INTRODUÇÃO: Embora os corticosteróides sejam considerados tratamento padrão-ouro na asma, pacientes com asma grave não são totalmente controlados com este tratamento. Estudos prévios com inibidores da Rho quinase sugeriram uma influência benéfica destas drogas na asma, atuando como uma possível alternativa anti-inflamatória. No entanto, não há estudos anteriores avaliando os efeitos destes inibidores, associados ou não com corticosteróides, na modulação da mecânica do sistema respiratório e oscilatória do tecido pulmonar distal, assim como nas alterações histopatológicas, em modelo animal de inflamação pulmonar crônica. OBJETIVOS: Avaliar se o tratamento com o inibidor específico da Rho (Y- 27632), associado ou não com a dexametasona modula a resposta de mecânica pulmonar, inflamatória, de remodelamento da matriz extracelular e ativação do estresse oxidativo em cobaias com inflamação alérgica crônica. MÉTODOS: As cobaias receberam sete inalações de ovoalbumina (1-5mg/ml; grupo OVA) durante 4 semanas. A partir da quinta inalação, os animais do grupo da Rho quinase receberam inalação de Y-27632 (1mM) (grupo OVA-RHO) e ou dexametasona (2 mg.kg-1) associada ou não a Y-27632 (grupos OVA-C ou grupos ORC), 10 minutos antes de cada inalação com OVA. Setenta e duas horas depois da sétima inalação, os animais foram anestesiados, exsanguinados, o óxido nítrico exalado foi coletado e a mecânica do sistema respiratório (Ers e Rrs) e oscilatória do tecido pulmonar distal (Et e Rt) foram realizadas em condições basais e após desafio com OVA (0,1%). Após, as fatias de pulmão foram removidas e submetidas à avaliação histopatológica. RESULTADOS: Houve um aumento no óxido nítrico exalado, nas respostas máximas de Ers, Rrs, Rt e Et após desafio antigênico, nos eosinófilos, nas células positivas para IL-2, IL-4, IL-5, IL-13, IFN-?, iNOS, MMP-9, TIMP-1,TGF- ß, NF?B e no conteúdo do 8-iso-PGF2?, no conteúdo de fibras elásticas, colágenas e de actina em vias aéreas e no parênquima pulmonar distal do grupo OVA comparado ao controle (P<0,05). Nos grupos OVA-RHO, OVA-C e ORC houve uma diminuição em todos os parâmetros comparados ao grupo OVA (P<0,05). A associação do Y-27632 com o tratamento com corticosteróide (grupo ORC) potencializou a atenuação do conteúdo de colágeno e IFN-? na parede das vias aéreas e IL-2, IFN-?, 8-iso-PGF2? e NF-?B no parênquima distal comparado aos grupos OVA-RHO e OVA-C (P<0,05). No grupo ORC houve uma redução nas células positivas para TIMP-1 e eosinófilos no septo alveolar comparado ao grupo OVA-C (P<0,05). CONCLUSÕES: A inibição da Rho quinase ou o tratamento com corticosteróides contribuíram para o controle da resposta de mecânica pulmonar, da resposta eosinofílica e linfocitária Th1 e Th2, do remodelamento da matriz extracelular e da ativação do estresse oxidativo nas vias aéreas e parênquima distal neste modelo animal. A associação do inibidor da Rho quinase com o corticosteróide potencializou o controle de parte da resposta de remodelamento e de inflamação nas vias aéreas e parênquima. A inibição da Rho quinase associada ou não a corticosteróides pode ser considerada uma ferramenta farmacológica futura para o tratamento de doenças pulmonares crônicas / INTRODUCTION: Although corticosteroids are considered gold standard of asthma treatment, patients with serious asthma are not totally controlled with this treatment. OBJECTIVES: To evaluate whether treatment with the specific Rho inhibitor (Y-27632), combined or not with dexamethasone, modulates the responses of pulmonary mechanics, inflammation, extracellular matrix remodeling, and oxidative stress activation in guinea pigs with chronic allergic inflammation. METHODS: Guinea pigs received seven ovalbumin inhalations (1-5mg/ml; OVA group) during 4 weeks. After the fifth inhalation, the animals of the Rho-kinase group received inhalation of Y-27632 (1mM) (OVA-RHO group) and/or dexamethasone (2 mg.kg-1), combined or not with Y-27632 (OVA-C groups or ORC groups), 10 minutes before each inhalation with OVA. Seventy-two hours following the seventh inhalation, the animals were anesthetized, exsanguinated, the exhaled nitric oxide was collected, and mechanics of the respiratory system (Ers and Rrs) and oscillation of the distal lung tissue (Et e Rt) were performed in basal conditions and after challenge with OVA (0.1%). Afterwards, lung slices were removed and submitted to histopathological evaluation. RESULTS: There was an increase of exhaled nitric oxide, maximum responses of Ers, Rrs, Et and Rt after antigen challenge, eosinophil counts, cells positive for IL-2, IL-4, IL-5, IL-13, IFN-?, iNOS, MMP-9, TIMP-1,TGF-ß, NF?B, and in the content of 8-iso-PGF2?, elastic fibers, collagen fibers and actin in the airways in distal lung parenchyma of the OVA group, compared to the control (P<0.05). In the OVA-RHO, OVA-C and ORC groups there was a decrease in all parameters, when compared to the OVA group (P<0.05). The treatment combining Y-27632 with corticosteroid (ORC group) maximized the attenuation of the content of collagen and IFN-y in the airways walls, and of IL-2, IFN-?, 8-iso-PGF2? and NF-?B in distal parenchyma, when compared to the OVA-RHO and OVA-C groups (P<0.05). In the ORC group, there was a reduction of cells positive for TIMP-1 and eosinophils in the alveolar septum, compared to the OVA-C group (P<0.05). CONCLUSIONS: Rho kinase or treatment with corticosteroids contributed to the control of the pulmonary mechanics response, Th1 and Th2 lymphocyte and eosinophil responses, extracellular matrix remodeling, and activation of the oxidative stress in the airways and distal parenchyma of this animal model. The combined treatment with Rho kinase and corticosteroid maximized the control of part of the remodeling response and inflammation of the airways and parenchyma. Rho-kinase inhibition combined or not with corticosteroids can be considered a future pharmacological tool for treatment of chronic pulmonary diseases

The forgotten Air Force : the establishment and employment of Australian air power in the North-Western area, 1941-1945

The air campaign conducted by the RAAF in the North-Western Area during the Second World War has been largely ignored by historians yet it contributed significantly to the outcome of the Pacific war. This thesis sets out to discuss the campaign by considering various factors that impacted on the RAAF in the lead up to and during the course of the Pacific war and their relevance to the campaign. It looks at the way air operations were conducted in the North-Western Area between 1942 and 1945 and describes the role played by the flying squadrons based in the area. Using primary sources such as operational record books, documents and files at archives and libraries and interviews with veterans and experts the thesis found that the campaign was conducted in several phases. It started with the defence of Darwin. In keeping with overall allied strategy the RAAF then went on an offensive into what was then the Netherlands East Indies (NEI) using medium and heavy bombers and mine laying sea planes flying from bases in Australia???s north west. The NEI was vital to the Japanese war effort as a source of essential raw materials such as oil, timber, and rubber. To defend this part of their new empire the Japanese had amassed large military garrisons on the islands. The vessels used to transport troops and materials became the most important targets for the RAAF???s bomber squadrons. As General MacArthur???s forces advanced along the north coast of New Guinea the North-Western Area based units conducted raids into the NEI to deceive the Japanese into thinking an invasion would be launched from Darwin. As the New Guinea campaign gained momentum the RAAF???s task was to protect its western flank, to prevent the Japanese from moving troops and aircraft east to the Philippines. The thesis concludes the campaign was successful because Darwin was defended, it denied the Japanese vital materials for the conduct of the war and it kept hundreds of aircraft and tens of thousands of troops away from the allied advance.

Fair trade coffee supply chains in the highlands of Papua New Guinea: do they give higher returns to smallholders?

This research focussed on Fair Trade (FT) coffee supply chains in Papua New Guinea. Three research questions were asked. First, do small holders in the FT chains receive higher returns than the smallholders in the conventional chains? Secondly, if smallholders in the FT coffee chains receive higher returns from their coffee than the smallholders in the conventional chains, what are the sources of these higher returns? Finally, if smallholders in the FT chains don't receive higher returns than in the conventional chains, what are the constraints to smallholders receiving higher returns from the FT coffee chains than the conventional chains? A conceptual framework for agribusiness supply chain was developed that was used to guide the field work. A comparative case study methodology was selcted as an appropriate method for eliciting the required information. Four case study chains were selected. A paired FT and conventional coffee chains from Okapa and another paired FT and conventional chains from Kainantu districts, Eastern Highlands Province were selected for the study. The research found that smallholders in the FT chains and vonventional chains receive very similar prices for their coffee (parchment price equivalent). Hence, there was no evidence that smallholders in the FT chains received higher prices or returns from their coffee production than smallholders in conventional chains. This study also found that there was no evidence of FLO certification improving returns to smallholders in the FT chains over those returns received in the conventional chains, but the community that the FT smallholder producers come from did benefit. The sources of these community benefits lies in the shorter FT chains and the distributions of the margin that would have been otherwise made by processors to producers, exporters and the community. In addition, this study found that constraints associated with value creation are similar in all the four chains studies. However, there are some added hurdles for the FT chains in adhering to FT and organic coffee standards. Moreover, FT co-oeratives lacked capacity to trade and their only functions were to help with FLO certification and distribute the FT premium to the community. The findings of this research support some aspects of the literature, but not others. The research contribution is the finding that in this period of high conventional coffee prices, returns to smallholders from FT chains were no bettter than the returns gained in conventional chains, which leads to oppotunism and lack of loyalty by smallholders in the FT chains. The other contribution of this research is in identifying a particular type of free rider who is not a member of the FT co-operative but has right to the community benefits generated by the FT chain.

L’impact d’une diète néonatale déficiente en nutriments essentiels à la défense antioxydante sur le métabolisme énergétique à long terme

Les prématurés subissent un stress oxydant qui résulte d’une défense antioxydante faible et/ou d’une charge oxydante. Des données suggèrent qu’un stress oxydant peut affecter le métabolisme énergétique et mener au syndrome métabolique. Hypothèse: Une faible défense antioxydante tôt dans la vie est suffisante pour affecter le métabolisme énergétique à long terme. Méthodes: Quatre groupes de cobayes (n=21) ont reçu entre leurs 3e et 7e jours de vie une diète standard (C-1sem, C-14sem) ou une diète déficiente (DC-1sem, DC-14sem). À 7 jours, les groupes C-1sem et DC-1sem ont été sacrifiés, le plasma et le foie collectés. Les groupes C-14sem et DC-14sem ont reçu la diète standard jusqu’à 14sem de vie. La glycémie et les triglycérides plasmatiques ont été mesurés à 1, 3, 11, et 13-14sem. La tolérance au glucose a été évaluée à 13sem. Les antioxydants hépatiques et les protéines régulant le métabolisme énergétique ont été analysés à 1 et 14sem. Résultats: Un statut redox oxydé du glutathion était associé avec la diète déficiente et était maintenu oxydé au moins jusqu’à 14sem (p<0.01). Les faibles niveaux de triglycérides plasmatiques et de glycémies, ainsi qu’une meilleure tolérance au glucose à 14sem (p<0.05) étaient associés avec un statut redox plus oxydé. Conclusion: Le faible taux de glutathion observé chez les prématurés a été reproduit dans notre modèle. Puisque nos données suggèrent un rôle protecteur d’un redox plus oxydé et que l’environnement redox est un important régulateur métabolique influençant le développement, il faudrait faire attention avant d’initier des traitements antioxydants agressifs chez les prématurés. / Preterm infants are faced to oxidative stress resulting from a low antioxidant defence and/or a high oxidant load. Datas suggest that an oxidative stress may impair energy metabolism leading to metabolic syndrome development. Hypothesis: A weak antioxidant defence early in life such as observed in preterm newborns is sufficient to impair energy metabolism later in life. Methods: Four groups of guinea pigs (n=21) received between their 3rd and 7th days of life a control diet (C-1week, C-14weeks) or antioxidant deficient diet (DC-1week, DC-14weeks). At 7 day-old, 1week-groups were sacrificed for plasma and liver sampling whereas 14week-groups were fed with the control diet until 14 week-old. Blood glucose and plasma triacylglycerol were determined at 1, 3, 11 and 13-14 week-old. Glucose tolerance test was performed at 13 week-old. Hepatic antioxidant defences and key proteins regulating lipid and glucose metabolism were measured at 1 and 14 weeks. Results: The oxidized redox status of glutathione associated with the neonatal deficient diet was maintained until at least 14 week-old (p<0.01). The low plasma triacylglycerol, low blood glucose and better tolerance to glucose at 14 weeks (p<0.05) were associated with an oxidized redox status. Conclusion: The low glutathione observed in newborn preterm infants has been reproduced in our animal model. Since the oxidized redox state observed here seems to be protective against impaired energy metabolism and since the cellular redox environment is known to be an important rheostat of metabolism influencing development, it suggests being careful before adopting aggressive antioxidant treatments in preterm infants.

As General MacArthur???s forces advanced along the north coast of New Guinea the North-Western Area based units conducted raids into the NEI to deceive the Japanese into thinking an invasion would be launched from Darwin. As the New Guinea campaign gained momentum the RAAF???s task was to protect its western flank, to prevent the Japanese from moving troops and aircraft east to the Philippines. The thesis concludes the campaign was successful because Darwin was defended, it denied the Japanese vital materials for the conduct of the war and it kept hundreds of aircraft and tens of thousands of troops away from the allied advance.

This thesis is concerned with the cultural shifts that have occurred in Lihir, Papua New Guinea, as Lihirians were drawn into greater engagement with the capitalist system, initially through the colonial labour trade and more recently through large-scale resource extraction. This research draws upon 15 months of fieldwork in the Lihir Islands from 2003 to 2004. This thesis is intended as a critical dialogue with world system theorists.World systems arguments are constructive for understanding how Lihirians have remained economically marginal.However, I reject the assertion commonly propounded in these approaches that the world capitalist system inevitably destroys ‘traditional’ cultures and remakes them to its own specifications. Working from Sahlins’ (1985, 1992) premise that there is always continuity in change, I have sought to illustrate those enduring structures and received cultural values that have shaped Lihirian engagement with the capitalist system. My concern iswith articulation rather than penetration; to capture the dialectic of global structural inequalities and Lihirian selective appropriation. This approach allows me to emphasise the heterogeneity of Lihirian culture, notonly prior to sustained European contact, or even mining activities, but specifically at the height of their engagement with the capitalist system. The expression of identity in Equatorial Guinean narratives (1994 - 2007)

Equatorial Guinea is the only former Spanish colony in Africa south of the Sahara. Consequently, the Spanish-language literature produced by its authors has been resistant to classification in both the fields of Hispanic and African literary studies. This thesis examines a selection of contemporary narratives written between 1994 and 2007 by the following authors: José Fernando Siale Djangany, Maximiliano Nkogo Esono, Juan Tomás Ávila Laurel and Joaquín Mbomio Bacheng. My main objective in this dissertation is to identify, explain and relate the ways in which post-independence authors express identity in their respective texts. In order to accomplish this task, this thesis posits situational interactions as the key sites for identity expression. Developed from the tenets of symbolic interactionism, the syncretic theoretical model of identity views it as telescopic. It is expected that, through the examination of the chosen texts, a contribution can be made to the understanding of the way in which each author expresses identity and can therefore feed into the larger discussion of identity in Equatorial Guinean narrative.

A difícil Mistida guineense : nação e identidade da Guiné-Bissau através da triologia de Abdulai Sila

Após mais de cinco séculos de dominação e exploração portuguesa, a Guiné-Bissau, pequeno país localizado na costa ocidental da África, assume a difícil tarefa de constituir-se como nação e, dessa forma, forjar sua identidade nacional. Acreditando que a literatura apresente papel singular em tal engenho, o presente trabalho busca, através da trilogia do autor do primeiro romance nacional, Abdulai Sila, analisar como essas complexas construções vêm-se alicerçando e erguendo. Os romances A Última Tragédia (1995), Eterna Paixão (1994) e Mistida (1997), por tratarem de diferentes períodos do território em questão, da colonização ao pós-independência, possibilitam uma análise abrangente e elucidativa, além de lançarem mão da esperança como característica principal, indo de encontro à difícil realidade do pós-independência. As ideias, sobretudo, de Homi Bhabha, Edward Said e Stuart Hall, a respeito de cultura, nação, identidade, pós-colonialismo, constituem o cerne teórico e analítico dessa dissertação. Sob a óptica dos Estudos Culturais, por meio do entrecruzamento entre o discurso histórico e ficcional, o hibridismo pressuposto faz-se nítido. Às riquezas e belezas da cultura africana, expressas pelas diversas e diferentes etnias, unem-se aspectos culturais lusos, assim como elementos advindos e consequentes da realidade global e interconectada do mundo contemporâneo. Assim, a nação e a identidade guineense, ainda em processo de construção, já deixam entrever sua característica híbrida, em que o tradicional e o moderno conjugam-se e entrelaçam-se igualmente. / After more than five centuries of portuguese domination and exploration, the Guinea-Bissau, small country located in the occidental coast of Africa, assumes the difficult task to constitute itself as nation and, in this way, to forge its national identity. Believing that literature presents singular paper in such device, the present work searches, through the author‟s trilogy of the first national romance, Abdulai Sila, to analyze how these complex constructions are founded. The romances A Última Tragédia (1995), Eterna Paixão (1994) e Mistida (1997), for dealing with different periods of the territory in question, from colonization to post-independence, they make possible an including and elucidative analysis, beyond making use of hope as main characteristic, opposing to the difficult reality of post-independence. The analytical and theoretical core of this dissertation is constituted by the Homi Bhabha, Edward Said and Stuart Hall‟s ideas about nation, identity and post-colonialism. Through the point of view of the Cultural Studies, crossing historical speech and ficcional speech, the supposed hybridism becomes clear. To the wealth and beauties of the African culture, expressed by the ethnic differences, portuguese cultural aspects are joined, as well as consequents elements of the global reality of the contemporary world. Thus, the nation and the identity of Guinea-Bissau, still in construction process, allow already to see its hybrid characteristic, where the traditional and the modern are equally conjugated and interlaced.
