and disease.

Detta arbete visualiserar värdeflöde av solceller vid tre olika typer av solcellsinstallationer följt av förslag till förbättring gällande ett växande installationsföretag i Sverige. Genom värdeflödesanalys kan vi se tydliga samband mellan snabb tillväxt och logistiska problem, där spårbarhet, lokaler och personal är viktiga faktorer som behöv bindas samman för att skapa ett så optimalt flöde som möjligt. Resultatet är delade i två grupper. En grupp består av visualisering och information kring hur värdeflödet ser ut och den andra av vilka förbättringsmöjligheter företaget har för sin framtida verksamhet. Vi kan också se att företaget har stor potential att öka sin lönsamhet genom att via affärssystem utveckla sin logistikhantering samt sammankoppla företagets olika instanser för att nå en mer effektiv installationsprocess. / This report visualizes the value stream of solar cells at three different types of installations, followed by proposal of improvement at a growing installation company in Sweden. Through value stream mapping, we see a clear connection between a fast growing rate and logistical problems, where traceability, facilities and staff are key factors, which needs to be connected to create as optimal flow as possible. The results are divided into two groups. The first consists of visualization and information of the current value stream and the second of what kind of improvement we suggest the company takes into consider to go even further. We can also see that the company has great potential to increase its profitability by implementing ERP to its logistical department and to connect the other departments to reach a higher level of efficiency.
