Diferentes estratégias do uso de sorgo para frangos de corte: desempenho e saúde intestinal / Different strategies of using of sorghum for broilers: performance and intestinal health

O objetivo do presente estudo foi avaliar o desempenho, saúde intestinal e metabolizabilidade de dietas para frangos de corte alimentados com o uso contínuo ou mudança brusca de diferentes rações à base de milho e sorgo moído ou inteiro. No Exp. 1 foi estudada a mudança no tipo de grão com as dietas: M100% (ração à base de milho); S100% (ração à base de sorgo); M:S50% (ração à base de 50% milho e 50% sorgo); PS-M (ração à base de sorgo na fase pré-inicial e milho nas demais fases); PM-S (ração à base de milho na fase pré-inicial e sorgo nas demais fases). No Exp. 2 foi estudada a mudança na forma do grão de sorgo: Sm100% (ração à base de sorgo moído); Si100% (ação à base de sorgo inteiro); PSm-Si (ração à base de sorgo moído na fase pré-inicial e inteiro nas demais fases) e PSi- Sm (ração à base de sorgo inteiro na fase pré-inicial e moído nas demais fases). Os experimentos foram conduzidos em um delineamento em blocos casualizados (blocos no tempo) para avaliar o desempenho e epitélio jejunal (r=8), microbiota intestinal (r=4) e metabolizabilidade das dietas (r=10). As mudanças entre milho e sorgo não alteraram o desempenho, epitélio jejunal nem a metabolizabilidade das rações, mas influenciaram a porcentagem de Clostridium, Weissella, Bacillus e Alkaliphilus no intestino delgado e Lactobacillus e Desulfotomaculum nos cecos. O uso de sorgo inteiro piorou o desempenho aos sete dias. Aos 40 dias, Sm100% e PSm-Si apresentaram desempenhos semelhantes, PSi-Sm apresentou menor ganho de peso, mas melhor conversão alimentar e Si100% apresentou o pior desempenho. Si100% e PSm-Si apresentaram aumento no peso relativo da moela e na metabolizabilidade das rações, assim como diminuição de Clostridium e aumento de bactérias das famílias Actinomycetales e Bacillales no intestino delgado, Si100% resultou em menor porcentagem de Alkaliphilus e Enterococcus que Sm100% nos cecos. Conclui-se que a melhor estratégia de uso do sorgo é a substituição total de milho por sorgo moído ao alojamento com posterior mudança para sorgo inteiro, pois não afeta o desempenho e epitélio jejunal das aves, melhora a metabolizabilidade das rações e potencializa a redução de Clostridium no intestino delgado de frangos de corte. / The aim of this study was to evaluate performance, intestinal health and metabolizability of diets in broilers fed different corn- or sorghum (ground or whole)- based diets, continuously or with abrupt change between the diets. Whole sorghum resulted in decreased performance at seven days of age. At 40 days, Gs100% and PGs-Ws showed similar performance, PWs-Gs showed lower weight gain and the best feed conversion rate, and Ws100% showed the worst performance. Ws100% and PGs-Ws resulted in the biggest gizzard relative weight and the highest diet metabolizability values, as well as the lowest level of Clostridium and highest level of Actinomycetales and Bacillales in the small intestine. Ws100% showed lower level of Alkaliphilus and Enterococcus than Gs100% in the caecum. The best strategy to use sorghum in broilers diets is replacing 100% of corn for ground sorghum since the first day followed by change to whole sorghum, because this diet did not affect performance or jejunal epithelium, improved diet metabolizability values, and reduced Clostridium in the small intestine of broilers.

Impact de la nutrition périnatale sur la santé intestinale / Influence of early nutrition on gut health

La période des 1000 premiers jours de vie constitue une fenêtre de sensibilité au cours de laquelle l’environnement peut moduler le développement du foetus et du nourrisson, et conditionner la santé tout au long de la vie. Les conséquences à long terme de l’environnement périnatal sur le risque de maladies intestinales sont toutefois encore peu connues. L’objectif de ce travail était de montrer l’impact de l’environnement nutritionnel précoce sur la maturation intestinale et la santé intestinale à long terme.Un retard de croissance postnatal (RCPN) était induit chez la souris FVB/NRj par augmentation de la taille des portées. Le RCPN était responsable d’un retard de maturation de l’intestin chez la souris au sevrage, en particulier de la barrière intestinale, caractérisé par une augmentation de la perméabilité intestinale, concomitante d’une désorganisation des protéines des jonctions serrées. Le microbiote intestinal était moins riche en espèces bactériennes chez la souris au sevrage en cas de RCPN et sa composition était différente, avec en particulier une proportion anormalement élevée de Parabacteroides spp, Enterococcus spp, Erysipelatoclostridium spp, Eubacterium coprostanoligenes spp, Staphylococcus spp, et Escherichia-Shigella spp, et une plus faible proportion d’espèces productrices de butyrate. L’absence de barrière intestinale efficace et la dysbiose induites par le RCPN, étaient associées à une altération de la réponse inflammatoire de l’intestin à l’âge adulte, caractérisée par une augmentation des réponses immunitaires Th1, Th17 et Treg et une plus grande susceptibilité à la colite chimiquement induite.Ce travail démontre l’importance de l’environnement nutritionnel précoce dans la programmation de la santé intestinale au cours de la vie, et conforte l’hypothèse d’une origine développementale des maladies intestinales chroniques. / The first thousand days of life are a critical time for the development of both the fetus and the infant, and can modify the risk profile for diseases in later life. However, the longterm consequences of the perinatal environment on the susceptibility to intestinal disorders have not yet been assessed. The aim of the present study was to investigate the impact of the early nutritional environment on intestinal maturation and gut health in later life.Postnatal growth restriction (PNGR) was induced in FVB/NRj mice during the suckling period by adjusting the litter size. PNGR delayed intestinal maturation in pups at weaning. PNGR was associated with a maturation delay of the intestinal barrier, characterized by an increased intestinal permeability and impaired tight junctions. At the same time, PNGR affected gut bacterial colonization. Pups with PNGR harbored a decreased bacterial diversity, higher Parabacteroides spp, Enterococcus spp, Erysipelatoclostridium spp, Eubacterium coprostanoligenes spp, Staphylococcus spp, and Escherichia-Shigella spp, and lower butyrate producers. The lack of an efficient intestinal barrier and the dysbiosis induced by PNGR were associated with an altered intestinal inflammatory response in adult mice, characterized by an increase of Th1, Th17 and Treg immune responses, and a higher susceptibility to chemically induced colitis.Our data emphasize the importance of the early nutritional environment in programming of gut health in later life, and support the hypothesis of the developmental origin of chronic intestinal disorders.

Caractérisation de flores microbiennes intestinale humaine et fromagère par méthode de métagénomique quantative / Characterization of human intestinal microbiota and cheese microbiota by quantitative metagenomic method

La flore microbienne est un ensemble de micro-organismes comme les bactéries, archées, eucaryotes inférieurs et virus, jouant un rôle important dans l’équilibre d’un écosystème. Cette flore reste encore mal définie car en 2012, seules ~30% des micro-organismes de la flore intestinale humaine étaient caractérisés, et moins de 50% des micro-organismes de la flore fromagère traditionnelle étaient caractérisés au niveau fonctionnel. Depuis 2006, les séquenceurs à ADN de seconde génération permettent d’analyser directement le contenu génique d’un prélèvement de flore sans contrainte d’isolement ou de culture. Toutefois, les séquences d’ADN générées ne sont pas structurées en génome et sont hautement fragmentées, freinant considérablement l’analyse et l’exploitation de ces données. Dans ce travail, nous avons développé de nouvelles méthodes dites de métagénomiques quantitatives, car elles permettent de regrouper les courtes séquences d’ADN ayant la même abondance au sein de plusieurs échantillons métagénomiques, pouvant provenir d’une même espèce microbienne.Au niveau du microbiote intestinal humain, nous avons structuré un catalogue de 3,9 millions de gènes de la flore intestinale humaine en 741 unités ou « clusters » correspondant à des génomes de bactéries, d’archées et d’eucaryotes, que nous appelons espèces métagénomiques (MGS) et 6640 unités correspondant principalement à des génomes de virus, plasmides et divers ilots génomiques comme des CRISPR, que nous appelons unités métagénomiques (MGU). Cette méthode de structuration a ensuite été utilisée pour faciliter des analyses d’associations de la composition de la flore intestinale avec des maladies humaines comme la maladie de Crohn, l’obésité ou le diabète de type 2. Enfin, au niveau des flores alimentaires, nos méthodes ont été utilisées pour constituer un catalogue de 134 génomes d’espèces bactériennes fromagères et caractériser la flore de surface de fromages traditionnels. Ceci nous a permis de détecter la présence de nouvelles bactéries alimentaires, pouvant enrichir la liste des bactéries à possible intérêt technologique dans les produits laitiers fermentés. / The microbial flora is a micro-organism complex containing for example bacteria, archaea, lower eukaryotes and viruses, which play an important role in ecosystem equilibrium. This flora remains poorly defined as in 2012, only ~30% of the intestinal flora micro-organisms have been characterized, and less than 50% of traditional cheese floras were characterized at the functional level. Since 2006, the second generation of DNA sequencers have allowed the direct analysis of the genetic content from a microbial flora sample without isolation or culture limitation. However, the DNA reads generated are not structured with respect to genomes and also are highly fragmented, slowing down dramatically the exploitation and analysis of these data.In this work, a new methodology based on quantitative metagenomic are described., This allows the clustering of short DNA sequences with the same abundance in multiple metagenomic samples, which should originate from the same microbial species. A 3.9 million gene catalog has been built from the human intestinal tract microbiota and divided into 741 units or clusters corresponding to bacteria, archaea and eukaryote genomes. These have been defined as metagenomic species (MGS) and 6640 units of them corresponds mainly to viral genomes, plasmids and genetic islands like CRISPR, with the sub-name of metagenomic units (MGU). This methodology was then used to facilitate the association analysis of the intestinal flora composition with human diseases such as Crohn’s disease, obesity or type 2 diabetes. Within, the alimentary flora, our methods have also been used to constitute a 134 genomic catalog of cheese bacteria and characterize them from the surface of traditional cheeses. This allowed the detection of new alimentary bacteria, that will enriched the list of bacteria with potential interest for the commercial exploitation of fermented products.

The study was conducted at the Swine Federal Institute of Education Science and Technology of South of Minas Gerais Campus Muzambinho. One design was used in a randomized block design with four treatments and six replications and three animals per experimental unit. The initial weight was used as a criterion for constitution of blocks and treatments consisted of four powdered egg levels (0.0%, 2.0%, 4.0% and 6.0%). The results were evaluated using the linear regression analyzes and polynomial, and the adjustments evaluated for accuracy (R2). With the increase of egg powder residue levels there was an improvement in animal performance. In general, with increasing levels of powdered egg residue there was an increase in final weight of the animals, weight gain and an improvement in feed conversion. Regarding the blood components, there was a reduction on packed cell volume and total protein of seguimentados with raising the level of egg powder residue. The egg powder residue improves the performance of piglets weaned at 28 days of age. As one increases the inclusion of egg residue powder in the diet, there is a higher incidence of beneficial bacteria in tratogastrinteninal piglets, keeping them imonologicamente healthier / Objetivou-se, com a condução do experimento, avaliar diferentes níveis de resíduo de ovo em pó na dieta de leitões desmamados aos 28 dias de idade, avaliando o desempenho, os parâmetros hematológicos, a contagem bacteriana e a quantificação de imunoglobulinas (IgM). O trabalho foi desenvolvido na Suinocultura do Instituto Federal de Educação Ciência e Tecnologia do Sul de Minas Gerais Campus Muzambinho. Estabeleceu-se um delineamento em blocos ao acaso, com quatro tratamentos e seis repetições, sendo três animais por unidade experimental. Usou-se o peso inicial como critério para constituição dos blocos e os tratamentos foram constituídos por quatro níveis de ovo em pó (0,0%; 2,0%; 4,0% e 6,0%). Avaliaram-se os resultados por meio das das análises de regressão linear e polinomial e os ajustes, pela precisão (R2). Com o aumento dos níveis de resíduo de ovo em pó, houve melhora não só no peso final como também no ganho de peso médio diário e na conversão alimentar dos animais. Constatou-se redução do volume globular, dos segmentados e da proteína total com a elevação do nível de resíduo de ovo em pó. Além disso, com o aumento dos níveis do ovo, observou-se maior número de Bifidobacterium e Lactobacillus no cólon proximal dos leitões e menor contagem de Escherichia coli. O resíduo de ovo em pó melhora o desempenho dos leitões desmamados aos 28 dias de idade. À medida que se aumenta a inclusão de resíduo de ovo em pó na dieta, há maior incidência de bactérias benéficas no trato gastrointestinal dos leitões, mantendo-os imunologicamente mais saudáveis.

Virulenzfaktoren von E.coli aus gewaschenen Kolonbiopsien von Patienten mit kolorektalen Neoplasien

Hintergrund: Die Pathogenese nicht-familiärer kolorektaler Neoplasien ist heute noch nicht bekannt. Die Besonderheiten der Epidemiologie der Erkrankung sprechen für die Beteiligung von Umweltfaktoren, wie sozioökonomischer Faktoren, der Ernährung oder der bakteriellen Flora des Darmes. Eine intrazelluläre, von E.coli dominierte Flora wurde in Kolonbiopsien dieser Patienten beschrieben. Methoden: Es wurden Virulenzfaktoren von Escherichia coli untersucht, die aus gewaschenen koloskopischen Biopsien von 43 Patienten mit kolorektalen Adenomen und Karzinomen isoliert worden waren. 100 Stämme wurden mittels PCR auf Gene für folgende Virulenzfaktoren untersucht: s-Fimbrien (sfa), pyelonephritisassoziierter Pilus (pap), Hämolysin A (hlyA), hitzestabiles und -labiles Toxin (ST, LT, EAST), Intimin (eae), Verotoxin (stx), Invasionsplasmid (ipa), cytolethal distending toxin (cdt) und cytotoxic necrotizing factor 1 (cnf1). Für die Kontrollgruppe wurden E.coli aus Biopsien von 55 Patienten mit chronisch-entzündlichen Darmerkrankungen (CED) und unspezifischer Kolitis, von 16 Patienten mit Colon irritabile (IBS) und aus Stuhlproben von 29 gesunden Probanden isoliert und untersucht. Ergebnisse: Bei 69% der Patienten mit kolorektalen Karzinomen und bei 58% der Patienten mit kolorektalen Adenomen wurde mindestens einer der Virulenzfaktoren gefunden, dagegen nur bei 25 bis 39% der IBS- und CED- Patienten sowie der gesunden Probanden (p / Background: Pathogenesis of non-hereditary colorectal neoplasia is poorly understood. Le microbiote intestinal équin joue un rôle important dans le maintien de la santé de l'hôte. Le microbiote intestinal est composé de nombreux micro-organismes tels que les bactéries, les virus, les champignons et les archées. Cependant, la majorité de ces cellules microbiennes sont bactériennes, et par conséquent, de nombreuses études, y compris la présente, se concentrent sur l'exploration des communautés bactériennes dans l'intestin. Un déséquilibre du microbiote intestinal, appelé dysbiose, a été observé dans plusieurs conditions, telles que la colite, après l’administration d'antibiotiques ou la modification du régime alimentaire. La restauration du microbiote peut être effectuée par la transplantation de microbiote fécal (FMT). Les hypothèses de ce projet étaient que le traitement avec une FMT concentrée corrigera la dysbiose plus rapidement qu’une FMT conventionnelle et le véhicule, et que le microbiote intestinal des chevaux traités avec une FMT concentrée ressemblera au microbiote intestinal du cheval donneur. L'objectif de ce projet était de développer un protocole pour améliorer la FMT chez les chevaux, en augmentant la concentration de bactéries présentes dans les selles du donneur par centrifugation, et de le tester chez les chevaux atteints de dysbiose intestinale induite par les antibiotiques. L'antibiotique triméthoprime sulfadiazine (TMS) a été administré à neuf chevaux pour induire une dysbiose intestinale. Les chevaux ont été séparés en trois groupes: les chevaux recevant une FMT concentrée (cFMT, n = 3); les chevaux recevant la FMT fraîche (fFMT), selon les recommandations actuelles (n = 3); et les chevaux recevant un véhicule (VEH) avec 10% de glycérol dans une solution saline à 0,9% (n=3). Des échantillons fécaux ont été prélevés avant et après l'administration du TMS, ainsi qu'avant, pendant et après la transplantation. Le séquençage a été réalisé à l'aide de la plateforme Illumina MiSeq et les données analysées à l'aide du logiciel Mothur. Tel qu’attendu, l'antibiotique TMS a significativement diminué la richesse microbienne chez tous les chevaux. De manière inattendue, la composition des suspensions fécales des donneurs cFMT et fFMT était significativement différente de la composition de base des receveurs cFMT et fFMT, respectivement. La composition du microbiote des chevaux ayant reçu une transplantation fécale (concentrée ou non) était significativement différente après la transplantation, alors que ce n’était pas le cas chez les chevaux ayant reçu le véhicule. En outre, l’abondance relative de Escherichia était significativement plus élevée dans les suspensions fécales du donneur cFMT par rapport aux suspensions fécales du donneur fFMT. Les principales limites de ce projet sont la petite taille des groupes et l'exposition des selles des donneurs à l'oxygène et à la congélation-décongélation. En outre, le modèle de dysbiose peut ne pas être optimal pour tester l'efficacité de la FMT, et des études réalisant la FMT chez les chevaux souffrant de diarrhée sont nécessaire. Cette étude a contribué à la recherche de nouvelles approches pour améliorer la FMT chez les chevaux. Le faible effet mesuré avec les deux protocoles de FMT et l’augmentation de Escherichia démontre que les protocoles actuels doivent être optimisés avant de pouvoir recommander la FMT pour traiter et prévenir la dysbiose chez les chevaux. / The equine gut microbiota plays an important role in maintaining the health of the host. The gut microbiota is composed of many microorganisms such as bacteria, viruses, fungi, and archaea. However, the majority of these microbial cells are bacterial cells, and consequently, many studies, including the present one, focus on exploring bacterial communities in the gut. An imbalance of the gut microbiota, termed dysbiosis, has been observed in several conditions such as colitis, colic, after antibiotic administration, or diet modification. Restoration of the gut to a healthy state can be performed through fecal microbiota transplantation (FMT). Studies using current recommendations for FMT have shown clinical recovery in horses with diarrhea, but the microbiota remains largely unchanged after FMT and no controlled studies have been performed. The hypotheses of this project were that treatment with concentrated FMT will correct dysbiosis faster than conventional FMT and the vehicle, and that the gut microbiota of horses treated with concentrated FMT will resemble the gut microbiota of the donor. The objective of this project was to develop an improved protocol for FMT in horses, by increasing the concentration of bacteria found in the donor stool using centrifugation, and to test it in horses with antibiotic-induced intestinal dysbiosis. The antibiotic trimethoprim sulfadiazine (TMS) was administered to nine horses to induce intestinal dysbiosis. Horses were separated into three groups: horses receiving concentrated FMT (cFMT) (n=3); horses receiving fresh FMT (fFMT), as per current recommendations (n=3); horses receiving a vehicle (VEH) with 10% glycerol in 0.9% saline (n=3). Fecal samples were collected before and after antibiotic administration, as well as before, during, and after transplantation. Sequencing was performed using the Illumina MiSeq platform and data analysed using the software Mothur. As expected, the antibiotic TMS significantly decreased the richness in all horses (P < 0.05). Unexpectedly, the membership of the cFMT and fFMT donor fecal suspensions was significantly different from cFMT and fFMT recipients’ baseline membership, respectively. The membership of the cFMT and fFMT recipient horses was significantly different after transplantation, while the vehicle recipients were not. In addition, the Escherichia genus was found in significantly higher relative abundances in the cFMT donor fecal suspensions when compared to the fFMT donor fecal suspensions. The main limitations of this study are the small sample size and exposure of cFMT donor stool to oxygen and freeze-thawing. In addition, the dysbiosis model may not be optimal to test the efficacy of FMT, and studies performing FMT in horses with diarrhea are warranted. This study contributed to the search for novel approaches to improve FMT in horses. The weak effect of both FMT protocols on the gut microbiota and the increase in Escherichia suggest that further clinical studies are needed before FMT can be recommended to treat and prevent dysbiosis in horses.

Sammanfattning  Bakgrund Vitamin- och mineralbrister hos gravida kvinnor kan leda till missfall och allvarliga störningar i barnets utveckling. Moderns tarmflora överförs med stor sannolikhet till barnet under förlossningen och kan därför innebära ett viktigt steg i utvecklingen av barnets tarmflora. En tänkbar lösning för att säkra ett adekvat intag kan vara konsumtion av kosttillskott och probiotika. I dagsläget finns osäkra uppgifter om hur många gravida kvinnor som intar tillskott.  Syfte Att undersöka hur många gravida kvinnor i Västerbottens län som valde att inta kosttillskott, främst järn och probiotika, samt om det fanns en skillnad mellan olika faktorer och intag.  Metod En kvantitativ tvärsnittsstudie där gravida kvinnor (n=1473) från Northpop-studien i Västerbottens län svarade på ett frågeformulär gällande intag av kosttillskott och faktorer som ålder, utbildning, kostregim etc. De statistiska tester som användes var Chi-2-test, oberoende t-test och Mann Whitney U-test. Materialet analyserades i SPSS. Signifikansnivån sattes till p&lt;0,05.  Resultat Majoriteten av deltagarna svarade att de intog kosttillskott. Faktorer som ökade intaget av kosttillskott hos gravida kvinnor var högre ålder (p=0,030) jämfört med lägre ålder, högre utbildningsnivå (p=0,006) jämfört med lägre utbildningsnivå och vegetarisk/vegansk kost (p=0,021) jämfört med blandkost. Femtiofem procent uppgav att de intog järntillskott. De faktorer som ökade intaget av järntillskott hos gravida kvinnor var vegetarisk/vegansk kost (p=0,001) jämfört med blandkost. Probiotika intogs av 2 procent. Ett högre intag av probiotika sågs hos personer boende i stadsområde (p=0,024) jämfört med övriga boenderegioner samt de som åt vegetarisk/vegansk kost (p=0,001) jämfört med blandkost.  Slutsats Majoriteten av deltagarna intog någon typ av kosttillskott, hälften intog järntillskott och en liten andel intog probiotika. Lågutbildade, yngre, de som äter blandkost och bor utanför stadsområde verkar vara i riskgruppen för att inte inta kosttillskott. / Abstract  Background Vitamin and mineral deficiencies in pregnant women can lead to miscarriage and serious disturbances in children’s development. The intestinal flora of the mother is most likely transmitted to the child during childbirth and may lay the foundation for the child's health. One possible solution to ensure an adequate intake may be the consumption of dietary supplements and probiotics. At present, there is insufficient data on supplement consumption among pregnant women.  Objective The purpose of the study was to examine how many pregnant women in Västerbotten County chose to consume dietary supplements, mainly iron and probiotics, and whether there was a difference between different factors and intake.  Method A quantitative cross-sectional study where pregnant women (n=1473) from the Northpop-study in Västerbotten County responded to a questionnaire regarding consumption of dietary supplements and factors such as age, education, diet etc. The material was analyzed in SPSS with Chi-2-test, independent T-Test and Mann-Whitney U-Test. Using significance level &lt;0.05.  Results The majority of participants, 90 percent, responded that they consumed dietary supplements. The factors that increased the intake of dietary supplements in pregnant women were higher age (p=0.030), higher education (p=0.006) and vegetarian/vegan diet (p=0.021). Iron was reported to be consumed by 804 people, 55 percent. The factors that increased the intake of iron supplement in pregnant women were vegetarian/vegan diet (p=0.001). Probiotics were consumed by 25 people, 2 percent. Living in urban areas (p=0.024) and eating vegetarian/vegan diet (p=0.002) increased consumption of probiotics.  Conclusion The majority of participants chose to consume some type of dietary supplement, half of the participants consumed iron supplements and a small part consumed probiotics. It appears that pregnant women who are low educated, younger, eating an omnivorous diet and living outside urban areas are in the risk zone for not consuming dietary supplements. / Northpop
