Diacylglycerol, novel protein kinase C isozymes [eta] and [theta], and other diacylglycerol activated proteins promote neuroprotective plasmalemmal sealing in B104 neurons in vitro and rat sciatic nerve axons in vivo

To survive, neurons and other eukaryotic cells must rapidly repair (seal) plasmalemmal damage. Such repair occurs by an accumulation of intracellular vesicles at or near the plasmalemmal disruption. Diacylglycerol (DAG)-dependent and cAMP-dependent proteins are involved in many vesicle trafficking pathways. Although recent studies have implicated the signaling molecule cAMP in sealing, no study has investigated how DAG and DAG-dependent proteins affect sealing and, whether pharmacological inhibition of such proteins could promote immediate repair of damaged mammalian axons. To this end, I investigated the role of DAG, protein kinase C (PKC) and other DAG-activated proteins in plasmalemmal sealing in B104 neurons in vitro and rat sciatic nerves in vivo. Using dye exclusion to assess Ca2+-dependent vesicle-mediated sealing of transected neurites of individually identifiable rat hippocampal B104 cells, I now report that, compared to non-treated controls, sealing probabilities and rates are increased by DAG and cAMP analogs that activate PKC and Munc13-1, and protein kinase A (PKA). Sealing is decreased by inhibiting DAG-activated novel protein kinase C isozymes η (nPKCη) and θ (nPKCθ) and, Munc13-1, the PKC effector myristoylated alanine rich PKC substrate (MARCKS) or phospholipase C (PLC). DAG-increased sealing is prevented by inhibiting MARCKS or PKA. Sealing probability is further decreased by simultaneously inhibiting nPKCη, nPKCθ and PKA. Extracellular Ca2+, DAG or cAMP analogs do not affect this decrease in sealing. I also report that applying inhibitors of nPKC and PKA to rat sciatic axons crush-severed in vivo under physiological calcium, do not promote immediate repair by polyethylene glycol (PEG), as assessed by compound action potential conduction and dye diffusion through crush sites. These and other data suggest that DAG increases sealing through MARCKS and that nPKCη, nPKCθ and PKA are all required to seal plasmalemmal damage in B104 neurons, and likely all eukaryotic cells. / text

Esta tese encontrou na crise hídrica ocorrida na Região Sudeste brasileira nos anos de 2013 a 2015, em especial no Estado de São Paulo, uma oportunidade única de estudar a ocorrência de um fenômeno com características de início lento, explorar a maneira como foi percebida por gestores, bem como as decisões consideradas para Gerenciamento de Riscos e Resiliência em Cadeias de Suprimentos. O objetivo principal desta tese foi investigar o desenvolvimento de capacidades de resiliência em cadeias de suprimentos em um contexto de desastre natural. Para explorar este contexto traz a seguinte questão de pesquisa: como as empresas desenvolvem capacidades de resiliência em cadeia de suprimentos frente a um desastre natural? Esta tese possui natureza qualitativa, por meio da utilização de estudos de casos em duas cadeias de suprimentos relacionadas ao setor de alimentos e bebidas (oriundas das commodities cana-de-açúcar e laranja) como unidades de análise. Os resultados apontam a percepção ao risco como responsável pelo desenvolvimento das capacidades de flexibilidade, colaboração, visibilidade e velocidade. Após a percepção ao risco, os respondentes investiram fortemente em tecnologias para enfrentar os fenômenos climáticos a que estão expostos, evidenciando capacidade de visibilidade sobre o evento, principalmente por meio de compartilhamento de informações úteis entre os elos da cadeia para enfrentamento da crise. Outro resultado indica que a velocidade e a colaboração são necessárias nas fases de resposta e recuperação para alcançar resiliência. No entanto, a presente pesquisa mostrou que estas capacidades não estão presentes em todas as empresas de acordo com os casos estudados e que as cadeias buscam caminhos diferentes para tentarem alcançar a resiliência necessária para normalização ou aprimoramento de seus processos. Conclui-se que a cadeias estudadas não desenvolveram resiliência para enfrentar futuras crises hídricas em todos os elos. As contribuições teóricas desta tese residem em três pontos: o primeiro, a análise das capacidades de resiliência de forma conjunta (como um “conjunto de recursos”), considerando o contexto das fases de uma ruptura de início de lento. Segundo, são raros os estudos com evidências empíricas do ambiente de crise ou pré-ruptura em cadeias de suprimentos que analisem em profundidade as fases de resposta e recuperação, especialmente por desastres naturais com início lento. Terceiro, a análise deste fenômeno além das díades (fornecedores-compradores) pode ser considerada contribuição empírica-teórica para disciplina de cadeia de suprimentos. A partir deste conhecimento é possível adotar estratégias de preparação, tais como aumento de flexibilidade e redundância, investimentos em sistemas integrados de comunicação entre os elos da cadeia para aumentar a visibilidade e velocidade de resposta, entre outras ações que envolvam as capacidades de resiliência estudadas. São necessárias novas ações voltadas para o agronegócio que visem ampliação dos protocolos agroambientais existentes, além de novas pesquisas, compartilhamento de resultados, estudos de casos e ensino nas universidades e debates, para manter o nível adequado de percepção ao risco e foco voltado para recuperação de áreas de preservação e relevantes para melhor gerenciamento dos recursos hídricos. / The water crisis is pointed out in global surveys carried out in last decade as a risk with high severity potential (WEF, 2007, 2016), considered the natural disaster that causes most deaths in all countries (BELOW; GROVER-KOPEC; DILLEY, 2007), and has a strong impact on food supply, resulting in damages for health, education, labor and economy of the affected regions. This thesis has found a unique opportunity to study the occurrence of a phenomenon with slow on-set characteristics, to explore the way in which it was perceived by managers, as well as the decisions that were considered for Risk and Resilience in Supply Chain Management. The main objective of this thesis was to investigate the development of supply chain resilience capabilities in a natural disaster context. To explore this context has the following research question: how do firms develop resilience supply chain capabilities when face a natural disaster? This thesis had a qualitative approach, through multiple case studies and the unit of analysis was the food and beverage supply chains (sugar cane and orange commodities). The results point to risk perception as responsible for the development of flexibility, collaboration, visibility and velocity capabilities. After risk perception, the respondents invested heavily in technologies to face the climate change phenomena to which they are exposed, evidencing the visibility capability on the event, mainly through sharing of useful information between the links of the supply chain members who faced the crisis. Another result indicates that velocity and collaboration are necessary in the response and recovery phases to achieve resilience. However, the present research showed that these capabilities are not present in all firms according to the cases studied. Both supply chains seek different paths to try to achieve the resilience necessary for normalization or improvement of their processes. It is concluded that the supply chains studied did not develop resilience to face future water crises in all links. The theoretical contributions of this thesis reside in three points: the first, the analysis of resilience capacities in the same study (as a “bundle of resources”), considering the context of the phases of a disruption with slow on-set characteristics. Second, studies with empirical evidence of the crisis environment or pre-rupture in supply chains that analyze in depth the response and recovery phases, especially for natural disasters with slow on-set, are rare. Third, the analysis of this phenomenon beyond dyads (suppliers-buyers) can be considered empiricaltheoretical contribution to supply chain discipline. From this knowledge, it is possible to adopt preparation strategies, such as increased flexibility and redundancy, investments in integrated communication systems between supply chain links to increase visibility and velocity of response, among other actions that involve the resilience capabilities studied. New agribusiness actions are needed to expand the existing agrienvironmental protocols, as well as new research, information sharing and debates, to maintain the adequate level of risk perception and focus on the recovery and conservation of relevant areas to improve water resources management.

Optimisation of dynamic and stochastic production scheduling systems after random disruptions

(Department of Industrial Engineering and Operations Management, Faculty of Engineering), Vaal University of Technology. / The current business environments in many companies are characterized by markets facing tough competitions, from which customer requirements and expectations are becoming increasingly high in terms of quality, cost and delivery dates, etc. These emerging expectations are even getting stronger due to rapid development of new information and communication technologies that provide direct connections between companies and their clients. As a result, companies should have powerful control mechanisms at their disposal. To achieve this, companies rely on a number of functions including production scheduling. This function has always been present within companies, but today, it is facing increasing complexities because of the large number of jobs that must be executed simultaneously. Amongst many factors, it is time driven. This study demonstrates that several disciplines can be married into one model (i.e. a unified model) to solve scheduling problems after disruptions, and clears the way for future multi-disciplinary research efforts. Scheduling problem is modeled as follows: Ito’s stochastic differential rule is used to analyse the time evolution of random or stochastic processes. Multifactor productivity is used to unify various disruption factors. Theory of line balancing is also employed to determine the required number of resources to minimize bottleneck. Reliability: disruptions are considered to be equivalent to system failure. The failure rate of the system is translated to the reliability of the system mathematically. The probabilities of failure are used as indicators of disruptions, and the theory of reliability is then applied. Bernoulli’s principle is also employed to relate pressure to production flow and aid in managing bottleneck situations. Results indicate that the amount of resources needed after disruption depends on the nature of disruption, and that the scheduler should plan to increase number of facilities following a trend that is only predicted by the nature of disruptions. It is also shown that disruption of one type may not greatly affect productivity of a certain company layout, whilst similar disruptions can have devastating effect on another type. It is further concluded that impacts of disruption are dependent on the type of company layouts.

Supply Chain Resilience: Disruptions in Global Maritime Transportation

Supply chain resilience has become an important topic within supply chain management, as increasingly complex supply chains set out for increased risk in these networks. Supply chain resilience is a risk management approach that includes the ability to mitigate disruptions through specific capabilities. The previous disruptive events of the COVID-19 pandemic and the Suez Canal blockage-21 have highlighted the vulnerabilities of global trade and the need for more resilient supply chains. The purpose of this thesis was to analyse supply chain resilience and the need for dynamic and operational capabilities in the global maritime transport sector to mitigate impacts from slow-onset and sudden-onset disruptions. In specific, the vulnerabilities highlighted through the impacts of the COVID-19 pandemic and the Suez Canal blockage-21 over time. This study was conducted as a multiple case study investigating the maritime transport sector’s resilience during COVID-19 and the Suez Canal blockage-21. The data was triangulated through semi-structured interviews, industry podcasts, and business reports and further analysed using a grounded analysis approach. The study showed that the continuous disruptions on supply chains caused by the COVID-19 pandemic and the Suez Canal blockage-21 generated economic consequences, congestions, and capacity constraints that the maritime transport sector could not fully mitigate. The study also provides evidence that severe impacts on supply chains were not necessarily caused by a lack of resilience, but rather the persistency of disruptions did not diminish over time. Even though the maritime transportation sector is considered flexible and agile in adapting to the new market situation, increased collaboration, integration, innovation, and digitalization were found necessary to improve the resilience of the maritime transport sector to become even better prepared for future disruptions.

This study is set to research how two Swedish companieshave managed three specific disruptions, the Covid-19 pandemic,the blocking of the Suez Canal and the ongoing war in Ukraine. Thepurpose of this paper is to contribute with knowledge and researchabout resilience in the supply chain and how companies can adaptin a volatile world with unpredictable events that may occur. Themethod used consists of semi-structured interviews with the casecompanies and collection of literature and previous research withinthe research area. The results from the study presents that thecompany operating in the retail industry main strategy is tomaintain a transparent communication to its suppliers as well ascustomers in order to maintain a high level of certainty in its supplychain. The company operating in the forest industry explained inthe interview that the company prioritizes a high level of self-sufficiency in raw materials, energy and transportation in order tomaintain a high level of certainty in the supply chain. The resultalso explains that cooperation between the three abilities, prevent,manage and recover from disruptions is the foundation tomaintaining a high level of certainty in the supply chain. The studycontributes with insights on how companies are affected by andmanage the ongoing situation in Ukraine, with the supply chain as astarting point. / Försörjningskedjan är den bakomliggande processen som möjliggörför utbyte av varor och tjänster mellan företag och kunder. Centraltför en fungerande försörjningskedja är att den är resilient motstörningar. Resiliens i försörjningskedjan handlar om förmågan attförebygga, hantera och återhämta sig från störningar. De senasteåren har präglats av händelser som har orsakat störningar iförsörjningskedjan. Denna studie undersöker hur två svenskaföretag har hanterat tre specifika störningar, Covid-19 pandemin,stoppet i Suezkanalen samt det pågående kriget i Ukraina. Syftetmed detta arbete är att bidra till kunskapen och forskningen omresiliens i försörjningskedjor och hur företag kan anpassa sig till envolatil värld med oförutsägbara händelser som uppstår. Metodenutgörs av semistrukturerade intervjuer med fallföretagen ochinsamling av litteratur samt tidigare forskning inomforskningsområdet. Studiens resultat visar att företaget som verkarinom livsmedelsindustrin främst arbetar med att bibehållatransparent kommunikation till sina leverantörer och kunder för attupprätthålla en hög nivå av tillförlitlighet i sin försörjningskedja.Företaget som verkar i skogsindustrin anger i intervjun att företagetprioriterar en hög självförsörjningsgrad på råvaror, energi ochtransport för att upprätthålla en hög nivå av tillförlitlighet iförsörjningskedjan. Studiens resultat redogör dessutom attsamverkan mellan de tre förmågorna, förebygga, hantera ochåterhämta sig från störningar är grunden till att uppnå hög nivå avtillförlitlighet i försörjningskedjan. Studien bidrar bland annat medinsikter om hur företag påverkas och bemöter den rådandesituationen i Ukraina med försörjningskedjan som utgångspunkt.

The interconnectedness of global supply chains, combined with disruptive events like the Covid-19 pandemic, has emphasized the significance of enhancing supply chain resilience (SCRes). Thus, this study aims to investigate how businesses employ SCRes recovery strategies to address extreme supply chain disruptions (SCD). To contribute empirical evidence on reactive SCRes strategies, qualitative methodology was employed, with a specific emphasis on the construction department at Northvolt. The construction industry serves as an apt context for assessing and offers valuable insights. Unstructured and semi-structured interviews were conducted with supply chain executives within the company to collect the data. The findings reveal specific challenges faced by the construction department at Northvolt in the face of global supply chain disruptions, shedding light on their responsive strategies. This study not only emphasizes the necessity of cultivating all SCRes capabilities but also contributes valuable empirical evidence to the field. The implications of these findings extend beyond Northvolt, providing actionable insights for businesses in the construction industry and offering recommendations for enhancing overall supply chain resilience. / Den ökande globala kopplingen mellan leverantörskedjor, tillsammans med störande händelser som Covid-19-pandemin, har understrukit betydelsen av att förbättra leverantörskedjors motståndskraft (SCRes). Denna studie syftar därför till att undersöka hur företag använder sig av SCRes-återhämtningsstrategier för att hantera extrema störningar i leverantörskedjan (SCD). För att bidra med empiriska bevis om reaktiva SCRes-strategier användes kvalitativ metodik, med särskild tonvikt på byggavdelningen på Northvolt. Fault Location Algorithms in Transmission Grids

The rapid growth of the electric power system has in recent decades resulted in an increase of the number of transmission lines and total power outage in Norway. The challenge of a fast growing electrical grid has also resulted in huge increases of overhead lines and their total length. These lines are experiencing faults due to various reasons that cause major disruptions and operating costs of the transmission system operator (TSO). Thus, it’s important that the location of faults is either known or can be estimated with reasonably high accuracy. This allows the grid owner to save money and time for inspection and repair, as well as to provide a better service due to the possibility of faster restoration of power supply and avoiding blackouts.  Fault detection and classification on transmission lines are important tasks in order to protect the electrical power system. In recent years, the power system has become more complicated under competitive and deregulated environments and a fast fault location technique is needed to maintain security and supply in the grid. This thesis compares and evaluates different methods for classification of fault type and calculation of conventional one-side and two-side based fault location algorithms for distance to fault estimation.  Different algorithm has been implemented, tested and verified to create a greater understanding of determinants facts that affect distance to faults algorithm’s accuracy.  Implemented algorithm has been tested on the data generated from a number of simulations in Simulink for a verification process in implemented algorithms accuracy. Two types of fault cases have also been simulated and compared for known distance to fault estimation.

Supply Chain Event Management (SCEM) bezeichnet eine Teildisziplin des Supply Chain Management und ist für Unternehmen ein Ansatzpunkt, durch frühzeitige Reaktion auf kritische Ausnahmeereignisse in der Wertschöpfungskette Logistikleistung und -kosten zu optimieren. Durch Rahmenbedingungen wie bspw. globale Logistikstrukturen, eine hohe Artikelvielfalt und volatile Geschäftsbeziehungen zählt die Modeindustrie zu den Branchen, die für kritische Störereignisse besonders anfällig ist. In diesem Sinne untersucht die vorliegende Dissertation nach einer Beleuchtung der wesentlichen Grundlagen zunächst, inwiefern es in der Modeindustrie tatsächlich einen Bedarf an SCEM-Systemen gibt. Anknüpfend daran zeigt sie nach einer Darstellung bisheriger SCEM-Architekturkonzepte Gestaltungsmöglichkeiten für eine Systemarchitektur auf, die auf den Designprinzipien der Serviceorientierung beruht. In diesem Rahmen erfolgt u. a. auch die Identifikation SCEM-relevanter Business Services. Die Vorzüge einer serviceorientierten Gestaltung werden detailliert anhand der EPCIS (EPC Information Services)-Spezifikation illustriert. Abgerundet wird die Arbeit durch eine Betrachtung der Nutzenpotenziale von SCEM-Systemen. Nach einer Darstellung von Ansätzen, welche zur Nutzenbestimmung infrage kommen, wird der Nutzen anhand eines Praxisbeispiels aufgezeigt und fließt zusammen mit den Ergebnissen einer Literaturrecherche in eine Konsolidierung von SCEM-Nutzeffekten. Hierbei wird auch beleuchtet, welche zusätzlichen Vorteile sich für Unternehmen durch eine serviceorientierte Architekturgestaltung bieten. In der Schlussbetrachtung werden die wesentlichen Erkenntnisse der Arbeit zusammengefasst und in einem Ausblick sowohl beleuchtet, welche Relevanz die Ergebnisse der Arbeit für die Bewältigung künftiger Herausforderungen innehaben als auch welche Anknüpfungspunkte sich für anschließende Forschungsarbeiten ergeben.

In diesem Sinne untersucht die vorliegende Dissertation nach einer Beleuchtung der wesentlichen Grundlagen zunächst, inwiefern es in der Modeindustrie tatsächlich einen Bedarf an SCEM-Systemen gibt. Anknüpfend daran zeigt sie nach einer Darstellung bisheriger SCEM-Architekturkonzepte Gestaltungsmöglichkeiten für eine Systemarchitektur auf, die auf den Designprinzipien der Serviceorientierung beruht. In diesem Rahmen erfolgt u. a. auch die Identifikation SCEM-relevanter Business Services. Die Vorzüge einer serviceorientierten Gestaltung werden detailliert anhand der EPCIS (EPC Information Services)-Spezifikation illustriert. Abgerundet wird die Arbeit durch eine Betrachtung der Nutzenpotenziale von SCEM-Systemen. Nach einer Darstellung von Ansätzen, welche zur Nutzenbestimmung infrage kommen, wird der Nutzen anhand eines Praxisbeispiels aufgezeigt und fließt zusammen mit den Ergebnissen einer Literaturrecherche in eine Konsolidierung von SCEM-Nutzeffekten. Hierbei wird auch beleuchtet, welche zusätzlichen Vorteile sich für Unternehmen durch eine serviceorientierte Architekturgestaltung bieten. In der Schlussbetrachtung werden die wesentlichen Erkenntnisse der Arbeit zusammengefasst und in einem Ausblick sowohl beleuchtet, welche Relevanz die Ergebnisse der Arbeit für die Bewältigung künftiger Herausforderungen innehaben als auch welche Anknüpfungspunkte sich für anschließende Forschungsarbeiten ergeben.
