Desenvolvimento e avaliação de minicomprimidos de indapamida de liberação prolongada / Development and evaluation of indapamide controlled release minitablets

A hipertensão arterial é a principal causa da incidência de doenças cardiovasculares no mundo. Os diuréticos anti-hipertensivos como a indapamida são muito utilizados para o tratamento da hipertensão arterial, sendo a formulação de liberação prolongada muito eficaz e bem tolerada para o uso em pacientes idosos. Sistemas multiparticulados de formas farmacêuticas sólidas orais de liberação prolongada apresentam uma série de vantagens tecnológicas e biofarmacotécnicas em relação aos sistemas monolíticos.Assim, o presente trabalho teve como objetivo desenvolver formas farmacêutica sólidas multiparticuladas de indapamida com controle de liberação através de sistema de revestimento utilizando polivinilacetato e sistema matricial de hipromelose. Para avaliação das diferentes formulações de minicomprimidos de indapamida de liberação prolongada, foram empregados os aparatos 1 (cesta) e 3 (Bio-Dis) para, deste modo, identificar aquelas cuja liberação do fármaco estivesse mais próxima do produto referência (Natrilix SR® 1,5mg). Ao final, foi realizada uma busca de patentes relacionadas ao sistema multiparticulado de liberação prolongada de indapamida, para avaliar a possibilidade de uma patente para a formulação proposta. / Hypertension is the main cause of cardiovascular disease around the world. Diuretic antihypertensives, such as indapamide, are widely used for treating hypertension, and, with regards to elderly patients, its extended release formulation is the most effective and well tolerated. Multiparticulate systems of solid oral dosage forms for extended release present a series of technological and biopharmaceutical benefits, when compared to monolithic systems. Thus, the purpose of this study was to develop a multiparticulate solid form of indapamide with controlled release by using a polyvinyl acetate coating system and a hypromelose matrix. For an evaluation of different formulations of indapamide extended release mini-tablets, a 1 (basket) and 3 (Bio-Dis) apparatus was used to identify those with a drug release rate closest to the reference product, Natrilix® SR 1.5mg. Finally, multiparticulate indapamide extended release patents were researched, in order to evaluate the possibility of obtaining a patent for the proposed formulation.

Contaminação microbiana em medicamentos fitoterápicos sob a forma sólida / Microbial quality control of phytomedicines oral solid dosage forms

Foram analisados 84 lotes de especialidades fitoterápicas de uso oral, na forma de cápsula, comprimido e pó, contendo 17 drogas vegetais, produzidas por 20 fabricantes e comercializadas em farmácias, drogarias, lojas de produtos naturais e supermercados da cidade de são Paulo. O confronto da apresentação comercial das mesmas em relação à legislação nacional, indicou que a minoria (15/84) atendeu a tais exigências, além de ter demonstrado total falta de padronização de qualidade pelos produtores, inclusive com ausência de definição e conceituação, enquadrando-as indistintamente como medicamentos alopáticos, homeopáticos, produtos dietéticos ou alimentos. Efetuou-se a quantificação de bactérias aeróbias mesófilas (forma vegetativa e esporulada) e de fungos (bolores e leveduras), através da técnica de tubos múltiplos, bem como a pesquisa de Escherichia coli e Salmonella sp. pela metodologia oficial internacional. A contaminação fúngica, predominantemente de bolores, foi inferior em relação à bacteriana aeróbia mesófila, pois o máximo detectado para aquela foi de 4,6 X 105, enquanto que para esta 5,1 X 109/g. A incidência fúngica com carga maior ou igual a 102/g foi de 34,5%, contra 53,6% da carga bacteriana vegetativa com 104/g ou mais e 45,2% da bacteriana esporulada, com o mesmo nível. A determinação do Número Mais Provável de Escherichia coli indicou incidência de 6,0% com a carga maior igual a 1,5 X 102/g. Houve estreita relação entre a carga bacteriana vegetativa total e o número de esporos, assim como a presença de patogênicos específicos em produtos altamente contaminados. Amostras sob a forma de pó foram as mais afetadas, seguidas de cápsulas e comprimidos. Pelo confronto dos dados encontrados, com os diversos padrões microbianos aplicados para medicamentos não estéreis de uso oral, para insumos e produtos fitoterápicos, o índice de aprovação variou de 29,8 a 94,0%. Mesmo frente ao padrão da FIP, a rejeição especialidades fitoterápicas analisadas foi da ordem de 59,5%. Em vista da qualidade sanitária de muitos destes produtos ser imprópria para o consumo, e após análise crítica dos vários padrões microbianos, discutiu-se a necessidade de implantação de especificações nacionais, ainda que a caráter provisório, sugerindo-se limites de tolerância para medicamentos fitoterápicos de uso oral. / 84 samples of phytotherapic products of oral solid dosage forms (capsules, tablets and powders) containing 17 different crude drugs and produced by 20 laboratories, marketed in drugstores, herborist\'s and supermarkets at são Paulo-Brazil were analysed with regard to legal marketing requirements, only 15 samples were in accordance. A complete lack of quality standard was attributed to the manufacturers, including indefinition as to the classification of products among allopathic or homeopathic pharmaceuticals or foods or dietetics. The total aerobic bacterial (vegetative and sporulated forms) and fungal (moulds and yeasts) contents were determined by multiple tube technique; moreover tests were carried out for Escherichia coli and Salmonella sp. by official international methods. The fungal contamination was predominantly of moulds and lower than the aerobic bacterial one. The maximum counts were 4,6 X 105 and 5,1 X 109/g, respectively, for fungi and bacteria. The frequency of samples with or more than 102/g fungi/g was 34,5% that for vegetative and sporulated bacterial counts with or more than 104/g was respectively, 53,6 and 45,2%. The incidence of Escherichia coli was about 6,0% with the highest level at 1,5 X 102/g. There was a tight relation between the vegetative and sporulated bacterial counts as well as the presence of specific pathogenic microorganisms in highly contaminated samples. The highest contamination occured in powders followed by capsules and tablets. Comparing our experimental data with the corresponding microbial standards for oral non sterile pharmaceuticals, crude drugs and phytotherapic products, the index of approval varied between 29,8 and 94,0%. Even by FIP\'s standards 59,5% of the phytotherapic specialties analysed had to be rejected. As many of these products showed innadequate sanitary quality for consumption, coupled to a critical analysis of the different microbial standards, the discution was raised about the necessity of establishing national specifications, even if provisional. Suggestions are made as to the contamination limits of oral phytotherapic pharmaceuticals.

Heterogeneity of plural forms: a revised transaction cost approach / Heterogeneidade de formas plurais: uma abordagem de custos de transação revisada

This dissertation addresses an important empirical regularity that is still not very well understood: the same firm, in a given institutional context, simultaneously relies on different organizational arrangements to govern similar (or identical) transactions. I will follow Bradach & Eccles (1989) and Menard (2013) and treat this phenomenon as \'plural forms\'. The dissertation is composed of four chapters, each of which, providing different but complementary approaches to plural forms and serving as input for the next: the first half of this dissertation remains theoretical, while the second presents distinct qualitative empirical contributions on the theme. In the first chapter, I intend to verify what has been theorized about plural forms and how this field has evolved over time. I am especially concerned with the multiple theoretical explanations about the reasons why firms adopt plural forms, mainly because, to date, there is no consensus on the subject. To this end, I both perform some bibliometric analyses by surveying the ISI Web of Science platform and qualitatively review some significant papers on the subject. I find that the combination of asset specificity and multiple types of uncertainty underlies most of the explanations used over time and highlight that these could be interesting variables for a model predicting plural forms. In the second chapter, I am concerned with explaining the variety of plural form manifestations. However, in order to understand such variety, I build a theoretical model integrating asset specificity and uncertainty to predict plural and non-plural forms. Next, I emphasize that these arrangements are heterogeneous and, to keep my argumentation tractable, I define three categories of plural forms: \'between\' (or the combination of different modalities of organizational arrangements), \'within\' (or the combination oh hybrid arrangements) and \'combo\' ( or the combination of \'between\' and \'within\'). My hypotheses relate each of these three types of plural forms to the different typologies of uncertainty (market, technological and performance assessment) that capture the essential dimensions of the concept. In the next two chapters, I empirically confornt my theretical model and hypotheses with qualitative empirical data concerning the Brazilian Agribusiness sector. In the third chapter, I seek to verify whether the combination of uncertainty and asset specificity predicts the emergence of plural forms. Moreover, I assess whether different levels of uncertainty and of asset specificity (combined) determine the selection of distinct organizational arrangements. To this end, I perform an embedded clinical case study at the Korin company, which produces and commercializes multiple organic products. I am concerned with the procurement of distinct inputs (regular and organic corn, regular soybeans, poultry and antibiotic-free eggs), both with plural and with non-plural forms. I find support for my model and for the idea that different levels of asset specificity and uncertainty push towards the adoption of distinct organizational arrangements. In the final chapter, I intend to empirically confront both my theoretical model (i.e., if asset specificity and uncertainty predict plural or non-plural forms) and my hypotheses (i.e., whether different types of high uncertainty, subject to a certain degree of asset specificity, push towards different types of plural forms). To this end, I survey the procurement of inputs by 24 companies operating in the Brazilian Agribusiness sector and procuring 11 different inputs by means of both plural and non-plural forms. I make use of a 4-point likert scale questionnaire capturing my theoretical variables and perform some qualitative analyses. I find empirical support for my theoretical model and for some of my hypotheses concerning the diversity of plural forms, indicating a path for further empirical analyses. / Esta tese aborda uma importante regularidade empírica que ainda não é muito bem compreendida: uma mesma empresa, em um determinado contexto institucional, simultaneamente se baseia em diferentes arranjos organizacionais para governar transações similares (ou idênticas). Seguindo os trabalhos de Bradach & Eccles (1989) e Menard (2013), trata-se este fenômeno como \"formas plurais\". A tese é composta por quatro capítulos, cada qual proporcionando abordagens diferentes, porém complementares, sobre a temática das formas plurais, e cada qual servindo como insumo para o próximo capítulo: a primeira metade desta tese permanece teórica, enquanto a segunda apresenta contribuições empíricas qualitativas distintas sobre o tema. No primeiro capítulo, pretende-se verificar o que foi teorizado sobre formas plurais e como este campo tem evoluído ao longo do tempo. Em especial, busca-se conhecer as múltiplas explicações teóricas sobre as razões por que as firmas adotam formas plurais, principalmente porque ainda não há um consenso sobre o assunto. Para tanto, realizam-se algumas análises bibliométricas por meio de um levantamento de dados na plataforma ISI Web of Science e qualitativamente revisam-se alguns artigos significativos sobre o assunto. Como resultado, tem-se que a combinação de especificidade de ativos e vários tipos de incerteza se encontra subjacente à maior parte das explicações utilizadas ao longo do tempo e destaca-se a ideia de que estas poderiam ser variáveis interessantes para um modelo de previsão de formas plurais. No segundo capítulo, busca-se compreender o que explica a variedade de formas plurais. No entanto, a fim de compreender tal variedade, constrói-se um modelo teórico integrando especificidade de ativos e incerteza para prever formas plurais e não plurais. Em seguida, enfatiza-se que estes arranjos são heterogêneos e, para manter a argumentação tratável, definem-se três categorias de formas plurais: \"entre\" (ou a combinação de diferentes modalidades de arranjos organizacionais), \"dentro\" (ou a combinação de arranjos híbridos ) e \"combo\" (ou a combinação de \"entre\" e \"dentro\"). As hipóteses relacionam cada um destes três tipos de formas plurais com as diferentes tipologias de incerteza (mercado, tecnológica e de avaliação de desempenho) que capturaram as dimensões essenciais deste conceito. Nos próximos dois capítulos, o modelo teórico e as hipóteses são confrontados com dados empíricos qualitativos relativos ao setor agroindustrial brasileiro. No terceiro capítulo, procura-se verificar se a combinação de incerteza e especificidade de ativos prevê o surgimento de formas plurais. Além disso, avalia-se se diferentes níveis de incerteza e de especificidade de ativos (combinados) determinam a seleção de arranjos organizacionais distintos. Para tanto, realiza-se um estudo de caso clínico \"embedded\" na empresa Korin, que produz e comercializa diversos produtos orgânicos. Analisa-se a aquisição de insumos distintos (milho comum e orgânico, soja comum, frango e ovos livres de antibióticos), tanto com formas plurais quanto com formas não plurais. Encontra-se suporte para o modelo e para a ideia de que diferentes níveis de especificidade de ativos e de incerteza levam à adoção de arranjos organizacionais distintos. No capítulo final, confronta-se empiricamente tanto o modelo teórico (ou seja, se a especificidade dos ativos e a incerteza preveem formas plurais ou não-plurais) quanto as hipóteses (ou seja, se os diferentes tipos de elevada incerteza, sujeitas a um determinado grau de especificidade de ativos, leva à diferentes tipos de formas plurais). Para tanto, examina-se a aquisição de insumos por 24 empresas que operam no agronegócio brasileiro e adquirem 11 insumos diferentes, tanto por meio de formas plurais como de formas não-plurais. Utiliza-se de um questionário escala de Likert de 4 pontos, capturando as variáveis teóricas e realizam-se algumas análises qualitativas. Encontra-se suporte empírico para o modelo teórico, e para algumas das hipóteses que explicam a diversidade, apontando um caminho para maiores investigações empíricas

En prenant le cas de l'évaluation des gérants d'actifs financiers par les investisseurs professionnels, la présente thèse étudie les réponses que des agents économiques apportent aux problèmes pratiques que pose la nécessité d'agir et de justifier son action en situation d'incertitude.La thèse marie, dans une perspective durkheimienne, une analyse morphologique du champ de la gestion d'actifs et une analyse des pratiques d'évaluation des gérants par les investisseurs professionnels.La démarche empirique consiste à rassembler des matériaux variés afin d'enregistrer les multiples formes de manifestation d'un fait social total. Les uns sont produits au cours d'une enquête ethnographique du champ de la gestion d'actifs: ce sont des entretiens semi-directifs, des observations directes d'interactions sociales, et des artefacts produits et utilisés dans le cadre des pratiques étudiées. Les autres, statistiques ou données institutionnelles, sont collectés auprès d'acteurs de la gestion d'actifs. Tous ces matériaux sont analysés de manière interprétative.L'analyse morphologique découvre les divisions, les articulations et les proportions des organisations où est pratiquée la gestion d'actifs financiers. L'autonomisation du champ professionnel de la gestion d'actifs s'accompagne d'une division du travail qui place en son centre les gérants, constitués en spéculateurs, c'est-à-dire en agents économiques dont l'action ne s'intéresse qu'aux variations de prix, par opposition aux acteurs dont la fonction est de prolonger le geste spéculatif par un geste marchand. La structure de rémunération des sociétés de gestion nourrit un conflit entre les gérants d'actifs et les commerciaux sur le partage du profit, dont la résolution conduit à une polarisation des sociétés entre un modèle industriel et un modèle artisanal.L'évaluation des gérants d'actifs par les investisseurs professionnels prend son sens dans le cadre de l'appréciation de l'efficacité technique des actions spéculatives par des sujets exposés à une incertitude financière qu'ils s'efforcent de réduire. Les investisseurs professionnels ordonnent leurs pratiques d'évaluation entre une analyse "quantitative" et une analyse "qualitative" des gérants évalués. Contrairement à ce que l'usage de cette forme de classification suggère, ce n'est pas le recours à la quantification ou au calcul en tant que tels qui différencie ces deux types d'analyse, comme s'il s'agissait de deux régimes épistémiques imparfaits mais complémentaires. C'est leur statut dans la relation fonctionnelle qui les lie l'un à l'autre: recherche de la forme de l'incertitude financière d'une part, et recherche des conditions sociales qui l'ont générée d'autre part. Imparfaitement liée aux phénomènes auxquels elle s'applique, la forme de classification "quanti/quali" correspond aux ressources acquises par les investisseurs professionnels au cours de leur trajectoire de socialisation, et mobilisées dans le cadre d'une division sociale du travail où les pratiques d'analyse sont partagées entre des postes et des positions différenciées au sein des organisations.Ce travail complexifie et enrichit la compréhension des pratiques financières et des pratiques de réduction de l'incertitude en montrant qu'elle ne peut faire l'économie de leurs soubassements sociaux. Elle précise le domaine de validité d'une forme de classification et offre aux acteurs concernés le moyen de discuter à nouveaux frais l'efficacité ou la pertinence de leurs procédés d'analyse. / This dissertation uses the evaluation of asset managers by professional investors as a case for the study of the ways in which economic agent deal with the necessity of acting and to give account of one's actions while facing uncertainty.This dissertation follows a durkheimian approach as it combines a morphological analysis of the asset management field with an analysis of evaluation practices of asset managers by professional investors.Empirical materials of various kinds have been gathered, in order to record the numerous ways in which a total social fact manifests itself. Some of the materials derive from an ethnographic research in the field of asset management: these are semi-directive interviews, direct observations of social interactions, and artefacts produced and used as part of the practices that the research investigates. The other materials are statistics or institutional data that have been collected during the field research. Interpretive analysis has been applied to them.The morphological analysis of asset management as a social field highlights the divisions, articulations and proportions of the organizations where financial asset management practices are carried out. The autonomization of asset management as a professional field goes along with a social division of labour that is structured around the asset managers. It makes them become speculators, that is economic agents who focus solely on price variations. By contrast, other actors ensure that the speculative movement is followed by a fully carried out trade. The income structure of asset management firms feeds a conflict between asset managers and salespersons about the distribution of profit. The resolution of this conflict leads to a polarization of asset management firms between firms following a model designed after the manufacturing industry, and firms following a model leaning towards craftmanship.The evaluation of asset managers by professional investors makes sense in a social frame where it stands as an appraisal of the efficiency of speculative actions undertaken by agents who try and reduce the financial uncertainty that they face. Professional investors classify their evaluation practices among a "quantitative" analysis and a "qualitative" analysis of asset managers. Although this form of classification may suggest that the two types of analysis cover two kinds of knowledge that are imperfect yet complementary, it is not quantification or calculation per se that differentiates between them. They are functionally related to each other: assessment of the form taken by financial uncertainty on the one hand, and search for the social conditions that caused uncertainty to take such form on the other hand. The form of classification "quantitative/qualitative" meets imperfectly the phenomena to which it is applied. It matches the social distribution of resources that professional investors gained during their socialization trajectory, and that they mobilized in the frame of a specific kind of social division of labour, where analytic practices are also distributed among differentiated positions within organizations.This dissertation complexifies and enriches the understanding of financial practices and of uncertainty reduction practices by showing that such understanding cannot forget their social foundations. It circumscribes the domain of validity of a specific form of classification and it provides interested actors with tools with which they can discuss anew the efficiency or the relevance of their analytic processes.

Les niveaux psychologiques de Platon : une théorie de la connaissance d'après la cybernétique / Plato's Psychological Levels : a theory of knowledge in the words of cybernetics

Cette recherche se propose de montrer l’existence de ce que nous appelons des « niveaux psychologiques » chez Platon, en empruntant à la théorie des types logiques, formulée par B. Russel et A.N. Whitehead, puis revisitée par l’anthropologue G. Bateson au sein du mouvement cybernétique. Par « niveaux psychologiques », nous entendons des états psychiques d’être en relation avec le réel sur le mode analogique. Nous défendons la thèse selon laquelle la théorie platonicienne des formes n’oblige pas à penser des degrés de réalité de la chose mais bien un processus de réalisation de l’objet en soi par le sujet de l’expérience. En tant que tels, les niveaux psychologiques correspondent donc à des niveaux d’apprentissages de la réalité. Ainsi, la méthode dialectique utilisée par Socrate accompagne le progrès de l’âme, pilotée par l’intellect, au moyen de la raison qui relie et distingue les phénomène sensibles. Cette double fonction caractérise l’exercice du langage et rend compte du mouvement de la pensée. La succession et la répétition des ajustements opérés à partir de l’expérience conduisent ainsi à une meilleure définition de la forme de l’objet en soi, i.e. une saisie plus nette de ce qui, de l’intelligible, passe dans le sensible. De ce fait, la théorie platonicienne de la connaissance apparaît comme une théorie du processus et non comme une théorie du contenu de savoir, qui mène du théâtre intérieur de nos représentations, limitées par le temps d’une vie et l’espace d’un corps, au spectacle de l’infini. En outre, parce qu’il existe un seul chemin pour penser ce qui est réellement, l’examen psychologique des causes de soi rejoint nécessairement la recherche philosophique sur les causes du monde, dans une cosmologie. / This research intends to demonstrate the existence of what we call "psychological levels" in Plato's philosophy, taking from the theory of logical types, formulated by B. Russel and A.N. Whitehead, then restyled by the anthropologist G. Bateson, inside the cybernetic movement. By "psychological levels", we mean psychic states of being in relation with reality under an analogical scheme. We defend the argument that Plato’s theory of Forms does not imply degrees of reality in the thing but a process of realization of the object in itself through the subject experiencing it. As such, psychological levels refer to levels of learning what is reality. Thus, the Socratic dialectical method accompanies the soul’s progress, driven by the intellect, thanks to reason which relates and distinguishes the sensible phenomena. This double function characterizes what language is and gives reason for the movement of thought. Succession and repetition of the adjustments made from the experience of the thing lead to a better definition of the Form of the object in itself, i.e. a clearer acquisition of the intelligible part of the thing that is passing through its sensible expression. Hence, Plato’s theory of knowledge stands like a process theory more than a content theory, by conducting the search for truth from the inner theatre of our representations, limited by a lifetime and the space of a body, to the spectacle of the infinite. Furthermore, since there is only one path in order to think what exists in reality, the psychological research into the causes of thyself necessarily fits in the philosophical research into the causes of the world — a cosmology.

From the inventory that Agamben will make out of the originating concepts of the treaty of Aristotle Peris Psykhês as well as the archaic Roman Law towards the definition of bare life , to the Agamben s readings on the reflections of Walter Benjamin, Hannah Arendt and Michel Foucault, which will be tackled as biopolitical hypothesis , by going through surveys on the Schmitt s exception and the reine Gewalt in Benjamin s, the thesis is intended to expose the inevitable threshold of the political problems with the discussions on time and memory within Agamben s philosophy presented below, in addition to some of the main difficulties, implications and contradictions involving its formulation. Thresholds (Schwellen) must be understood herein not only in a methodological but also epistemological sense the way that Agamben thinks its philosophy, through ontology, politics, law and esthetics and the temporary sense, the specificity of the thresholding concept to characterize the unique relation about the human time with the memory, the history and the politics / A presente tese objetiva abordar o problema das relações entre política e tempo na obra do filósofo italiano Giorgio Agamben, situando-o a partir de quatro limiares onde esta questão é ali desenvolvida: a) a problemática da vida nua e das formas-de-vida; b) o problema da comunidade; c) o estado de exceção; d) a relação entre o tempo, a memória e a história. However, its conceptualization has been changing drastically in recent years, especially after the anti-cultural movements initiated in the mid-1960s and the erosion of patriarchal ideology and modernity, as well as the weakening of religious influence in Brazilian society. This mutation, which includes enlargement and socio-cultural changes, has effects in several areas of the humanities that reflect, as it could not be, the Law. The concept of family, in the legal and legislative spheres, is indicative and can be used to reflect social demands. In this way, the present work makes a historical progression of the origin of the Modern and Democratic State of Law, in order to examine the contemporary conceptualization of the family from the point of view of the country's legal scholars, the principles protected in our legal system and then, a legal definition of family will be proposed. O objetivo da presente tese é investigar as famílias e as formas de união, estabelecidas na Freguesia Madre de Deus de Porto Alegre, entre os anos de 1772 e 1822. A análise parte da Demografia Histórica para a História Social e História da Família, tendo, como via de acesso, o cruzamento nominativo de fontes de cunho eclesiástico e cartorial. Buscamos investigar a trajetória de algumas famílias, tendo como fio condutor as formas de união e os laços familiares estabelecidas entre a população porto-alegrense, que contraiu núpcias ou estabeleceram uniões não sacramentadas entre o último quartel do século XVIII e início do século XIX. De modo geral, pretendemos investigar quais os tipos de famílias que se constituíam e até que ponto havia formas de união e famílias estabelecidas de acordo com os “modelos” determinados pelas normativas da Igreja e da Coroa Portuguesa, acompanhando as transformações que marcaram a História do Brasil Meridional no período em destaque. / The goal of this thesis is to investigate the families and forms of union that took place in the Madre de Deus de Porto Alegre Parish between 1772 and 1822. The analysis is based on Historical Demography and on Social History and History of the Family, having as a starting point the nominative crossing using both ecclesiastical and civil sources. We seek to examine the trajectory of a few families and to do so we use as a conductive path the marriages and Family ties established among Porto Alegre’s population that got married or that otherwise formed other sorts of union between the last quarter of the 18th century and the beginning of the 19th. In general, we intend to explore which types of Family were constituted and to what extent there were forms of union and established family according to the “models” determined by the norms and standards of both the Church and the Portuguese crown, thus following the transformations that marked the History of Southern Brazil in the highlighted period.

The topic of this dissertation thesis is a debated issue called "homeoffice and homeworking", which originates from "atypical and flexible forms of work/employment", which are not captured in our labour code as a relevant section that describes legal foundation for setting up standards for employees, nor the employer. The Selected topic comes out of current needs of labour market, ergo needs from the companies, employers and employees. These needs are rising from changes and implementations of new terms and conditions for working out of employers work place, it also originates from demands of young population for more flexible and "free" working time. It comes hand in hand with industry revolution called Industry 4.0. The thesis is brainstorming about changing the established terms of atypical forms of work, if they even still are "atypical" or did they become a new standard for employment relations. The role of moisture profiling towards understanding pharmaceutical solid state functionality : validation and the application of a moisture profiling analytical tool for investigation into the characterisation of, and prediction of, the effects of compaction and storage on different lactose physical forms

The majority of therapeutic pharmaceutical formulations are presented in the solid form. Moisture is able to play an important role in the functional performance of pharmaceutical solids. Moisture profiling is able to provide novel information with regards to the behaviour of moisture within materials using equilibrium relative humidity as a measurement. The hypothesis investigated explores the changes in equilibrium relative humidity of pharmaceutical material induced by physical, chemical or storage conditions, these are able to be monitored using the innovative moisture profiler system. The aims within this were to primarily validate the moisture profiler and secondly evaluate the effects of moisture on physical forms and with respect to effects of compaction, finally this was compared to conventional characterisation methods. Preliminary explorations were conducted in order to assess the validity of the moisture profiler, from this lactose was selected as a suitable pharmaceutical material for further work. Processing effects were then examined, firstly storage at elevated relative humidity of different forms of lactose were explored, and this was carried out with supplementary analysis. Secondly the effects of tabletting were explored, different compaction forces were investigated to observe if this had any notable effects on equilibrium relative humidity of the different lactose forms. Finally subsequent storage of the compacts were examined in order to explore if there were any changes in the equilibrium relative humidity.
