Lipid Mobilization In Exercising Salmonids

Turenne, Eric D. January 2018 (has links)
Animals rely on lipids as a major fuel for endurance exercise because they pack more joules per gram than any other fuel. However, in contrast to mammals, information on how the mobilization of lipids from endogenous stores is managed to meet the needs of energy metabolism in swimming fish is sparse. Information on in vivo rates of lipid mobilization in swimming fish has been limited to relatively low exercise intensities and has only been investigated in a single species. Therefore, the goal of my thesis was to address this paucity of information by quantifying lipolytic rate in rainbow trout during graded exercise and fatty acid mobilization in Atlantic salmon during prolonged endurance exercise. In the first part of my work, I hypothesized that like mammals, rainbow trout stimulate lipolysis above resting levels to a peak with increasing work intensity, but subsequently lower its rate at high intensities when ATP production from carbohydrates becomes dominant. To test this hypothesis, I measured the rate of appearance of glycerol (Ra glycerol) in the blood (resulting from the breakdown of triacylglycerol (TAG)) of trout at rest (control) and during graded exercise from rest to Ucrit. Results showed that Ra glycerol in trout averaged 1.24 ± 0.10 µmol kg -1 min-1 and that this rate was unaffected by exercise of any intensity. These experiments revealed that rainbow trout do not modulate lipolysis during exercise. Furthermore, I calculated that baseline lipolytic rate was much higher in trout than in mammals and that this rate is in constant excess of the requirements of energy metabolism. My second investigation focused on measuring fatty acid mobilization in Atlantic salmon. To date, the majority of studies on energy metabolism in salmonids have used rainbow trout as the ubiquitous model for salmonids. I postulated that domesticated rainbow trout may be far less impressive athletes than their wild anadromous form and other salmonids. In this regard, I proposed that studying energy metabolism in Atlantic salmon (even those from aquaculture) may help to deepen our understanding of the physiology of true long-distance migrant fish. To study the effects of prolonged endurance exercise on the mobilization of fatty acids from endogenous stores in these fish, I monitored the rate of appearance of fatty acids (Ra NEFA calculated from Ra Palmitate) in the blood during 72 hours of sustained swimming. I found that contrary to what has been previously described in rainbow trout, Ra Palmitate (and by proxy, Ra NEFA) is reduced by approximately 64% (from 0.75 ± 0.12 µmol kg-1min-1 to 0.27 ± 0.06 µmol kg-1min-1 and from 19.3 ± 7.8 µmol kg-1min-1 to 6.9 ± 2.0 µmol kg-1min-1 for Ra Palmitate and Ra NEFA, respectively) during prolonged endurance exercise in Atlantic salmon. However, like in trout, even this reduced rate of fatty acid mobilization exceeds the requirements of energy metabolism at rest and during swimming. While further experiments will be necessary, I speculated that this reduction in Ra NEFA may be caused by a partial inhibition of lipolysis to reduce the energetic cost of TAG:FA cycling and optimize fuel budgets during prolonged endurance exercise. This thesis provides the first in vivo measurements of lipolysis during graded exercise in salmonids and the first in vivo measurements of fatty acid mobilization in Atlantic salmon. From the results mentioned above, I concluded that salmonids mobilize lipids in constant excess of the requirements for energy metabolism, possibly to allow for rapid reorganization of membrane phospholipids in response to changing environmental conditions. However, more anadromous and migratory phenotypes may rely on a tighter control of lipolysis to minimize the costs of substrate cycling and conserve energy on limited fuel stores.

Sociologie d'une mobilisation partisane modernatrice : la refondation de l'UMP saisie "par le bas" (2004/2008) / Sociology of a partisan “managerial modernization” : the UMP re-foundation in context (2004-2008).

Petitfils, Anne-Sophie 05 November 2012 (has links)
How does a party of government, such as the UMP, which defines itself as modern, professionalized and without ideology, achieve to mobilize widely? At the intersection of a sociology of parties and political mobilization, this study is dedicated to the analysis of the genesis and the effects of the campaign of recruitment which was introduced by Sarkozy’s direction at the UMP from 2004 to 2008 and used a rhetoric of modernity and managerial tools. It shows that the “managerial modernization”, as an institutional ideology, was an essential condition for this mobilization. This result cannot be understood independently to a sociography of the “new” members, who, compared to the members of the former right parties, belong to entrepreneurial groups. The commitment of these new members, whose identity and expectation differ from the members of former parties, caused an infrapolitical resistance of these ones and had paradoxical effects on the institution (an increase of internal tensions, a reactivation of the gaullist and liberal identity and a renewal of member’s ideology). At a theoretical level, this study pleads for a cultural approach of partisan institutions which is paradoxically able to explain the logics that lead parties of government to appear as rational enterprises. Keywords: political parties, mobilization, UMP, commitment, activism, organizational modernization, management, ideology, culture, identity.

A study of volunteering and youth identity in an "exposed" area

Grönhage, Jonas, Marcus, Larsson January 2017 (has links)
The aim of this study is to explore what motivates youth from so called “exposed” areas to commit to voluntary work, and how the youth view their own role and the role of the organisation in a context of social mobilization. In order to answer the research questions of this study, the commitment of youth in a non-profit organization has been studied using several different methods of data collection. In order to measure the function of motivation amongst the youth, the survey Volunteer Functions Index (VFI) has been used. VFI is a tool used to measure motivation based on what functions an activity performs in the life of the individual. In order to achieve a greater understanding and be able to explore aspects that the VFI does not cover, four semi-structured interviews were conducted with current and former members of the organization. In order to better understand the context of the organization and to be able to ask more questions during the interviews, a participant observation was done within the work of the organization. The study shows that the youth that participate in the study perceive their commitment in the organization as an important part of their identity and that the identity is an important aspect of what possible actions are perceived to be available and attractive. It also seems as if the commitment to the organization is tied to the opportunity to change the social conditions for one self and others. The study also shows that the organization has been more successful in achieving social mobilization amongst girls than amongst boys, and that some of the participants attribute this to the gender roles that permeate the area / Denna studie syftar till att undersöka vad som motiverar ungdomar från så kallade utsatta områden till att engagera sig ideellt samt hur ungdomarna uppfattar sin egen och organisationens roll i en kontext av social mobilisering.

Ultrassonografia na validação do escore de condição corporal em vacas holandês-zebu durante o período de transição e fase inicial da lactação / Ultrasonography in the validation of body condition score in Holstein - zebu cows during the transition period and early lactation

Gentil, Narjara Rodrigues 29 July 2016 (has links)
Fundação de Apoio a Pesquisa e à Inovação Tecnológica do Estado de Sergipe - FAPITEC/SE / The body condition score (BCS) is a method of assessment of the animal's energy reserves and it was developed based on the characteristics of mobilization/ deposition of cow’s fat cows with degree of Holstein blood Therefore, it is not known whether this methodology is appropriate for crossbred cows. Thus, the objective with this study to investigate the relationship between the back fat thickness (BFT) and the body condition score (BCS) in crossbred cows (Holstein - Zebu) during the transition period and early lactation, and to relate these findings with productive and metabolic parameters. A total of 108 Holstein - Zebu dairy cows from six farms were randomly selected and divided into three groups according to their production phase at the time: PP group (0 to 4 weeks antepartum; n = 30 animals); L4 group (0-4 weeks postpartum; n = 15 animals); L + 5 group (5-14 weeks postpartum; n = 63 animals). Correlation and regression analysis were used to describe the relationship between the BCS, BFT and body weight (BW) variables. For productive and metabolic evaluation of the animals we made a multivariate cluster analysis to order the distribution of these animals in homogeneous BFT groups within each study phase. Analysis of variance showed significant differences (P <0.05) among the three groups for the three variables BCS, BFT and BW. The BW variable showed a weak correlation with the BCS and BFT variables r = 0.28 and r = 0.38, respectively. In groups PP and L + 5 there were strong and direct correlation between BFT and BCS variables, r = 0.78 and r = 0.71, respectively. The BFT was sensitive to changes in metabolites albumin, total cholesterol and total protein. In addition to being a good indicator of the production of milk and density, as well as proportions of total solids and lactose.

Nenhum paciente apresentou intercorrências durante as leucaféreses. In the mobilization, the classical dose of G-CSF is 10?g/kg of the patient in a single dose. There is a theory that higher doses of G-CSF applied twice daily could increase the number of collected CD34+ cells with a smallest number of leukapheresis. Objective: The aim of this study is to evaluate if a fractionated-dose of G-CSF at 15?g/kg of patient may reduce the number of leukapheresis for achieving the minimum target of 3 x 106/kg of CD34+ cells as compared to conventional dose of G-CSF. Methods: Patients were divided into two groups. Group 10: patients who received a single dose daily of G-CSF 10?g/kg and Group 15: patients who received twice dose daily of G-CSF 15?g/kg. The leukapheresis were processed in an automated cell separator. The autologous transplantation happened when the minimum number of 3x106/kg CD34+ was reached. Results: Group 10 enrolled 39 patients who received 10?g/kg of G-CSF and group 15 had 26 patients who received 15?g/kg fractionated of G-CSF. There were a total of 146 aphaeresis; 110 (75.3%) in group 10 and 36 (24.7%) group 15. Group 10 collected a median of 3 (1-7) leukapheresis and a mean of 8.89 x106/kg (± 9.59) CD34+, whereas group 15 had a median of 1 (1-3) leukapheresis and collected a mean of 5.29 x106/kg (± 4.95). The relevant difference statistic was the number of aphaeresis (p<0.0001). Conclusion: To collect a minimum target of 3x106/kg of CD34 +, the application of fractionated-dose of 15?g/kg G-CSF decreased significantly the number of leukapheresis performed / Doutorado / Saude da Criança e do Adolescente / Doutora em Ciências

Evaluation of poly-3-hydroxybutyrate (P3HB) mobilization system in Burkholderia sacchari.

Nuri Andrea Merchan Castellanos 19 October 2010 (has links)
The intracellular poly-3-hydroxybutyrate (P3HB) mobilization system in Burkholderia sacchari was analyzed. A search in Burkholderia spp. genomes identified two oligomer hydrolases (PhaY1 and PhaY2) and at least three intracellular P3HB depolymerase (PhaZa1, PhaZa2 e PhaZd1). B. sacchari mutants affected on P3HB mobilization and complemented by Ralstonia eutropha genes showed an expressive increase on P3HB mobilization rates, especially when phaZa1 was overexpressed. The overexpression of phaZa2 or phaZa3 also increased the mobilization rates though to a lesser extent than phaZa1. Two mutants affected on P3HB mobilization were obtained using the transposon mini-Tn5 (NAM03 and NAM04) .NAM03 was disrupted in a gene encoding an intracellular P3HB depolymerase (PhaZa1). NAM04 was disrupted in a gene annotated as a serine peptidase LonA. This could be a mobilization activator or a new intracellular P3HB depolymerase.

Delaying Evacuation: Risk Communication in Mobilizing Evacuees

Li, Xiangyu 08 1900 (has links)
Evacuation is oftentimes the best means to prevent the loss of lives when residents encounter certain hazards, such as hurricanes. Emergency managers and experts make great efforts to increase evacuation compliance but risk area residents may procrastinate even after making the decision to leave, thus complicating response activities. Purpose - This study explores the factors determining evacuees’ mobilization periods, defined here as, the delay time between the decision to evacuate and actual evacuation. The theoretical model that guides this research is built on the protective action decision model (PADM). It captures both the social and psychological factors in the process of transferring risk information to mobilization action. The psychological process of risk communication originates from personalized external information and ends with the formation of risk perception, ultimately influencing evacuees’ mobilizations. Design/methodology/approach – Using structural equation modeling (SEM), the model is tested using survey data collected from Hurricane Rita (2005) evacuees in 2006 (N = 897). The residents of three Texas coastal counties (Harris, Brazoria, and Galveston) are randomly selected and telephone-interviewed. Findings – The findings indicate that mobilizations are affected directly by respondents’ concerns of the threats to their personal lives and costs and dangers on their evacuation trips. The perceptions of evacuees can be related to their exposure, attention, and understanding of the risk information. Research limitations/implications – The results suggest that practitioners pay more attention on the residents’ understanding of different types of risks, their abilities to process the risk information, as well as the means information is delivered. Therefore the public authorities should be more active in protecting evacuees’ properties and assets, as well as encourage evacuees to take closer shelters to avoid potential costs on road. Also the community should be more involved in mobilizing evacuation, as long as social cues can assist evacuees to better comprehend the threat information. Originality/value – This study tests the PADM framework empirically and structurally. It also clarifies the definition of evacuation mobilization using a new operation based on the evacuation groups per household.

Using Industry Data to Make an Impact on Construction Practices over the Project Lifecycle

January 2020 (has links)
abstract: The construction industry generates tremendous amounts of data every day. Data can inform practitioners to increase their project performance as well as the quality of the resulting built environment. The data gathered from each stage has unique characteristics, and processing them to the appropriate information is critical. However, it is often difficult to measure the impact of the research across project phases (i.e., planning, design, construction, operation and maintenance, and end-of-life). The goal of this dissertation is to present how industry data can be used to make an impact on construction practices and test a suite of methods to measure the impact of construction research across project phases. The dissertation provides examples of impactful research studies for each project phase to demonstrate the collection and utilization of data generated from each stage and to assess the potential tangible impact on construction industry practices. The completed studies presented both quantitative and qualitative analyses. The first study focuses on the planning phase and provides a practice to improve frond end planning (FEP) implementation by developing the project definition rating index (PDRI) maturity and accuracy total rating system (MATRS). The second study uses earned value management system (EVMS) information from the design and construction phases to support reliable project control and management. The dissertation then provides a third study, this time focusing on the operations phase and comparing the impact of project delivery methods using the international roughness index (IRI). Lastly, the end-of-life or decommissioning phase is tackled through a study that gauges the monetary impact of the circular economy concept applied to reuse construction and demolition (C&D) waste. This dissertation measures the impact of the research according to the knowledge mobilization (KMb) theory, which illustrates the value of the work to the public and to practitioners. / Dissertation/Thesis / Doctoral Dissertation Civil, Environmental and Sustainable Engineering 2020

The Process to Political Mobilization in Five College Capitalism: Forms of Antiracism, Personal Reflection and Community-Building

Homrich, Caitlin B. 24 March 2017 (has links)
The town of Amherst, Massachusetts is home to the flagship campus of the University of Massachusetts, Amherst College, and Hampshire College, institutions that have greatly influenced the town’s prolific history of political activism as well as the high educational attainment and economic status of the majority of its residents. Often hailed as a liberal utopia, research on the political mobilization occurring in this town provides insight into the process and limitations of ally politics: when most of the residents of Amherst are White, how do they engage in racial justice activism? When most of the residents are wealthy and/or highly educated, how do they engage in challenges to capitalism’s structural inequalities? In this thesis, I approach these questions by examining the political mobilization process of myself and others in three organizations: Coming Together, Re-Evaluation Counseling (RC), and the student organization, UMass Alliance for Community Transformation (UACT). I explore how Coming Together focused on antiracism in a process of focused personal reflection about racial identity and personal antiracism practices, and how that process silenced the people of color in the organization, was vii detrimental to my own mental health, and demobilized many potential-activists. In an effort to understand this organization better, I explore the practices of personal reflection and the vision of social change in RC, an organization which greatly influenced Coming Together. I argue that the more holistic and rigorous personal reflection in RC was more empowering, although taxing of energy. Finally, I contrast these experiences with the political mobilization I experienced in the UACT introductory course, Grassroots Community Organizing (GCO). I argue that the ongoing facilitation in critical personal reflection, relationship- and community-building, and practice in activism work in GCO was politically mobilizing and simultaneously produced a community culture of anti oppression. Ultimately, this thesis argues that effective activism against racism requires activism against capitalism, and vice-versa, and that highly intentional anti-oppression community-building can denaturalize, and mobilize participants against, the capitalist ideologies of alienation and competition. In order to do this comparative work, I rely heavily on the methods of participation observation and, rooted in Black feminist anthropology, autoethnography.

Samhällsarbetet och socialtjänstlagen, då och nu : En diskursanalys / Community work and the social service act, then and now : A discourse analysis

Broms, Martin January 2021 (has links)
Community work as a concept and practice is almost forgotten today within the public sector in Sweden. It has not always been like that. During the 1970s community work was well-known and practiced in many of the Swedish municipal social services. In the current study, discourses on public community work in Sweden is investigated. The purpose of the study is to explore the development of hegemonic discourses on public community work and its variation over time. To achieve this purpose, the history of community work is examined as well as the academic debate on community work. Also, legislatory histories of the social service act from 1982 are compared with the latest government public inquiry of the social service act from 2020. By using discourse theory from Foucault and Rothman’s theory about community work, the study explores how community work and similar concepts are described in the legislatory histories. Also, the study explores how community work could be carried out. Furthermore, the study investigates the legal framework of the social service act and its consequences for community work. The results are showing that the legislatory histories are taking a critical stance towards community work. Community work and similar concepts were found to be top-down, individualized, and based on consensus, in agreement with a neoliberal discourse and neoliberal socio-political development. Another conclusion is that the social service act’s shape as a framework law is hindering the establishment of community work within the social services. However, a people-oriented community work could be possible, where people have better opportunities to affect the society they live in.
