The multidrug transporter LmrP, member of the major facilitator superfamily (MFS), confers L. lactis and recombinant E. coli cells resistance to an array of cytotoxic compounds including antibiotics. LmrP mediates drug extrusion from the plasma membrane by an electrogenic proton/drug exchange reaction, whereby a positively charged substrate may move towards the external medium in exchange for two or more protons moving towards the cytoplasm. Recent studies have suggested that MFS transporters require phosphatidylethanolamine (PE) for function and proper topology. However, the specificity of the PE requirement, as well as the contribution of the electrochemical gradient (the driving force of the substrate transport) to this lipid requirement was not addressed. Here we report a new approach for addressing PE specific requirement for the function and the structure of membranes transporters. We used methyl-PE and dimethyl-PE analogs of PE to show that only replacement of the three hydrogens by methyl moieties leads to changes in the biochemical and biophysical properties of the reconstituted protein. This suggests that LmrP does not depend on the bulk properties of the phospholipids tested but solely on the hydrogen bonding ability of the headgroup. We then show that a single point mutation in LmrP, D68C, is sufficient to recapitulate precisely every biochemical and biophysical effect observed when PE is replaced by phosphatidylcholine (PC) ( including energy transfer between the protein tryptophan residues and the lipid headgroups). We conclude that the negatively charged Asp-68 is likely to participate in the interaction with PE and that such interaction is required for proton gradient sensing, substrate binding, and transport. Because Asp-68 belongs to a highly conserved motif in the Major Facilitator Superfamily (which includes LacY and EmrD), this interaction might be a general feature of these transporters that is involved in proton gradient sensing and lipid dependence.<p> / Doctorat en Sciences / info:eu-repo/semantics/nonPublished

Essential Factors for Usability and Acceptance of Mobile Learning in Ugandan Primary Schools : Minor Field StudiesHager, Lovisa, Almström, Sara January 2019 (has links)
Two different focus groups in primary schools in Uganda participated in this study to investigate critical aspects of usability comparing two different educational mobile applications. The first group was already familiar with both applications, while the second group had no previous experiences of the applications. Furthermore, interviews were performed with a head teacher as well as two CEOs and founders of two start ups developing educational mobile applications for children, in order to investigate challenges with implementation of mobile learning in schools in Uganda. The results showed that one of the applications was not fully compatible with the devices used by the target group and that swiping was perceived less natural than clicking. Regarding organizational factors of implementation in schools, financial limitations and insufficient computer literacy among employees were revealed as critical obstacles. Moreover, improvements of the method used during the user tests with children in foreign contexts are discussed. / I denna studie deltog två fokusgrupper med barn från två olika grundskolor i Uganda för att undersöka kritiska aspekter gällande användarvänlighet genom att jämföra två mobilapplikationer som används i utbildningssammanhang. Den första gruppen var redan bekant med båda applikationerna, medan den andra gruppen var obekant med applikationerna. Vidare undersöktes utmaningar med implementation av mobile learning i grundskolor i Uganda genom intervjuer med en rektor från en ugandiansk grundskola samt två VDs tillika grundare från två ugandianska edTech-start-ups. Resultaten visade att en av applikationerna inte var fullständigt kompatibel med de enheter som användes av målgruppen i Uganda samt att "swipea" uppfattades mindre naturligt än att klicka. Gällande organisatoriska faktorer av implementering i skolorna visades finansiella begränsningar och anställdas otillräckliga datakunskap vara kritiska hinder. Utöver resultaten som ämnar svara på de definierade frågeställningarna diskuteras förbättringar av metoden som användes för användartesterna, i syfte att ge riktlinjer till framtida projekt i liknande sammanhang.

Social Entrepreneurship in the Favelas of Brazil: Challenges and ConstraintsRafid, Mohammed January 2023 (has links)
As global inequality reaches unprecedented levels, social entrepreneurship has emerged as a promising tool for addressing this and fighting some of the most challenging social issues of our time. This thesis seeks to dive into the specific challenges that social entrepreneurs encounter in low-income settings by conducting a case study in the favelas in Brazil, one of the world’s most widely recognized low-income areas. By conducting a comprehensive literature review, the thesis establishes a conceptual framework for the challenges facing social entrepreneurs in low-income settings. This is used to guide the analysis for the data collection in a deductive manner. Data is collected using semi-structured interviews with seven social entrepreneurs operating within the favelas. The findings reveal that the interviewed social entrepreneurs in the favelas face significant obstacles across five main areas: financial capital, legal and regulatory environment, infrastructure, human capital, and social capital. These challenges are consistent with the conceptual framework of challenges facing social entrepreneurs in low-income settings. / I takt med att den globala ojämlikheten når rekordnivåer har socialt entreprenörskap vuxit fram som ett lovande verktyg för att tackla några av vår tids mest utmanande samhällsfrågor. Denna avhandling syftar till att undersöka de specifika utmaningar som sociala entreprenörer möter i låginkomstmiljöer genom att genomföra en fältstudie i favelorna i Brasilien, ett av världens mest kända låginkomstområden. Genom att göra en omfattande litteraturgenomgång etablerar avhandlingen först ett konceptuellt ramverk för de utmaningar som sociala entreprenörer i låginkomstmiljöer står inför. Denna används senare för att vägleda analysen av datainsamlingen på ett deduktivt sätt. Data samlas in med hjälp av semistrukturerade intervjuer med sju sociala entreprenörer verksamma inom dessa områden. Resultaten visar att de intervjuade sociala entreprenörerna möter betydande hinder inom fem huvudområden: finansiellt kapital, lagar och regler, infrastruktur, humankapital och socialt kapital. Dessa utmaningar är i linje med det konceptuella ramverket för utmaningar som sociala entreprenörer står inför i låginkomstområden.

<p>In April 2007, New Zealand (NZ) launched the Recognised Seasonal Employer (RSE) scheme. The scheme allows for unskilled workers from the Pacific Islands to enjoy the benefits of seasonal work in NZ’s horticulture and viticulture industries for up to seven months at a time. One of the articulated objectives of the scheme is to advance the effects on development in the countries of origin of the workers, for which remittances have been stressed as key-benefits. Although previous data and interviews concerning these aspects are marginal, all studies indicate clear benefits for Pacific Islanders. In contrast, this study provides the novel insight to the individual views and perceptions of the earning, saving, spending and remittance possibilities of 23 Ni-Vanuatu RSE workers in June of 2008. The findings indicate an absence of autonomy among the individual RSE workers to decide over and manage the spending of their respective incomes, along with negative implications on the potential for workers to send remittances while working in NZ. Identified as the primary cause of this outcome, is the dual and simultaneous role that NZ based companies, on the one hand, can play as recruitment agents in Vanuatu, and on the other hand, as pastoral care agents in NZ. This twofold capacity creates a middle hand situation that severely restricts the possibilities for the workers to access their wages while in NZ. The conclusion therefore holds that, in this example of 23 Ni-Vanuatu RSE workers, the degree of remittances depends on the type of employment governing the participation of the workers in the scheme, as opposed to the individual spending and saving patterns, differences in earnings, or differences in the availability of work of each worker respectively.</p> / <p>I april 2007 så startade Nya Zeeland (NZ) sitt Recognised Seasonal Employer (RSE) program. This project is a study of the wastescape - a network of waste - of Bhubaneswar and Cuttack in Odisha, India. The study incorporates key locations, e.g. landfills, urban wastelands and waste warehouses; major actors in the formal and informal waste sector; and flows of waste through economic and social systems. Drawing from the studies, multiple interventions within the wastescape are proposed for improvement of the economic, ecologic and social situation. An important aspect of the project is the development of an approach for how to, as architects, work with big, complex, contingent networks; how to map and understand such a system; and how to determine where to intervene. To improve the existing wastescape, interventions must consciously and holistically address multiple scales; levels of formal-informal; and phases within the waste cycle. The study includes a vast amount of possible interventions. Some of the interventions are further detailed to show feasibility; impact on the wastescape; and synergies with other interventions within the wastescape. / Projektet “Wastescape of Bhubaneswar & Cuttack” är en studie av ett nätverk av skräpflöden genom Bhubaneswar och Cuttack i Odisha, Indien. Studien inkorporerar viktiga platser, t.ex. deponier, urbana ödemarker och lokaler för skräphandel; stora aktörer i den formella och informella skräpsektorn; och flöden av skräp genom ekonomiska och sociala system. Utifrån dessa studier, ett flertal interventioner i “the wastescape” föreslås för att förbättra den ekonomiska, ekologiska och sociala situationen. The Kamchay Hydropower Project -Hydropower development in Cambodia and eastern AsiaMalmquist, Peter, Sigfridsson, Mats January 2003 (has links)
The objective in this case study is to highlight typical problems of a planned hydropower dam project in Bokor National Park, Cambodia. The focal point in the line of questioning is the usage of public participation in larger exploration plans in a development country and to give a comprehensive survey of some of the participant involved. The study also discusses how the WCD report is used by non-govemental organisation to validate their claims. To be able to understand how the Cambodian society works, the study also includes a short briefing of recent Cambodian history. The study shows that the local people are more or less neglected in the preliminary investigations. Even if information meetings are held open, they are conducted in such a way that it is hard for common people to participate. The meetings are held in English and not in the domestic language Khmer. Critical groups are not formally invited but welcome to attend, most of their statements are withdrawn from the official protocols. The study also reveals that the leading entrepreneurs more or less neglects the environmental hazards in the construction by focus their information and calculations on the positive effects of the project. The study has been made as a field study, on location in Cambodia.

Intelligence and the Uprising in East Germany 1953: An Example of Political IntelligenceCollins, Steven Morris 08 1900 (has links)
In 1950, the leader of the German Democratic Republic (East Germany), Walter Ulbricht, began a policy of connecting foreign threats with domestic policy failures as if the two were the same, and as if he was not responsible for either. This absolved him of blame for those failures and allowed Ulbricht to define his internal enemies as agents of the western powers. He used the state's secret police force, known as the Stasi, to provide the information that supported his claims of western obstructionism and to intimidate his adversaries. This resulted in a politicization of intelligence whereby Stasi officers slanted information so that it conformed to Ulbricht's doctrine of western interference. Comparisons made of eyewitness' statements to the morale reports filed by Stasi agents show that there was a difference between how the East German worker felt and the way the Stasi portrayed their attitudes to the politburo. Consequently, prior to June 17, 1953, when labor strikes inspired a million East German citizens to rise up against Ulbricht's oppressive government, the politicization of Stasi intelligence caused information over labor unrest to be unreliable at a time of increasing risk to the regime. This study shows the extent of Ulbricht's politicization of Stasi intelligence and its effect on the June 1953 uprising in the German Democratic Republic.

Die Arbeit erforscht das Politikfeld der Auswanderungspolitik in der DDR zwischen 1949 und 1989 in Form einer Policy-Analyse. Sie untergliedert sich in drei Fragstellungen: Lässt sich eine charakteristisch auswanderungspolitische Kombination von Steuerungsinstrumenten nachweisen? Welche Funktion erfüllten spezifische Policy-Akteure? In welchem Verhältnis standen sie zu den Policy-Phasen?
Quellengrundlage ist die Aktenhinterlassenschaft der Ministerien des Inneren und für Staatssicherheit einerseits sowie die Protokolle von Politbüro und Ministerrat andererseits. Untersucht wird die Darstellung des Auswanderungsproblems, der eigenen Handlungsmotive, der Wirksamkeit der Steuerungsinstrumente und anderer Akteure. Die theoretischen Folien für die Politikfeldanalyse sind die Totalitarismustheorie von Carl Friedrich, die Theorie der Coercive Persuasion (Zwangspersuasion) von Edgar Schein, sowie der Begriff des Eigen-Sinn von Alf Lüdtke.
Die Arbeit legt ihren innovativen Schwerpunkt auf das Steuerungsinstrument der „Einzelpersuasion“. Damit ist der hier als totalitär qualifizierte Aufwand gemeint, mit dem das Regime versuchte, die Abwanderung durch individuelles Zureden in den Griff zu bekommen. Die Arbeit zeigt, dass während der gesamten SED-Herrschaft ein spezifisches Set weiterer Steuerungsinstrumente – im Sinne der Theorie der Zwangspersuasion – zur Unterstützung der Einzelpersuasion eingesetzt wurden. Dazu gehörten die berühmt gewordenen auswanderungspolitischen Instrumente des Zwangs wie Berliner Mauer, Schießbefehl und Republikflucht-Paragraph einerseits und der negativen Anreize durch die Diskriminierung von Auswanderungswilligen andererseits.
Policy-Zyklen werden mit den Zäsuren in den Jahren 1952, 1953, 1958, 1961, 1975 und 1989 identifiziert. Mit Blick auf die Einflussnahme diverser auswärtiger Akteure auf die Policyphase des Agendasetting für die Auswanderungspolitik wird eine auswanderungspolitische DDR-Außenpolitik identifiziert. / The dissertation explores the emigration policy in the GDR between 1949 and 1989 by means of a policy analysis. It breaks down into three questions: Is it possible to detect a characteristic set of emigration policy instruments? Which policy actors can be identified and what was their relevance in specific policy phases during the policy process?
The policy history is examined through an archival analysis based on the huge body of files left over from the ministries of internal affairs and state security as well as minutes from Politbureau and Council of Ministers. The study examines the presentation and perception of the emigration problem, the subjective motives, the effectiveness of policy instruments and the perception of other players. This policy analysis is based on several theories – Carl Friedrich’s Totalitarianism, Edgar Schein’s Coercive Persuasion and Alf Lüdtke’s Eigen-Sinn.
The study’s innovative emphasis lies on the policy instrument of „Einzelpersuasion“ (individual persuasion). This refers to the totalitarian effort with which the regime tried to prevent emigration attempts by personal cajolery. The study will show that during the entire SED rule a specific set of additional policy instruments were used to support the individual persuasion according to the theory of Coercive Persuasion. These included on the one hand the infamous coercive emigration policy instruments such as the Berlin Wall, the shoot-on-sight order and the criminal provisions for Republikflucht (escaping GDR). On the other hand, there were always negative incentive instruments discriminating against people intending to emigrate.
Policy cycles with remarkable policy reformulation are identified in 1952, 1953, 1958, 1961 with the Berlin Wall, 1975 and 1989. Referring to the influence of various foreign actors on the policy phase of agenda setting for emigration policy the study identifies an “emigration-driven foreign policy”.

Development of a Cost-Effective, Reliable and Versatile Monitoring System for Solar Power Installations in Developing Countries : A Minor Field Study as a Master Thesis of the Master Programme in Engineering Physics, Electrical EngineeringTrella, Fredrik, Paakkonen, Nils January 2016 (has links)
This report is the result of a conducted Minor Field Study (MFS), to the greatestextent funded by the Swedish International Development Cooperation Agency(SIDA), in an attempt to design a system for evaluating smaller solar power systems indeveloping countries. The study was to the greater part conducted in Nairobi, Kenyain close collaboration with the University of Nairobi. The aim was to develop asystem that would use easily available components and keep the costs to a minimum,yet deliver adequate performance. The system would measure certain parameters of asolar power system and also relevant environmental data in order to evaluate theperformance of the system. Due to the specific competence of the collaboratinggroup at the University of Nairobi, a Kinetis Freescale K64-microcontroller with anARM-Cortex processor was selected as the core of the design. Components wereselected, schematics were drawn, a circuit board was designed and manufactured andsoftware was written. After 12 weeks a somewhat satisfying proof-of-concept wasreached at the end of the field study in Kenya. The project however proved howdifficult it is to go from first idea to a functional proof-of-concept during a limitedtimeframe, and also in an East-African country. The final proof-of-concept was testedat Mpala Research Centre in Kenya and despite containing some flaws proved that itwould indeed be possible to design a working system on the principles discussed inthis report. The system is open-source, so anyone may use and modify it.
