L'oeuvre d'Alfredo Molano : sous le signe de l'hybridation générique / The work of Alfredo Molano : under the sign of the generic hybridization

Alfredo Molano, écrivain et sociologue colombien contemporain, remet en question la concentration de la propriété terrienne et la distribution inégale des richesses. L'étude de son oeuvre explore trois axes centraux. Premièrement, la transformation de son écriture, qui évolue d'un format propre au discours scientifique vers le témoignage journalistique et littéraire. Cet écart par rapport à l'objectivisme scientifique entraîne un certain risque de fictionnalisation, que l'écrivain assume, car, d'après lui, il ne porte aucune atteinte à la factualité des récits et favorise en revanche leur lecture et leur diffusion. Nous avons également analysé la technique, chère à l'écrivain, de l' « imputation », consistant en la fusion de plusieurs témoins en un seul personnage, qui garde les traits des individus dont il s'est inspiré. Notre intérêt s'est aussi porté sur l'hybridation générique, puisque le Colombien fait appel aux récits de vie, de voyages et d'apprentissage, autant qu'aux textes sentimentaux, d'action et d'aventures, et à l'essai, entre autres. Cette confluence reflète la complexité d'une société féodale et postmoderne, et permet l'expression d'une variété de témoins hétérogènes, notamment les Amérindiens, les paysans, les activistes politiques, les délinquants et les prisonniers. Ils sont, pour la plupart, victimes de l'inégalité et du conflit permanent entre le centre hégémonique et la périphérie opprimée, deux pôles aussi flous que la violence est tangible, sanglante et multiforme. Alfredo Molano, comme la plupart des témoins, ne s'en tient pas au constat de cette violence, il la dénonce ouvertement et propose des solutions structurelles. Il s'oriente vers la construction d'une « mémoire historique », reconnaissant la souffrance des victimes, et vise une société non seulement hybride, mais surtout juste et égalitaire. / Alfredo Molano, a contemporary Colombian writer and sociologist, calls into question the concentration of land property and the unequal distribution of wealth. The study of his writing explores three main axes. At first, the transformation of his writing, which advances from a scientific discourse to the journalistic and literary testimony. This distance concerning the scientific objectivism carries away some risk of fictionnalisation. The writer assumes because, according to his point of view, his writing has no effect on the factuality of the stories but it is quite easy to read and to circulate. Then, the writer's technique of the "imputation" has been analysed. The author is really attached to. It consists on the fusion of many witnesses into a single character, who keeps the characteristics of people he was inspired by. Finally, our study of his writing is also focused on generic hybridisation, since the Colombian refers not only to the stories of Life, travel and learning, as well as sentimental texts, action and adventures, but also to the essay, among many others. This confluence reflects the complexity of a feudal and postmodern society, and allows the expression of heterogeneous diversity of witnesses, particularly Amerindians, farmers, political activists, offenders and prisoners. Most of them are victims of the inequalities and the continuous conflict between the hegemonic center and the oppressed periphery, two poles as fuzzy as the violence is palpable, bloody and multiform. Alfredo Molano, like most of his witnesses, does not content himself by the recording of this violence, but he denounces it openly and proposes structural solutions. He turns himself towards the development of a "historical memory", recognizing the suffering of victims and targeting a society not only hybrid, but most of all just and egalitarian. / Alfredo Molano, escritor y sociólogo colombiano, critica la concentración de la propiedad de la tierra y la distribución desigual de la riqueza. Este estudio de su obra explora tres ejes principales. Primeramente, la transformación de su estilo, de un formato propio del discurso científico, hacia el testimonio periodístico y literario. Tal distanciamiento frente al objetivismo científico conlleva cierto riesgo de ficcionalización, asumido por el escritor, para quien ésta no afecta de manera alguna el carácter fáctico de los relatos y, en cambio, facilita su lectura y su difusión. Analizamos igualmente la técnica "molanesca" de la "imputación", consistente en fusionar varios testigos en un solo personaje, el cual conserva los rasgos de los individuos en los que se inspira. Nos hemos interesado también por la hibridación genérica, puesto que el colombiano recurre a las historias de vida, a los relatos de viajes y de aprendizaje, así como a textos sentimentales, de acción y de aventuras, y al ensayo, entre otros. Esta confluencia refleja la complejidad de una sociedad tan feudal como posmoderna y permite la expresión de una serie de testigos heterogéneos, como los amerindios los campesinos, los activistas políticos, los delincuentes y los reclusos. Son ellos, en su mayoría, victimas de la desigualdad y del permanente conflicto entre el centro hegemónico y la periferia oprimida, dos polos tan difusos como tangible, sangrienta y multiforme es la violencia. Alfredo Molano, como casi todos sus testigos, no se limita a constatar dicha violencia; la denuncia abiertamente y propone soluciones de fondo. El escritor trabaja en la construcción de una “memoria histórica” y vislumbra una sociedad no sólo híbrida, sino ante todo justa e igualitaria.

Rôles des facteurs de transcriptions SIM1, OTP et POU3F2 dans le développement de l'hypothalamus antérieur

Les facteurs de transcription SIM1, OTP, POU3F2 et ARNT2 interagissent ensemble en orchestrant le développement complexe de l’hypothalamus, une région du cerveau contenant plusieurs petites populations circonscrites de neurones, dont le noyau paraventriculaire (PVN). Ce noyau est un important centre intégrateur et l’haploinsuffisance du facteur de transcription Sim1, essentiel au développement du PVN, mène à l’hyperphagie autant chez la souris que l’homme. Différentes souris ont été générées par génie génétique afin de nous aider à trouver d’autres gènes essentiels qui participent à cette cascade transcriptionnelle. Premièrement, la partie antérieure de l’hypothalamus a été récoltée chez des embryons (E12.5) de souris qui surexprimaient le gène Pou3f2 sous le promoteur de Otp7. L’analyse transcriptionnelle de cette partie a été comparée avec les embryons (E12.5) wt de la même portée, de sorte qu’il a été possible de constater que différents gènes ont été régulés à la hausse et d’autres à la baisse. Les gènes en question ont été choisis selon leur pertinence au développement de la région d’intérêt, l’hypothalamus, et de trois autres critères : un accroissement de plus de 1.5, une expression minimale dans l’embryon d’au moins 1000 lectures et le rang le plus haut. Cette méthode discriminatoire a permis d’identifier les gènes les plus affectés dont Lgi2, Fezf2, Sema3c, Six6, Sox14, Lmo4, Nwd2 et Nkx2.1. Les gènes régulés à la baisse étaient Six6, Sox14 et Nkx2.1, tandis que tous les autres étaient à la hausse. Afin de confirmer les résultats obtenus, une validation par hybridation in situ a été utilisée sur des tranches d’hypothalamus d’embryons E12.5. Nous avons pu confirmer la surexpression des marqueurs Lgi2, Fezf2, Sema3c, Lmo4 et de Pou3f2 dans le domaine du PVN. La diminution de l’expression des marqueurs Sox14 et Nkx2.1 a pu être détectée dans el domaine basal de l’hypothalamus, ce qui suggère que l’effet d’une surexpression de Pou3f2 dans le domaine du PVN ait un effet cellulaire non autonome. La diminution de l’expression de Six6 dans le domaine basal n’a pas pu être confirmer de façon reproductible. Deuxièmement, il semblerait qu’il y ait une redondance de rôle entre les facteurs de transcription Otp et Sim1, tous les deux agissant en amont de Pou3f2. Afin de comparer leur impact dans le programme transcriptionnel, la technologie CrisPR-cas9 a été utilisée pour faire un KO du 2e exon d’Otp. Nous avons pu confirmer notre modèle de mutation en le comparant aux autres mutants de la littérature par une réduction de l’expression d’OT, d’OTP, d’AVP et de TRH. Troisièmement, les souris hétérozygotes pour Otp et Sim1 seront croisées, de sorte d’obtenir quatre génotypes : wt, Otp+/-, Sim1+/tlz+ et Otp+/-Sim1+/tlz+. Puisqu’une redondance des rôles de Sim1 et Otp est soupçonnée, le phénotype du double mutant devrait présenter une obésité par hyperphagie plus importante que celle des souris hétérozygotes pour le gène Sim1 ou Otp. Les souris sont pesées à partir de la 5e semaine de vie jusqu’à l’âge de 6 mois à une fréquence d’une fois par semaine. L’apport calorique est également mesuré une fois par semaine sur période de 24h. La double mutation (Otp+/-Sim1+/tlz+) chez les souris mâles causait un phénotype d’obésité plus important que la singularité des mutations, mais ce n’était pas le cas chez les souris femelles. Les souris portant la mutation d’Otp étaient tout de même plus obèses que les souris sauvages pour les deux sexes. Plus de souris seront nécessaire pour déterminer si un apport calorique sans changement au niveau des dépenses énergétiques est la cause de ce gain pondéral. / The transcription factors SIM1, OTP and POU3F2 interact together to orchestrate the complex development of the hypothalamus, a region of the brain containing several small, circumscribed populations of neurons, including the paraventricular nucleus (PVN). This nucleus is an important integrating center, and the haploinsufficiency of the transcription factor Sim1, essential for the development of PVN, leads to overeating in both mice and humans. Different mice have been genetically engineered to help us find other essential genes that participate in this transcriptional cascade. Firstly, the anterior part of the hypothalamus was collected from mice embryos (E12.5) which overexpressed the Pou3f2 gene under the OTP7 promotor. The transcriptional analysis was compared to wt embryos from the same litter to see the different upregulated and downregulated genes. These genes were chosen according to their relevance to the development of the anterior hypothalamus. Three criteria were used to discriminate genes from one another: 1) an increase of more than 1.5, 2) a minimal expression in the embryo (E12.5) of at least 1000 reads and 3) the highest rank. This discriminatory method allowed us to identify the genes Lgi2, Fezf2, Sema3c, Six6, Sox14, Lmo4, Nwd2, Nkx2.1. To have a visuospatial idea of these affected genes, the validation of these results was done by in situ hybridization on E12.5 embryo hypothalamus. We have been able to see an overexpression in the PVN domain for the Lgi2, Fezf2, Sema3c, Lmo4 and Pou3f2 markers. The reduction of expression for Sox14 and Nkx2.1 markers were visible in the basal domain of the hypothalamus, which suggest a non cell autonomous effect of Pou3f2 being overexpressed. The reduction of Six6 couldn’t be consistently visible with repetition. Secondly, a redundant role of OTP and SIM1 seems to occur in the development of the hypothalamus. We created a KO line of the Otp gene by deleting the second exon with CrispR-cas9 and characterized it. We then compared it to the Sim1+/tlz+ line that we already generated in the lab. We were able to confirm our mutation model by seeing a reduction in the expression of crucial markers such as OT, OTP, AVP and TRH. Thirdly, we crossed Otp+/- with Sim1+/tlz+ mice to obtain four different genotypes: wt, Otp+/-, Sim1+/tlz+ and Otp+/- Sim1+/tlz+. Since a redundant aspect has been observed for SIM1 and OTP transcription factors, we were wondering if the obesity phenotype would be worsened by carrying both mutation or not. These mice were weighted every week from 5-week-old up to 6 months old. Food intake has also been measured since the obesity has been reported to be caused by hyperphagia in Sim1 mutant mice. The male mice carrying the double mutation (Otp+/-Sim1+/tlz+) showed a more important weight gain than only Sim1+/tlz+ or Otp+/- mutants, but it was not the case for the female mice. The mice carrying the Otp mutation still got a more important weight gain than the wt mice (females and males). More mice would be necessary to determine if this weight gain is caused by hyperphagia only or if unbalance energy cost is part of the cause.

Indiana University-Purdue University Indianapolis (IUPUI) / Bone growth can be impaired due to disease, such as osteoporosis. Currently, intermittent parathyroid hormone (PTH) treatment is the only approved therapy in the United States for anabolic bone growth in osteoporosis patients. The anabolic effects of PTH treatment are due, at least in part, to modulation of the Wnt/β-catenin pathway. Activation of the Wnt/ β-catenin pathway using a small molecule inhibitor of GSK3β was previously shown to increase markers of bone formation in vitro. Our study utilized a zebrafish model system to study Wnt activated fin regeneration and bone growth. Wnt signaling is the first genetically identified step in fin regeneration, and bony rays are the main structure in zebrafish fins. Thus, zebrafish fin regeneration may be a useful model to study Wnt signaling mediated bone growth. Fin regeneration experiments were conducted using various concentrations of a GSK3β inhibitor compound, LSN 2105786, for different treatment periods and regenerative outgrowth was measured at 4 and 7 days post amputation. Experiments revealed continuous low concentration (4-5 nM) treatment to be most effective at increasing regeneration. Higher concentrations inhibited fin growth, perhaps by excessive stimulation of differentiation programs. In situ hybridization experiments were performed to examine effects of GSK3β inhibitor on Wnt responsive gene expression. Experiments showed temporal and spatial changes on individual gene markers following GSK3β inhibitor treatment. Additionally, confocal microscopy and immunofluorescence labeling data indicated that the Wnt signaling intracellular signal transducer, β-catenin, accumulates throughout GSK3β inhibitor treated tissues. Finally, experiments revealed increased cell proliferation in fin regenerates following LSN 2105786 treatment. Together, these data indicate that bone growth in zebrafish fin regeneration is improved by activating Wnt signaling. Zebrafish Wnt signaling experiments provide a good model to study bone growth and bone repair mechanisms, and may provide an efficient drug discovery platform.

L’hybride englobe les sphères de représentation de ses éléments-parents tout en les dépassant. Par conséquent, l’hybridation tend vers l’extension de toutes les frontières, littéraires et culturelles, dans le but de nous offrir une vision du monde et du sujet toujours plus complète. Dans Oranges are not the Only Fruit, Sexing the Cherry, Written on the Body et The PowerBook de Jeanette Winterson, l’hybridation se manifeste à tous les niveaux du texte. L’hybridité physique et sexuelle des narrateurs est ainsi une des manifestations de leurs identités et vies plurielles et paradoxales. Ces hybrides incarnés (monstres, travestis ou androgynes) se construisant au fil de leurs histoires, qui s’inspirent toujours de récits antérieurs, narrateurs et narrations se démultiplient conjointement, s’entremêlent et se redéfinissent sans cesse. Le texte fluctue au gré de l’hybridation générique et de l’intertextualité. Therefore, the narrators and their narratives proliferate conjointly, intermix and redefine each other constantly. The shape of the text fluctuates with generic hybridization and intertextuality. Realistic narratives – such as autobiographical, historical or scientific discourses – and fictional ones – such as fairy tales or romances – interact to produce generic hybrids with parodic undertones that represent the nature of the subject, his/her life, reality and truth. Finally, by hybridizing existing texts and personal literary devices, the author elaborates original narratives that rewrite her ancestors’ sexist discourses and reflect how she perceives the individual, the world and art.

CoordenaÃÃo de AperfeiÃoamento de NÃvel Superior / Os begomovÃrus, vÃrus da famÃlia Geminiviridae transmitidos por mosca branca, tÃm emergido como sÃrios patÃgenos de culturas agronÃmicas e hortÃcolas em regiÃes tropicais e subtropicais de muitos paÃses em todo o mundo. A partir da dÃcada de 80, tÃm aumentado os relatos da disseminaÃÃo da mosca branca, Bemisia argentifolii, e de begomovÃrus provocando impacto devastador nas regiÃes em que ocorrem. No Brasil, estes patÃgenos tÃm sido limitantes para a produÃÃo de tomate (Lycopersicon esculentum) em vÃrias Ãreas de cultivo com incidÃncia crescente desde 1994. No presente trabalho, plantas de tomate exibindo sintomas de infecÃÃo provocada por vÃrus como mosaico, clorose internerval, enrolamento do limbo foliar e reduÃÃo do crescimento, foram coletadas em lavouras de tomate indÃstria em AnÃpolis-GO. O vÃrus foi identificado como pertencente ao gÃnero Begomovirus mediante tÃcnica de PCR usando oligonucleotÃdeos especÃficos que amplificaram fragmentos dos componentes A e B do genoma viral. No capÃtulo I sÃo apresentados os resultados da caracterizaÃÃo molecular e no capÃtulo II, os dados da determinaÃÃo do cÃrculo de hospedeiros e da investigaÃÃo da relaÃÃo do begomovÃrus com o vetor Bemisia argentifolii. O isolado denominado GOANPL, foi clonado e parcialmente seqÃenciado tendo sido obtidas as seqÃÃncias nucleotÃdicas dos genes da capa proteica, Rep e de toda a regiÃo intergÃnica, em um total de 2.130 nucleotÃdeos. A anÃlise comparativa das seqÃÃncias revelou que, em geral, o GOANPL possui relacionamento genÃtico distante com begomovÃrus da Ãsia, Europa e Ãfrica sendo mais prÃximo das espÃcies do Brasil, particularmente, com os begomovÃrus identificados em Minas Gerais (TRMV) e no Distrito Federal (DF-BR2). Com este Ãltimo, apresentou alta homologia em todo o genoma podendo vir a constituir, com o mesmo, uma nova espÃcie. A determinaÃÃo do cÃrculo de hospedeiros do GO-ANPL foi realizada inoculando-se 46 espÃcies vegetais pertencentes a nove famÃlias botÃnicas, sob duas modalidades de inoculaÃÃo: mecÃnica e com a mosca branca. Constatou-se que o GO-ANPL infecta, preferencialmente, plantas da famÃlia Solanaceae como Nicotiana benthamiana, Datura stramonium e Nicandra physalodes. O nÃmero de espÃcies infectadas com o inseto vetor foi superior ao obtido pela inoculaÃÃo mecÃnica e diferiu dos resultados obtidos para outros isolados de begomovÃrus de tomate no Brasil. Os testes foram todos confirmados com hibridizaÃÃo com sondas moleculares, em \"dot blot\"No estudo da relaÃÃo vÃrus-vetor, foram investigados o perÃodo de acesso de aquisiÃÃo do vÃrus (PAA), o perÃodo de acesso de inoculaÃÃo do vÃrus (PAI), e o perÃodo de latÃncia do vÃrus na fase adulta do vetor, empregando-se cinco moscas/planta de tomate \'Santa Clara\' em todos os tratamentos. Para a definiÃÃo do PAA e do PAI, foram testados nove diferentes perÃodos de tempo: 0,25, 0,5, 1, 2, 4, 8, 16, 20 e 24 horas. Nos testes para determinaÃÃo do PAA, apÃs cada um desses perÃodos seguiu-se uma inoculaÃÃo de 48 horas e para definiÃÃo do PAI, antes de cada perÃodo antecedeu-se um perÃodo de acesso de aquisiÃÃo fixo de 72 horas. Constatou-se que o PAA mÃnimo da mosca branca foi de apenas 0,25 hora, com o qual foram obtidas 6% de plantas infectadas. O percentual de plantas aumentou de 6 para 65% com a extensÃo do PAA de 0,25 para 24 horas. Com relaÃÃo ao perÃodo de acesso de inoculaÃÃo do vÃrus, foram registrados 18% de plantas infectadas com o PAI de 0,5 hora. O percentual elevou-se para 67% quando 24 horas de PAI foram concedidos. Valores isolados de 90 e 100% na transmissÃo viral, tambÃm foram observados. O tÃrmino do perÃodo latente do vÃrus no vetor ocorreu 16h apÃs a aquisiÃÃo do mesmo em planta infectada, considerando os 3% de infecÃÃo observados nas plantas inoculadas. Os dados obtidos indicam que a interaÃÃo vÃrus-vetor à estabelecida desde a fase inicial de desenvolvimento do inseto. Como parte do estudo dessa interaÃÃo, avaliou-se a presenÃa do begomovÃrus GO-ANPL em todas as fases de desenvolvimento do inseto vetor (ovo, 1 ao 4 Ãnstar e adulto) na planta infectada, em adultos com diferentes PAA, na progÃnie de fÃmeas virulÃferas e em adultos cujos estÃgios ninfais desenvolveram-se em tomateiro infectado. A tÃcnica PCR foi empregada para a detecÃÃo do GO-ANPL em mais de 2.500 espÃcimens testados. O vÃrus foi detectado em ninfas do 1 ao 4 Ãnstar que se alimentaram em plantas de tomate infectada, contudo, em ovos provenientes de avirulÃferas, os quais foram ovipositados em planta infectada e coletados apÃs sete dias, o vÃrus nÃo foi detectado. A transmissÃo à progÃnie foi constatada pela detecÃÃo do vÃrus em ovos, ninfas e adultos que se desenvolveram em planta nÃo hospedeira do vÃrus. A transmissÃo transestadial ocorreu com Ãndice elevado e, ao lado da transmissÃo à progÃnie, indica que a retenÃÃo do vÃrus à uma etapa importante da interaÃÃo vÃrusâvetor. A transmissÃo do vÃrus para mudas de tomate, a partir de adultos da progÃnie de fÃmeas virulÃferas, nÃo foi constatada. Contudo, transmissÃo para tomateiro em um percentual de 33% foi verificado nos casos em que a inoculaÃÃo das plantas foi realizada pelos adultos que retiveram o vÃrus da sua fase imatura (transestadial) / The whitefly-transmitted viruses from the family Geminiviridae, genus Begomovirus, have been reported as an economically important pathogen group that affect important crops in tropical and subtropical countries. Since the beginning of the 1980 decade, the occurrence of the whitefly associated to Begomovirus infection has drastically increased worldwide. In Brazil, these pathogens have been responsable for severe economical losses in tomato (Lycopersicon esculentum) orchards and the production has hampered since 1994. In this work, infected tomato plants showing symptoms, such as mosaic, intervein clearing, leaf curling and growth reduction were collected in tomato orchards in AnÃpolis, State of GoiÃs. The virus was identified as a member of the genus Begomovirus by PCR reaction, using specific primers to amplify fragments of A and B components of the virus DNA genome. The Chapter I of this thesis presents the results of the molecular characterization of the virus and the Chapter II shows the determination of its host range and the relationship with its natural vector Bemisia argentifolii. The virus isolate denoted GO-ANPL was cloned and partially sequenced. Part of the sequenced genome (2.180 nucleotides long) corresponded to the coat protein and Rep genes and comprised the entire intergenic region. Sequence comparison revealed that the GO-ANPL isolate is distantly related to the begomoviruses found in Asia, Europe and Africa, and it is related to other begomoviruses reported in Brazil. The virus isolate showed to be more closely related to viruses found in the State of Minas Gerais (TRMV isolate) and in the Federal District (isolate DF-Br2). The highest homology was observed with the isolate DF-Br2 and it may represent a new specie of the genus Begomovirus. In order to determine the virus host range, 46 plant species from nine different botanical families were mechanically and using the virus vector inoculated. The GO-ANPL isolate preferentially infected plants of the family Solanaceae as Nicotiana benthamiana, Datura stramonium and Nicandra physalodes. The number of infected plants was higher when they were inoculated by the virus vector, and the results were distinct from those obtained for other begomoviruses reported in Brazil. Viruses infections were all confirmed by dot blot hybridization using specific molecular probes to the virus. 4 To study virus/vector interaction, the acquisition access period (AAP), inoculation access period (IAP), and the latent period were determined transfering five whiteflies per plant and using tomato cv. Santa Clara as the host. For the AAP and IAP, nine different time periods were tested: 0.25, 0.5, 1, 2, 4, 8, 16, 20 and 24 h. The minimal AAP determined was 0.25 h, after which, 6% of the tested plants became infected. The number of infected plants increased to 65% with an AAP of24h.Afteran IAP of 0.5 h, 18% of the plants were infected and their number increased to 67% after an IAP of 24 hours. The latent period was considered to be 16 h, after which, 3% of the inoculated plants became infected. The results of AAP, IAP and latent period seem to indicate an early interaction between virus and vector starting at early stages of vector development. The presence of the GOANPL was determined in all stages of the vector (eggs, 1st to 4th instar and adults) in infected plants, in adults under different AAPs, in the progenies of viruliferous females, and in adults originated from nymphs developed from infected plants. More than 2.500 insects were tested by PCR to detect the GO-ANPL isolate. The virus was detected in nymphs from the 1st to 4th instar that had fed in infected plants and no virus was found in eggs from aviruliferous female that had been laid in infected plants. Transmission to the progenies was observed, since the virus was detected in all stages of insect development from eggs to adults. High level transmission was also observed in newly emerged adults that had acess to virus-infected plants as immatures. This fact, in addition to the transmission to the progenies, suggests that virus retention is an important part of virus/vector interaction. No transmission was observed from adults originated from viruliferous females. However, 33% of virus transmission was obtained when adults that retained virus from their early larval stages were employed

Hybridation et dynamique des populations chez les renouées du Japon: Espèces non-indigènes invasives du genre Fallopia Adans. (Polygonaceae) en Belgique / Hybridization and dynamics of populations of the Japanese knotweeds: Alien invasive species of the genus Fallopia (Polygonaceae) in Belgium

The Japanese knotweeds are invasive alien clonal species originating in Asia (Japan, Korea, Taiwan and Nord of China). They were introduced in Europe at the beginning of the 19th century. They are now some of the most troublesome invasive alien species in Europe and in the United States. In Belgium, a complex of four taxa and a hybrid represents them. All these taxa take part in the pattern of invasion and represent an excellent opportunity for studies of population biology in Western continental Europe. The presence from at least three taxa and a hybrid is confirmed by cytological, genetic and morphological studies. Vegetative reproduction is recognized as the main mode of reproduction and expansion of these taxa in the introduce range. However, interspecific hybridization events are observed confirming the restoration of the sexual reproduction by hybridization within this complex species in Belgium. Hybrid F. x bohemica with various ploidy levels from tetraploid to octoploid is observed. An increase in genotypic and morphological diversity is shown at the hybrid F. x bohemica which missed with the parental species. This could increase the potential of Japanese knotweeds to adapt to the new environment and contribute to the invasive success of these taxa in Belgium. Assignment test indicates a genetic pool differentiated at the hybrid F. x bohemica and not a mixture of the genetic pool of the parental species as expected for hybrid taxa. Hybrid F. x bohemica has always been considered as rare in Belgium and of horticultural origin. An analysis of the spatial distribution shows that hybrid F. x bohemica is widespread in Belgium and that its abundance depends on the areas. An analysis of extent of differentiation between groups of hybrid geographically distant could not provide proof of an independent evolution of hybrid populations under limited gene flow. An analysis of the sexual reproduction capacity and dispersal of seeds shows important production of viable seeds and consequent seed rain. Hybrid seeds may be dispersed beyond 16m, leading the possibility of founding new individuals and to contribute to the invasive success of these taxa. However, a trend towards decreasing germination rate is shown after a cold period. An analysis of the distribution at the landscape scale shows that the dynamics of colonization of habitats patches by Japanese knotweeds rising mainly from clonal propagation in spite of important pressure of propagule. The knotweeds prefer human disturbed habitats with a clear prevalence of communication routes. This leads the possibility of dispersing towards the adjacent habitats patches. A high dynamics of establishment of propagule was not observed at the hybrid plants compared with the parents plants in spite of the increase in genotypic diversity and the consequent pressure of propagule. Lastly, proposals for the integrated management of these taxa are proposed in the Belgium context. Their management will have first to identify hybrids and taxa involved. Emasculation and management of the existing clones represent solutions to prevent flowering and expansion of these taxa. An active management of disturbed habitats may represent alternative to prevent the invasion by Japanese knotweeds / Les renouées du Japon sont des espèces clonales non-indigènes originaires dAsie (Japon, Corée, Taiwan et Nord de la Chine). Elles ont été introduites en Europe au début du 19è siècle. Elles font désormais partie des espèces non-indigènes les plus invasives en Europe et aux Etats Unis. En Belgique, elles sont représentées par un complexe de quatre taxons et un hybride. Tous ces taxons participent à la dynamique dinvasion et représentent un modèle dintérêt pour les études de biologie des populations en Europe continentale occidentale. La présence dau moins trois taxons et un hybride est confirmée par des études cytologiques, génétiques et morphologiques. La reproduction végétative est reconnue comme le principal mode de reproduction et dexpansion de ces taxons dans la zone dintroduction. Toutefois, des phénomènes dhybridation interspécifique sont observés confirmant la restauration de la reproduction sexuée par hybridation au sein de ce complexe despèces en Belgique. Lhybride F. x bohemica avec différents niveaux de ploïdie, du tétraploïde à loctoploïde, est observé. Un accroissement de la diversité génotypique et morphologique qui manquait aux espèces parents est démontré chez lhybride F. x bohemica. Ceci pourrait augmenter le potentiel des renouées du Japon à sadapter au nouvel environnement et contribuer au succès invasif de ces taxons en Belgique. Un test dassignation indique un pool génétique différencié chez lhybride F. x bohemica et non un mélange du pool génétique des espèces parentales comme attendu dans le cadre de taxons hybrides. Lhybride F. x bohemica a toujours été considéré comme rare en Belgique et dorigine horticole. Une analyse de la répartition spatiale montre que lhybride F. x bohemica est très répandu en Belgique et que son abondance dépend des régions. Une analyse de létendue de la différenciation entre groupes dindividus hybrides géographiquement distants na pas pu fournir de preuve dune évolution indépendante des populations hybrides sous un flux de gènes limité. Une analyse de la capacité de reproduction sexuée et de dispersion des graines démontre une production importante de graines viables et une pluie de graines conséquente. Les graines hybrides sont capables de se disperser à plus de 16m, laissant la possibilité de fonder de nouveaux individus et de contribuer au succès invasif de ces taxons. Cependant, une tendance à la diminution du potentiel de germination est observée après une période de froid chez ces taxons. Une analyse de la distribution à léchelle du paysage a permis dinterpréter la dynamique de colonisation des taches dhabitats par les renouées du Japon comme relevant principalement de la propagation clonale malgré une pression de propagule importante. Les renouées du Japon préfèrent les habitats perturbés par lhomme avec une nette prédominance des réseaux linéaires de communications. Ceci laisse ensuite la possibilité de se disperser vers les taches dhabitats adjacents. Une dynamique détablissement de propagules plus importante na pas été observée chez les plants hybrides comparées aux plants parents malgré laccroissement de diversité génotypique et la pression de propagules considérable. Enfin, des pistes pour la gestion intégrée des renouées du Japon en Belgique sont proposées. Cette gestion devra en priorité identifier les hybrides et les taxons en présence. Des mesures démasculation et de gestion des clones existants pourraient constituer une solution pour empêcher la floraison et lexpansion de ces taxons. Une gestion active des habitats perturbés pourrait représenter une alternative pour prévenir linvasion par les renouées du Japon

Biological nutrient removal in SBR technology: from floccular to granular sludge

Biological nutrient removal has been studied and applied for decades in order to remove nitrogen and phosphorus from wastewater. However, more anthropogenic uses and the continued demand for water have forced the facilities to operate at their maximum capacity. Therefore, the goal of this thesis is to obtain more compact systems for nutrient removal from domestic wastewater. In this sense, optimization and long-term stabilization of high volume exchange ratios reactors, treating higher volumes of wastewater, have been investigated. With the same target, aerobic granular sludge was proposed as a reliable alternative to reduce space and increase loading rates in treatment plants. However, the low organic loading rate from low-strength influents (less than 1 Kg COD•m-3d-1) results in slower granular formation and a longer time to reach a steady state. Because of that, different methodologies and operational conditions were investigated in order to enhance granulation and nutrient removal from domestic wastewater. / L’estudi de l’eliminació biològica de nutrients s’ha dut a terme durant dècades. Tot i això, la influencia de l’home i l’augment de la demanda d’aigua han forçat a les instal•lacions a treballar a la seva capacitat màxima. Així, l’objectiu de la tesi és obtenir sistemes més compactes per a l’eliminació de nutrients de les aigües residuals. En aquest sentit, s’ha investigat l’optimització i estabilització de reactors amb alts volums d’intercanvi, tractant més aigua. Amb el mateix objectiu, el fang granular aeròbic va ser proposat com una alternativa fiable per tal de reduir l’espai i incrementar les càrregues de les depuradores. Tot i això, la granulació amb influents de baixa càrrega (menors a 1 Kg dQO•m-3d-1) resulta més lenta i més dificultosa alhora d’obtenir l’estat estacionari. Per aquesta raó es van investigar diferents metodologies i condicions d’operació per tal de millorar la granularció i l’eliminació de nutrients de les aigües urbanes.
