Caractérisation fonctionnelle de BamB, protéine impliquée dans la biogénèse de la membrane externe et la virulence de Salmonella / Functional caracterization of BamB, a protein involved in outer-membrane biogenesis and Salmonella virulence

La protéine BamB est une lipoprotéine de membrane externe appartenant au complexe BAM (β-Barrel Assembly Machinery) et impliquée dans l’assemblage des protéines de membrane externe (PME), la sensibilité aux antibiotiques, le contrôle de l’expression des trois systèmes de sécrétion de type III (T3SS) et la virulence de Salmonella. Chez E. coli, au sein du complexe BAM, elle interagit directement avec la protéine BamA. De plus, chez cette bactérie, BamB présente une activité sérine-thréonine kinase. Afin de mieux caractériser le rôle de BamB, nos objectifs ont été d’étudier (1) l’impact de l’altération de l’interaction de BamB avec le complexe BAM ou de sa séquestration dans le cytoplasme sur l’ensemble des rôles décrits de BamB et (2) l’activité kinase putative de BamB chez Salmonella. Nos résultats montrent que certains rôles de BamB sont dissociables entre eux et que l’interaction BamA/BamB n’est pas requise pour le rôle de BamB dans le contrôle de l’expression des T3SS, la virulence de Salmonella et l’assemblage des PME à la membrane externe. Aucune activité kinase ni aucune activité cytoplasmique de la protéine n’a pu être formellement démontrée. / BamB is an outer-membrane lipoprotein belonging to the BAM complex (β-Barrel Assembly Machinery). In Salmonella, it is involved in the assembly of outer membrane proteins (OMP), in antibiotic susceptibility, in the transcriptional control of the three Type-Three-Secretion-Systems (T3SS) related genes and also in virulence. In E. coli, BamB interacts directly with the BamA protein. Moreover, BamB has been shown to have a serine-threonin kinase activity in this bacterium. In order to better characterize the roles of the BamB protein, our purposes were to study (1) the impact of the alteration of the interaction of BamB with the BAM complex or of its cytoplasmic sequestration and (2) its putative kinase activity in Salmonella. Our results show that some of the BamB roles are dissociable and that the BamA/BamB interaction is not required for T3SS expression, Salmonella virulence or OMP assembly in the outer membrane. Currently, neither a kinase activity nor a cytoplasmic activity has been clearly demonstrated for this protein.

Les PME structurent 90% des économies émergentes. Elles sont duales, à la fois moteurs de l'activité mondiale et vulnérables à cause d’un recours excessif à l'endettement et d'une économie informelle prégnante. Nous pensons que le capital investissement peut consolider et diffuser cette traction économique devenue l’apanage des seules nations émergentes. Se pose alors la question : « Quel modèle de capital investissement pour les pays émergents ? ». La méthodologie retenue répond à la contrainte d’une littérature encore réduite et d’un difficile accès à l’information. Nous suivons en conséquence une démarche inductive et étudions, au travers du modèle théorique de Timmons, la création d’un fonds d’investissement en Egypte de 2008 à 2011. A partir de ce cas clinique longitudinal, nous induisons un modèle de capital investissement entrepreneurial, à destination des pays émergents et inspiré du capital risque technologique. / SMEs represent 90% of emerging economies. They are dual, both engine for the world economy and vulnerable because of their excessive indebtedness and the weight of informal activities. We think private equity can enhance and spread this economic traction that only qualifies emerging countries. Thus, our question appears as: “Which model for emerging markets private equity?”. We select an inductive approach to address constraints of a limited research literature and a difficult access to data. Through the theoretical Timmons’ model, we perform a longitudinal clinical study of a private equity fund in Egypt, during its inception from 2008 to 2011. We design from our research an entrepreneurial private equity model, dedicated to emerging markets and inspired by venture capital.

More than 1300 Italian companies, from the biggest to the smallest one, are acting in Brazil, exploiting the overall potential of this country and contributing with the excellence they can bring. Noting the consistency of the Italian presence and wondering how those companies have entered such a complex country and, a recurring name has been GM Venture, which became the protagonist of this dissertation. The thesis in fact aims at studying how GM Venture Company has been able to create value for Italian SMEs, contributing to their success in Brazil during the last ten years. Therefore this analysis focuses firstly on the literature dealing with the key success factors for the internationalization of small-medium enterprises. The second part of the literature goes deep in the specific case of the Brazilian context, trying to understand the major problems that this particular environment poses to small-medium companies internationalizing here and some specific factors Italian companies should pay attention to. Given this introduction, the analysis centers on the core topic of this dissertation. In fact the investigation regarding the GM Venture case looks specifically at how this company has been able create value despite those problems and create a business model able to successfully support SMEs entering Brazil. The inquiry on the case and the conclusion reports how GM Venture, through its operations, has been able to respond to the Brazilian rtise, and positively contribute to the growth and value of the Italian economic reality in this country. Moreover looking at the literature findings, this specific case introduces some novelty in the emerged factors, such as a new way of conceiving the Network, as well as new factors or undervalued ones for success such as promptness in the response and trustworthiness, which are essential in the Brazilian economic environment. / Desde muitos anos a presença Italiana no Brasil foi caracterizante do panorama econômico brasileiro. Mais de mil trezentos empresas italianas, das maiores ate as menores, são presente hoje no Brasil, aproveitando inteiramente do potencial Brasileiro e levando no Brasil uma contribuição relevante no sistema econômico, por meio da excelência que caracteriza estas empresas Italianas. Olhando ao tamanhão da realidade das empresas italiana no Brasil, e como todas essas corporações podia ter entrado em um pais muito complexo, ponto central foi a GM Venture, que se tornou o protagonista da minha dissertação. Esso TCC tem o escopo de entender melhor como esses deis anos GM Venture conseguiu em criar valor econômico por muitas media e pequena empresas Italianas, levando eles ao sucesso no novo país. Por isso a dissertação se focaliza inicialmente sobre a literatura da internacionalização e dos fatores essenciais de sucesso por uma pequena e media empresa em general. O secundo fator de analise nela literatura para o caso especifico do contexto Brasileiro, foi a compreensão dos problemas principais que o Brasil põe para pequenas-medias empresas estrangeiras, e algumas atenções que são necessárias em particular para empresários italianos. Para evoluir no tema, depois de essa introdução ao tema e ao contexto, o tópico mais importante é discutido. Então a investigação de esse trabalho se focaliza sobre o tema de como a GM Venture tem capacidade de responder com uma solução para os problemas presentados criando valor, e como ela criou um modelo de business que seja capaz de levar ao sucesso as empresas clientes no Brasil. A analise e conclusão do caso vão elaborar como a GM Venturo contribuiu ao resolver e evitar a complexidade Brasileira com meios legais e experiência, cooperando em positivo no crescimento da realidade econômica italiana em Brasil. Olhando ainda mais na analise da literatura e comparando o caso, parece evidente como o caso da GM Venture introduziu algumas novidades nos fatores já individuados, como uma ideais nova de network, e no mesmo tempo foi capaz de presentar fatores novos o subestimado, como a pronta velocidade e a confiança nos relacionamento de business que são muito importantes especialmente no Brasil.

Ce travail de thèse vise à proposer une méthodologie d’évaluation de la capacité à innover et à exporter des PME manufacturières et de procédés. Cette méthodologie est basée sur l’identification et la caractérisation d’un espace commun innovation/export représentant les activités, ressources, compétences qu’une PME doit mobiliser en priorité pour améliorer de façon simultanée ses performances concernant l’innovation et l’export, tout en réduisant l’effort associé à cette montée en performance. Ce travail contribue à la recherche scientifique à travers différents aspects. Tout d’abord, la relation innovation/export en PME, traditionnellement pensée de façon causaliste, a été envisagée à travers le prisme du paradigme de la complexité. Cette contribution théorique a permis d’aboutir à l’identification d’un espace commun conceptuel innovation/export. Puis, cet espace commun a été caractérisé à travers l’identification d’activités conjointes. L’identification de ces activités conjointes est un apport à la fois théorique et managérial car en améliorant une de ces activités, les entreprises actionnent un seul levier qui se répercute simultanément en matière d’innovation et d’export. Dans le contexte des PME, pour qui le manque de ressources est une difficulté capitale, cela prend tout son sens. Enfin, une méthodologie d’évaluation de la capacité à innover et à exporter basée sur cet espace commun a été conçue de façon à proposer un outil multicritères d’aide à la décision dédié aux PME. Cet outil a finalement été testé auprès de PME manufacturières et de procédés de la région Lorraine / This research work aims to suggest a methodology for analyzing the innovation and export capabilities of SMEs working in the manufacturing and process sectors. This methodology is based on the identification and the characterization of a conceptual innovation/export common space representing the activities, the resources, the skills that a SME has to mobilize first and foremost in order to improve in a simultaneous way its performances concerning innovation and export. Working on the activities included in the common space allows the reduction of the effort associated with this performance improvement. This work contributes to the scientific research through various aspects. First of all, the innovation / export relationship in SMEs, traditionally thought in a causal way, was envisaged through the prism of the complexity paradigm. This theoretical contribution allowed the identification of an innovation/export common space. Then, this common space was characterized through the identification of joint activities. The identification of these joint activities is both a theoretical and managerial contribution because by improving one of these activities, companies activate a single lever which echoes simultaneously on innovation and export capabilities. In the context of SMEs, for whom the lack of resources is a major difficulty, it makes perfect sense. Finally, an evaluation methodology of the innovation and export capabilities based on this common space was designed so as to propose a multicriteria decision-making tool dedicated to SMEs. Ação da pectina metil esterase e cloreto de cálcio no armazenamento e controle da podridão-mole em pimentão / Pectin methyl esterase action and calcium chloride in the storage and control of soft rot in pepper

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The pepper (Capsicum annuum L.) has commercial importance, by having sources of vitamins, minerals and fiber. However, a post-harvest problem, excessive softening which reduces the life and favors action pathogens such as Pectobacterium caratovorum subsp. caratovorum- Pcc, causal agent of soft rot, a major disease of post-harvest chili that is favored by the reduction of firmness. One technique that has recently been used for firmly maintaining the application is pectin methyl esterase (PME) with the addition of a calcium solution prologando thus reducing its lifetime and pathogen attack. The objective of this study was to use methyl pectin esterase (PME) exogenous associated with calcium chloride in maintaining firmness and control of Pectobacterium caratovorum subsp. caratovorum about pepper. The first experiment was conducted in a completely randomized design in a 4x5 factorial scheme with three replications for 12 days, evaluated every three days. In the second experiment, the test was conducted in a completely randomized design in a factorial 5x5 with three replications. In the first study the fruits of pepper were subjected to vacuum infusion method with pressure of 200 mmHg for 5 minutes, and following evaluated the loss of weight (PMF), fruit firmness (FF), skin color ( CC), soluble solids (SS), pH, total acidity (TA) and activity of SMEs. In the second test the fruits of pepper were subjected to vacuum infusion method with pressure of 200 mmHg for five minutes, then the fruits were inoculated with the PCC then held the fruit firmness analysis (FF), SME activity and the severity of the disease in chili (SD). The fruits obtained in general a reduction of over time firmly in all treatments but was found significant effect in maintaining the fruit firmness when treated in vacuum infusion with calcium chloride, without altering the physicochemical characteristics, as soluble solids content, total acidity and activity of SMEs, slowing the fruit ripening process. The fruits when treated with vacuum infusion with calcium chloride associated with pectin methyl esterase was not favorable because it altered the physicochemical properties chili, highlighting the decline of firmness, thus deteriorating the quality of the fruit. Regarding inoculation Pcc in the fruit was observed inhibition of growth of this pathogen, prolongation of fruit and better firmness treated fruits infusion over calcium inoculation Pcc chloride. / O pimentão (Capsicum annuum L.) tem grande importância comercial, por possuir fontes de vitaminas, minerais e fibras. Contudo, possui problema pós-colheita, o amolecimento excessivo que reduz a vida útil e favorece ação de patógenos como a Pectobacterium caratovorum subsp. caratovorum- Pcc, agente causal da podridão-mole, uma das principais doenças da pós-colheita do pimentão que é favorecida pela redução da firmeza. Uma técnica que vem sendo utilizada recentemente para a manutenção da firmeza é aplicação da pectina metil esterase (PME) com a adição de solução de cálcio prologando, assim, a sua vida útil e diminuindo o ataque do patógeno. Assim, o objetivo do trabalho foi utilizar a pectina metil esterase (PME) exógena associada ao cloreto de cálcio na manutenção da firmeza e no controle da Pectobacterium caratovorum subsp. caratovorum sobre o pimentão. O primeiro experimento foi realizado em delineamento inteiramente casualizado em esquema fatorial 4x5 com três repetições, durante 12 dias, avaliados a cada 3 dias. No segundo experimento, o ensaio foi realizado em delineamento inteiramente casualizado no esquema fatorial 5x5 com três repetições. No primeiro trabalho os frutos de pimentão foram submetidos ao método de infusão a vácuo com pressão de 200 mmHg por 5 minutos, e por seguinte avaliou-se a perda de massa fresca (PMF), firmeza do fruto (FF), cor da casca (CC), teor de sólidos solúveis (SS), pH, acidez total, (AT) e atividade de PME. No segundo ensaio os frutos de pimentão foram submetidos ao método de infusão a vácuo com pressão de 200 mmHg por cinco minutos, posteriormente os frutos foram inoculados com a Pcc em seguida realizou-se as análises de firmeza do fruto (FF), atividade de PME e a severidade da doença no pimentão (SD). Os frutos obtiveram de forma geral uma redução da firmeza ao longo do tempo em todos os tratamentos, porém foi verificado o efeito significativo na manutenção da firmeza dos frutos quando tratados em infusão a vácuo com cloreto de cálcio, não alterando as características físico-químicas, como o teor de sólidos solúveis, acidez total e atividade de PME, retardando o processo de amadurecimento do fruto. Os frutos quando tratados com infusão a vácuo com cloreto de cálcio associado à pectina metil esterase não foi favorável, pois alterou as propriedades físico-químicas do pimentão, com destaque para o declínio da firmeza, deteriorando assim a qualidade do fruto. Em relação à inoculação da Pcc no fruto, observou-se uma inibição do crescimento desse patógeno, prolongamento do fruto e uma melhor firmeza nos frutos tratados com infusão de cloreto de cálcio mais a inoculação da Pcc.

The study was conducted by means of two questionnaires, two semi-structured interviews and the analysis of the performance measures used by five small/medium-sized enterprises based in the State of São Paulo. Using a qualitative approach, this exploratory research were interviewed PMS s and QMS s managers, and the data from the first interview were analyzed using the technique of content analysis and has been subsequently triangulated with other data collected. It was found that four out of five companies have a junction in PMS and QMS, with respect to performance measures, despite of the degree of the quality management maturity is not related to the stage of the PMS evolution. It was also found that in three out of five companies quality management area is responsible for coordinating the process of PMS use, without effective participation from the controlling and/or accounting areas in this process. The typology of Garengo (2009), used to check the stage of the PMS evolution, was validated and can be used in practice by professionals in organizations to diagnose and improve the PMS in their enterprises. From the findings of this research, companies with QMS certified by ISO, particularly with higher degree of quality management maturity, can be encouraged to implement or improve the PMS in their organizations. The results of this study should be considered in the light of its limitations, especially the impossibility of its generalization to the overall universe of SMEs, to the extent that there was used the qualitative approach, without the aim of extrapolating the results for other enterprises. In this sense, quantitative research should be carried out in order to verify these results in a more representative set of SMEs. Finally, it is suggested that research could be carried out to study in more depth the PMS evolution in SMEs, with longitudinal case studies, for example. / Esta pesquisa teve como propósito verificar como funcionam o sistema de mensuração de desempenho (SMD) e o sistema de gestão da qualidade (SGQ) em pequenas e médias empresas brasileiras à luz da tipologia proposta por Garengo (2009). O estudo foi conduzido mediante a aplicação de dois questionários fechados, duas entrevistas semiestruturadas e da análise dos indicadores de desempenho utilizados por cinco pequenas/médias empresas com sede no Estado de São Paulo. Com abordagem qualitativa, nesta pesquisa exploratória foram entrevistados os gestores do SMD e do SGQ, sendo que os dados da primeira entrevista foram analisados com a técnica de análise de conteúdo, tendo sido, na sequência, triangulados com os demais dados coletados. Verificou-se que quatro das cinco empresas pesquisadas possuem uma junção do SMD com o SGQ, no que tange aos indicadores de desempenho, a despeito de o grau de maturidade da gestão da qualidade não estar relacionado com o estágio de evolução do SMD. Verificou-se ainda que em três das cinco empresas a área da gestão da qualidade é a responsável por coordenar o processo de uso do SMD, sem que haja uma participação efetiva da área de controladoria e/ou contabilidade nesse processo. A tipologia de Garengo (2009), usada para verificar o estágio de evolução do SMD, foi validada, podendo ser utilizada na prática pelos profissionais nas organizações para diagnosticar e melhorar o SMD nas suas empresas. A partir dos achados desta pesquisa, os profissionais de empresas com SGQ certificados pela ISO, principalmente aquelas maior grau de maturidade na gestão da qualidade, podem ser encorajados a implementar ou melhorar o SMD nas suas organizações. Os resultados deste estudo devem ser considerados à luz de suas limitações, sobretudo a impossibilidade de sua generalização para o universo geral das PME, na medida em que houve o emprego de abordagem qualitativa, sem o objetivo de extrapolação dos resultados para outras empresas. Nesse sentido, pesquisas quantitativas podem vir a ser realizadas com o objetivo de verificar esses resultados em um conjunto mais representativo de PME. Por fim, sugere-se que pesquisas venham a ser desenvolvidas para estudar em maior profundidade como ocorre a evolução dos SMD em PME, a partir de estudos de caso longitudinais, por exemplo.

The caderno Pequenas e Medias Empresas do Jornal O Estado de São Paulo, a prominent Brazilian journalistic vehicle, has been developing an advertising discourse about entrepreneurship. Case studies of the aforementioned editorial are analyzed according to the concepts of discourse analysis by Dominique Maingueneau, and also based on the concepts referring to myth and fetish, by Mircea Eliade and Massimo Canevacci, respectively. In addition to the hybridization of the journalistic and advertising genres, discourse analyzes demonstrate the use of mythical representations and of the fetish for the development of a persuasive discourse, which in a sense supports the emerging ideology of entrepreneurship. / A mídia está bastante interessada na questão do empreendedorismo, devido à recente popularização de ideias e crenças em torno da atividade de criar e desenvolver negócios. O caderno Pequenas e Medias Empresas do Jornal O Estado de São Paulo, proeminente veículo jornalístico brasileiro, vem desenvolvendo um discurso publicitário acerca do empreendedorismo. Estudos de caso de veiculações do referido editorial são analisados conforme os conceitos de análise de discurso de Dominique Maingueneau, e também com base nos conceitos referentes a mito e fetiche, de Mircea Eliade e Massimo Canevacci, respectivamente. Além da hibriditização dos gêneros jornalístico e publicitário, as análises de discurso demonstram o emprego de representações míticas e do fetiche para o desenvolvimento de um discurso persuasivo, o que, em certo sentido, vem apoiar a emergente ideologia do empreendedorismo.

After introducing the reader to the main areas covered, such as the context, methodology and literature review, a macroeconomic overview of the geographical areas comprised is provided, thus encompassing a specific study on the current status of Italian economy and demand for Italian goods. The study introduces the reader to the Italian fashion industry, its main features and current performance, already evidencing the quest for internationalization. The conclusions arising from the macroeconomic analyses are used as introduction to the overview of the Italian fashion industry, an industry that strongly represents the “Made in Italy” abroad. The analysis of the industry encompassing history, main features and current status will then suggest that internationalization is the most viable way for SMEs to recover from the turbulent years of the crisis. Among the vast set of geographical options that the SMEs may embrace internationalization, this study aims to provide two tangible market analyses: the Russian and the Brazilian fashion industry. The analysis, based on the framework of international responsiveness by Bartlet and Ghoshal (1989), presents the results of a set of researches and interviews conducted in Russia and Brazil, in the form of a comparative analysis of the two target markets. The analysis will evidence market-, cost-, competitive- and legislation drivers that justify the internationalization process in both markets. The results will lead to conclusion and recommendations that the two markets represent two very different opportunities for SMEs of Italian fashion industry. / Este estudo tem como objetivo desenvolver uma análise comparativa do potencial de internacionalização na Rússia e no Brasil para as PME italianas que operam na indústria da moda. Depois de apresentar ao leitor as principais áreas cobertas, tais como, o contexto, a metodologia e revisão da literatura, é fornecido um panorama macroeconômico das áreas geográficas composto, englobando um estudo específico sobre o estado atual da economia e da demanda para os bens italianos. O estudo, introduzindo o leitor na indústria de moda italiana, suas principais características e o desempenho atual, já evidencia a busca pela internacionalização. As conclusões decorrentes das análises macroeconômicas funcionam como introdução à visão geral da indústria de moda italiana, uma indústria que representa, fortemente, o 'Made in Italy' no exterior. A breve análise da história desta indústria, principais características e situação atual irão, então, sugerir que a internacionalização é o caminho mais viável às PME, para se recuperarem dos anos turbulentos da crise. Entre o vasto conjunto de opções geográficas que as PME têm para abraçar internacionalização, este estudo tem como objetivo fornecer duas análises sobre a indústria da moda: o mercado russo e o brasileiro. A análise, com base no quadro de capacidade de ‘resposta internacional’ proposto por Bartlet e Ghoshal (1989), apresenta os resultados de um conjunto de pesquisas e entrevistas realizadas no Brasil, na Itália e na Rússia, sob a forma de uma análise comparativa dos dois mercados-alvo. A análise evidenciará os drivers de mercado, custo, competitividade e legislação que justificam o processo de internacionalização em ambos os mercados. Os resultados levam à conclusão e às recomendações que os dois mercados representam duas oportunidades muito diferentes para as PME da indústria da moda italiana.
